Evaluation of fibroblast-mediated gene therapy in a feline model of mucopolysaccharidosis type VI.

Fibroblast-mediated ex vivo gene therapy was evaluated in the N-acetylgalactosamine 4-sulfatase (4S) deficient mucopolysaccharidosis type VI (MPS VI) cat. Skin biopsies were obtained at birth from severely affected MPS VI kittens and used to initiate fibroblast outgrowths for retroviral transduction with the 4S cDNA. 4S gene expression in transduced cells was under the transcriptional control of the MoMLV long terminal repeat promoter or the cytomegalovirus (CMV) immediate-early promoter. Characterisation of gene-transduced fibroblasts demonstrated the cells to be over-expressing 4S activity.

Nucleotide excision repair and the link with transcription.

Nucleotide excision repair (NER) uses the products of about 30 genes to remove a damage-containing oligonucleotide from cellular DNA. The transcription factor TFIIH is an essential component of NER. In man, defects in NER can result in three distinct genetic disorders, whose features can be ascribed to abnormalities in DNA repair or transcription.

Turner syndrome and female sex chromosome aberrations: deduction of the principal factors involved in the development of clinical features.

Although clinical features in Turner syndrome have been well defined, underlying genetic factors have not been clarified. To deduce the factors leading to the development of clinical features, we took the following four steps: (1) assessment of clinical features in classic 45,X Turner syndrome; (2) review of clinical features in various female sex chromosome aberrations (karyotype-phenotype correlations); (3) assessment of factors that could lead to Turner features; and (4) correlation of the clinical features with the effects of specific factors. The results indicate that the clinical feature

Intrafamilial correlation in idiopathic torsion dystonia.

Age of onset and severity of idiopathic torsion dystonia (ITD) were studied in 100 British families containing 107 index cases and 79 secondary cases. Analysis of variance of these clinical features did not suggest that ITD is genetically heterogenous, and they were similar in Jewish and non-Jewish patients. Intrafamilial correlation for age of onset was low, particularly between parents and their offspring, suggesting that the ITD phenotype may be determined in part by nongenetic factors or an allelic modifying gene.

Molecular basis of hexavalent chromium carcinogenicity: effect on gene expression.

The carcinogenicity of chromium(VI) compounds is explained in terms of the "uptake-reduction" model. According to this model, chromium (VI) enters cells as the anionic tetrahedral species chromate, CrO4(2-), using normal cellular anion transport systems, such as the sulfate transport system. Redox-active enzymes and small molecules reduce chromium (VI) intracellularly and produce "reactive intermediates" capable of damaging cellular constituents. DNA is presumed to be a critical target within the cell; however, chromium (VI) itself is unreactive toward DNA under physiological conditions in vit

Cross-sectional geometry of Pecos Pueblo femora and tibiae--a biomechanical investigation: I. Method and general patterns of variation.

Variation in long bone cross-sectional geometry can be given a more precise functional interpretation using engineering beam theory. However, difficulties in measurement technique have generally prevented studies of large samples of cross sections in this way. In the present study, an automated system utilizing an electronic digitizer and computer software was used to analyze cross-sectional geometric properties of 11 femoral and tibial locations in 119 individuals from the Pecos Pueblo, New Mexico site. The data generated allow identification of clear differences in geometric properties betwe

Improved detection of disease-associated variation by sex-specific characterization and prediction of genes required for fertility.

Despite its great potential, high-throughput functional genomic data are rarely integrated and applied to characterizing the genomic basis of fertility. We obtained and reprocessed over 30 functional genomics datasets from human and mouse germ cells to perform genome-wide prediction of genes underlying various reproductive phenotypes in both species. Genes involved in male fertility are easier to predict than their female analogs. Of the multiple genomic data types examined, protein-protein interactions are by far the most informative for gene prediction, followed by gene expression, and then

Genetics of Vitiligo: A Review.

Vitiligo is a common acquired depigmentation skin disease with obvious family aggregation. About 25-50% of patients have positive family history, which belongs to polygenetic disease. In recent years, through candidate genes and genome-wide association studies, multiple susceptibility gene loci have been found, and studies also show that there is genetic heterogeneity among different populations. Environmental factors can also interact with genetic factors to trigger diseases through various mechanisms. The risk assessment model based on genetic and environmental factors provides a new directi

The heritability of eating disorders: methods and current findings.

Family, twin, and adoption studies of anorexia nervosa (AN), bulimia nervosa (BN), binge-eating disorder (BED), and the proposed purging disorder presentation (PD) have consistently demonstrated that genetic factors contribute to the variance in liability to eating disorders. In addition, endophenotypes and component phenotypes of eating disorders have been evaluated and provide further insight regarding genetic factors influencing eating disorders and eating disorder diagnostic criteria. Many of these phenotypes have demonstrated substantial heritability. This chapter reviews biometrical gene

Familial predisposition to filarial infection--not linked to HLA-A or-B locus specificities.

Two hundred and twenty-five Polynesians were selected from a larger study population for the evaluation of potential genetic influences on the susceptibility to bancroftian filariasis. Analysis showed that there was significantly familial clustering of patients with filariasis and that this clustering was most compatible with genetic transmission of disease susceptibility. The data best fit a model in which the hypothetical gene for filariasis was recessive with a frequency of 0.82 +/- 0.15 in the population and a penetrance of 0.62 +/- 0.14. The alternative hypothesis that susceptibility was

A Genetic Risk Adoption Design for Psychiatric and Substance Use Disorders.

IMPORTANCE: Traditional adoption studies examine disorder-to-disorder parent-offspring transmission. The role of parental genetic risk in offspring disorder transmission can capture indirect genetic effects from parental genotype to parental phenotype to offspring risk. OBJECTIVE: To assess the relative importance of genetic and rearing effects from paternal family genetic risk scores (FGRSs) in 3 pairs of disorders: internalizing (major depression [MD] and anxiety disorders [AD]), substance use (alcohol use disorder [AUD] and drug use disorder [DUD]), and severe (bipolar disorder [BD] and sc

Sex differences in the genetic architecture of cognitive resilience to Alzheimer's disease.

Approximately 30% of elderly adults are cognitively unimpaired at time of death despite the presence of Alzheimer's disease neuropathology at autopsy. Studying individuals who are resilient to the cognitive consequences of Alzheimer's disease neuropathology may uncover novel therapeutic targets to treat Alzheimer's disease. It is well established that there are sex differences in response to Alzheimer's disease pathology, and growing evidence suggests that genetic factors may contribute to these differences. Taken together, we sought to elucidate sex-specific genetic drivers of resilience. We

Microevolutionary models and multi-omics analysis uncover the cross-drug resistance characteristics of capreomycin-selected Mycobacterium tuberculosis.

UNLABELLED: The drug-resistant tuberculosis (TB) is a major public health problem worldwide. Capreomycin (Cm) has been used as an effective drug to treat multidrug-resistant tuberculosis (MDR-TB) since 1960s. Although Cm and amikacin (Am) are historically grouped as second-line injectable drugs (SLIDs) with shared resistance mechanisms (e.g., rrs mutations), recent clinical and genetic data suggests divergent evolutionary pathways. It remains unclear how Mtb evolves from sensitive to resistant phenotypes under Cm pressure. In this study, we generate in vitro drug-resistant Mtb models to system

Should GLP-1 receptor agonist therapy be used to treat obesity in Bardet-Biedl syndrome?

Bardet-Biedl syndrome (BBS) is a complex genetic condition that can affect multiple organ systems, frequently causing pigmentary retinopathy, renal abnormalities, polydactyly, and obesity. Metabolic disturbances including obesity, unsuppressed appetite, and an increased risk of type 2 diabetes (T2D) present clinical management challenges. In this issue of the JCI, Singh et al. present a mouse model of a specific BBS subtype with genetic deletion of the Bbs5 gene. The model recapitulates many of the clinical features observed in patients living with BBS5 and sheds light on adipocyte biology, as

The multifactor pelvis: An alternative to the adaptationist approach of the obstetrical dilemma.

The obstetrical dilemma describes the competing demands that a bipedally adapted pelvis and a large-brained neonate place on human childbirth and is the predominant model within which hypotheses about the evolution of the pelvis are framed. I argue the obstetrical dilemma follows the adaptationist program outlined by Gould and Lewontin in 1979 and should be replaced with a new model, the multifactor pelvis. This change will allow thorough consideration of nonadaptive explanations for the evolution of the human pelvis and avoid negative social impacts from considering human childbirth inherentl

Gene drive mosquitoes can aid malaria elimination by retarding Plasmodium sporogonic development.

Gene drives hold promise for the genetic control of malaria vectors. The development of vector population modification strategies hinges on the availability of effector mechanisms impeding parasite development in transgenic mosquitoes. We augmented a midgut gene of the malaria mosquito Anopheles gambiae to secrete two exogenous antimicrobial peptides, magainin 2 and melittin. This small genetic modification, capable of efficient nonautonomous gene drive, hampers oocyst development in both Plasmodium falciparum and Plasmodium berghei. It delays the release of infectious sporozoites, while it si

Triploid genetic algorithm for convolutional neural network-based diagnosis of mild cognitive impairment.

The diagnosis of mild cognitive impairment (MCI), which is deemed a formative phase of dementia, may greatly assist clinicians in delaying its headway toward dementia. This article proposes a deep learning approach based on a triploid genetic algorithm, a proposed variant of genetic algorithms, for classifying MCI converts and non-converts using structural magnetic resonance imaging data. It also explores the effect of the choice of activation functions and that of the selection of hyper-parameters on the performance of the model. The proposed work is a step toward automated convolutional neur

A bone paradigm challenging the standard model of myeloma oncogenesis.

The standard model of multiple myeloma (MM) oncogenesis from monoclonal gammopathy of undetermined significance (MGUS) relies on genetic instability in the normal counterparts of MM cells. However, the importance of both MGUS-associated and MM-induced bone changes has been recently re-appraised, emphasizing the bone microenvironment (BME) as a tissue of significance. In this review, we propose that early BME alterations (bone senescence and inflammation, i.e., bone inflamm'aging) at the pre-MGUS stage could be causal, and not simply permissive, and creative of phenotypic instability and geneti

Juxtaposition of chemical and mutation-induced developmental defects in zebrafish reveal a copper-chelating activity for kalihinol F.

A major hurdle in using complex systems for drug screening is the difficulty of defining the mechanistic targets of small molecules. The zebrafish provides an excellent model system for juxtaposing developmental phenotypes with mechanism discovery using organism genetics. We carried out a phenotype-based screen of uncharacterized small molecules in zebrafish that produced a variety of chemically induced phenotypes with potential genetic parallels. Specifically, kalihinol F caused an undulated notochord, defects in pigment formation, hematopoiesis, and neural development. These phenotypes were

Phylodynamic inference of the contribution of transmission routes in infectious disease outbreaks.

Understanding which types of contact drive pathogen transmission is critical for outbreak control. Traditionally, inference of transmission events with contact tracing or network analyses is used to identify high-risk contacts. Inference based on genetic data might offer higher accuracy on identifying the true events, but is rarely used to evaluate the contribution of different types of contact to transmission. We extended the Bayesian phylodynamic model phybreak to incorporate data on different types of contacts between cases. The model estimates the fractions of transmission events attributa
