HTLV-1-associated myelopathy/tropical spastic paraparesis (HAM/TSP) inflammatory network.

HTLV-1 associated myelopathy/ tropical spastic paraparesis (HAM/TSP) is a systemic immune-mediated inflammatory disease and tissues other than nervous can be damaged, mainly ocular, rheumatic and dermatologic. Over 90% of HTLV-1-infected individuals remain lifelong asymptomatic and this retrovirus persists indefinitely in their CD4+ T-lymphocytes. The infection is maintained due to the proliferation of lymphocytes that harbor a provirus and express HTLV-1 proteins, particularly Tax, promoting an active and selective expansion of infected T cells. High proviral load is related to disease progre

Necrotizing fasciitis with toxic shock syndrome in a child: a case report and review of literature.

INTRODUCTION: Necrotizing fasciitis was described as early as the fifth century BC. It showed an increased incidence worldwide in the past several years. CASE PRESENTATION: An 8-year-old Arabian boy was referred for admission as a case of cellulitis of the left thigh. Ten days prior to admission he had a cat scratch to his left thigh and the parents did not seek medical advice at that time. The child was again examined by orthopedic surgeon and a diagnosis of cellulites was made at that time.Physical examination on admission revealed a very toxic appearing weak child with cold extremities and

Pseudotumors due to IgG4 immune-complex tubulointerstitial nephritis associated with autoimmune pancreatocentric disease.

Autoimmune pancreatitis (AIP) is a mass-forming chronic fibroinflammatory condition centered on the pancreatobiliary system and characterized by predominant immunoglobulin G4 (IgG4)-positive plasma cells. Recent reports have brought to light the multiorgan involvement of this disease. We describe a series of 5 cases of tubulointerstitial nephritis (TIN) associated with AIP and characterize the clinical, pathologic, ultrastructural, and immunopathologic features of TIN. The specimens consisted of 4 biopsies and 1 nephrectomy. The average patient age was 64 years (range 45 to 78) and the male to

Maternal plasma hypertonicity is accentuated in the postterm rat.

OBJECTIVE: In humans and rats, pregnancy-associated maternal plasma volume expansion and plasma hypotonicity may facilitate maternal-to-fetal water transfer. Although reduced amniotic fluid volume occurs commonly in postterm pregnancy, the mechanisms are unknown. We previously demonstrated a reversal of pregnancy-induced maternal plasma hypotonicity that occurs in the near term (20 days) pregnant rats. We sought to determine whether the relative maternal plasma hypertonicity continues in the postterm period. STUDY DESIGN: Rat gestation (normal, 21 days) was prolonged with subcutaneous progest

The first report of extraosseous Ewing's sarcoma in the rectovaginal septum.

AIMS AND BACKGROUND: To report an extremely rare case of Ewing's sarcoma located in the rectovaginal septum. Ewing's sarcoma is a highly malignant neoplasm of bone, which usually occurs during childhood. Common extraosseous localizations of Ewing's sarcoma include the trunk, extremities, uterus, cervix and vagina. METHODS: A 45-year-old woman presented to us with a six-month history of pain in the lower abdomen during intercourse. Pelvic examination was performed and a palpable mass was found. The mass had a size of 9 x 6 cm, a soft tissue consistency, was partially movable and the patient fe

Fas-deficient lpr mice are more susceptible to graft-versus-host disease.

The Fas/Fas ligand (FasL) pathway is involved in a variety of regulatory mechanisms that could be important for the development of graft-vs-host disease (GVHD) after bone marrow transplantation (BMT), such as cytolysis of target cells by cytotoxic T cells, regulation of inflammatory responses, peripheral deletion of autoimmune cells, costimulation of T cells, and activation-induced cell death. To further evaluate the role of Fas/FasL in the complex pathophysiology of GVHD, we used Fas-deficient B6.lpr mice as recipients in a MHC-matched minor histocompatibility Ag-mismatched murine model for G

Fetal liver hematopoietic stem cells as a target for in utero retroviral gene transfer.

Retroviral-mediated gene transfer into hematopoietic precursors often results in only short-term gene transduction in vivo. Loss of the transduced genetic material over time may be caused by the limited ability of retroviral infection to transduce genes into early, pluripotent hematopoietic stem cells. Because fetal liver contains actively proliferating multipotential stem cells that should be more susceptible to retroviral-mediated gene transfer than quiescent cells derived from adult bone marrow, these cells may be an ideal target for gene transduction. Furthermore, physiologic expansion of

Phenotypic and genotypic heterogeneity of peripheral T-cell lymphoma.

A series of 21 phenotypically characterised T-cell lymphomas histologically defined as lymphocytic, lymphoblastic, immunoblastic, AILD type, pleomorphic, T-zone and Lennert's T-cell lymphoma, were investigated for T-cell receptor (TcR) and immunoglobulin (Ig) gene rearrangements. Phenotypic analyses of frozen sections and cell suspensions were heterogeneous and in many cases no single T-cell marker recognised all of the malignant cells. Data derived by staining with antibodies reactive with antigens in paraffin embedded tissue were consistent with T NHL in all cases except lymphoblastic lympho

Role of tumor promotion in affecting the multi-hit nature of carcinogenesis.

The assessment of risk from low-level exposure to radiation and chemicals is hindered by the basic lack of scientific understanding of the complex nature of the multiple levels of protective or synergistic interactions. Radiation and chemicals have the potential of inducing mutations, cell death and altered gene expression. The biological consequences of each of these effects is again complex, since many factors can enhance or mask the effect of a single mutation, cytotoxic or gene modulatory change. Carcinogenesis, representing but one chronic disease state, is a multi-step process, involving

Visceral Obesity and Its Complications: The Role of Bioelectrical Impedance Analysis in Longevity Medicine.

Background: Visceral obesity is increasingly recognised not as a simple excess of adipose tissue, but as a systemic pathological condition characterised by profound metabolic, endocrine, and immune dysregulation. Visceral adipose tissue (VAT) operates as an autonomous neuro-immune-endocrine organ whose dysfunctional expansion drives insulin resistance, atherogenesis, and accelerated cellular ageing through mechanisms converging on chronic low-grade sterile inflammation, referred to as inflammaging. Objectives: This narrative review integrates evidence across four domains: (1) the multi-system

Targeting the Jun-Irf8-CD36 axis attenuates fibrotic scar formation and promotes functional recovery after spinal cord injury.

BACKGROUND: Fibrotic scar formation constitutes a significant pathological obstacle that impedes neural regeneration and long-term functional recovery following spinal cord injury (SCI). However, the spatial distribution of key pro-fibrotic mediators within lesion scars and the upstream regulatory mechanisms driving fibroblast activation remain inadequately defined. This study aims to characterize CD36-associated fibrotic remodeling after SCI and to determine whether targeting the c-Jun-Irf8-CD36 axis could attenuate scar formation, improve the regenerative microenvironment, and promote functi

Perimenopause as an obesogenic sensitive period: Contributions to elevated cardiovascular risk.

BACKGROUND: Perimenopause, encompassing the period of progressive menstrual irregularity preceding a woman's final menstrual cycle and extending 12 months thereafter, is associated with an acceleration in atherosclerotic cardiovascular disease (CVD) risk, attributable in part to redistribution of body fat. During perimenopause, even in the context of minimal-to-modest weight gain, women experience an expansion of visceral adipose tissue. Concomitantly, women often experience a reduction in gluteofemoral subcutaneous adipose tissue. This pattern of fat redistribution is associated with a greate

Bone Marrow Aspirate Concentrate With Two-Staged-Revision ACL Reconstruction.

BACKGROUND: The incidence of anterior cruciate ligament (ACL) injuries, reconstructions, and re-ruptures has rapidly increased. Patients with failed ACL reconstructions have been reported to suffer from far worse outcomes as compared with those with primary reconstructions, prompting the advancement of surgical and biologic techniques. Effective treatment of re-tears has been shown to be achieved either utilizing a 1-stage or 2-staged approach, with the latter preferred if the patient presents with significant bone loss, previously malpositioned tunnels, or unacceptable tunnel expansion. A telomere-to-telomere genome reveals arachidonic acid as a key regulator of hepatic lipid metabolism during hibernation.

Elucidating the molecular mechanisms governing mammalian hibernation is crucial for understanding energy homeostasis and physiological adaptation to extreme environments. In this study, we present the first high-quality telomere-to-telomere (T2T) reference genome (2.69 Gb) of the Daurian ground squirrel (Spermophilus dauricus), a typical hibernating mammal. The assembled genome comprises 19 chromosomes, nine of which are completely gapless chromosomes, and exhibits a scaffold N50 of 160 Mb. Comparative genomic analysis with 17 hibernating and non-hibernating mammals revealed lineage-specific e

Feeder-free generation of functional dendritic cells from human pluripotent stem cells.

BACKGROUND: The scarcity of primary conventional dendritic cells (cDCs) and the limited functionality of monocyte-derived dendritic cells (moDCs) have long hindered mechanistic and translational studies in human dendritic cell (DC) biology and immunotherapy. METHODS: We developed a feeder-free differentiation platform to generate CD1c+CD141+ human pluripotent stem cell-derived conventional dendritic cells (hPSC-cDCs) to provide a scalable source of DCs with defined properties. A Design-of-Experiments (DoE) optimization strategy was applied to refine cytokine and serum conditions, with the goa

Single-cell profiling reveals MIF-mediated immune evasion and CD8 + T cell exhaustion in relapsed multiple myeloma.

Multiple myeloma (MM) is a heterogeneous hematologic cancer marked by clonal plasma cell expansion in the bone marrow. Although treatments have improved, relapse remains common due to clonal evolution, tumor microenvironment changes, ultimately leading to drug resistance. This study utilized single-cell RNA sequencing data analysis to explore immune microenvironment dynamics during MM progression and relapse, aiming to uncover key pathways and cellular interactions correlated with disease relapse. ScRNA-seq data from 20 MM patients (10 primary, 2 remission, 8 relapsed) from the GEO dataset GSE

HEATR1 deficiency enhanced gastric cancer cell PANoptosis to boost anti-tumor immunity.

BACKGROUND: Despite the promise of immune checkpoint inhibitors (ICIs) for gastric cancer (GC) treatment, limited cytotoxic T cell infiltration remains a major barrier to therapeutic efficacy. This study investigates whether targeting HEAT repeat containing 1 (HEATR1) can induce immunogenic PANoptosis to enhance anti-tumor immunity. METHODS: Bioinformatics analysis of TCGA-STAD cohort, syngeneic mouse models, and patient-derived organoids (PDOs) were employed to assess HEATR1 function. Mechanistic studies examined STAT1/IRF1 signaling, PANoptosis markers, and dendritic cell (DC) activation. T

The global outbreak of Oropouche virus: surveillance of global trends.

Oropouche virus (OROV) is a re-emerging arthropod-borne virus of the genus Orthobunyavirus (family Peribunyaviridae) that poses an increasing public health concern in South and Central America and the Caribbean. First identified in Trinidad in 1955, OROV has caused recurrent outbreaks, with recent resurgence associated with geographic expansion and increased urban transmission. In 2024 alone, more than 11,600 confirmed cases were reported in the Region of the Americas, with continued transmission documented in early 2025. The virus is primarily transmitted in urban settings by the biting midge

Lymphoid Organs Remodeling in Non-Obese Diabetic Goto-Kakizaki Rats Immunized With SARS-CoV-2 Antigens.

The COVID-19 pandemic revealed reduced vaccine efficacy and worse outcomes in people with type 2 diabetes mellitus (T2DM). However, how T2DM, independent of obesity, alters immune responses and lymphoid organ architecture remains unclear. We evaluated antibody production and lymphoid organ morphology following SARS-CoV-2 antigen immunization in Goto-Kakizaki (GK) rats, a non-obese T2DM model, to evaluate the isolated effect of diabetes. Ten-week-old male Wistar and GK rats received three intramuscular injections of SARS-CoV-2 antigens at 15-day intervals. Two antigen formulations were used: ch

Establishment and functional analysis of an inducible Apoc2-knockout mouse model to investigate the role of Apoc2 in normal hematopoiesis.

Apolipoprotein C2 (APOC2) has been identified as a potential therapeutic target for acute myeloid leukemia, but its impact on normal hematopoiesis remains unclear. Here we generated a global inducible Apoc2 knockout (KO) mouse model by breeding R26-CreERT2 mice with mice homozygous for Apoc2 lox sites (Apoc2fl/fl) over three generations. Global Apoc2 KO was induced in 4-8-week-old mice by 5 consecutive days of intraperitoneal administration of tamoxifen. Successful Apoc2 KO was confirmed by measuring Apoc2 mRNA levels, showing 80-99% reduction in the liver, bone marrow, spleen and blood tissue
