High endothelial cells (HECs) control lymphocyte homing for adaptive immunity. They are defined by plump morphology, reflecting endoplasmic reticulum (ER) and Golgi expansion, and are by display of sulfo-sialomucins of the peripheral node addressin (PNAd) critical for lymphocyte recruitment. How these features are linked was unclear. Here, we found that HEC genes for sulfoglycoprotein synthesis and organelle expansion are enriched in binding motifs for ER stress-response transcription factors XBP1 and CREB3L2. These factors bound and drove expression from conserved elements in sulfo- and fucos

Köhlmeier-Degos disease (Degos disease [DD]) is a rare vasculopathy with characteristic skin lesions and life-threatening gastrointestinal and cerebrovascular involvement. Although DD is often viewed as a thrombo-obliterative disorder, its immunopathology remains poorly defined. Here, single-cell RNA and T cell receptor sequencing of skin, blood, and cerebrospinal fluid from DD patients reveal pervasive type I and type II interferon activation, with an interferon-γ-biased program compared with systemic lupus erythematosus. Cytotoxic CD8A+ T cells show interferon-responsive activation and restr

Spinal cord injury (SCI) triggers a multifaceted cascade of cellular and molecular events that profoundly influence the extent of secondary damage. Central to this process, microglia-the innate immune cells of the central nervous system-display a range of programmed cell death pathways that have significant implications for injury outcomes. This article mainly focuses on three key programmed cell death modalities that have emerged in SCI: ferroptosis, autophagy, and pyroptosis. Ferroptosis, characterized by iron-dependent lipid peroxidation, autophagy, which can serve dual roles in cell surviv

Oncogenic KRAS mutations underlie some of the deadliest human cancers. Genetic or pharmacological KRAS inactivation produces mixed outcomes and frequent relapse. Mechanisms of tumor resistance to KRAS inhibition remain poorly understood. We present evidence that STAT3 supports tumor growth following KRAS depletion. Using a conceptual framework of pancreatic ductal adenocarcinoma, we show that cancer cells that survive CRISPR-mediated ablation of mutant KRAS are dependent on STAT3 function to maintain tumorigenicity. Mechanistically, the combined loss of mutant KRAS and STAT3 disrupts a core tr

Immunotherapy efficacy is hindered by the immunosuppressive metabolism of cancer cells and tumor-associated macrophages (TAMs), yet their opposite metabolic programs complicate synchronized modulation of tumor microenvironment. Here, we report an acid-activated Fe-Zn nanocomplex (FZNC) that transforms into spiky FeOOH nanoparticles within the tumor microenvironment. This transformation enhances cellular uptake and enables selective scavenging of hydrogen sulfide (H2S)-a metabolite that promotes glycolysis in cancer cells and oxidative phosphorylation (OXPHOS) in TAMs. Local H2S depletion by FZ

Natural killer (NK) cells are innate lymphocytes that play a critical role in host defense against viral infection. In addition to rapid effector cytokine production and direct cytotoxicity, NK cells exhibit features of adaptive immunity, including the capacity to undergo robust antigen-specific clonal proliferation and to generate immunological memory1-3. However, the transcriptional programs and regulators governing dynamic NK cell responses to viral infection have not been fully uncovered. In this study, we identified transcription factor 19 (TCF19) as a key driver of NK cell proliferation

It is possible that parasites may influence the course of COVID-19 infection, as either risk factors or protective agents; as such, the current coronavirus pandemic may affect the diagnosis and prevention of parasitic disease, and its elimination programs. The present review highlights the similarity between the symptoms of human parasitoses and those of COVID-19 and discuss their mutual influence. The study evaluated selected human parasitoses with similar symptoms to COVID-19 and examined their potential influence on SARS-CoV-2 virus invasion. The available data suggest that at least several

How tissue-specific anatomical distribution and phenotypic specialization are linked to protective efficacy of memory T cells against reinfection is unclear. Here, we show that lung environmental cues program recently recruited central-like memory cells with migratory potentials for their tissue-specific functions during lethal respiratory virus infection. After entering the lung, some central-like cells retain their original CD27hiCXCR3hi phenotype, enabling them to localize near the infected bronchiolar epithelium and airway lumen to function as the first line of defense against pathogen enc

The interaction of programmed death-1 (PD-1) with its ligand, programmed death ligand-1 (PD-L1), has been considered to play a key role in the negative regulation of immune responses. Patients with diffuse cutaneous systemic sclerosis (SSc) had higher levels of soluble PD-1 (sPD-1) than those with limited cutaneous SSc and healthy individuals. Serum sPD-1 levels positively correlated with the severity of skin sclerosis. In contrast, serum sPD-L1 levels were significantly increased in patients with SSc compared with healthy individuals. Moreover, serum sPD-L1 levels were not associated with the

In latent tuberculosis infection (LTBI) spread of the bacteria is contained by a persistent immune response, which includes CD4(+) T cells as important contributors. In this study we show that TB-specific CD4(+) T cells have a characteristic chemokine expression signature (CCR6(+)CXCR3(+)CCR4(-)), and that the overall number of these cells is significantly increased in LTBI donors compared with healthy subjects. We have comprehensively characterized the transcriptional signature of CCR6(+)CXCR3(+)CCR4(-) cells and found significant differences to conventional Th1, Th17, and Th2 cells, but no m

Although hematopoietic cell transplantation (HCT) is an effective treatment option for patients with life-threatening blood, immune system, or genetic disorders, many barriers besides a lack of suitably matched donors exist and can have an adverse impact on access and outcomes of HCT. In 2008, the National Marrow Donor Program, through its Office of Patient Advocacy, convened a diverse group of experts and transplantation survivors to identify persistent patient barriers throughout the transplantation process and to make recommendations for programs and initiatives to address these barriers, i

This study identified factors impacting adherence to human immunodeficiency virus (HIV) therapies in a sample of 46 persons diagnosed with HIV/acquired immune deficiency syndrome and severe mental illness who were receiving care through a day care program in Northern California. Comprehensive descriptions of factors and circumstances that influence adherence behaviors were obtained through critical incident interviews, and a taxonomy of adherence factors was determined. A chi2 analysis that examined differences between patient characteristics and major adherence factors found that planning was

Failure of acute inflammation to resolve leads to persistence of the inflammatory response and may contribute to the development of chronic inflammation. Thus, an understanding of inflammatory resolution will provide insight into the etiology of chronic inflammation. In an acute pleurisy, polymorphonuclear leukocytes (PMNs) were found to predominate at the onset of the lesion but decreased in number by undergoing apoptosis, the principal mechanism by which PMNs died in this model. PMNs were progressively replaced by monocytes, which differentiated into macrophages. As with PMNs, macrophages al

In Japan, a nationwide prevention program against mother-to-infant infection by hepatitis B virus (HBV) started in 1985. This program consists of double screenings of pregnant women and prophylactic treatment to the infants born to both hepatitis B surface antigen (HBsAg) and hepatitis B e antigen (HBeAg) positive mothers. These infants are treated with two injections of hepatitis B immune globulin (HBIG) and at least three injections of plasma derived hepatitis B vaccine. We sent questionnaires about the numbers of each procedure or examination during nine months of investigation period to ea

BACKGROUND: Anti(α)-programmed cell death-1 (PD-1)/programmed death-ligand 1 (PD-L1) monotherapy fails to provide durable clinical benefit for most patients with carcinoma. Recent studies suggested that strategies to reduce immunosuppressive cells, promote systemic T-cell responses and lymphocyte trafficking to the tumor microenvironment (TME) may improve efficacy. N-809 is a first-in-class bifunctional agent comprising the interleukin (IL)-15 superagonist N-803 fused to two αPD-L1 domains. Thus, N-809 can potentially stimulate effector immune cells through IL-15 and block immunosuppressive PD

BACKGROUND: Maternal immune activation (MIA) is associated with increased risk for neurodevelopmental disorders, including schizophrenia and autism spectrum disorder, yet its long-term impact on intestinal immune-microbial programming remains incompletely understood. Here, we investigated whether lipopolysaccharide (LPS)-induced MIA drives persistent alterations in the offspring mucosal immune environment, with a focus on intraepithelial lymphocytes, mucosa-associated microbiota, systemic cytokines, and prefrontal cortical immune-related readouts.
METHODS: MIA was induced in wild-type BALB/cB

Early embryonic loss remains a major constraint to reproductive efficiency in cattle, with a large proportion of failures occurring during early gestation, particularly across the peri-implantation period encompassing maternal recognition of pregnancy (MRP). During this critical window, conceptus-derived interferon tau (IFNT) acts as a key spatiotemporal regulator of immune and endocrine adaptation required for pregnancy establishment. Beyond its classical luteotropic role in preventing prostaglandin F₂α-mediated luteolysis, IFNT exerts cell- and compartment-specific immunomodulatory actions a

Dendritic cells (DCs) critically regulate immune dynamics within the tumor microenvironment (TME), yet the mechanisms governing the balance between effective anti-tumor immunity and immune tolerance remain incompletely defined. Existing reviews have largely examined DC biology, tumor immunology and neuroinflammation within disease-specific frameworks, leaving insufficient attention to how DC dysfunction may be compared across chronic pathological microenvironments. Here, we propose DC-driven immune rebalancing as a context-dependent framework for understanding how antigen-presenting cell state

BACKGROUND: IL-17-producing Type 17 T (T17) cells are central drivers of inflammation in both psoriasis and hidradenitis suppurativa (HS), as evidenced by the clinical efficacy of IL-17-targeting therapies. However, therapeutic responses differ substantially between diseases, raising the possibility that the composition and regulation of T17 states are disease context dependent.
OBJECTIVE: To define disease-specific T17 subset composition and regulatory T-cell (Treg) transcriptional programs in psoriasis and HS at single-cell resolution, and to generate hypothesis framework regarding their po

Pulmonary tuberculosis (TB), caused by lung infection with Mycobacterium tuberculosis (Mtb), often manifests both alveolar pneumonia and aggregation of immune cells containing Mtb, termed granulomas, within the same lung. Here, we integrate spatial transcriptomics, single-cell RNA sequencing, high-resolution imaging, and functional assays of lung biopsies from four men with pulmonary TB to compare alveolar pneumonia with adjacent granulomas. We find that alveolar TB pneumonia is enriched for TREM2⁺ lipid-laden macrophages characterized by increased lipid metabolism gene expression, reduced T c
