ACC1 inhibition enhances BCG-induced trained immunity by reprogramming acetyl-CoA metabolism.

Trained immunity enhances long-term innate immune responsiveness through metabolic and epigenetic rewiring. Using Bacille Calmette-Guérin (BCG) as a model, we demonstrate that inhibition of acetyl-CoA carboxylase 1 (ACC1) enhances BCG-induced trained immunity by increasing intracellular acetyl-CoA (ACoA) availability. This shift redirects ACoA from lipid biosynthesis toward enhanced tricarboxylic acid cycle flux and histone acetylation, reinforcing metabolic and epigenetic programs that sustain trained immunity. ACC1 inhibitors amplify cytokine production, mitochondrial respiration, and glutam

Combined machine learning identification and experimental validation of NDUFS8 and SUCLG1 as potential biomarkers for mechanical asphyxia.

INTRODUCTION: Mechanical asphyxia (MA) is a major form of violent death caused by external forces that impair respiration and gas exchange. Despite its frequency in forensic casework, its diagnosis remains challenging because autopsy findings are often non-specific. METHODS: In this exploratory study, we combined data-independent acquisition (DIA) proteomics with machine-learning methods to identify candidate myocardial markers of MA. A discovery cohort of human left ventricular myocardium was profiled to identify nominally differentially abundant proteins. The leading candidates were confirm

Use of computer-assisted tomography for diagnosis and standing surgical reduction of progressive ethmoidal hematoma in a mare.

Progressive ethmoidal hematoma (PEH) is a rare, nonneoplastic mass occurring in the sinonasal passages of horses. An 11-year-old, pregnant Thoroughbred mare was presented with a history of intermittent nasal discharge and stertorous respiration. On examination, radiography revealed increased opacity in the sinus and nasal cavities and endoscopy identified a mass emerging from the left ethmoidal turbinate. Computed tomography (CT) showed the range and origin of the mass, indicative of PEH. A standing frontonasal flap was made under sedation, following temporary tracheostomy to secure the airway

Macrophage adaptation to hypoxia in the tuberculous granuloma potentiates mycobacterium-induced mitochondrial damage and granuloma necrosis.

Granulomas, organized macrophage-rich aggregates, are pathological hallmarks of tuberculosis. Characteristically, as Mycobacterium tuberculosis replicates within macrophages, the granuloma enlarges, and then the center undergoes necrosis, promoting mycobacterial growth in the debris (caseum). This sequence results in large, mycobacterium-rich granulomas that can rupture into airways, facilitating disease transmission. Here, using the zebrafish-Mycobacterium marinum tuberculosis model, we find that both stages - granuloma enlargement and necrosis - depend on the mycobacterial virulence factor,

Dietary soy fiber limits Salmonella infection by disrupting the self-reinforcing loop between pathogen virulence and host inflammation.

The global threat of Salmonella Enteritidis (S. Enteritidis) is exacerbated by its ability to exploit host inflammation for colonization. Here, we find that dietary soy fiber (SF) confers colonization resistance by suppressing S. Enteritidis virulence and host inflammatory amplification. We identify harmaline (HAL), derived from Akkermansia, as a key effector. On the pathogen side, HAL disrupts tetrathionate respiration, induces oxidative stress, and inactivates Fur, thereby suppressing HilD-dependent virulence. On the host side, HAL enhances type I interferon signaling and promotes SUMOylatio

A ratiometric fluorescent reporter of mitochondrial sodium.

Cells cope with salt stress, hypoxia or elevated cytosolic Ca2+ by regulating their mitochondrial Na+ levels. The discovery of the mitochondrial Na+/Ca2+ exchanger and its disease relevance has revealed the need to map mitochondrial Na+ in situ. Here we describe a ratiometric fluorescent reporter for Na+, denoted MitRatiNa, that reports mitochondrial Na+ levels independent of membrane potential and in diverse cell lines. Na+ in individual mitochondria varies greatly and, depending on cell type, can be as low as 1-5 mM or as high as 40 mM on average. We demonstrate that mitochondrial Na+ increa

Transcriptional immune programs underlying inadequate response to tumor necrosis factor inhibitors in ankylosing spondylitis.

Inadequate response to tumor necrosis factor inhibitors (TNFi) in ankylosing spondylitis (AS) remains common. As the immunoregulatory and immunometabolic alterations across TNFi inadequate responder (TNFi-IR) subsets remain unexplored, we characterized molecular profiles across distinct immune subsets in TNFi-IRs and identified signatures linked to TNFi-IR. We compared subset-specific expression in 16 TNFi-IRs and 13 TNFi responders (TNFi-Rs). Gene expression demonstrated fair-good discriminative power (area under the curve: 0.70-0.87). TNFi-IRs displayed distinct transcriptomic profiles and a

Solid-Pseudo papillary Tumor of the Pancreas (Frantz Tumor): Case Report of a Rare Pancreatic Tumor in Ethiopia.

Solid-pseudopapillary tumor (SPT) of the pancreas is an extremely rare neoplasm in children, with an unknown origin. It accounts for 8% to 12% of primary pancreatic tumors and predominantly affects young females (91%), with very few cases reported in males. Malignant potential is minimal, particularly in adolescents. We report a 12-year-old child presenting with abdominal distension, nausea, vomiting, appetite loss, weight loss, pruritus, jaundice, dark urine, and back pain. Examination revealed a distended abdomen moving with respiration and a firm, tender 15 cm × 20 cm mass in the right uppe

Delayed Emergence Due to Severe Respiratory Acidosis Following Prolonged Spine Surgery: A Case Report.

Delayed emergence from anesthesia is a frequent challenge in surgical settings. Common etiologies include residual drug effects, metabolic derangements, or neurological insults. We report a case of a 55-year-old woman who underwent an elective six-hour lumbar laminectomy and cervical implantation surgery. Despite an uneventful intraoperative course and routine neuromuscular blockade reversal, the patient exhibited somnolence and ventilatory failure after reversal and extubation. Arterial blood gas (ABG) analysis revealed severe respiratory acidosis. The patient required re-intubation and contr

Multi-organ acute and delayed effects following exposure to neutron radiation.

Neutron radiation exposure is a life-threatening consequence of nuclear disasters. To determine the effect of high dose neutron exposures on late responding organs, we assessed survival and characterized organ-specific injuries in adult male and female rats exposed to whole-body 2-10 Gy high linear energy transfer fast neutrons with 5%-8% bone marrow shielding. Survival through the acute and delayed sequelae of organ radiation injury was monitored for 1 year. Longitudinal secondary outcomes, including body weight, hematology, respiration, blood urea nitrogen, urinary proteomics and metabolomic

Molecular pathogenesis and gene therapy-based intervention of GTPBP3-related mitochondrial disease.

Mitochondrial transfer RNA (mt-tRNA) modification determines organelle translation and function. GTPBP3 and MTO1 catalyze 5-taurinomethyluridine (τm5U) modification at wobble uridine of five mt-tRNAs. τm5U hypomodification causes mitochondrial encephalomyopathy, but the underlying pathogenesis and intervention strategy due to GTPBP3 mutations are lacking. In this study, we identify two genetic variants (c.689 A > C (p.Q230P) and c.1120 A > G (p.N374D)) of GTPBP3 in a Chinese proband with metabolic disorders and multisystem dysfunction. Mechanistically, Q230P and N374D mutations induce protein

In vivo CRISPRi screens reveal Escherichia coli functional adaptations in the mouse gut.

Escherichia coli exhibits remarkable genetic diversity that enables it to adapt to the intestinal environment. Here we establish an in vivo CRISPR interference platform that leverages bacterial gene fitness as a high-resolution functional reporter of E. coli adaptations within mice harbouring a defined minimal microbial community (OligoMM12). The screen revealed that diet profoundly shapes the metabolic landscape of E. coli and the essential gene profile identified cross-feeding interactions. Comparison between a laboratory strain (MG1655), a uropathogenic strain (CFT073) and an adherent-invas

Single-cell transcriptomic atlas of frontoinsular cortex reveals molecular correlates of selective neuronal vulnerability in FTD.

Frontotemporal dementia (FTD) is characterized by selective neuronal vulnerability, yet the features that predispose specific neuron types to degeneration remain unclear. We performed single-nucleus RNA sequencing of frontoinsular cortex, a region affected early in behavioral variant FTD, across individuals with C9orf72-associated and sporadic FTD-MND spectrum disease. By enriching for large projection neurons, we resolved molecular subtypes of layer 5 extratelencephalic neurons, including von Economo neurons, and identified selective depletion of specific layer 2/3 and layer 5 neuron subtypes

YY1/Asprosin/PFKP axis regulates glycolytic metabolic and exacerbates pathological cardiac hypertrophy.

In hypertrophic and failing hearts, fuel metabolism is reprogrammed toward enhanced glycolysis. Here, we identify asprosin, an adipokine, as a critical regulator of this process in pathological cardiac hypertrophy. In patients, circulating asprosin levels correlate with NT-proBNP and EF. Cardiomyocyte-specific asprosin overexpression aggravates hypertrophy, abnormal glycolysis, and impairs mitochondrial ATP production in male mice, whereas its deficiency confers protection against TAC- or Ang II-induced remodeling in male/female mice. Mechanistically, asprosin binds PFKP and inhibits the K48-l

Oxidative phosphorylation and mitochondrial dynamics are regulated by Sestrin2 to maintain cellular function.

The stress-inducible protein Sestrin2 (SESN2) has recently emerged as an orchestrator of mitochondrial signaling. The regulation of mitochondria-related pathways, such as aerobic respiration, is thought to be mediated by SESN2, but the underlying mechanisms are not fully understood. Here, we characterized mitochondria in Sesn2-knockdown myoblasts under physiological conditions using oxygen consumption rate measurements, fluorescence microscopy, and protein content analysis. We discovered that SESN2 is essential for sustaining oxidative phosphorylation and maintaining the mitochondrial network

Cardiolipin remodelling in mitochondrial therapeutics: translational evidence chains from elamipretide to emerging strategies.

Mitochondrial therapeutics have repeatedly fallen short of disease modification, in part because inner-membrane architecture constrains both molecular access and the reversibility of bioenergetic failure. Cardiolipin (CL), the signature phospholipid of the inner mitochondrial membrane, provides a mechanistically cohesive axis that links membrane mechanics to cristae organisation, respiratory-chain supercomplex stability and redox vulnerability. This Review summarises how CL biosynthesis and remodelling shape tissue-specific lipid species and how pathological remodelling can amplify oxidative i

Caspase-3 regulates RNA splicing and mitochondrial dynamics in microglia during Parkinson's disease.

Caspase-3, a cysteine-aspartic protease canonically known for its role in apoptotic cell death, has been implicated in several non-apoptotic functions, particularly in microglia, the brain-resident macrophages. These novel functions appear to depend on distinct levels of caspase-3 activation; however, the role of basal caspase-3 activity remains unclear. Here, we show that basal caspase-3 regulates RNA splicing in microglia, and its deficiency in a Parkinson´s disease (PD) model leads to splicing dysregulation. Loss of basal caspase-3 activity also increases double-stranded RNA (dsRNA) accumul

A bioluminescence-based chemical screen identifies a bactericidal naphthalen-1-ylmethanamine scaffold targeting MmpL3 in Mycobacterium abscessus.

Treatment of Mycobacterium abscessus pulmonary disease (Mabs-PD) is a growing global health challenge. The lack of bactericidal antibiotics effective at therapeutically relevant concentrations underscores an urgent need for drug discovery. Targeting cell wall synthesis is a promising strategy for drug discovery, as exemplified by the clinical success of broad-spectrum β-lactam antibiotics. Here, we employ a bioluminescence-based whole-cell assay optimized to identify compounds that disrupt both cell wall synthesis and oxidative phosphorylation. A focused drug library screen against Mabs reveal

Simulated microgravity enhances intracellular energy metabolism and electricity generation by electrochemically active bacteria for in situ space bioenergy production.

Electrochemically active bacteria (EAB) characterized by extracellular respiration is promising for bioregenerative life support systems and elemental cycling at extraterrestrial planetary bases. However, it remains unclear how microgravity modulates genes related to electricity generation and reshapes intracellular energy metabolism. In this study, the impact of simulated microgravity on EAB was systematically analyzed through electricity generation, gene enrichment, energy metabolism, and central carbon metabolic reprogramming. The results demonstrate that simulated microgravity increases th

Non-coding RNAs at the intersection of epigenetics and cancer metabolism.

Cancer cells are primarily characterized by uncontrolled proliferation, which is closely linked to an increased energy demand. According to this, cancer cells preferentially utilize the glycolytic pathway for energy production. Metabolic regulation in cancer occurs at multiple levels, including post-translational modifications, which are rapid and energy-efficient, as they do not require enzyme degradation. In this context, non-coding RNA-mediated control plays a crucial role, as these small RNAs can directly modulate the mRNAs of key metabolic enzymes or induce epigenetic modulations, ultimat
