Chronic diabetic wounds are at lifelong risk of developing diabetic foot ulcers owing to severe hypoxia, excessive reactive oxygen species (ROS), a complex inflammatory microenvironment, and the potential for bacterial infection. Here we develop a programmed treatment strategy employing live Haematococcus (HEA). By modulating light intensity, HEA can be programmed to perform a variety of functions, such as antibacterial activity, oxygen supply, ROS scavenging, and immune regulation, suggesting its potential for use in programmed therapy. Under high light intensity (658 nm, 0.5 W/cm2), green HE

Aberrant expression of the E26 transformation-specific (ETS) transcription factors characterizes numerous human malignancies. Many of these proteins, including EWS:FLI1 and EWS:ERG fusions in Ewing sarcoma (EwS) and TMPRSS2:ERG in prostate cancer (PCa), drive oncogenic programs via binding to GGAA repeats. We report here that both EWS:FLI1 and ERG bind and transcriptionally activate GGAA-rich pericentromeric heterochromatin. The respective pathogen-like HSAT2 and HSAT3 RNAs, together with LINE, SINE, ERV, and other repeat transcripts, are expressed in EwS and PCa tumors, secreted in extracellu

Further advances in cell engineering are needed to increase the efficacy of chimeric antigen receptor (CAR) and other T cell-based therapies1-5. As T cell differentiation and functional states are associated with distinct epigenetic profiles6,7, we hypothesized that epigenetic programming may provide a means to improve CAR T cell performance. Targeting the gene that encodes the epigenetic regulator ten-eleven translocation 2 (TET2)8 presents an interesting opportunity as its loss may enhance T cell memory9,10, albeit not cause malignancy9,11,12. Here we show that disruption of TET2 enhances T

Twelve regulated cell death programs have been described. We review in detail the basic biology of nine including death receptor-mediated apoptosis, death receptor-mediated necrosis (necroptosis), mitochondrial-mediated apoptosis, mitochondrial-mediated necrosis, autophagy-dependent cell death, ferroptosis, pyroptosis, parthanatos, and immunogenic cell death. This is followed by a dissection of the roles of these cell death programs in the major cardiac syndromes: myocardial infarction and heart failure. The most important conclusion relevant to heart disease is that regulated forms of cardiom

Programmed death-ligand 1 (PD-L1) down-modulates various immune responses by engaging the co-inhibitory receptor programmed death-1. Expression of PD-L1 and programmed death-1 is elevated in the salivary glands of patients with Sjögren's syndrome (SS). The objective of this study is to define the role of endogenous PD-L1 in SS pathogenesis in non-obese diabetic (NOD) mouse model of this disease. We inhibited endogenous PD-L1 function by intraperitoneal administration of a blocking antibody to 6 week-old female NOD/ShiLtJ mice repeatedly during a 9-day period. PD-L1 blockade accelerated leukocy

Chronic infections and infestations represent major causes of cancer. Overall, Helicobacter pylori, HPV, HBV, and HCV are estimated to account for 15% of all human cancers. We have estimated that cancers associated with 6 pathogens in Italy account for 31,000 yearly cases, 42.0%of which is attributable to H. pylori, 34.7%to HBV and HCV, 19.8%to HPV, 2.9%to KSHV, and 0.2% to EBV. These figures represent 8.5% of all incident cases of cancer in Italy. The implementation of anti-HBV vaccination programs in countries with high endemicity resulted in a significant impact on the incidence of hepatoce

Persistent infection by EBV is explained by the germinal center model (GCM) which provides a satisfying and currently the only explanation for EBVs disparate biology. Since the GCM touches on every aspect of the virus, this chapter will serve as an introduction to the subsequent chapters. EBV is B lymphotropic, and its biology closely follows that of normal mature B lymphocytes. The virus persists quiescently in resting memory B cells for the lifetime of the host in a non-pathogenic state that is also invisible to the immune response. To access this compartment, the virus infects naïve B cells

The study's objective was to identify HPA 1a-negative women and to offer them an intervention program aimed to reduce morbidity and mortality of neonatal alloimmune thrombocytopenia (NAIT). HPA 1 typing was performed in 100 448 pregnant women. The HPA 1a-negative women were screened for anti-HPA 1a. In immunized women, delivery was performed by Cesarean section 2 to 4 weeks prior to term, with platelets from HPA 1a-negative donors reserved for immediate transfusion if petechiae were present and/or if platelet count was less than 35 x 10(9)/L. Of the women screened, 2.1% were HPA 1a negative, a

Immune checkpoint blockade targeting the PD-1/PD-L1 axis has revolutionized cancer therapy, yet the frequent emergence of resistance limits its clinical efficacy. Elucidating the mechanisms underlying resistance and developing effective strategies remain critical challenges in tumor immunotherapy. This study identifies kinase suppressor of Ras 2 (KSR2) as a driver of resistance to anti-PD-1 therapy in lung cancer. Transcriptomic analysis of an anti-PD-1-resistant mouse model and public clinical datasets revealed upregulation of KSR2 in resistant tumors. In vivo functional studies demonstrated

BACKGROUND: Heterodimeric IL-15 (NIZ985) shows rapid and indirect effects on different myeloid cells but may upregulate PD-1 (programmed cell death-protein 1) on CD8 + T cells, limiting its therapeutic potential. Combining IL-15 with an anti-PD-1 antibody is known to enhance antitumor immune response and prolong survival in mice.
METHODS: This was a phase I/Ib, open-label, multicenter study (NCT04261439) of NIZ985 single agent (SA) or with an anti-PD-1/CPI agent in patients with advanced solid tumors or lymphoma (responded and progressed on prior anti-PD-1) in a dose escalation (SA/with spart

Immune checkpoint inhibitor (ICI) myocarditis is a rare but frequently fatal immune-related adverse event of cancer immunotherapy. Understanding the mechanisms of this toxicity is critical to balancing treatment with maintenance of antitumor immunity. Using integrated spatial and single-cell analyses in a pharmacological murine model, we identified regional infiltration of Ly6C+ monocytes and PD-1+ CD8+ T cells in the heart that organize into fibroblast-rich immune structures, which we term tertiary T cell niches (TTCNs). TTCNs serve as hubs for T cell activation, sharing features of tertiary

Combination therapy with targeted agents and immune checkpoint inhibitors (ICIs) represents a major advance in the treatment of advanced hepatocellular carcinoma (HCC), yet clinical efficacy remains modest. In the phase III COSMIC-312 trial, cabozantinib plus atezolizumab prolonged progression-free survival but did not improve overall survival, indicating the presence of tumor-intrinsic resistance mechanisms. In this study, we investigated the therapeutic and immunological effects of cabozantinib, celecoxib, and anti-programmed death-1 (anti-PD1), administered individually or in combination, i

Uncovering novel targets that synergize with immune checkpoint blockade (ICB) is an urgent clinical priority. While cancer cell-intrinsic CD28 facilitates immune escape by functioning as a non-classical RNA-binding protein to stabilize CD274 (PD-L1) mRNA, inhibiting this pathway in cancer cells without impairing essential T cell CD28 costimulation remains a major structural challenge. Here, we report the development of a 16:0 LPC-modified SM102-based lipid nanoparticle (LPC-LNP) that exploits altered tumor lipid metabolism for highly selective cancer cell transfection. By leveraging elevated l

BACKGROUND: Currently, predictive biomarkers for the efficacy of immunotherapy in metastatic gastric cancer (mGC) during the era of immune checkpoint inhibitors are still under evaluation. The aim of this study is to investigate the predictive value of early C-reactive protein (CRP) kinetics on the efficacy of programmed cell death protein-1(PD-1) inhibitors in mGC patients with microsatellite stable (MSS).
METHODS: This retrospective study included 59 mGC patients with MSS who had been treated with PD-1 inhibitors as first-line therapy at a tertiary hospital. Based on the "CRP-flare response

BACKGROUND: While immune checkpoint inhibitors (ICI) have transformed the landscape of clinical oncology, their application in rare malignancies such as anaplastic large cell lymphoma (ALCL) remains limited.
CASE: We report a patient with relapsed/refractory (r/r) ALCL treated at the University Hospital Krems who achieved sustained complete remission under PD-1 blockade after multiple prior lines of therapy. In addition, we performed a literature review and identified three previously published cases. Across the four reported patients, all achieved complete remission and remained progression-

OBJECTIVE: The optimal order of treatment with programmed cell death protein 1 (PD-1) inhibitor immunotherapy and stereotactic radiosurgery (SRS) in malignant gliomas remains unclear. This study examined whether the timing of PD-1 inhibitor administration relative to SRS affects patient neurological outcomes and survival in a multi-institutional propensity score-matched cohort.
METHODS: A retrospective analysis was conducted using the TriNetX Research Network database (2005-2022), identifying adults with malignant gliomas who received SRS and at least one PD-1 inhibitor. Patients were stratif

BACKGROUD: Different anti-PD-1 combination therapies are used to improve response rate and combat drug resistance for melanoma in the real world of China. While the reported safety data has remained scarce, especially for the controversial use of interferon. Identification of risk factors and comparative safety of different therapies will be beneficial for physicians to make decisions on rational usage of immunotherapy.
METHODS: A nationwide retrospective cohort study consecutively collected advanced melanoma patients treated by anti-PD-1 based therapies between July 1, 2018 and June 30, 2023

INTRODUCTION: Immune checkpoint inhibitors (ICIs) have transformed cancer treatment; however, their use in patients with pre-existing autoimmune diseases (PAD) remains controversial. This study evaluated the real-world safety and effectiveness of ICIs in older adults with metastatic non-small cell lung cancer (mNSCLC) and PAD.
MATERIALS AND METHODS: This retrospective cohort study used SEER-Medicare data to investigate individuals aged ≥66 years diagnosed with mNSCLC between 2015 and 2017, receiving either immunotherapy/chemoimmunotherapy (IT/CIT) or chemotherapy (CT). Outcomes were time to h

BACKGROUND: China launched a universal campaign of monovalent-inactivated EV-A71 vaccine in 2016. However, the monovalent-inactivated vaccine is ineffective against HFMD with other etiologies. To date, little is known about the epidemiological patterns and the replacement of the predominant pathogens that induce HFMD after the universal vaccination campaign.
METHODS: Using HFMD surveillance and VP1 sequencing data in pre- and post-vaccination periods, time series analysis, and Wavelet approaches, we identified pathogen replacement, changes in the viral spectrum and epidemiological patterns of

BACKGROUND: Maintaining high vaccination coverage requires understanding parental psychological factors, such as readiness conceptualized by the 7C model and descriptive norms, that shape decisions about childhood vaccinations. This study aimed to validate the Japanese short version of the Children-7C (C7C) scale, and examine how parental readiness and descriptive norms relate to vaccination status.
METHODS: This cross-sectional study analyzed data from 5148 parents in a self-reported online survey. Structural validity and internal consistency were assessed using confirmatory factor analysis
