Molecular and genetic analyses of Actinomyces spp.

Members of the genus Actinomyces are predominant primary colonizers of the oral cavity and play an important role in initiating plaque development. These bacteria have evolved unique mechanisms that favor colonization and persistence in this micro-environment. The expression of cell-surface fimbriae is correlated with the ability of these bacteria to adhere to specific receptors on the tooth and mucosal surfaces, and to interact with other plaque bacteria. The elaboration of sialidase is thought to enhance fimbriae-mediated adherence by unmasking the fimbrial receptors on mammalian cells. The

Host-microbe interaction in the gastrointestinal tract.

In order for an infection to occur, the target organ must come in contact with sufficient microbes, the microbe must possess specific virulence factors, these virulence factors must be expressed, and the defenses of the organ system must be overcome. This dynamic process, which is ongoing in all living entities, can be described by the following relationship: [formula: see text] The establishment of infection first occurs in a particular organ. This phenomenon is known as tissue trophism and the association of microbes with organ systems governs the practice of clinical microbiology and infect

The root surface: an illustrated review of some scanning electron microscope studies.

This review paper highlights how the advent of a new type of surface microscopy in the late 1960s, scanning electron microscopy (SEM), was responsible for a fresh appraisal of the structure of the root surface. Details of the formation, resorption and repair of cementum, all surface phenomena, and the varied relationships and mineralization patterns of the two sets of fibres within cementum - the hall mark of the tissue - could be seen in a way and at a range of magnifications hitherto impossible. The major interpretational advances were made rapidly using secondary electron imaging of anorgan

Microbe-mineral interactions control fluoride mobilization from aluminum-substituted goethite into groundwater.

The genesis of geogenic fluoride (F-)-contaminated groundwater is frequently attributed to abiotic processes, while the roles of biotransformation kinetics of redox-sensitive iron (Fe) oxides have largely been underestimated. Aluminum (Al) substitution alters the structure and surface properties of natural Fe oxides, driving dynamic shifts between F- sinks and source by modulating F- mobilization-immobilization during redox-driven Fe-oxide transformation, but the influence of Al substitution on microbial Fe(III)-oxide reduction and F- turnover remains unclear. Through field investigations, mic

Short communication: effects of dietary inulin and animal density on stress in pigs.

Cortisol is a key glucocorticoid hormone involved in the physiological stress response of animals. Its secretion, largely regulated by the hypothalamic-pituitary-adrenal (HPA) axis, is essential for animal welfare and growth. Recent studies have shown that this stress-regulatory system operates in coordination with the gut microbiota, communicating with the brain through the gut-brain axis. Given that the diet can modulate microbial composition, dietary interventions thus offer potential in modulating HPA activity. Inulin is a prebiotic that promotes beneficial gut microbiota such as Lactobaci

Alpha-linolenic acid ameliorates T2DM via reshaping gut-liver axis and inflammatory GPR120-NF-κB/NLRP3 pathway in mouse and rat models.

BACKGROUND: The gut-liver axis, pivotal in managing glucose balance and insulin responsiveness, is central to the development of type 2 diabetes mellitus (T2DM). Research has highlighted the regulatory effects of dietary alpha-linolenic acid (ALA), but it remains unclear how ALA modulates gut microbiota and liver inflammation in T2DM. PURPOSE: This study aimed to systematically investigate ALA's influence on liver inflammation, intestinal barrier integrity, gut microbial composition, and metabolic homeostasis in T2DM, with a focus on the underlying molecular mechanisms. Determination of the effect of pyocyanin and Saccharomyces boulardii on gut microbiota and TLR4/MyD88/NF-κB and NLRP3 signaling pathways in sepsis induced by cecal ligation and puncture in rats.

Sepsis is a life-threatening systemic inflammatory condition marked by a dysregulated host response to infection. The intestinal microbiota plays a pivotal role in maintaining immune homeostasis and epithelial barrier integrity, whereas dysbiosis significantly contributes to the pathogenesis of sepsis. This study investigates the effects of the Pseudomonas aeruginosa-derived metabolite Pyocyanin and the probiotic Saccharomyces boulardii (S. boulardii) on microbial composition and the TLR4/MyD88/NF-κB/NLRP3 signaling axis in a cecal ligation and puncture (CLP)-induced rat model of sepsis. Candida albicans Oropharyngeal Infection Is an Exception to Iron-Based Nutritional Immunity.

Candida albicans is a commensal of the human gastrointestinal tract and a common cause of human fungal disease, including mucosal infections, such as oropharyngeal candidiasis and disseminated infections of the bloodstream and deep organs. We directly compared the in vivo transcriptional profile of C. albicans during oral infection and disseminated infection of the kidney to identify niche specific features. Overall, 97 genes were differentially expressed between the 2 infection sites. Virulence-associated genes, such as hyphae-specific transcripts, were expressed similarly in the 2 sites. Gen

Deoxycholic acid-chitosan coated liposomes combined with in situ colonic gel enhances renal fibrosis therapy of emodin.

BACKGROUND: Renal fibrosis is the final common pathological feature of various chronic kidney diseases (CKD). Despite recent advances, development of new treatments strategy is needed. Emodin (EMO), an important ingredient of Chinese medicine, rhubarb (Polygonaceae Rheum palmatum l.), has been reported to inhibit the development of renal fibrosis effectively. However, the poor oral bioavailability of EMO and the insufficient monotherapy therapy compromise its efficacy. PURPOSE: In order to enhance renal fibrosis therapy of emodin, an innovative combination therapy based on deoxycholic acid-ch

Systems Analysis of Gut Microbiome Influence on Metabolic Disease in HIV-Positive and High-Risk Populations.

Poor metabolic health, characterized by insulin resistance and dyslipidemia, is higher in people living with HIV and has been linked with inflammation, antiretroviral therapy (ART) drugs, and ART-associated lipodystrophy (LD). Metabolic disease is associated with gut microbiome composition outside the context of HIV but has not been deeply explored in HIV infection or in high-risk men who have sex with men (HR-MSM), who have a highly altered gut microbiome composition. Furthermore, the contribution of increased bacterial translocation and associated systemic inflammation that has been describe

Contemporary approaches for identifying individual risk for periodontitis.

Key breakthroughs in our understanding of the etiology and principles of predictable treatment of patients with chronic periodontitis first emerged in the late 1960s and carried on into the mid-1980s. Unfortunately, some generalizations of the evidence led many to believe that periodontitis was a predictable result of exposure to bacterial plaque accumulations over time. For a brief period, the initial plaque concept was translated by some to implicate specific bacterial infections, with both concepts (plaque exposure and specific infection) being false assumptions that led to clinical outcome

Electrolyte transport pathways induced in the midgut epithelium of Drosophila melanogaster larvae by commensal gut microbiota and pathogens.

KEY POINTS: The digestive tract of larval and adult Drosophila is an excellent analogue of the mammalian gut. Enterocytes of the posterior midgut are separated by septa, with no paracellular path, and therefore perform both immune and transport functions. Using microperfusion electrophysiology, we show that larvae emerging from the embryo into sterile medium have symmetrical apical and basal membrane conductances while larvae emerging into non-sterile medium have apical membranes fivefold more conductive than basal membranes. The channels inserted into the apical membranes could originate in m

Gut Microbiota-Derived Extracellular Vesicles in Cancer: Friends or Foes?

The gut microbiota has emerged as a key regulator of cancer biology through its dynamic interaction with host immunity, metabolism, and cellular signaling. Recent evidence highlights that gut microbiota-derived extracellular vesicles (GMEVs), nanosized bi-layered particles secreted by gut bacteria, play a pivotal role in mediating microbiota-host communications. Unlike whole bacteria, GMEVs can cross epithelial and vascular barriers, circulate systemically, and deliver bioactive cargo including proteins, nucleic acid, and metabolites to distant organs exerting profound effects on tumor develop

Effect of Diosmetin on Gut Microbiota and Serum Metabolites in Acute Pancreatitis Mice: A Metagenomic and Metabolomic Study.

Diosmetin is a bioactive flavonoid that exhibits well-documented antioxidant, anti-inflammatory, and anti-tumor properties. However, its potential to attenuate acute pancreatitis (AP) progression through gut microbiota modulation has not yet been elucidated. In this study, mice were pretreated with varying oral doses of diosmetin for 1 week before AP induction via intraperitoneal (i.p.) caerulein injections. The therapeutic efficacy and optimal dosage were determined through histopathological analysis of pancreatic tissue and serological biomarker assessment. Additionally, transcriptomic profi

Research progress on regulatory mechanisms of mucosal barriers and their applications in allergic diseases.

The mucosal barrier, as a critical interface of the body's defense system, is central to the pathogenesis of allergic diseases, with its structural integrity (epithelial cells, tight junctions, mucus layer, basement membrane) and functional homeostasis being key factors. This paper systematically elucidates the dynamic regulatory network constituted by five major signaling pathways: Wnt/β-catenin, TGF-β/Smad, RhoA/ROCK, MAPK, and JAK-STAT. These pathways interact through cross-talk (for example, Smad7 inhibits TGF-βRI to enhance Wnt signaling, and the β-catenin/Smad4 complex synergistically ac

Sequential adsorption enabled by dye-saturated biogenic vaterite: A waste-to-material strategy for enhanced methylene blue removal.

The coexistence of multiple dyes in industrial wastewater poses significant challenges for conventional adsorption treatment due to competitive and synergistic interactions. Herein, we present a sequential adsorption strategy using biogenic vaterite (BV), a microbially precipitated calcium carbonate, to treat mixed-dye wastewater. BV was first saturated with Congo red (CR) to form BV-CR, which was then used for methylene blue (MB) removal. CR pre-loading significantly enhanced MB adsorption capacity from 452.47 to 836.57 mg/g, with equilibrium reached within 10 min. Mechanistic analyses (XPS,
