Generation of a three-dimensional retinal-like structure using a laminin-111-modified alginate layer.

AimOver the past few decades, the prevalence of visual impairments has increased substantially, and some are incurable. This study aimed to present a co-culture method that promotes the differentiation of retinal progenitor cells (RPCs) in the presence of retinal pigment epithelium (RPE) cells.MethodsWe implemented a three-dimensional culture system using a modified alginate substrate to culture human embryonic stem cell-derived RPCs on top of an RPE layer, mimicking an in vivo environment. The characteristics of RPCs in experimental and control groups were analyzed at the RNA and protein leve

Poly-l-lactic acid electrospun membrane with specific morphology promotes bone regeneration through macrophage reprogramming.

Host immune response to bone biomaterials plays a crucial role in influencing the outcome of bone regeneration. In this research, electrospun membranes composed of poly-l-lactic acid (PLLA) with different configurations (aligned and random) and sizes (nanoscale and microscale) were created, and the molecular mechanisms through which PLLA electrospun membranes facilitate bone regeneration were investigated. Firstly, when various material groups were inserted into the rat skull defect model, the bone defect repair was most obvious in the aligned 600 nm group compared with other groups. Then, sin

Three-dimensional bioprinting for the construction of an in vitro gastrointestinal stromal tumor model.

Traditional two-dimensional (2D) cell culture systems are inadequate for maintaining numerous intrinsic cellular characteristics inherent to the native three-dimensional (3D) in vivo environment, which can result in misleading experimental outcomes. Hence, there is an urgent demand for innovative 3D tumor models that can be reliably employed in preclinical investigations. As an additive manufacturing technique, 3D bioprinting enables the fabrication of biomimetic 3D structures using a variety of materials, cells, and other small-molecule substances. This capability enables the fabrication of c

A Synthetic Hydrogel with Tunable Stiffness for Engineering Pancreatic Cancer Organoids and Drug Testing.

Pancreatic cancer organoids (PCOs) have gained extensive attention as promising in vitro models that can advance our understanding of translational cancer biology and biomedical research. To date, PCOs are mostly cultured in animal-derived matrices, which are limited by their low similarity with native tumors due to batch-to-batch variations, stringent operating conditions, and uncontrollable physicochemical properties. Here, we developed a more controllable hydrogel matrix comprising sodium alginate (NaA) and hyaluronic acid (HA) that can mimic the mechanical properties of native tumor tissue

Fc-Engineering Improves PET Imaging of Anti-Mesothelin VH-Fc across Multiple Tumor Mouse Models and Reveals Sex-Specific Renal Clearance.

Mesothelin (MSLN) is overexpressed in various malignancies, making it a promising target for molecular imaging and therapeutic strategies. Anti-MSLN VH-Fc fusion proteins show high tumor uptake as compared with monoclonal antibodies; however, elevated accumulation in Fc-rich organs (liver, spleen) can compromise tumor-to-background ratios and limit clinical applicability. To overcome this, we developed Fc mutant anti-MSLN VH-Fc fusion proteins incorporating G236R/L328R (GRLR) and L234A/L235A/P329G (LALAPG) mutations to eliminate FcγRs interactions. Engineered mutants exhibited high purity (>95

Jawbone-like organoids generated from human pluripotent stem cells.

Engineering jawbone tissues from pluripotent stem cells presents a challenge owing to the lack of protocols for selectively inducing the jawbone progenitor, the first pharyngeal arch (PA1) ectomesenchyme, and for recapitulating three-dimensional osteocyte networks. Here we present a method for generating jawbone-like organoids from human induced pluripotent stem cells through PA1 ectomesenchyme of the mandibular prominence (mdEM). A three-dimensional culture system enables sequential differentiation of induced pluripotent stem cells into neural crest cells and mdEM. The mdEM exhibits proximal-

New Advances in Transplants and Bioengineering Aid in Replacing the Womb.

When Kayla Edwards turned 13, she began to wonder if she was different. It started as a seed of suspicion when her friends began their menstrual cycles, and hers never arrived. Her grandmother was late, she learned, but for Edwards, it still seemed odd. She had hit puberty's other benchmarks-the hormones, the breasts-just no cycle.

Amniotic Fluid Stem Cells for the Treatment of Surgical Disorders in the Fetus and Neonate.

Over the past decade, amniotic fluid-derived stem cells have emerged as a novel experimental approach aimed at improving outcomes in children with congenital anomalies, including spina bifida, heart defects, and diaphragmatic hernia. Interest in these cells for the treatment of prenatally diagnosed diseases has arisen based on numerous studies demonstrating the relative ease of harvesting an abundant quantity of amniocytes from a small aliquot of fluid, the unique properties of amniocytes themselves, and the beneficial effects of amniotic fluid-derived stem cells in experimental animal models.

Acrylate-based materials for heart valve scaffold engineering.

Calcific aortic valve disease (CAVD) is the most frequent cardiac valve pathology. Its standard treatment consists of surgical replacement either with mechanical (metal made) or biological (animal tissue made) valve prostheses, both of which have glaring deficiencies. In the search for novel materials to manufacture artificial valve tissue, we have conducted a high-throughput screening with subsequent up-scaling to identify non-degradable polymer substrates that promote valve interstitial cells (VICs) adherence/growth and, at the same time, prevent their evolution toward a pro-calcific phenoty

Reconstruction of conjunctival epithelium-like tissue using a temperature-responsive culture dish.

PURPOSE: To study the feasibility of engineering conjunctival epithelial cell sheets on a temperature-responsive culture dish for ocular surface reconstruction. METHODS: Rabbit conjunctival epithelial cells (rCjECs) were cultured in DMEM/F-12 (1:1) medium. The morphology and phenotype of the rCjECs were confirmed with phalloidin staining, periodic acid-Schiff (PAS) staining, and immunocytochemistry. The rCjECs cultured on a temperature-responsive culture dish for 10 days produced confluent conjunctival epithelial cell sheets. Then, the phenotype, structure, and function of the conjunctival ep

The junctional epithelium originates from the odontogenic epithelium of an erupted tooth.

The junctional epithelium (JE) is an epithelial component that is directly attached to the tooth surface and has a protective function against periodontal diseases. In this study, we determined the origin of the JE using a bioengineered tooth technique. We transplanted the bioengineered tooth germ into the alveolar bone with an epithelial component that expressed green fluorescence protein. The reduced enamel epithelium from the bioengineered tooth fused with the oral epithelium, and the JE was apparently formed around the bioengineered tooth 50 days after transplantation. Importantly, the JE

Repair and regeneration of the respiratory system: complexity, plasticity, and mechanisms of lung stem cell function.

Respiratory disease is the third leading cause of death in the industrialized world. Consequently, the trachea, lungs, and cardiopulmonary vasculature have been the focus of extensive investigations. Recent studies have provided new information about the mechanisms driving lung development and differentiation. However, there is still much to learn about the ability of the adult respiratory system to undergo repair and to replace cells lost in response to injury and disease. This Review highlights the multiple stem/progenitor populations in different regions of the adult lung, the plasticity of

Increased afterload induces pathological cardiac hypertrophy: a new in vitro model.

Increased afterload results in 'pathological' cardiac hypertrophy, the most important risk factor for the development of heart failure. Current in vitro models fall short in deciphering the mechanisms of hypertrophy induced by afterload enhancement. The aim of this study was to develop an experimental model that allows investigating the impact of afterload enhancement (AE) on work-performing heart muscles in vitro. Fibrin-based engineered heart tissue (EHT) was cast between two hollow elastic silicone posts in a 24-well cell culture format. After 2 weeks, the posts were reinforced with metal b

In vitro localization of human neural stem cell neurogenesis by engineered FGF-2 gradients.

The development of effective stem cell-based therapies for treating brain disorders is keenly dependent upon an understanding of how to generate specific neural cell types and organize them into functional, higher-order tissues analogous to those of the cerebral cortex. Studies of cortical development have revealed that the proper formation of the human cerebral cortex results from specific intercellular interactions and soluble signaling between the highly-proliferative region occupied by dividing neural stem cells and an adjacent region of active neurogenesis and neural migration. However, t

Keratin films for ocular surface reconstruction.

Human amniotic membrane (AM) is frequently used as a substrate for ocular surface reconstruction. Its disadvantages (e.g., reduced transparency and biomechanical strength, heterogeneity depending on donor) create the need for standardized alternatives. Keratin from hair or wool has been proposed as an appropriate material for producing films or cell cultivation scaffolds. The current study was performed to develop transparent, stable and transferable films based on human hair keratin that support cellular adhesion and proliferation. The films were engineered by a multi-step procedure including

In vivo bioprinting for computer- and robotic-assisted medical intervention: preliminary study in mice.

We present the first attempt to apply bioprinting technologies in the perspective of computer-assisted medical interventions. A workstation dedicated to high-throughput biological laser printing has been designed. Nano-hydroxyapatite (n-HA) was printed in the mouse calvaria defect model in vivo. Critical size bone defects were performed in OF-1 male mice calvaria with a 4 mm diameter trephine. Prior to laser printing experiments, the absence of inflammation due to laser irradiation onto mice dura mater was shown by means of magnetic resonance imaging. Procedures for in vivo bioprinting and res

Converting human pluripotent stem cells into beta-cells: recent advances and future challenges.

PURPOSE OF REVIEW: The transplantation of insulin-producing beta-cells derived from human embryonic stem cells and induced pluripotent stem cells (collectively termed pluripotent stem cells or PSCs) holds great promise for therapy of diabetes mellitus. The purpose of this review is to summarize recent advances in this area, emphasizing the importance of studies of endocrine pancreas development in efforts to direct PSC differentiation into endocrine cells, as well as to outline the major challenges remaining before transplantation of PSC-derived beta-cells can become a reality. Pancreatic endoderm derived from human embryonic stem cells generates glucose-responsive insulin-secreting cells in vivo.

Development of a cell therapy for diabetes would be greatly aided by a renewable supply of human beta-cells. Here we show that pancreatic endoderm derived from human embryonic stem (hES) cells efficiently generates glucose-responsive endocrine cells after implantation into mice. Upon glucose stimulation of the implanted mice, human insulin and C-peptide are detected in sera at levels similar to those of mice transplanted with approximately 3,000 human islets. Moreover, the insulin-expressing cells generated after engraftment exhibit many properties of functional beta-cells, including expressio
