Photobiomodulation therapy: Location-dependent effects on memory and recovery in male adult rats with spinal cord injury.

Spinal cord injuries (SCIs) can cause significant physical and emotional challenges for individuals as a result of inflammation and cell death. This study investigates the effectiveness of photobiomodulation therapy (PBMT) in reducing inflammation and memory impairment through the use of 3 specific treatment protocols. Forty male rats (180-200 g) were divided into 5 groups: Control; SCI model; and SCI with 3 different PBMT protocols. The rats underwent behavioral tests (Novel Object Recognition and Shuttle Box), histological examination (H&E and Nissl staining), and protein analysis (HDAC2, NF

Innovation, Inhibition and Flexibility in Rhesus Macaques (Macaca mulatta).

Innovation is a key component of behavioral flexibility. When individuals are presented with novel problems, their ability to behave flexibly often relies upon their exploratory tendencies, motivational states and intrinsic traits. Studies of repeated innovation pose additional benefits to understand mechanisms of behavioral flexibility, including measuring persistence, exploration and inhibitory control when learned solutions are blocked. The multi-access box (MAB) paradigm tests repeated innovation without prior training and minimal habituation. We tested fifty adult captive female rhesus ma

Adsorption analysis of Cr(VI) by ZnCl2-bentonite-comodified cyanobacterial bloom-derived biochar.

Biochar has been utilized to treat industrial wastewater containing high Cr concentrations. The collection of large amounts of nauseous algae sludge from lakes where cyanobacterial blooms frequently occur and the conversion of this sludge into biochar is a valuable strategy for optimizing resource utilization. In this study, the copyrolysis of bentonite and ZnCl2 with waste cyanobacterial bloom biomass produced biochar (ZBMC) with a high cation exchange capacity (CEC) and specific surface area (SSA), enabling the effective remediation of high Cr concentrations in industrial wastewater. The mor

Expanding the CarD interaction network: CrsL is a novel transcription regulator in actinobacteria.

Bacterial transcription regulation is critical for adaptation and survival. CarD is an essential transcription factor in mycobacteria involved in the regulation of gene expression. We searched for CarD interaction partners in Mycobacterium smegmatis and identified a novel uncharacterized protein, named CrsL (MSMEG_5890). CrsL is a 5.7 kDa protein shown by NMR to be intrinsically disordered. CrsL homologs are present in actinobacteria, including pathogenic species such as Mycobacterium tuberculosis. CrsL interacts directly with CarD, adopting an ordered structure in the complex, and also binds

DNA methylation and transcriptome signatures of the FOXO1 gene in ankylosing spondylitis.

BACKGROUND: Ankylosing spondylitis (AS) is an inflammatory autoimmune disease with complex etiology. The forkhead box O (FOXO) 1 is an important transcription factor related to proliferation, homeostasis and metabolism. Notably, the involvement of methylation and the expression of mRNA in the promoter region of the FOXO1 gene in relation to AS is still not understood. METHODS: A two-stage case-control study enrolled 60 AS patients and 60 healthy controls (HCs) for integrated demographic and clinical evaluation and DNA methylation profiling. Subsequently, FOXO1 mRNA expression was comparativel

The FBXW7-KMT2 axis in cancer-associated fibroblasts controls tumor growth via an epigenetic-paracrine mechanism.

F-box and WD repeat domain-containing 7 (FBXW7) is a tumor suppressor that targets various oncoproteins for degradation, but its role in modulating cancer-associated fibroblasts (CAFs) in the tumor microenvironment remains elusive. Here, we report that FBXW7 expression is gradually downregulated in CAFs during the progression of human pancreatic and lung cancers. Mechanically, FBXW7 inhibits histone lysine methyltransferase 2 (KMT2) methyltransferase activity via retinoblastoma binding protein 5 (RbBP5) binding, whereas FBXW7 depletion abrogates the binding to activate KMT2, leading to increas

SIRT3 mitigates dry eye disease through the activation of autophagy by deacetylation of FOXO1.

Dry eye disease (DED) is a complex ocular condition characterized by oxidative stress, inflammation, and apoptosis. An increasing number of studies suggest that Sirtuin3 (SIRT3), a mitochondrial deacetylase, may offer protection against related pathologies. Despite these indications, the precise function and underlying mechanisms of SIRT3 in the context of DED have not been fully elucidated. Here, we observed a decline in SIRT3 expression in human corneal epithelial cells (HCE-Ts) and the corneal conjunctiva of mice as the disease advanced. Overexpression of SIRT3 in HCE-Ts reduced the accumul

Early detection in oral cancer: Are we prepared for artificial intelligence-driven precision medicine?

Oral cancer, particularly oral squamous cell carcinoma, remains a serious health concern, with a poor prognosis and a late diagnosis. Leukoplakia, erythroplakia, lichen planus, and submucous fibrosis are examples of oral potentially malignant disorders that must be detected early but are not always so by traditional, laborious, and subjective diagnostic techniques. In oral cancer, artificial intelligence (AI) and precision medicine are becoming game-changing technologies that enhance individualized care, treatment planning, and diagnostic precision. Complex imaging and histopathology data may

CILP1 interacting with YBX1 promotes hypertrophic scar formation by suppressing PPARs transcription.

Hypertrophic scar (HS) represents the most prevalent form of skin fibrosis, significantly impacting the quality of life. Despite this, the molecular mechanisms driving HS formation remain largely undefined, impeding the development of effective treatments. The study showed that Cartilage Intermediate Layer Protein 1 (CILP1) was predominantly expressed in myofibroblasts and was up-regulated in various forms of skin fibrosis, including human hypertrophic and keloid scars, and in animal models of HS. Notably, we detected elevated serum levels of CILP1 in fifty-two patients with HS compared to twe

Scalable extracellular expression of tag-free MPT64 protein in E. coli via pelB signal optimization: a step toward tuberculosis diagnostic antigen preparation.

Tuberculosis (TB) remains a global health burden, requiring affordable and scalable diagnostic tools. The MPT64 protein is a secreted biomarker specific to Mycobacterium tuberculosis and holds promise for rapid antigen-based diagnostics. However, existing recombinant expression systems often yield low extracellular amounts, complicating purification and limiting downstream application. This study aimed to optimize the extracellular, tag-free expression of MPT64 protein in Escherichia coli by employing the pelB signal peptide in combination with Response Surface Methodology (RSM). Cutaneous Melanocytic Tumor With CRTC1::TRIM11 Fusion: A Case Report.

Cutaneous melanocytic tumor with CRTC1::TRIM11 fusion (CMTCT) is a rare and recently recognized melanocytic neoplasm with fewer than 50 reported cases. These tumors can be diagnostically challenging due to overlapping histologic features with malignant melanoma and clear cell sarcoma (CCS). This report adds to the limited literature by describing a 50-year-old male with an atypical inflamed cystic lesion on the shin. Biopsy revealed a dermal proliferation of atypical epithelioid cells within a desmoplastic stroma. Immunohistochemistry was positive for SRY-box transcription factor 10 (SOX-10) a

Oligodendrocyte Precursor Cell-Specific HMGB1 Knockout Reduces Immune Cell Infiltration and Demyelination in Experimental Autoimmune Encephalomyelitis Models.

Infiltration and activation of peripheral immune cells are critical in the progression of multiple sclerosis and its experimental animal model, experimental autoimmune encephalomyelitis (EAE). This study investigates the role of high mobility group box 1 (HMGB1) in oligodendrocyte precursor cells (OPCs) in modulating pathogenic T cells infiltrating the central nervous system through the blood-brain barrier (BBB) by using OPC-specific HMGB1 knockout (KO) mice. We found that HMGB1 released from OPCs promotes BBB disruption, subsequently allowing increased immune cell infiltration. The migration

Targeted Delivery of Arctigenin Using Sialic Acid Conjugate-Modified Liposomes for the Treatment of Breast Cancer.

Arctigenin (ATG) is a broad-spectrum antitumor drug with an excellent inhibitory effect on malignant tumors such as breast cancer, glioblastoma, liver cancer, and colon cancer. However, the clinical application of ATG is limited by its poor water solubility and quick hydrolysis in the liver, intestine, and plasma, which might hinder its application. Sialic acid (SA) recognizes selectin receptors overexpressed on the surface of tumor-associated macrophages. In this study, SA was conjugated with octadecylamine (ODA) to prepare SA-ODA, which was employed to prepare SA functionalized nanoliposomes

Validation of Pelvis and Trunk Range of Motion as Assessed Using Inertial Measurement Units.

Trunk and pelvis range of motion (ROM) is essential to perform activities of daily living. The ROM may become limited with aging or with neuromusculoskeletal disorders. Inertial measurement units (IMU) with out-of-the box software solutions are increasingly being used to assess motion. We hypothesize that the accuracy (validity) and reliability (consistency) of the trunk and pelvis ROM during steady-state gait in normal individuals as measured using the Opal APDM 6 sensor IMU system and calculated using Mobility Lab version 4 software will be comparable to a gold-standard optoelectric motion c

Assessing the utility of Regenerative Peripheral Nerve Interfaces (RPNIs) in providing referred sensations in a functional task in a virtual environment.

Individuals who use upper limb prostheses receive limited feedback from their devices. Researchers have attempted to elicit sensation through direct stimulation of peripheral nerves or through stimulation of reinnervated skin or muscle. Previous research found that electrical stimulation of Regenerative Peripheral Nerve Interfaces (RPNIs) elicited sensations that were referred to the phantom hand. The purpose of this study was to determine if this sensation could be used to improve performance of a functional task. Two participants with upper limb loss completed the Box and Blocks Test in a vi

Sox6 and ALDH1A1 Truncation by Asparagine Endopeptidase Defines Selective Neuronal Vulnerability in Parkinson's Disease.

Dopaminergic neurons in the substantia nigra pars compacta (SNpc) demonstrate regionally selective susceptibility in Parkinson's disease (PD) compared to those in the ventral tegmental area (VTA). However, the molecular mechanism for this distinct vulnerability remains unclear. Here, it is shown that Legumain, also known as asparagine endopeptidase (AEP), is activated in a subgroup of SRY-box transcription factor 6 /Aldehyde dehydrogenase 1 family member A1, (Sox6+/ALDH1A1+) neurons in the ventral tier of the SNpc and cleaves Sox6 and ALDH1A1, leading to repression of Special AT-rich sequence
