Genome-wide CRISPR/Cas9 screen identified MCL1 as a senolytic target for clearing palbociclib-induced senescent and PD-L1-positive cells in colorectal cancer.

Colorectal cancer (CRC) is the most prevalent digestive system malignancy worldwide. The development of targeted therapeutics specifically effective for CRC is currently in dire need. Preclinical studies showed that CDK4/6 inhibitor palbociclib suppressed the growth of CRC, but whether this effect is durable is unclear. In this study, we aimed to evaluate the roles of palbociclib-induced senescence and find a new strategy to maximize its effectiveness in CRC treatment. Animal and cellular experiments revealed that palbociclib-induced senescence and the senescence-associated secretory phenotype

A NIR-Ⅱ fluorescent probe for real-time visualization and early assessment of responses to CDK4/6 inhibitors in breast cancer.

The evaluation of therapeutic response to cyclin-dependent kinase 4/6 (CDK4/6) inhibitors in hormone receptor-positive (HR+), HER2-negative (HER2-) breast cancer currently relies on anatomical imaging, which suffers from significant delays. To enable early and direct visualization of drug activity, we developed a near-infrared-II (NIR-II) fluorescent molecular probe, HSA-ICi, by conjugating a CDK4/6 inhibitor (Palbociclib or Ribociclib) with ICG and facilitating its self-assembly with human serum albumin. This probe demonstrated specific targeting to the CDK4/6-cyclin D complex, excellent bioc

Targeting histamine H4 receptor improves anti-tumoral response in a murine model of breast cancer.

BACKGROUND: Breast cancer is one of the most common malignant tumors in women worldwide. Histamine (HIS) has been associated with either pro-tumor or anti-tumor effects, depending on the receptor evaluated. It exerts its physiological and pharmacological functions through four receptors (H1R-H4R). In breast cancer, it promotes tumor growth by activating H1 and H2 receptors. Furthermore, HIS released into the tumor microenvironment can promote inflammation associated with tumor development and immunosuppression of the immune response. Several years ago, breast cancer was shown to express the H4

Structure-Based Design, Synthesis, and Biological Evaluation of Oxadiazole-Morpholine Hybrids as Potent PARP-1 Inhibitors Inducing Apoptosis in Breast Cancer Cells.

Poly(ADP-ribose) polymerase-1 (PARP-1) plays a central role in the repair of DNA single-strand breaks and represents an established therapeutic target in cancer treatment. In this study, a series of novel oxadiazole-morpholine hybrid compounds was designed and synthesized using a structure-based drug design approach to target the catalytic domain of PARP-1. The synthesized compounds were evaluated for their cytotoxic activity against breast and ovarian cancer cells, PARP-1 inhibitory potency, and apoptosis-inducing effects. Among the tested compounds, 12a exhibited potent cytotoxic activity ag

Repurposing T-type calcium channel blocker lomerizine as a therapeutic strategy for glioblastoma.

Glioblastoma (GBM) is the most malignant primary brain tumor. The presence of glioma stem/initiating cells (GICs) is known to cause strong treatment resistance; therefore, GICs are a major target for GBM therapy, although there are no therapies targeting GICs clinically. To identify novel treatments for GBMs, we performed drug repurposing screening using GICs and identified the T-type calcium channel blocker lomerizine - a migraine prophylactic drug. Lomerizine inhibited proliferation, migration, invasion, and cell cycle progression and induced apoptosis in GICs and differentiated glioma cells

Sequenced treatment alternatives to relieve adolescent depression: A pragmatic clinical trial.

BACKGROUND: Adolescent major depressive disorder (MDD) is a widespread mental health condition for which treatment outcomes remain suboptimal. For adolescents, it remains unclear whether fluoxetine monotherapy or fluoxetine combined with cognitive-behavioral therapy (CBT) is more effective as an initial treatment. Additionally, no research has compared augmentation versus switching strategies after initial fluoxetine failure. METHOD: We conducted a multistep, multicenter, pragmatical clinical trial with a partially randomized design. In step 1, patients had the opportunity to choose treatment

Efficacy of dolutegravir plus lamivudine in treatment-naïve people with HIV with baseline transmitted drug-resistance mutations.

BACKGROUND: Dolutegravir plus lamivudine (DTG+3TC) is a recommended first-line regimen for people with HIV (PWH), based on its efficacy and safety. However, pivotal trials excluded individuals with transmitted resistance-associated mutations (tRAMs), even when these did not affect regimen activity. The impact of such mutations on DTG+3TC efficacy remains unknown. METHODS: This was a preplanned subanalysis of the D2ARLING trial, a randomized, open-label, phase IV study comparing DTG+3TC vs. DTG+tenofovir disoproxil fumarate with emtricitabine or lamivudine (DTG+TDF/XTC) in antiretroviral-naïve

PARP-1 inhibition attenuates tumor pathology in a novel murine model of diabetes-associated colitis-induced colorectal cancer.

Colorectal cancer is a potentially fatal form of mutagenesis, which is known to be aggravated by other gut associated complications such as ulcerative colitis. In addition, there is emerging evidence to suggest a close correlation between the progression of colorectal cancer and the occurrence of metabolic disorders like diabetes mellitus. Despite these clinical findings, there is a lack of relevant models and pharmacological interventions targeting these complications for the mitigation of overall health benefits. To address this knowledge gap, we designed a study to recapitulate these co-mor

Targeting CDK12 rescues C/EBPβ-mediated platinum and PARP inhibitor resistance in ovarian cancer.

Despite multimodality treatment efforts, resistance to platinum and PARP inhibitors represents a primary impediment to improve prognosis of ovarian cancer. Here, we found that ovarian cancer tissues had higher C/EBPβ expression compared with normal tissues and high C/EBPβ expression predicted unfavorable survival outcomes. Elevated C/EBPβ expression enhanced cisplatin resistance and olaparib resistance. C/EBPβ could affect DDR signals of ovarian cancer. CDK12, serving as a C/EBPβ-regulated DDR-related gene, was directly targeted by and bound with C/EBPβ. C/EBPβ could promote CDK12 expression a

Brief communication: virological outcomes and dolutegravir resistance mutations in HIV-infected patients: a multicenter retrospective cohort study in Mozambique.

The global HIV epidemic remains a major public health challenge, with DTG playing a key role in ART regimens due to its efficacy and tolerability. This study evaluated virological outcomes and resistance mutations in patients on DTG in Mozambique through a retrospective cohort study in seven DREAM centers. Data from 29,601 patients (98.1% on DTG) revealed a virological suppression rate of 95% (27,622/29,051). Factors positively associated with suppression included age > 50, longer ART duration, and being female. Of 17 resistance tests, 8 showed major mutations, including G118R and E138K. Resul

Palbociclib in endocrine-resistant metastatic breast cancer patients: gene polymorphism- based study.

BACKGROUND: Despite the proven benefit of adding palbociclib to endocrine therapy (ET) as demonstrated in pivotal clinical trials (e.g., PALOMA-2 and PALOMA-3), many patients with hormone receptor-positive (HR+) and human epidermal growth factor-2-negative (HER2−) metastatic breast cancer (MBC) still experience early disease progression. This study aimed to identify predictive biomarkers for long-term response and evaluate the impact of adenosine triphosphate-binding cassette subfamily B member 1 (ABCB1) gene polymorphisms in MBC patients who had progressed on prior ET.
