Maxillary sinus schwannoma.

Schwannoma (neurilemmoma) is a benign tumour that arises from the Schwann cells which cover the nerve sheaths of the peripheral or autonomic nervous system especially the sympathetic system. Reported cases of schwannoma arising from the sinonasal tract are rare. We report a case of a 28-year-old man who presented to otolaryngology-head and neck surgery clinic with left cheek swelling and left-sided nasal obstruction for 1-year period. Endoscopic examination and high-resolution CT scan revealed a homogenous expansile mass occupying the left maxillary antrum extending to the left orbit and left

[Acute Chagas cardiopathy. A systematic study of the intracardiac excitatory-conduction and autonomic nervous systems in an autochthonous case in Acre].

The histopathology of the heart is described in an acute case of Chagas' disease (DC). Lesions involving the conducting system (SC) and the autonomic intracardiac nervous system (SNAIC) are emphasized. Light microscopy showed acute pan-carditis with plenty of Trypanosoma cruzi amastigotes within heart muscle cells. Multiple inflammatory foci were found in the SC with parasitic nests within the atrioventricular node and left his bundle. There were also severe atrial periganglionitis and perineuritis with or without peripheral involvement of those structures. Apparently there was no cardiac neur

[An autopsy case of olivocerebellar and thalamic degeneration, diffuse sclerosis and hypertrophic neuropathy: infantile system degeneration? (author's transl)].

The brain of a 21-year-old man with imbecility and spastic tetraplegia since early childhood showed a diffuse sclerosis of the cerebral white matter and symmetrical degeneration of the thalamus and olivocerebellar system. The spinal nerve roots as well as proximal branches of the spinal nerves were hypertrophic and there was seen a marked fibrosis with some onion-bulb formations of Schwann cells. In spite of a possible relation to an unknown infection at the age of 3 months this case could preferably be regarded as a peculiar form of the combined system degeneration in the early childhood, inc

Cannabinoids in the Cardiovascular System.

Cannabinoids are known to modulate cardiovascular functions including heart rate, vascular tone, and blood pressure in humans and animal models. Essential components of the endocannabinoid system, namely, the production, degradation, and signaling pathways of endocannabinoids have been described not only in the central and peripheral nervous system but also in myocardium, vasculature, platelets, and immune cells. The mechanisms of cardiovascular responses to endocannabinoids are often complex and may involve cannabinoid CB1 and CB2 receptors or non-CB1/2 receptor targets. Preclinical and some

The Antioxidative Role of Autophagy in Hearing Loss.

Autophagy, a highly conserved cellular mechanism, plays an essential role in the development and pathology of many central and peripheral nervous system diseases. The auditory system, especially hair cells (HCs) and spiral ganglion neurons (SGNs) in the inner ear, are postmitotic cells, which are extremely reliant on cellular homeostasis and energy supply. Therefore, autophagy may be involved in contributing to and facilitating the normal function of inner ear cells. Recently, studies on hearing loss induced by ototoxic drugs, noise exposure and other factors have revealed that autophagy could

Interferon Control of Neurotropic Viral Infections.

Interferons (IFNs) comprise a pleiotropic family of signaling molecules that are often the first line of defense against viral infection. Inflammatory responses induced by IFN are generally well tolerated during peripheral infections; yet, the same degree of inflammation during infection of the central nervous system (CNS) could be catastrophic. Thus, IFN responses must be modified within the CNS to ensure host survival. In this review, we discuss emerging principles highlighting unique aspects of antiviral effects of IFN protection following neurotropic viral infection, chiefly using new tech

Modelling the HIV persistence through the network of lymphocyte recirculation in vivo.

Human Immunodeficiency Virus (HIV) is able to persist in cellular and/or anatomical viral reservoirs, despite the effective inhibition of virus replication by the antiretroviral therapy (ART). Here we develop a mathematical model to gain some insights of HIV persistence relevant to the lymphocyte recirculation network of immune system and the central nervous system (CNS). Our simulations and analyses illustrate the role of the CNS as a virus reservoir to prevent antiretroviral drugs from penetrating the blood-brain (or blood-testis) barrier, and we examine the long-term impact of this reservoi

Pathogenesis of neuroimmunologic diseases. Experimental models.

Animal models of autoimmune diseases have greatly improved our current understanding of the pathogenesis of human autoimmunity and have provided the potential for therapies based on manipulation of the immune system. In our laboratory, we have investigated the immunopathogenesis of autoimmune diseases of the nervous system and muscle. We have developed immune-based approaches for the suppression of experimental autoimmune encephalomyelitis (EAE), a model for multiple sclerosis (MS), and experimental autoimmune neuritis (EAN), a model for the Guillain-Barré syndrome (GBS). These approaches incl

Immunomodulation by two potent analogs of met-enkephalin.

Met-enkephalin (Tyr-Gly-Gly-Phe-Met) and its more stable analogs, Tyr-D-Ala-Gly-MePhe-Met-NHC3H gamma-iso (1) and Tyr-D-Ala-Gly-MePhe-Gly-NHC3H gamma-iso (2) significantly enhanced human T-cell proliferation in vitro after 5 days of incubation in the absence of mitogen. The activity was completely inhibited by naloxone, an opioid antagonist. These peptides significantly enhanced human active T-cell rosette (CD2R) also on in vitro treatment. Furthermore, these analogs stimulated interleukin-2 production by human peripheral blood mononuclear cells in vitro which was completely inhibited by nalox

Functional alpha 1-adrenergic receptors on leukocytes of patients with polyarticular juvenile rheumatoid arthritis.

During the last decade it has been shown that the central nervous system can influence the immune system. In healthy individuals, catecholamines can inhibit the production of pro-inflammatory cytokines like interleukin-6 (IL-6) and tumor necrosis factor alpha (TNF-alpha) via interaction with beta 2-adrenergic receptors. In contrast, we show here that catecholamines can stimulate the production of the interleukin-6 (IL-6) in children with the chronic inflammatory disease polyarticular juvenile rheumatoid arthritis (JRA). The induction of IL-6 is mediated by triggering of alpha 1-adrenergic rece

The acute effects of central- and peripheral-acting dopamine antagonists on plasma HVA in schizophrenic patients.

The short-term effects of fluphenazine on plasma HVA concentrations were compared with the effects of fluphenazine and concurrent administration of debrisoquin, a monoamine oxidase inhibitor which does not cross the blood brain barrier and is used to enhance the CNS contribution to circulating plasma HVA concentrations. Fluphenazine significantly increased plasma HVA with or without debrisoquin 24 hours following the initiation of treatment. Domperidone, a butyrophenone dopamine antagonist which acts only in the peripheral nervous system, failed to alter plasma HVA concentrations. These data s

Non-reproductive systems and the pineal gland.

The paper reviews the data accumulated on the interrelationship between the pineal gland and the central nervous system as evidenced by induced neuroanatomical, biochemical and hormonal changes taking place in one system following the activation or inhibition of the other. The possible role of the pineal in modifying behaviour and its involvement in mental impairment is consequently discussed. Further questions dealt with are, whether in addition to the pineal effect via the hypothalamic-pituitary axis on the peripheral non-reproductive glands, thyroid and adrenals, a direct feedback mechanism

Exogenous opioids influence the microcirculation of injured peripheral nerves.

Local microvessels of peripheral nerve trunks (vasa nervorum) dilate following capsaicin-induced inflammation or local nerve trunk injury. In previous work, we observed that morphine blocked capsaicin-induced dilation of vasa nervorum presumably through the action of local opioid receptors. In the present work, we studied injury-related hyperemia of the rat sciatic vasa nervorum using laser Doppler and hydrogen clearance microelectrode measurements of local perfusion. Systemic morphine reversed hyperemia by vasoconstricting both extrinsic and intrinsic microvessels supplying 48-h-old "neuroma"

Peripheral nerve injection injury: an experimental study.

In an attempt to answer questions regarding nerve injection injuries, we injected 11 agents in current use and commonly administered by intramuscular injection into the sciatic nerves of adult Wistar rats. Equal volumes of normal saline were used as control. We harvested the sciatic nerves at various times after injection and examined them by both light and electron microscopy. We performed myelinated nerve fiber counts and constructed histograms. Any impairment of motor function was also noted. We gave injections to 79 animals a total of 158 times; 116 injections were directly into the nerve

Efficacy of non-pharmacological interventions for chemotherapy-induced peripheral neuropathy: a systematic review and network meta-analysis for randomized controlled trials.

BACKGROUND: Chemotherapy-induced peripheral neuropathy (CIPN) is a prevalent adverse effect linked to neurotoxic chemotherapeutic agents. Current pharmacological treatments exhibit limited efficacy and notable adverse effects. The clinical effectiveness of non-pharmacological therapies, like acupuncture, physical exercise (PE), cryotherapy (CR), and compression therapy, requires systematic comparison. This study employs a network meta-analysis (NMA) to appraise the efficacy and preventive effects of various non-pharmacological interventions on CIPN. METHODS: The study adhered to the PRISMA gu

Mitochondrial transfer as a therapeutic target for peripheral neuropathy.

Satellite glial cells transfer mitochondria to sensory neurons via myosin 10-dependent tunneling nanotubes. Ji et al. show that this transfer is impaired in diabetic neuropathy, causing energy failure. Restoring it via cell or mitochondrial transplantation alleviates pain and promotes nerve regeneration, revealing a new therapeutic strategy for peripheral neuropathy.

Monoallelic POLR3A Variants Cause Early-Onset Peripheral Neuropathy.

OBJECTIVE: Biallelic variants in genes encoding the RNA polymerase III complex (Pol III) cause a spectrum of neurological disorders primarily affecting the central nervous system. Monoallelic variants have been reported in the POLR3B subunit only, associated with neurodevelopmental disorder, epilepsy, and peripheral neuropathy. We describe a novel Pol III-related disorder caused by monoallelic variants in POLR3A and presenting primarily with peripheral neuropathy. METHODS: We performed clinical and genetic investigation of the affected families. Biophysical characterization of mutant proteins

Unraveling GDAP1: Bridging Mitochondrial Biology and Peripheral Neuropathy.

The mitochondrial outer membrane (OMM) plays a crucial role in maintaining cellular homeostasis by regulating mitochondrial dynamics, organelle interactions, and stress responses. In peripheral neurons-cells with high metabolic demands and long axons-the OMM acts as a vital platform for coordinating bioenergetics, calcium signaling, and redox balance. Ganglioside-induced differentiation-associated protein 1 (GDAP1), an OMM-anchored protein, has emerged as a key regulator of mitochondrial fission and transport, redox homeostasis, and mitochondrial membrane contact sites (MCSs). Genetic variants

Touch and manual action in chemotherapy-induced peripheral neuropathy: a mixed-methods study.

Chemotherapy induced peripheral neuropathy (CIPN) is a frequent side effect of a number of chemotherapeutic agents which are widely used in the treatment of common cancers. Sensory symptoms primarily affect the fingers and toes and include numbness, tingling and pain. However, there is limited understanding of how these touch impairments may impact manipulation skills. Questionnaire and focus group methods were used to describe the experience of CIPN side effects impacting manual activities in 25 self-selected participants recruited from a cancer charity web site advertisement.
