Microarray-based Detection of Critical Overexpressed Genes in the Progression of Hepatic Fibrosis in Non-alcoholic Fatty Liver Disease: A Protein-protein Interaction Network Analysis.

BACKGROUND: Non-alcoholic fatty liver disease (NAFLD) is a prevalent cause of chronic liver disease and encompasses a broad spectrum of disorders, including simple steatosis, steatohepatitis, fibrosis, cirrhosis, and liver cancer. However, due to the global epidemic of NAFLD, where invasive liver biopsy is the gold standard for diagnosis, it is necessary to identify a more practical method for early NAFLD diagnosis with useful therapeutic targets; as such, molecular biomarkers could most readily serve these aims. To this end, we explored the hub genes and biological pathways in fibrosis progre

A Role for the VPS Retromer in Brucella Intracellular Replication Revealed by Genomewide siRNA Screening.

Brucella, the agent causing brucellosis, is a major zoonotic pathogen with worldwide distribution. Brucella resides and replicates inside infected host cells in membrane-bound compartments called Brucella-containing vacuoles (BCVs). Following uptake, Brucella resides in endosomal BCVs (eBCVs) that gradually mature from early to late endosomal features. Through a poorly understood process that is key to the intracellular lifestyle of Brucella, the eBCV escapes fusion with lysosomes by transitioning to the replicative BCV (rBCV), a replicative niche directly connected to the endoplasmic reticulu

Serotonergic cortico-limbic and executive network dysfunction in Parkinson's disease impulse control disorders: a PET-fMRI study.

Impulse control disorders (ICDs) affect up to 45% of Parkinson's disease (PD) patients, yet their neural mechanisms remain unclear. Using multimodal PET and resting-state fMRI in 23 PD patients (11 PDICD + , 12 PD-ICD-) and 14 healthy controls, we identified specific brain pathways underlying ICDs. PDICD+ patients showed steeper delay discounting and altered functional connectivity, including enhanced posterior parietal coupling within executive networks and disrupted salience-executive interactions. Critically, aberrant right supplementary motor area-amygdala connectivity was linked to ICD se

Prodh2-Mediated Mitochondrial Stress Drives TNF-α-Induced Myoblast Dysfunction and Sarcopenia in COPD.

Sarcopenia in chronic obstructive pulmonary disease (COPD) is driven by elevated tumor necrosis factor-alpha (TNF-α), yet the underlying mechanism remains unclear. Here, we identify the mitochondrial metabolic enzyme Prodh2 as a pivotal mediator. Prodh2 was significantly upregulated in sarcopenic COPD patients and a corresponding mouse model. Mechanistically, TNF-α induced Prodh2 transcription in myoblasts via a p53-dependent pathway. This upregulation triggered excessive mitochondrial reactive oxygen species, leading to the release of mitochondrial DNA (mtDNA). Cytosolic mtDNA subsequently ac

Regulation of T Cell Senescence in Health and Diseases.

Aging is accompanied by a progressive decline in immune function, a process termed immunosenescence, which affects both innate and adaptive immune compartments. Among these, the adaptive immune system-and particularly T cells-undergoes the most profound functional and phenotypic alterations, critically impairing host defense against infections, cancer, and vaccination responses. The age-associated decline of adaptive immunity is shaped by the divergent senescence pathways of CD4+ helper and CD8+ cytotoxic T cells. While both lineages enter a state of cell-cycle arrest, their distinct immunolog

Family function and problem behaviors among preadolescents: A mediation model of generalized trust beliefs and peer attachment.

Preadolescence marks the critical onset period for problem behaviors that shape later adjustment. The purpose of this study was to investigate whether family function is associated with preadolescent problem behaviors through the sequential mediation of generalized trust beliefs and peer attachment, while exploring potential gender differences in these pathways. A total of 371 preadolescents (Mage = 11.58 ± 0.58) completed self-reported questionnaires. Results of latent variable structural equation modeling showed that family function was negatively associated with internalizing problems large

"Pulling-Out" N2 from Diazo Compounds by Iron Complex for Carbene Formation.

One of the main pathways to (metal)carbene formation proceeds via the decomposition of diazo compounds, which can be triggered thermally, photochemically, or by transition-metal mediation. An alternative pathway for carbene generation from diazo compounds is the "pulling-out" N2 by different reagents. Herein, we report an iron-mediated strategy for the homolytic cleavage of C═N bond in diazo compounds, enabling the generation of free carbenes and iron-dinitrogen complexes under mild conditions. The corresponding Fe-diazo intermediates were isolated and fully characterized by X-ray crystallogra

Functional Autoantibodies in Myocardial Diseases.

Myocardial autoimmune responses result in distinctive cardiac diseases and are associated with poor clinical outcomes. Autoantibodies (AAbs) play a crucial role in myocardial humoral autoimmunity by targeting endogenous tissue, or "self-antigens." Myocardial tissue injury or myocyte disarray caused by AAbs triggers a cascade of defensive immunological responses that may lead to worsening biventricular remodeling. However, over the past decade, the concept of AAbs in myocardial diseases has shifted from viewing AAbs as uniformly deleterious to a more nuanced explanation of AAbs influencing down

Hyperglycemia differentially regulates osteoblast and osteoclast autophagy via AMPK/mTOR/p70 S6K signaling in diabetic osteoporosis.

Type 2 diabetes mellitus (T2DM) often induces diabetic osteoporosis (DOP) with impaired bone remodeling, yet its underlying mechanism remains elusive. This study identified the differential regulatory role of the AMPK/mTOR/p70 S6K signaling axis in bone cell function. In vivo, diabetes reduced AMPK phosphorylation, enhanced mTOR/p70 S6K activation, and diminished autophagy in rat femoral tissue. In vitro, HG exerted cell-type-specific effects via the AMPK signaling pathway: in osteoblasts, HG inhibited AMPK phosphorylation, activated mTOR/p70 S6K, suppressed autophagy, and impaired mineralizat

ANXA1-Derived Peptide Increases Melanoma Cell Sensitivity to Vemurafenib by Downregulating EphA2.

Vemurafenib (VEM) is a BRAF inhibitor that improves the prognosis of melanoma, but acquired resistance represents a key limitation to its clinical efficacy. This study investigated the potential of A11, an Annexin A1 (ANXA1)-derived peptide, to enhance VEM efficacy in both VEM-sensitive and VEM-resistant melanoma cells. We evaluated the effects of A11 in melanoma through in vitro assays (including MTT, clonogenic survival, apoptosis, and cell cycle analysis) and in vivo xenograft models in nude mice. Furthermore, we mechanistically interrogated A11-mediated suppression of EphA2 expression and

Multi-Omics Reveals Impact of Hypoxia on Lactate Metabolic Switch and Monoclonal Antibody Production in CHO Fed-Batch Cultivation.

Oxygen supply is critical for mammalian cell culture. Insufficient oxygen supply or oxygen inhomogeneity in large-scale bioreactors often leads to attenuated process performance, such as reduced cell growth, viability, and productivity. However, the understanding of how hypoxia affects process performance by modulating the underlying biological mechanisms of Chinese hamster ovary (CHO) cells remains limited. In this study, a comprehensive multi-omics analysis was performed to investigate the impact of hypoxia on intracellular biology. Systematic analysis of the multi-omics data revealed that m

EPDR1 promotes gastric cancer progression via STAT3-mediated fatty acid metabolic reprogramming.

EPDR1 has been shown to play critical regulatory roles in development of several types of cancer. Nevertheless, the role of EPDR1, a family of glycoproteins involved in cell-cell contact, to gastric cancer (GC) remain unknown. Here, we determined that EPDR1 is significantly upregulated in GC tissues and cell lines, and is associated with poor prognosis. In vitro and in vivo functional assays demonstrate that EPDR1 promotes the proliferation, migration, and invasion of GC cells. Mechanistically, EPDR1 regulates the expression level of CPT1A, thereby mediating metabolic reprogramming of the fatt

Src/FN1 pathway activation drives tumor cell cluster formation and metastasis in lung cancer: A promising therapeutic target.

Lung cancer remains the leading cause of cancer-related death globally, with metastasis driven by circulating tumor cells (CTCs)-particularly clusters-being a major treatment challenge. Despite their critical role, the biological differences between single CTCs and CTC clusters remain unclear. Here, we comprehensively compared their behavioral, transcriptomic, and proteomic profiles in lung cancer models. Compared with single cells, CTC clusters present enhanced metastatic potential, greater survival in the bloodstream and increased resistance to microenvironment. Mechanistically, the Src/FN1

Revealing rutaecarpine's promise: A pathway to parkinson's disease relief through PPAR modulation.

The pathological mechanisms of Parkinson's disease (PD) is complex, and no definitive cure currently exists. This study identified Rutaecarpine (Rut), an alkaloid extracted from natural plants, as a potential therapeutic agent for PD. To elucidate its mechanisms of action and specific effects in PD, network pharmacology, molecular docking, and experimental validation methods were employed. Our findings demonstrated the efficacy of Rut in ameliorating PD symptoms. Network pharmacology analysis indicated that Rut exerts its therapeutic effects through the PPAR signaling pathway and the lipid pat

Ferroptosis role in complexity of cell death: unrevealing mechanisms in Parkinson's disease and therapeutic approaches.

Parkinson's disease (PD), a common neurodegenerative disorder, is characterized by progressive loss of dopaminergic neurons, and accumulation of α-synuclein in the substantial nigra. Emerging evidence identifies ferroptosis as a regulated iron-dependent cell death mechanism marked by excessive lipid peroxidation (LPO) as a key contributor to PD pathogenesis. Ferroptosis is intertwined with critical disease processes such as aggregation of α-synuclein protein, oxidative stress generation, mitochondrial alteration, iron homeostasis dysregulation, and neuroinflammation. This mechanism disrupts ce

Regulation and therapy: the role of ferroptosis in DLBCL.

Diffuse large B-cell lymphoma (DLBCL) is the most common subtype of B-cell non-Hodgkin's lymphoma (NHL), up to 30%-40% of patients will relapse and 10%-15% of patients have primary refractory disease, so exploring new treatment options is necessary. Ferroptosis is a non-apoptotic cell death mode discovered in recent years. Its occurrence pathway plays an essential impact on the therapeutic effect of tumors. Numerous studies have shown that modulating critical factors in the ferroptosis pathway can influence the growth of tumor cells in hematological malignancies including DLBCL. This review hi

Zearalenone Regulates Energy Metabolism and Proliferation of Goat Endometrial Epithelial Cells through Estrogen Receptor 1.

Zearalenone (ZEN), an estrogenic mycotoxin inducing reproductive toxicity, disrupts endometrial cell function and contributes to pregnancy failure, although its endometrial toxicity mechanisms remain unclear. This study reveals that ZEN inhibits goat endometrial epithelial cells (EECs) proliferation through estrogen receptor 1 (ESR1) homodimers. ESR1 knockout reversed ZEN-induced mitochondrial structural/functional damage via the ESR1-OMA1 zinc metallopeptidase (OMA1) axis. Furthermore, ZEN suppresses glycolysis and mitochondrial pyruvate transport in goat EECs through the ESR1-pyruvate kinase
