Vaccination and antiviral treatment of neglected diseases caused by flaviviral infections.

Flaviviral infections have a re-emerging impact on the health situation in developing countries with several billions of people living at risk. In the present review, we focus on three members of the genus Flavivirus belonging to the Flaviviridae family. They are transmitted to humans by mosquito bites, namely those viruses leading to Dengue Fever, Yellow Fever and mosquito-borne Japanese encephalitis. All three virus groups have a spherical structure with a diameter of approximately 50 nm. Although sharing a similar genomic structure and intracellular life cycle, they show different clinical

HadA is an atypical new multifunctional trimeric coiled-coil adhesin of Haemophilus influenzae biogroup aegyptius, which promotes entry into host cells.

The Oca (Oligomeric coiled-coil adhesin) family is a subgroup of the bacterial trimeric autotransporter adhesins, which includes structurally related proteins, such as YadA of Yersinia enterocolitica and NadA of Neisseria meningitidis. In this study, we searched in silico for novel members of this family in bacterial genomes and identified HadA (Haemophilus adhesin A), a trimeric autotransporter expressed only by Haemophilus influenzae biogroup aegyptius causing Brazilian purpuric fever (BPF), a fulminant septicemic disease of children. By comparative genomics and sequence analysis we predicte

An arginine switch in the species B adenovirus knob determines high-affinity engagement of cellular receptor CD46.

Adenoviruses (Ads) are icosahedral, nonenveloped viruses with a double-stranded DNA genome. The 51 known Ad serotypes exhibit profound variations in cell tropism and disease types. The number of observed Ad infections is steadily increasing, sometimes leading to fatal outcomes even in healthy individuals. Species B Ads can cause kidney infections, hemorrhagic cystitis, and severe respiratory infections, and most of them use the membrane cofactor protein CD46 as a cellular receptor. The crystal structure of the human Ad type 11 (Ad11) knob complexed with CD46 is known; however, the determinants

Herpesvirus pan encodes a functional homologue of BHRF1, the Epstein-Barr virus v-Bcl-2.

BACKGROUND: Epstein-Barr virus (EBV) latently infects about 90% of the human population and is associated with benign and malignant diseases of lymphoid and epithelial origin. BHRF1, an early lytic cycle antigen, is an apoptosis suppressing member of the Bcl-2 family. In vitro studies imply that BHRF1 is dispensable for both virus replication and transformation. However, the fact that BHRF1 is highly conserved not only in all EBV isolates studied to date but also in the analogous viruses Herpesvirus papio and Herpesvirus pan that infect baboons and chimpanzees respectively, suggests BHRF1 may

Probing the pH-dependent structural features of alpha-KTx12.1, a potassium channel blocker from the scorpion Tityus serrulatus.

Potassium channels are widespread in living cells and are involved in many diseases. The scorpion toxin alpha-KTx(12.1) interacts with various K(+) channels, suggesting its capacity to match diverse channel pores. It is recognized that tissue injuries may affect the pH at toxins site of action, thereby modulating both protein conformation and activity. To better understand its molecular mechanism of action, we studied alpha-KTx(12.1) using pH as a tool to explore its plasticity and NMR in combination with MD calculations to detect it. The toxin solution structure consists of an alpha-helix and

New insights into the Rh superfamily of genes and proteins in erythroid cells and nonerythroid tissues.

The past decade has seen extensive studies of the erythrocyte Rh30 polypeptides and Rh-associated glycoprotein, which specify the clinically important Rh blood group system. Here we consider recent advances on these and other Rh homologues in the context of gene organization, molecular evolution, tissue-specific expression, protein structure, and potential biological functions. The Rh family is now known to contain a large number of homologues that form a unique branch in the eucarya life domain. The ancient origin and broad distribution imply central roles for the various Rh proteins in maint

Reversion of prion protein conformational changes by synthetic beta-sheet breaker peptides.

BACKGROUND: Transmissible spongiform encephalopathies are associated with a structural transition in the prion protein that results in the conversion of the physiological PrPc to pathological PrP(Sc). We investigated whether this conformational transition can be inhibited and reversed by peptides homologous to the PrP fragments implicated in the abnormal folding, which contain specific residues acting as beta-sheet blockers (beta-sheet breaker peptides). METHODS: We studied the effect of a 13-residue beta-sheet breaker peptide (iPrP13) on the reversion of the abnormal structure and properties

Heterogeneity of plasma von Willebrand factor multimers resulting from proteolysis of the constituent subunit.

In this report we demonstrate that proteolytic cleavage of the constituent subunit is one of the causes determining the heterogeneous size distribution of plasma von Willebrand factor (vWf) multimers. As shown by two-dimensional nonreduced/reduced agarose/polyacrylamide gel electrophoresis, the structure of circulating vWf molecules may deviate from that represented by assemblage of a variable number of identical subunits. Indeed, even though the largest multimers in normal plasma appear to be composed predominantly of intact 225-kD subunits, those of intermediate and smaller size contain also

Chemoreceptor complexes signal through a protein-ordering and stabilization mechanism.

Membrane protein complexes are critical to many cellular processes and have proven to be good targets for a variety of therapeutics. One such complex is the bacterial chemoreceptor signaling complex, which has potential as a target for novel antibiotics. This complex is responsible for sensing chemical signals in the environment to bias swimming of the bacterial cell toward more favorable conditions. While these complexes are well studied, the mechanisms of signal transduction and kinase control are still not fully understood. We have applied multiple thermal stability measurements and limited

TDP-43 multidomains and RNA modulate interactions and viscoelasticity in biomolecular condensates.

RNA-binding proteins form biomolecular condensates with RNA through phase separation, playing crucial roles in various cellular processes. Although intrinsically disordered regions (IDRs) are key drivers of phase separation, additional factors such as folded domains and RNA also influence condensate formation and physical properties. However, the molecular mechanisms underlying this regulation remain elusive. Here, using molecular dynamics simulations, we investigate how the multidomain structure of TDP-43, which consists of its IDR, RNA recognition motifs (RRMs), and N-terminal domain (NTD),

Design, synthesis and biological evaluation of triazolo[1,5-a]pyrimidinone derivatives as potent and orally efficacious WRN helicase inhibitors for MSI cancer therapy.

Microsatellite instability-high (MSI-H) cancers represent an unmet clinical need due to treatment resistance, and WRN helicase is a validated synthetic lethal target for this indication. Herein, we rationally designed, synthesized, and biologically evaluated a novel series of triazolo[1,5-a]pyrimidinone derivatives as potent WRN helicase inhibitors. Systematic structure-activity relationship (SAR) exploration identified optimal pharmacophores: 4-chlorobenzothiophene/2,2-difluoro-4-bromo-1,3-benzodioxole as R1, dihydro-2H-pyran as R2, and 3-hydroxypyridine/4-methyl-5-hydroxypyrimidines as R3. ADAPT-M: a workflow for rapid, quantitative in vitro measurements of enriched protein libraries.

Protein-protein interactions underpin most cellular processes, and engineered binders present powerful tools for probing biology and developing novel therapeutics. However, scalable, quantitative characterization of large numbers of candidates remains a major bottleneck. Here we show that ADAPT-M (Affinity Determination by Adaptation of ProTein binders for Microfluidics) enables rapid, parallel measurement of binding affinities and dissociation behavior directly from enriched display libraries in under one week, without requiring gene synthesis or hands-on protein purification. Engineered PCV2 VLPs for Antigen Delivery: A Platform for Next-Generation Veterinary Vaccines.

The capsid (Cap) protein of porcine circovirus type 2 (PCV2) self-assembles into virus-like particles (VLPs) that form stable, noninfectious nanoparticles with T = 1 icosahedral symmetry. Mimicking native virion architecture, these VLPs display antigens in a highly ordered array, conferring strong immunogenicity for both humoral and cellular responses. This review outlines the structural basis for their use as an antigen display platform, detailing molecular engineering strategies that target three defined sites, each with distinct functional roles: the N‑terminal for CD8+ T-cell activation, s

Discovery of a novel and potent KRASG12V-targeting peptide with antiproliferative activity against colorectal cancer cells.

Despite the clinical relevance of KRASG12V in colorectal cancer, KRASG12V-specific inhibitors remain limited. Through structure-based virtual screening of a 59,319-member peptide library, we identified four KRASG12V-targeting peptides, among which Peptide-1 showed the most favourable docking profile. MST assays confirmed Peptide-1 had the highest affinity among Peptides 1-4, with stronger binding to KRASG12V than to other KRAS mutants. Structural analysis, molecular dynamics simulations, and free-energy calculations indicated that Peptide-1 formed a stable and energetically favourable complex

Wood Carbon Sponge/SiO2 Aerogel Composites with Anisotropic Broadband Microwave Absorption, Elastic Resilience, and Thermal Insulation.

Biomass-derived carbon aerogels have attracted considerable interest for lightweight microwave-absorption owing to their hierarchical porosity and renewability. While the structural anisotropy of natural wood is well recognized, its consequences for electromagnetic response remain largely unexplored. Herein, we utilize SiO2 aerogel-incorporated wood-derived carbon sponge as a model system to investigate its electromagnetic anisotropy in the axial, radial, and tangential directions. Governed by the aligned cellular architecture, a robust permittivity ordering (tangential > radial > axial) is re

Cellular thermal shift assay of subcellular isolates to evaluate drug-membrane target interactions.

The cellular thermal shift assay (CETSA) is a tool for target identification and validation in drug discovery. It relies on thermal melting curves to indicate drug binding and is performed in cells, cell lysates, or purified protein. These approaches can disrupt the structural integrity of membrane proteins, hindering drug-target engagement. We describe the first application of CETSA in isolated mitochondria and show the effects of this approach on the analysis of the compound UK5099 and its known binding target, the mitochondrial pyruvate carrier (MPC), a mitochondrial inner membrane-localize

Molecular insights into the anti-browning effect of protocatechualdehyde.

Polyphenol oxidase (PPO) is a vital polyphenol-metabolizing enzyme responsible for the enzymatic browning of fruits and vegetables. This study assessed the anti-PPO activity of protocatechualdehyde (PCA), yielding an IC50 value of 94 μM (95% CI: 91.8 to 96.4 μM). Kinetic analysis revealed that PCA acted as a reversible and competitive inhibitor of PPO. Molecular docking indicated that hydrogen bonding and hydrophobic interactions were the main driving forces for PCA-PPO binding. Fluorescence spectroscopy further demonstrated that PCA binding altered the microenvironments around tyrosine and tr

Novel C3-Aminated Galantamine Derivatives as Selective Butyrylcholinesterase Inhibitors With Neuroprotective Effects.

To explore natural-inspired novel anti-AD bioactive molecules, a series of galantamine derivatives 6a-6u were synthesized via Pd-catalyzed Buchwald-Hartwig cross-coupling, with various nitrogen-containing functional groups installed at the C3 position of galantamine. Compared with galantamine, these compounds showed greatly enhanced selective BChE inhibition activity, suggesting that the C3 position of galantamine might be a key modifiable site for developing new selective BChE inhibitors. Among them, the derivative 6l which contained a 3,5-difluoroaniline structure moiety exhibited promising
