The design of nano-drug delivery vehicles responsive to tumor microenvironment stimuli has become a crucial aspect in developing cancer therapy in recent years. Among them, the enzyme-responsive nano-drug delivery system is particularly effective, as it utilizes tumor-specific and highly expressed enzymes as precise targets, leading to increased drug release at the target sites, reduced nonspecific release, and improved efficacy while minimizing toxic side effects on normal tissues. NAD(P)H:quinone oxidoreductase 1 (NQO1) is an important reductase associated with cancer and is overexpressed in

Dengue is a mosquito-borne infectious disease that is highly endemic in tropical and subtropical countries. Symptomatic patients can rapidly progress to severe conditions of hemorrhage, plasma extravasation, and hypovolemic shock, which leads to death. The blood tests of patients with severe dengue typically reveal low levels of high-density lipoprotein (HDL), which is responsible for reverse cholesterol transport (RCT) and regulation of the lipid composition in peripheral tissues. It is well known that dengue virus (DENV) depends on membrane cholesterol rafts to infect and to replicate in mam

BACKGROUND AND AIM: An essential aspect of ensuring availability and stability of mesenchymal stem/stromal cells (MSCs) products for clinical use is that these cells are cryopreserved before individual infusion into patients. Currently, cryopreservation of MSCs involves use of a cryoprotectant solution containing dimethyl sulfoxide (DMSO). However, it is recognized that DMSO may be toxic for both the patient and the MSC product. In this Production Assistance for Cellular Therapies (PACT) and Biomedical Excellence for Safer Transfusion (BEST) Collaborative study, we compared a novel DMSO-free s

Purinergic signalling plays a central regulatory role on cell proliferation, migration, differentiation, survival, and inflammation. However, the impact of purinergic modulators-based therapies on tissue repair is still overlooked. By using a systematic review framework, we investigated the therapeutic relevance of purinergic agonists and antagonists in animal models of skin wound healing. Following PRISMA guidelines, PubMed/Medline, Scopus, Web of Science, Embase, and CINAHL databases were searched, identifying 12 in vivo preclinical studies. Risk of bias was assessed using the SYRCLE tool. T

OBJECTIVE: Calcitonin gene related peptide (CGRP) is a central mediator in migraine and can trigger attacks in susceptible individuals. Therapies targeting CGRP or its receptor provide clinical benefit, but treatment responses vary. This suggests that local factors regulating CGRP availability and signalling within migraine relevant tissues may influence its biological effects. The extent and functional relevance of CGRP degradation at the dura mater, including possible effects on biomarker detection, remain insufficiently characterised.
METHODS: Proteomic analyses were used to identify human

Sepsis-associated acute kidney injury (SAKI) lacks mechanism-based therapies, with upstream ferroptosis regulators undefined. This study investigated growth differentiation factor 15 (GDF15) as a suppressor of TLR4-MyD88-NF-κB signaling and associated ferroptosis. Differential GDF15 expression was identified by integrated analysis of three GEO datasets (GSE232404, GSE30718, GSE44925) and validated in SAKI patient kidney tissue (n = 30 vs. controls n = 25) and in kidneys from cecal ligation and puncture (CLP) mice. CLP mice received recombinant GDF15 (rmGDF15) (50 ng/kg ip); renal function (Scr

Global prevalence of chronic kidney disease (CKD) has increased considerably in the recent decades. Overactivity of the renin-angiotensin-aldosterone system (RAAS), associated to renal inflammation and fibrosis, contributes to its evolution. The treatments currently employed to control CKD progression are limited and mainly based on the pharmacological inhibition of RAAS, associated with diuretics and immunosuppressive drugs. However, this conservative management promotes only partial deceleration of CKD evolution and does not completely avoid the progression of the disease and the loss of ren

Only a few cases of malignant peripheral nerve sheath tumour (MPNST) associated with Von Recklinghausen's disease or type I neurofibromatosis (NF-1) have so far been reported worldwide, yet the primary disease (NF-I ) does not seem rare even in Africa. We present a case of a 40 year old woman with MPNST of the left thigh associated with NF-1. The diagnosis was based on clinical, radiological and histopathological evidence. She presented with a 25 year history of painless, multiple, generalized skin nodules and hyperpigmented spots. She also noticed a gradually progressive, painless, redundant

Inflammation is known to preclude tolerance after transplantation. We have previously shown that systemic inflammation in xenograft recipients (SIXR) precedes activation of coagulation in the absence of T cell responses. Accordingly, SIXR may amplify innate and adaptive immune responses against xenografts after pig-to-primate xenotransplantation, even with efficient immunosuppressive therapy. We evaluated the impact of anti-inflammatory agents on pro-inflammatory cytokines and chemokines in pig artery patch and heart xenograft recipients. Baboons received an artery patch (Group1, n=8) or heart

Periodontitis (P) is one of the most common human dental diseases, with significant medical and social implications. According to the World Health Organization, more than 45% of the adult population worldwide exhibits clinical signs of periodontitis, with 10%-15% of cases becoming generalized with destruction of the alveolar bone. This review presents the results of modern research demonstrating that the etiopathogenesis of periodontitis is not limited to local microbial factors; systemic, social, and behavioral factors can play a significant role, determining the body's susceptibility to chro

Two billion people worldwide suffer from anemia, which can lead to the onset of cardiac disorders; nevertheless, the precise mechanisms remain unclear. There are at least three distinct mechanisms by which iron deficiency (ID) contributes to the development of cardiac disorders. First, ID increases concentrations of intact fibroblast growth factor-23 (iFGF-23), which promotes left ventricular hypertrophy. Additionally, individuals with ID typically have low circulating levels of vitamin D and an increased body burden of cadmium (Cd). Both factors-high Cd levels and a lack of vitamin D-elevate

INTRODUCTION: Adhesion G-Protein Coupled Receptors (aGPCRs) represent the second largest GPCR family and consist of 32 members in humans, of which 19 are orphan receptors. Despite making up a substantial portion of GPCRs, there are no clinically approved therapeutics targeting them or using them in clinical applications. aGPCRs have functional roles in numerous tissues and their dysregulation through mutations or expression is associated with a variety of diseases, including cancer. Therefore, there is strong interest in therapeutic targeting aGPCRs or their controlled pathways.

Digestive system tumors, including gastric cancer, hepatocellular carcinoma, pancreatic cancer, and colorectal cancer, represent one of the most prevalent and lethal malignancy groups worldwide, with high incidence and mortality rates. Despite the continuous development of chemotherapy in cancer treatment, this traditional therapy generally suffers from insufficient tumor specificity, drug resistance and limited efficacy. Precise targeting strategies such as antibody-drug conjugates (ADCs) and peptide-drug conjugates (PDCs) combine cell-targeting delivery with potent antitumor activity and hav

INTRODUCTION: Insulin-like Growth Factor Binding Protein 6 (IGFBP-6) is a member of the IGFBP family with unique and relevant biological properties. Emerging evidence suggests that IGFBP-6 plays a critical role in musculoskeletal homeostasis and pathology, insulin-like growth factor (IGF)-dependent mechanisms to include IGF-independent effects on cell proliferation, migration, and differentiation. IGFBP-6 is implicated in the regulation of inflammatory and fibrotic processes within articular tissues and tendons, influencing disease onset and progression.

Endometrial cancer (EC) is a gynecological malignancy classified into two biologically distinct subtypes, Type I and Type II EC. The more aggressive Type II EC exhibits a poor response to conventional platinum-based chemotherapy. Propofol, an anesthetic agent in gynecologic surgery, has been reported to exert antitumor effects beyond anesthesia; however, its role as a chemoadjuvant in EC remains insufficiently explored. In this study, we investigated the effects of propofol combined with carboplatin on drug resistance- and metastasis-associated microRNAs and genes using in vitro and in vivo Ty

P-glycoprotein (P-gp), a transmembrane efflux pump encoded by the ABCB1/MDR1 gene, is a major contributor to multidrug resistance in hematological malignancies. These malignancies, arising from hematopoietic precursors at various differentiation stages, can manifest in the bone marrow, circulate in the bloodstream, or infiltrate tissues. P-gp overexpression in malignant cells reduces the efficacy of chemotherapeutic agents by actively expelling them, decreasing intracellular drug concentrations, and promoting multidrug resistance, a significant obstacle to successful treatment. This review exa
