Prognostic significance and regulatory role of ACOT7 in the tumor immune microenvironment of breast invasive ductal carcinoma: a multi-omics analysis.

BACKGROUND: Invasive Ductal Carcinoma (IDC), which is the most common histological subtype of breast cancer, is highly aggressive and progresses rapidly. Acyl-CoA thioesterase 7 (ACOT7) is a key regulator of cell survival, the cell cycle, and lipid and glucose metabolism. However, the mechanism of ACOT7 in IDC is still unclear. Our study aims to investigate the clinical significance of ACOT7 in IDC. METHODS: A comparative analysis of ACOT7 expression in IDC and matched normal tissues was performed using the limma R package on datasets from GEO and TCGA. Prognostic evaluation was conducted usi

Advances in ferroptosis research in ovarian cancer: molecular mechanisms and therapeutic perspectives.

Ovarian cancer is one of the most aggressive malignancies in the female genital system and presents a poor prognosis. Its high rate of recurrence and emergence of acquired resistance to the current chemotherapeutic approaches significantly restrict its long-term therapeutic efficacy. While treatment strategies including cytoreductive surgery (CRS) plus platinum (Pt)-based chemotherapy and poly(ADP-ribose) polymerase inhibitors (PARP inhibitors) have brought clinical benefits to a proportion of patients, the majority eventually suffer cancer relapse post-treatment. This finding suggests that cl

Paederoside alleviates rheumatoid arthritis by targeting SMAD4 to suppress histone lactylation through inhibiting lactate transport in senescent macrophages.

BACKGROUND: Rheumatoid arthritis (RA) is a chronic autoimmune disease characterised by persistent synovial inflammation and progressive joint destruction. Senescent macrophages are key drivers of RA pathogenesis through metabolic reprogramming, leading to intracellular lactate accumulation and histone lactylation, which promotes the expression of pro-inflammatory cytokines and synovial hyperplasia. Paederoside, an iridoid glycoside from Paederia scandens, has demonstrated anti-inflammatory effects, but its precise molecular targets in RA remain unclear. PURPOSE: This study aimed to elucidate

Trophoblast Fusion Deficiency in Placenta-Related Pregnancy Disorders.

Trophoblast cell fusion is a vital developmental process that enables the formation of the multinucleated syncytiotrophoblast (STB), which plays a central role in placental function and maternal-fetal exchange. Fusion defects in this lineage are closely associated with pregnancy complications such as preeclampsia (PE) and fetal growth restriction (FGR). Although the fusion of trophoblasts is a highly coordinated event, it involves multiple interdependent steps, including transcriptional programming, membrane remodeling, cytoskeletal rearrangement, metabolic adaptation, and immune regulation. Gut mycobiota alteration contributes to the pathogenesis of Pneumocystis pneumonia.

BACKGROUND: Pneumocystis is an opportunistic fungal pathogen that causes life-threatening pneumonia in immunocompromised hosts, with increasing incidence in HIV-negative individuals. Although the gut mycobiota has emerged as a critical regulator of distal immunity, its role in HIV-negative Pneumocystis pneumonia (PCP) remains entirely unexplored. METHODS: We established a murine model of Pneumocystis murina infection and performed full-length internal transcribed spacer (ITS) sequencing to characterize longitudinal changes in gut fungal communities over five weeks. Untargeted metabolomics was

Myeloid HIF-1α couples glycolytic energy supply with NCF2-dependent oxidative killing to protect against Klebsiella pneumoniae pneumonia.

BACKGROUND: Hypervirulent Klebsiella pneumoniae (hvKp) induces severe pneumonia and sepsis. HIF-1α coordinates metabolic and immune responses in myeloid cells, but its role in hvKp-mediated pulmonary defense remains undefined. METHODS: Monocyte HIF-1α expression was assessed in patients with Klebsiella pneumoniae (K. pneumoniae) pneumonia. Myeloid-specific Hif-1α knockout mice and BMDMs were used to examine survival, bacterial burden, and macrophage function. RNA-seq, Seahorse flux analysis, and confocal microscopy were employed to investigate the regulatory effects of HIF-1α on phagocytosis

Vγ9Vδ2 T cells in tuberculosis: protective immunity and translational perspectives.

BACKGROUND: Vγ9Vδ2 T cells constitute the predominant human circulating γδ T cell subset and serve as key mediators of anti-mycobacterial immunity through major histocompatibility complex (MHC)-unrestricted phosphoantigen recognition and rapid effector activation. SUMMARY: This review evaluates Vγ9Vδ2 T cell biology in the context of tuberculosis (TB). We delineate the three-stage developmental pathway of Vγ9Vδ2 T cells in the postnatal thymus and the butyrophilin 3A1/2A1 (BTN3A1/BTN2A1)-mediated "inside-out" antigen sensing mechanism, noting the limitations of extrapolating in vitro findings

Metabolomics-Driven Integration of Traditional Chinese Medicine for Neurological Disorders: From Precision Diagnosis to Therapeutic Innovation.

Neurological disorders are leading causes of disability and death worldwide, yet many patients still face delayed diagnosis, limited disease-modifying options and substantial treatment-related adverse effects. Traditional Chinese medicine (TCM) provides holistic, multi-target interventions through acupuncture, herbal formulas and adjunctive therapies, but its mechanisms remain insufficiently defined. Metabolomics, which enables system-wide profiling of small-molecule metabolites, offers an objective way to characterise disease-related metabolic networks and quantify the global effects of TCM.

Paternal malnutrition programs breast cancer risk and tumor metabolism in offspring.

BACKGROUND: While many studies have shown that maternal factors in pregnancy affect the cancer risk for offspring, few studies have investigated the impact of paternal exposures on their progeny's risk of this disease. Population studies generally show a U-shaped association between birthweight and breast cancer risk, with both high and low birthweight increasing the risk compared with average birthweight. Here, we investigated whether paternal malnutrition would modulate the birthweight and later breast cancer risk of daughters. METHODS: Male mice were fed AIN93G-based diets containing eithe

Epigenetics: Linking Early Postnatal Nutrition to Obesity Programming?

Despite constant research and public policy efforts, the obesity epidemic continues to be a major public health threat, and new approaches are urgently needed. It has been shown that nutrient imbalance in early life, from conception to infancy, influences later obesity risk, suggesting that obesity could result from "developmental programming". In this review, we evaluate the possibility that early postnatal nutrition programs obesity risk via epigenetic mechanisms, especially DNA methylation, focusing on four main topics: (1) the dynamics of epigenetic processes in key metabolic organs during

Constitutive mTORC1 activation in skeletal muscle increases inflammation but is not sufficient to impair glucose tolerance.

Aberrant mechanistic target of rapamycin complex 1 (mTORC1) signaling in skeletal muscle has been implicated in aging and insulin resistance; however, it is not known whether chronic mTORC1 activation directly causes glucose intolerance. We tested the hypothesis that constitutive mTORC1 activation in mouse skeletal muscle impairs glucose homeostasis. Six-month-old female and male mice with tamoxifen-inducible, muscle-specific knockout of the DEP domain containing 5, GATOR1 subcomplex subunit gene (Depdc5), a key component of the GAP activity toward rag 1 (GATOR1) complex and negative regulator

AARS1-mediated AKR1B10 lactylation stabilizes an aerobic glycolysis-positive feedback loop to drive lenvatinib resistance in hepatocellular carcinoma.

BACKGROUND: Lenvatinib resistance (LR) represents a significant obstacle in hepatocellular carcinoma (HCC) treatment. Aldo-keto reductase family 1 member B10 (AKR1B10) is involved in tumour metabolic reprogramming; however, its role in LR remains unclear. METHODS: Bioinformatics analyses of public databases were integrated and validated in established LR HCC cell lines. Functional assays (CCK-8, flow cytometry and Seahorse XF analysis) were performed to assess proliferation, apoptosis and aerobic glycolysis. Post-translational modifications of AKR1B10 were characterized using co-immunoprecipi

Emerging strategies in senotherapeutics: from broad-spectrum senolysis to precision reprogramming.

Cellular senescence, originally described as a finite proliferative arrest in cultured somatic cells, has since been recognized as a central mechanism underlying aging and the development of age-associated disorders. The progressive accumulation of senescent cells (SnCs) promotes chronic inflammation through the senescence-associated secretory phenotype (SASP) and circumvents immune-mediated clearance by upregulating pro-survival and immune checkpoint pathways. Early "first-generation" senolytics, including navitoclax (ABT-263) and the dasatinib-quercetin (D + Q) combination, provided proof-of

Glutamate accumulation in myelofibrosis microenvironment rewires mesenchymal stromal cells metabolic and epigenetic profiles.

BACKGROUND: Myelofibrosis (MF) is a clonal myeloproliferative neoplasm characterized by bone marrow (BM) fibrosis, osteosclerosis, and a proinflammatory tumor microenvironment (TME). The interplay between malignant hematopoietic stem cells and mesenchymal stromal cells (MSCs) drives disease progression. Although glutamate has been implicated in various cancers, its contribution to MF pathogenesis remains unclear. METHODS: High-performance liquid chromatography (HPLC) profiling was performed on sera from MF patients and healthy controls (HCs) to identify alterations in amino acid abundance. Th
