Cyclic AMP as a possible mediator of dopamine stimulation of cockroach gland cells.

Isolated salivary glands of the cockroach Nauphoeta cinerea Olivier secrete fluid in response to nerve stimulation or application of dopamine, the acinar cells undergoing a hyperpolarization during secretion. The aim of the present work was to examine whether cyclic AMP acts as a second messenger in the acinar cells to cause the secretory and electrical responses to the transmitter dopamine. Cyclic AMP (10-500 microM) in the bathing solution of isolated glands caused a dose-dependent secretory response but no change in the membrane potential of acinar cells. The time courses and magnitudes of

The effect of diamide on transmitter release and on synaptic vesicle population at vertebrate synapses.

Diamide, a sulfhydryl-oxidizing agent, has previously been shown to cause acetylcholine release in two preparations in the absence of added Ca2+. Similarities in action between diamide and alpha-latrotoxin, a component of black widow spider venom which causes transmitter release with no added Ca2+, and which seems to require a disulfide bond for its action, led us to study further the transmitter-releasing properties of diamide. In rat cerebral cortical slices we show that diamide, like alpha-latrotoxin, released all transmitters studied; GABA, acetylcholine, norepinephrine and dopamine. The r

Conflicting electrophysiological and anatomical data from drug-impaired guinea pig cochleas.

In guinea pigs that had been treated with very large doses of the aminoglycoside amikacin (14 x 450 mg/kg/day, i.m.) clear, short-latency responses to various click stimuli could be recorded at the round window. When the same cochleas were examined histologically, no outer or inner hair cells could be found along the entire length of the basilar membrane, save for a very few outer hair cells remaining at the apex. The response patterns resembled that of the compound action potential, and various characteristics suggest that they were of neural origin. Vestibular function, investigated by elect

Effects of antiinflammatory agents and prostaglandins on acid and bicarbonate secretions in the amphibian-isolated gastric mucosa.

Effects of antiinflammatory agents and prostaglandins on H+ and HCO-3 secretions and electrical properties were investigated in the amphibian-isolated gastric mucosa. Gastric HCO-3 transport was studied in Rana temporaria fundus, in which H+ secretion had been inhibited with the histamine H2-receptor antagonists metiamide or cimetidine (10(-3) M), and in Necturus antrum, which secreted HCO-3 spontaneously. Hydrocortisone (100-500 microgram/ml) had no effect on H+ or HCO-3 secretion in the fundus. Indomethacin (10(-4) M) was a considerably more potent inhibitor of HCO-3 secretion than of H+ sec

[Pathogenesis of arrhythmias in acute myocardial infarction].

Over the last few years, much experimental work has been done to try and elucidate the pathogenesis of disorders of rhythm in myocardial infarction. The role of bradycardia during the first few hours has been discussed, and the use of atropine has been criticised. Some light has been thrown on the importance of the Purkinje network in the infarcted zone. Some new concepts have been applied to the disturbances in exchange across membranes. The authors present the results of a series of experiments on the action of lactate and that of acidosis. The latter is responsible for upturns in the descen

Effects of botulinum toxin on neuromuscular transmission in the rat.

1. Botulinum toxin (BoTx) type A partially blocks spontaneous transmitter release from nerve terminals in the rat. Minature end-plate potentials (m.e.p.p.s) are present at all end-plates, initially with a low frequency but increasing with time after posoning. Their amplitude distribution is at first skew with a predominace of very small m.e.p.p.s but, after a few days, larger than normal m.e.p.p.s appear. 2. Tetanic nerve stimulation, Black Widow Spider Venom, the Caionophore A 23187 or mechanical damage to nerve terminals increases the frequency of m.e.p.p.s and alters the amplitude distribut

A note of the mechanism by which inhibitors of the sodium pump accelerate spontaneous release of transmitter from motor nerve terminals.

1. The actions of 0-1 mM ouabain and of K-free Ringer have been examined at the frog neuromuscular junction. 2. After a delay of more than 30 min, ouabain produces an increase in the miniature end-plate potential (m.e.p.p.) frequency. This increase occurs unchanged in Ca-free Ringer containing 1 mM-EGTA and is therefore unlikely to be due to an entry of Ca into the motor nerve terminals. 3. If the nerve to the preparation is stimulated repetitively in Ca-free Ringer containing 0-1 mM ouabain and 1 mM-EGTA the response of the m.e.p.p. frequency depends on the timing of the tetanus relative to t

Organic Fouling Behavior and Mitigation Strategies in Pressure-Driven Distillation.

Pressure-driven distillation (PD) is a membrane-based water treatment process that uses applied pressure to drive vapor transport through a porous hydrophobic membrane. PD offers potential advantages in selectivity and chemical resilience, but its performance with feedwaters containing foulants remains poorly understood. In this study, we examine the organic fouling behavior of PD, isolate the effect of pressure on flux decline, and explore potential fouling mitigation strategies. We find the severity of organic fouling is highly dependent on foulant properties, with more hydrophobic humic aci

Membranolytic peptide programs immunogenic cell death for cancer therapy.

A rational design for facilitating innovative cell death modes can substantially aid advances in antitumour therapy1,2. Here we design and implement a unique mode of immunogenic membranolytic cell death (mLCD) in tumour cells, characterized by time-lagged rupture from the lysosomal to plasma membranes; this approach was found to robustly potentiate immune checkpoint blockade therapy. This mode of mLCD was induced by the synthetic-acid-responsive membranolytic peptide (aMP) aMPC16-CA50, which exhibits hierarchical responsiveness to the decreasing pH associated with the tumour extracellular envi

Reaction rate calculations indicate that α-tocopherol primarily acts as a membrane protein antioxidant in vivo.

Lipid peroxidation is a complex free radical chain reaction in which lipid peroxyl radicals serially attack other components of the lipid bilayer. As a condensed phase, the lipid bilayer contains numerous potentially reactive substrates beyond unsaturated fatty acids, including membrane proteins and dissolved small molecules like α-tocopherol, the major lipid-soluble antioxidant. Despite a long history of investigation, the reaction cascades that do succeed in vivo to produce tocopherol's pronounced antioxidant effect are still arguable. Here, we have revisited the tocopherol reaction cascade

In Situ Tracking of Membrane HER2 - Antibody-Drug Conjugate Interaction with Surface Plasmon Resonance Imaging.

Antibody-drug conjugates (ADCs) rely on specific recognition of tumor-associated membrane receptors to achieve targeted intracellular drug delivery, yet in situ characterization of their interaction dynamics remains limited. Here, we develop a single-cell plasmonic imaging platform to quantitatively resolve the interaction between the membrane human epidermal growth factor receptor 2 (HER2) and HER2-targeted therapeutics. This label-free approach enables continuous monitoring of the molecular interaction process, allowing real-time extraction of detailed binding kinetics. Using this platform,

ISL@mTOMV nanoparticles: a promising approach for VM-targeted therapy in breast cancer.

Vasculogenic mimicry (VM) in breast cancer is recognized as an important non-endothelium-dependent vascular mechanism that promotes tumor invasiveness, survival, and metastatic potential; however, effective and specific intervention strategies remain limited. In this study, we innovatively constructed a nanodelivery system based on hybrid membranes derived from bacterial outer membrane vesicles and breast cancer cell membranes, termed mTOMV, and loaded it with the natural flavonoid isoliquiritigenin (ISL) to enhance targeted antitumor activity. ISL@mTOMV nanoparticles exhibited a uniform spher

Myoferlin at the crossroad of vesicle trafficking and mitochondrial function: implications for pancreatic cancer progression and stromal reprogramming.

Myoferlin, a type 2 transmembrane protein in the ferlin family, is traditionally known for its role in membrane fusion during muscle development and repair. Recent research identifies myoferlin as a potential biomarker and a critical driver of cancer progression, particularly in breast cancer and pancreatic ductal adenocarcinoma. While its lack of specificity limits its use as a biomarker, its multifaceted role in cellular membrane dynamics makes it a promising therapeutic target. In cancer cells, myoferlin regulates the recycling and stability of receptor tyrosine kinases, thereby promoting i

Inhibiting AIDA suppresses VSMC-derived foam cell formation and atherosclerosis by hindering CD36 membrane translocation.

Vascular smooth muscle cell (VSMC)-derived foam cells critically drive atherosclerotic plaque progression, yet their regulatory mechanisms remain incompletely understood. This study aimed to elucidate the pathophysiological role of the VSMC-enriched factor axin interactor, dorsalization-associated (AIDA) in this process and evaluate its therapeutic potential. We utilized VSMC-specific AIDA knockout in male ApoE-/- mice and adenoviral shRNA silencing in oxLDL-stimulated murine aortic vascular smooth muscle cells (MOVAS), combined with co-immunoprecipitation (Co-IP), cholesterol flux assays, and

Aligned Electrospun PELA Fibers Promote the Neuronal Orientation and Attenuate Neurotoxicity.

Neural regeneration is one of the potential therapeutic approaches for Alzheimer's disease (AD). While aligned electrospun fibers have shown promise in guiding neural regeneration, conventional materials face limitations such as poor hydrophilicity and acidic degradation byproducts. The amphiphilic copolymer poly(lactic-co-glycolic acid)-block-polyethylene glycol (PELA) combines mechanical tunability with enhanced biocompatibility, but its potential as an aligned fibrous membrane for neural regeneration remains unexplored. Here, we fabricated aligned electrospun PELA fibrous membranes (AEF) by

Enhancing ethanol tolerance and metabolic activity of Saccharomyces cerevisiae during Rice fermentation by fixed-frequency ultrasound.

Ultrasound pretreatment shows potential for enhancing microbial fermentation efficiency, but its mechanochemical impact on S. cerevisiae during rice fermentation, particularly regarding ethanol tolerance and metabolic activity, requires systematic investigation. Therefore, the multi-dimensional influence of ultrasound on S. cerevisiae was studied from the aspects of biomass dynamics under ethanol stress, membrane biophysics, metabolic markers, and oxidative stress response. Following the ultrasound treatment (logarithmic metaphase, 28 kHz, and 45 W/L), the growth rate of S. cerevisiae was fast

Highly adhesive asynchronous drug-releasing membrane for facial care: Combined anti-acne and anti-inflammatory properties.

The facial skin of teenagers often suffers from problems such as acne and inflammation. In severe cases, it may even lead to skin ulcers. This not only affects their study and daily life, but also has a negative impact on their physical and mental health. However, conventional hydrocolloid dressings exhibit several limitations, such as poor adhesion in high-tension or contoured facial areas, lack of staged therapeutic functionality, and the inclusion of potentially irritating ingredients. To overcome these challenges, electrospun nanofiber membranes with a core-shell architecture have been dev
