Eucommia polysaccharides alleviate experimental colitis by reshaping colonic microbiota composition, metabolites, and modulating the IL-17 signaling pathway.

Intake of plant polysaccharides are associated with a reduced risk of ulcerative colitis (UC). Eucommia polysaccharides (EUPs) are promising nutritional supplements with notable antibacterial, anticancer, and anti-inflammatory properties. They exhibit a moderate molecular weight and are resistant to gastric acid degradation. Yet, the action mechanisms of microorganisms and metabolites in EUPs for UC treatment remain incompletely elucidated. The current study was formulated to assess the therapeutic efficacy of EUPs against fecal microbiota transplantation (FMT)-induced colitis, focusing on com

Diet and microbiome shape small-molecule cytokinin pools in mammals.

Cytokinins (CKs) are adenine-derived metabolites traditionally characterized as plant hormones, yet their origin, distribution, and functions in mammalian systems remain largely undefined. Using integrated metabolomics, microbiome, and metagenomics approaches, we provide a systematic characterization of CK occurrence and potential sources in mammals. Serum profiling across five animal species revealed consistent detection of multiple CK derivatives, with concentrations markedly lower than in plant tissue. The CK storage form, zeatin-O-glucoside, predominated in mammalian sera, followed by tran

Safety evaluation of the food enzyme AMP deaminase from the genetically modified Bacillus subtilis strain CCTCC M 2023264.

The food enzyme AMP deaminase (AMP aminohydrolase; EC 3.5.4.6) is produced with the genetically modified Bacillus subtilis strain CCTCC M 2023264 by Angel Yeast Co., Ltd. The production strain of the food enzyme contains multiple copies of three acquired antimicrobial resistance genes, therefore it does not fulfil the requirements of the qualified presumption of safety approach to safety assessment. However, considering the absence of viable cells and DNA from the production organism in the food enzyme, this is not considered to be a risk. As no other concerns arising from the microbial source

Constitutive sporulation in wild fission yeast enhances insect-mediated survival.

Sexual reproduction is generally considered costly for unicellular organisms and is typically induced in yeast only under nutrient limitation. However, wild strains of Schizosaccharomyces japonicus isolated from fruit flies in Japan exhibit constitutive sporulation even under nutrient-rich conditions. Here, we investigated the ecological and evolutionary significance of this unusual phenotype. We demonstrate that spores produced under nutrient-rich conditions survive passage through the digestive tract of Drosophila melanogaster, whereas vegetative cells do not. Although constitutive sporulati

Microbial Diversity and Hydrocarbon-Oxidizing Bacteria in Coastal Waters and Sands Contaminated by the Fuel Oil Spill in the Black Sea.

In 2024, an accident involving two tankers in the Kerch Strait resulted in the release of approximately 2400 tons of fuel oil into the Black Sea, causing significant contamination of seawater and the coastal zone. This study presents the first microbiological and molecular-ecological assessment of prokaryotic community composition and hydrocarbon-oxidizing bacteria (HOB) in coastal seawater and sand near Anapa (Russian Federation) following the spill. The taxonomic composition of nine samples was analyzed using high-throughput sequencing of 16S rRNA genes (V3-V4 regions), identifying Bacteria

Low-release biochar for improving coastal reclaimed Saline-Alkali soil: Short-Term physicochemical responses and exploratory microbial transcriptional associations under controlled pot conditions.

Coastal land reclamation is an important approach to expanding useable land, yet reclaimed soils are often constrained by low fertility, weak structural stability, and saline-alkaline stress. Considering traditional amendments may introduce environmental risks in sensitive coastal settings, this study evaluated the short-term responses of coastal reclaimed saline-alkali soil (Hengsha Island, Shanghai) under controlled pot conditions to a novel low-release biochar (BC-SR). Ten days after application, conventional biochar (BC-O) increased soil salinity to 2.5 times the initial level and raised p

Chemosynthesis enables microbial communities to flourish in a marine cave ecosystem.

Chemosynthesis, an ancient metabolism that uses chemical compounds for energy and biomass generation, occurs across the ocean. Although chemosynthesis typically plays a subsidiary role to photosynthesis in the euphotic ocean, it is unclear whether it plays a more important role in aphotic habitats within this zone. Here, we compared the composition, function, and activity of microorganisms colonising the sediment of a marine cave at mesophotic depth, across a transect from the entrance to the interior. Microbes thrived throughout this ecosystem, with interior communities having higher diversit

Endocytosis of the Saccharomyces cerevisiae Jen1 monocarboxylate-proton symporter under mildly alkaline conditions: a mechanism to prevent metabolite loss?

BACKGROUND: The Saccharomyces cerevisiae Jen1 transporter mediates electroneutral proton symport of lactate and pyruvate. In lactate-grown batch cultures, growth-coupled medium alkalinization was previously shown to coincide with endocytosis of Jen1. RESULTS: To investigate the physiological relevance of pH-dependent Jen1 endocytosis, S. cerevisiae was grown in carbon-limited continuous cultures on a mixed ethanol-lactate feed. When applying a linearly increasing pH (6.75–7.25) to these cultures, lactate and pyruvate concentrations in the external medium progressively increased. Up to a cultur

Adaptive evolution of thermotolerant xylose-utilizing Saccharomyces cerevisiae for lignocellulosic ethanol production via high-temperature simultaneous saccharification and co-fermentation.

Bioethanol serves as a pivotal renewable alternative to fossil fuels, with applications extending from energy production to use as a precursor for biomaterials. The temperature mismatch between enzymatic hydrolysis and microbial fermentation, combined with inefficient xylose metabolism under thermal stress, constitutes a major bottleneck for lignocellulosic ethanol production in simultaneous saccharification and co-fermentation (SSCF). In this work, thermotolerantSaccharomyces cerevisiaestrains were developed through adaptive laboratory evolution, enabling efficient xylose fermentation at 40°C

Postbiotics From Ligilactobacillus salivarius Enhance Mitochondrial Robustness Via TLR2 Signaling in Intestinal Macrophages.

Postbiotics, defined as preparations of inanimate microorganisms and/or their components, have attracted increasing attention because of their stability and functional benefits. Although lactic acid bacteria (LAB) modulate immune responses and oxidative stress, their direct effects on host mitochondrial homeostasis remain poorly understood. In this study, we investigated whether postbiotics derived from Ligilactobacillus salivarius enhance mitochondrial robustness in porcine intestinal macrophages (IPIMs). To establish a model of disrupted mitochondrial redox homeostasis, mitochondrial dysfunc

[Metabolic engineering of Escherichia coli for efficient production of nicotinamide riboside].

Nicotinamide riboside (NR), an essential precursor for nicotinamide adenine dinucleotide (NAD+), has garnered increasing attention for its anti-aging properties and diverse pharmacological activities. Developing an efficient, sustainable, and environmentally friendly synthesis approach is of great significance for the industrial production of NR. In this study, we developed a modular metabolic engineering strategy to construct an engineered strain capable of synthesizing NR from nicotinamide efficiently. First, six genes, including rihA, were deleted to block the degradation pathways of NR and

Perturbation of gut-liver axis by cerium dioxide nanoparticles (CeO2 NPs): Implications for intestinal flora and glycolipid metabolism in zebrafish (Danio rerio).

The increasing environmental release of cerium oxide nanoparticles (CeO2 NPs) raised concerns about their potential risks to aquatic organisms, yet their chronic effects remained poorly understood. This study investigated the impact of CeO2 NPs on the glucose and lipid metabolic homeostasis of the gut-liver axis and their bioaccumulation patterns in adult zebrafish. Exposure to 100 μg/L and 1000 μg/L CeO2 NPs for 26 days inhibited the growth of zebrafish and caused intestinal damage. The analysis of inductively coupled plasma mass spectrometry (ICP-MS) indicated that Ce was concentrated in the

Microbial-Mediated Coevolution of Vanadium and Ferric Iron Minerals in Sedimentary Environments.

Rising vanadium(V) contamination severely threatens environmental safety and human health. Microbial-mediated pentavalent V [V(V)] reduction, an effective route to V detoxification, can be influenced by coexisting redox-sensitive matters. However, the interaction between V(V) and widely distributed ferric iron [Fe(III)] minerals still remains largely unknown. This study investigated the coupled transformations of V(V) and Fe(III) minerals in biosystems with Bacillus subtilis. Fe(III) minerals, particularly low-crystallinity ferrihydrite, markedly enhanced V(V) reduction. Concurrently, B. subti

Efficient Myoinositol biosynthesis from glucose through gluconate bypass of glycolysis.

Microbial myo-inositol (MI) production typically requires deletion of conditionally essential genes such as phosphoglucose isomerase (pgi in E. coli) to increase pools of the precursor glucose-6-phosphate (G6P). This disrupts growth and has necessitated complex glycerol co-feeding strategies. Here, we address this limitation by implementing a synthetic gluconate-bypass (GBP) central metabolism that establishes an alternative route by which glucose-derived carbon enters central metabolism, allowing robust growth despite pgi deletion. However, pgi deletion alone was insufficient to improve MI pr

Biodegradation of tetracycline antibiotics: Advances and insights into microbial resources, enzymatic mechanisms, and remediation potential.

The extensive use of tetracycline antibiotics (TCs) in human therapy and animal husbandry has resulted in their persistent residues and accumulation across diverse environments, raising substantial global concern. These residues not only accelerate the emergence of antibiotic-resistant pathogens but also pose a long-term ecological risk through the horizontal transfer of antibiotic resistance genes (ARGs). Microbial biodegradation plays a crucial role in the elimination of TCs. Through long-term acclimation and targeted screening, numerous microorganisms exhibiting high degradation capacities

Multi-Omics Reveals Gut Microbiota Shifts and Hepatic Metabolic-Immune Alterations in "Short-Leg" Malformed Frog (Pelophylax nigromaculatus).

Amphibian malformation syndromes significantly impact both conservation efforts and aquaculture, yet their underlying systemic pathophysiological mechanisms remain poorly characterized. This study comprehensively examines the multi-level pathological processes associated with the "short-leg" malformation syndrome in the black-spotted frog (Pelophylax nigromaculatus) using an integrated methodology, encompassing morphological, histopathological, gut microbiome, and hepatic transcriptomic analyses. Affected frogs demonstrated shortened limbs, impaired motor function, and a distinctive metabolic

Recent advances in the defense mechanisms of arbuscular mycorrhizal fungi in antibiotic-contaminated soil ecosystems.

Arbuscular mycorrhizal fungi (AMF) are important soil symbionts that support plant nutrient acquisition, stress tolerance, and soil ecosystem stability. The increasing accumulation of antibiotic residues in agricultural soils may inhibit AMF spore germination, hyphal development, and root colonization, thereby weakening mycorrhizal functions and plant-soil feedback. This review summarizes recent advances in AMF responses to antibiotic stress in soil ecosystems and proposes a five-layer defense framework: external interfacial barriers, intracellular metabolic detoxification, antioxidant defense

Decoupling the "attract-and-kill" strategy: Independent zoospore attraction and ROS-executed killing synergistically drive disease-suppressive intercropping.

Soilborne Phytophthora diseases pose a major threat to agricultural sustainability. However, how nonhost roots disrupt the transmission of soilborne Phytophthora pathogens without relying solely on classical antimicrobial exudates remains poorly understood. Through a decade-long field study, we demonstrate that strip intercropping can sustainably suppress disease incidence by up to 46.85% by leveraging nonhost roots as ecological barriers that intercept zoospore transmission. Moving beyond the conventional focus on antimicrobial exudates, we resolve the "attract-and-kill" strategy into two dis

Changes in dysbiosis and gene expression in the gut of wharf roach (Ligia spp.) fed with expanded polystyrene.

The East Asian region, with its intensive human and fishery activity, suffers from serious marine plastic pollution along its coastlines. Wharf roaches (Ligia spp.) on the coast of western Japan frequently ingest expanded polystyrene (EPS) and excrete it as microplastic in their feces, yet the effects on their gut remain unclear. Under controlled laboratory exposure, we examined both the host gut transcriptome, and the gut microbiome of wharf roaches fed EPS versus a fasted control group. DESeq2 and PyDESeq2 yielded a high-confidence set of 25 differentially expressed genes, among which xenobi
