A comprehensive analysis of Atlantic salmon gonad and pituitary transcriptomes identifies novel players in sexual maturation.

BACKGROUND: Sexual maturation is a key developmental process important for reproductive success. Understanding the molecular mechanisms behind variation in sexual maturation can provide insights into reproductive biology and how life history variation is encoded in the genome. Atlantic salmon (Salmo salar) has become an excellent sexual maturation research model due to its diversity of life history strategies and its ecological and economic importance. A major challenge has been the lack of a comprehensive transcriptional investigation of reproductive tissues that captures the dynamic transcri

The TMEM43 S358L mutation affects cardiac, small intestine, and metabolic homeostasis in a knock-in mouse model.

The transmembrane protein 43 (TMEM43/LUMA) p.S358L mutation causes arrhythmogenic cardiomyopathy named as ARVC5, a fully penetrant disease with high risk of ventricular arrhythmias, sudden death, and heart failure. Male gender and vigorous exercise independently predicted deleterious outcome. Our systems genetics analysis revealed the importance of Tmem43 for cardiac and metabolic pathways associated with elevated lipid absorption from small intestine. This study sought to delineate gender-specific cardiac, intestinal, and metabolic phenotypes in vivo and investigate underlying pathophysiologi

Altered neuronal physiology, development, and function associated with a common chromosome 15 duplication involving CHRNA7.

BACKGROUND: Copy number variants (CNVs) linked to genes involved in nervous system development or function are often associated with neuropsychiatric disease. While CNVs involving deletions generally cause severe and highly penetrant patient phenotypes, CNVs leading to duplications tend instead to exhibit widely variable and less penetrant phenotypic expressivity among affected individuals. CNVs located on chromosome 15q13.3 affecting the alpha-7 nicotinic acetylcholine receptor subunit (CHRNA7) gene contribute to multiple neuropsychiatric disorders with highly variable penetrance. However, th

Downregulation of orosomucoid 2 acts as a prognostic factor associated with cancer-promoting pathways in liver cancer.

BACKGROUND: Liver cancer has a high mortality and morbidity rate throughout the world. In clinical practice, the prognosis of liver cancer patients is poor, and the complex reasons contribute to treatment failures, including fibrosis, hepatitis viral infection, drug resistance and metastasis. Thus, screening novel prognostic biomarkers is of great importance for guiding liver cancer therapy. Orosomucoid genes (ORMs) encode acute phase plasma proteins, including orosomucoid 1 (ORM1) and ORM2. Previous studies showed their upregulation upon inflammation, but the specific function of ORMs has not

Unveiling the cellular and molecular mechanisms of traditional Chinese medicine in hepatic fibrosis.

ETHNOPHARMACOLOGICAL RELEVANCE: Hepatic fibrosis (HF) is a prevalent consequence of chronic liver injury, resulting from the persistent interplay of inflammation, oxidative stress, dysregulated cell death, metabolic reprogramming, and remodeling of the immune microenvironment, which collectively establish a self-amplifying profibrotic network. In traditional medical systems, HF corresponds to syndromes such as hypochondriac pain, mass formation, blood stasis, and jaundice, indicating a chronic and systemically imbalanced disease trajectory. Guided by holistic and multi-target principles, tradi

Histone methylation defines c-Jun/Sox2/Hif1α axis that controls stemness and tumor progression in squamous cell carcinoma.

Squamous cell carcinomas (SCCs) originate from various sites that show poor survival due to the presence of cancer stem cells (CSCs), which impart therapy resistance. However, the crosstalk of molecular mechanisms regulating CSCs maintenance in skin and oral SCC is poorly understood. Here, we show that the whole-transcriptome profile of CSCs from skin SCC patients reveals upregulation in expression of genes associated with global hypermethylation, enhanced non-canonical Wnt signaling, and glycolysis, thereby activating the c-Jun/Sox2/Hif1α axis. Thus, it suggests crosstalk among epigenetics/si

Exploiting Honokiol-induced ER stress CHOP activation inhibits the growth and metastasis of melanoma by suppressing the MITF and β-catenin pathways.

There is increasing global incidence of highly metastatic melanoma and therapeutic strategies like those focusing on the downstream beta-catenin/MITF axis of invading melanoma cells are urgently needed. Targeting endoplasmic reticulum (ER) stress can promote cancer cell death and inhibit epithelial mesenchymal transition (EMT) in metastatic tumors. This study aimed to determine if Honokiol could promote ER stress-dependent apoptosis and regulate metastatic melanoma. The therapeutic efficacy of Honokiol was assessed using the highly metastatic melanoma xenograft mouse model for peritoneal metas

Micro RNA-214 contributes to proteasome independent downregulation of beta catenin in Huntington's disease knock-in striatal cell model STHdhQ111/Q111.

Role of beta catenin in Huntington's disease (HD) is not clear. Previous studies on HD reported varied levels of beta catenin. In the present study we showed that beta catenin is post transcriptionally down-regulated in mutant huntingtin knock-in cell model STHdhQ111/Q111. This in turn leads to decreased level of wnt/beta catenin responsive genes. We observed that Gsk3beta or Gsk3beta (phospho Ser 9) is unaltered in HD and this down-regulation of beta catenin is independent of proteasomal degradation. Finally, we showed that the overexpression of miR-214 leads to the down-regulation of beta ca

Preferential biological processes in the human limbus by differential gene profiling.

Corneal epithelial stem cells or limbal stem cells (LSCs) are responsible for the maintenance of the corneal epithelium in humans. The exact location of LSCs is still under debate, but the increasing need for identifying the biological processes in the limbus, where LSCs are located, is of great importance in the regulation of LSCs. In our current study we identified 146 preferentially expressed genes in the human limbus in direct comparison to that in the cornea and conjunctiva. The expression of newly identified limbal transcripts endomucin, fibromodulin, paired-like homeodomain 2 (PITX2) an

Moderate static magnetic field regulates iron metabolism and salvage bone loss caused by iron accumulation.

OBJECTIVE: Clinical studies, epidemiological investigations and animal experiments have demonstrated that iron overload lead to bone loss, especially postmenopausal osteoporosis. As a physiotherapy tool, electromagnetic fields already used in clinical treatment of osteoporosis and participates in bone remodeling by affecting the iron metabolism of organisms. As an electromagnetic field with constant magnetic flux density and direction, the mechanism of static magnetic field (SMF) regulating iron metabolism remains unclear. Therefore, the aim of this study was to investigate the effects of mode

Mindbomb 1, an E3 ubiquitin ligase, forms a complex with RYK to activate Wnt/β-catenin signaling.

Receptor-like tyrosine kinase (RYK) functions as a transmembrane receptor for the Wnt family of secreted protein ligands. Although RYK undergoes endocytosis in response to Wnt, the mechanisms that regulate its internalization and concomitant activation of Wnt signaling are unknown. We discovered that RYK both physically and functionally interacts with the E3 ubiquitin ligase Mindbomb 1 (MIB1). Overexpression of MIB1 promotes the ubiquitination of RYK and reduces its steady-state levels at the plasma membrane. Moreover, we show that MIB1 is sufficient to activate Wnt/β-catenin (CTNNB1) signalin

PP2A-activating drugs selectively eradicate TKI-resistant chronic myeloid leukemic stem cells.

The success of tyrosine kinase inhibitors (TKIs) in treating chronic myeloid leukemia (CML) depends on the requirement for BCR-ABL1 kinase activity in CML progenitors. However, CML quiescent HSCs are TKI resistant and represent a BCR-ABL1 kinase-independent disease reservoir. Here we have shown that persistence of leukemic HSCs in BM requires inhibition of the tumor suppressor protein phosphatase 2A (PP2A) and expression--but not activity--of the BCR-ABL1 oncogene. Examination of HSCs from CML patients and healthy individuals revealed that PP2A activity was suppressed in CML compared with norm

Secretome enriched with small extracellular vesicles derived from human gingiva-derived mesenchymal stem cells enhances rat tongue muscle regeneration.

BACKGROUND: Accumulating evidence demonstrates that the therapeutic effects of stem cells are most likely attributed to their secretome, composed of a myriad of bioactive factors, including small extracellular vesicles (EVs). Due to the potential benefits over cells in term of handling, preservation, stability, and safety, MSC-derived secretome is emerging as a novel cell-free therapeutic for regenerative therapy of various diseases. The purpose of this study is to optimize the xeno-free culture conditions to improve the secretome production by human gingiva-derived mesenchymal stem cells (GMS

A discovery protein panel for brain predicted age discordance using MRI in neurologically healthy individuals.

BACKGROUND AND OBJECTIVES: Brain age is a global measure that compares structural brain MRI with large reference datasets. Predicted age deviation (PAD) is the deviation between predicted brain age and chronological age, with positive values indicating advanced aging. Identifying blood-based biomarkers that approximate brain PAD could provide an accessible and cost-effective measure of brain health as an alternative to MRI, but no blood-based biomarkers have yet been identified. This study aimed to investigate novel blood-based biomarkers associated with accelerated PAD using an unbiased prote

Single-Cell RNA Sequencing Reveals Disrupted Stromal-Epithelial Crosstalk Impairs Gut Barrier in Depression.

BACKGROUND: Depressive disorder is commonly associated with gastrointestinal dysfunction, but the specific cellular and molecular alterations within the colonic mucosa remain poorly understood. The role of stromal-epithelial crosstalk in the colonic niche under depressive conditions is particularly unclear. METHODS: A chronic unpredictable mild stress (CUMS) mouse model of depression was established and validated behaviorally. Single-cell RNA sequencing (scRNA-seq) of colon tissues from CUMS and control mice was performed to define cellular composition and transcriptional changes. Pseudotime
