IRE1 regulates the proteostasis of TDP-43/TARDBP in ALS/FTD through ribosome-associated quality control.

Amyotrophic lateral sclerosis (ALS) and frontotemporal dementia (FTD) are progressive neurodegenerative disorders characterized by motor neuron degeneration, leading to muscle weakness, atrophy, and cognitive impairments. A defining pathological hallmark of ALS/FTD is the cytosolic mislocalization and accumulation of TAR DNA-binding protein 43 (TDP-43), highlighting its critical role in ALS pathogenesis. However, the molecular mechanisms underlying TDP-43 proteostasis remain poorly understood. Through a genetic screening approach, we identify inositol-requiring enzyme 1 (IRE1), an endoplasmic

Sleep consolidation of social memories and common ground.

Sleep consolidation effects are well established in the cognitive domains of memory and language learning, however very little is known regarding sleep effects in social contexts. Across three experiments, we investigated whether sleep enhances the accuracy of socially formed memories and communicative efficiency, including the use of partner-specific common ground between interlocutors in conversation. Participants completed a computerised (Experiment 1) or face-to-face (Experiment 2) matcher-director task with two other people before and after 12 h of sleep/wake. Both experiments showed more

Panax ginseng as a microbial ecosystem modulator: implications for systemic health via the gut-organ axes.

Panax ginseng C.A. Meyer, a renowned medicinal herb, exerts many of its systemic effects through intricate interactions with the gut microbiota, a relationship that also addresses the challenge of its own limited oral bioavailability. This review comprehensively examines the role of ginseng and its bioactive constituents in modulating gut microbiota and their subsequent influence on host health through key gut-organ axes. Based on an in-depth analysis of literature, we summarize how ginseng intervention is associated with a modulated gut microbial ecology-characterized by enriched beneficial t

Standard of care for rehabilitation in critical illness.

BACKGROUND: Rehabilitation is recognised as a cornerstone of intensive care, essential for optimising functional recovery and reducing long-term disability. Contemporary ICU populations, characterised by advanced age, multimorbidity, and prolonged stays, are at heightened risk of muscle wasting, immobility, frailty, cognitive decline, and functional dependence. Mitigation of these sequelae requires careful interprofessional collaboration for person-centred rehabilitation across the care continuum. CONTENT: This review synthesises evidence from randomised controlled trials, meta-analyses, and

The Relationship between Multisensory Sensitivity and Pain Interference in Adolescents with Menstrual Pain: The Mediating Roles of Pain Catastrophizing and Fatigue.

OBJECTIVE: Among adolescent girls, primary dysmenorrhea (ie,, menstrual pain without an identified underlying pathology) has been linked to heightened sensitivity to non-painful stimuli (eg,, light, touch, sound, etc.) and future widespread pain. However, less is known about how multisensory sensitivity is related to menstrual and non-menstrual pain interference, or which cognitive and psychophysiological factors contribute to these associations. This study examined the relationships between multisensory sensitivity and menstrual and non-menstrual pain interference, and whether pain catastroph

Ergothioneine attenuates whole-abdominal irradiation-induced multi-organ injury via the gut-heart-brain axis by modulating calcium voltage-gated channel subunit alpha1 C (Cacna1c) expression.

Despite the precise targeting of radiation therapy, collateral damage to adjacent healthy tissues remains an inevitable consequence. Currently, no effective clinical intervention exists to prevent or alleviate these adverse effects. To address this issue, our study established a whole-abdominal irradiation (WAI) model using C57BL/6J mice to investigate the systemic effects of ionizing radiation (IR) on the gut and other organs. Results confirmed that IR not only causes significant intestinal damage but also induces cardiac injury and cognitive dysfunction through remote effects. Ergothioneine

Effects of a Strength and Creative Dance Intervention on Brain Electrical Activity, Heart Rate Variability, and Dual-Task Performance in Women with Fibromyalgia: A Randomized Controlled Trial Protocol.

Fibromyalgia is a complex chronic disorder involving persistent widespread pain accompanied by functional limitations, cognitive impairments, and alterations in neural processing. Previous research indicates that exercise-based interventions can play a key role in alleviating symptom burden and enhancing physical performance; however, there is limited evidence regarding their impact on neurophysiological mechanisms. Creative dance, in combination with strength training, may stimulate both motor and cognitive systems, promoting brain plasticity and functional improvements. This study will analy

PIAS1-mediated GSK3β SUMOylation exacerbates tauopathy and cognitive deficits in Alzheimer's disease models.

Age-related neurodegenerative disorders, such as Alzheimer's disease (AD), are characterized by the accumulation of pathological Tau protein (tauopathy), which drives neurotoxicity and cognitive decline. Although SUMOylation significantly influences tauopathy, the underlying mechanisms remain largely elusive. Here, utilizing a bimolecular fluorescence complementation (BiFC) assay to monitor Tau-Tau aggregation and screen for SUMO E3 ligases, we identified PIAS1 as a critical driver of Tau pathology. Genetic analysis of the UK Biobank cohort revealed that single nucleotide polymorphisms in the

Adult-Onset Langerhans Cell Histiocytosis With Calvarial and Hypothalamic Involvement: Two Cases.

Langerhans cell histiocytosis (LCH) is a rare clonal proliferative disorder of bone marrow-derived dendritic cells that is uncommon in adults. Its clinical presentation varies depending on the organ involved. Central nervous system (CNS) involvement poses a critical therapeutic challenge because of permanent neurological dysfunction. Here, we report two contrasting cases of adult-onset LCH with distinct clinical outcomes. Case 1 involved a 36-year-old woman presenting with a persistent headache. Neuroimaging revealed a solitary osteolytic lesion in the left parietal bone with dural thickening.

Membrane-Coated DNA Nanoflowers Attenuate Cognitive Decline in Apolipoprotein E4 Mice.

APOE4-driven mitochondrial dysfunction is one of the important primary drivers of cognitive decline in neurodegenerative diseases. However, achieving mitochondrial-targeted drug delivery in the nervous system requires overcoming multiple barriers, necessitating the development of a safer and more efficient bionanoparticle drug delivery system. DNA nanoflowers (DNFs), generated via rolling circle amplification (RCA), offer programmable platforms for creating targeted drug delivery vehicles. Here, we designed a multifunctional DNF surface conjugated with ferrocene groups, loaded with resveratrol

Bipolar Disorder in Older Adults: A Comprehensive Overview of Pharmacotherapy Patterns and Clinical Considerations.

In older adults, bipolar disorder (BD) frequently presents atypically, with manic symptoms manifesting as irritability or confusion rather than euphoria. Cognitive impairments, including mild cognitive impairment and dementia, can obscure or mimic bipolar symptoms, complicating accurate diagnosis. Depressive episodes tend to be more frequent and prolonged, increasing misdiagnosis risk as unipolar depression. Pharmacological management requires careful consideration of age-related pharmacokinetic and pharmacodynamic alterations. Mood stabilizers including lithium, valproate, and lamotrigine are

CX3CL1/CX3CR1-dependent microglial phagocytosis of oligodendrocyte precursor cells contributes to sevoflurane-induced myelination impairments in neonatal mice.

General anesthesia exposure in early life may disrupt the normal progression of developmental myelination, but the underlying mechanisms remain unclear. Early postnatal microglia in developing white matter exhibit diverse transcriptional and functional states, including a population with pronounced phagocytic activity. This study aims to investigate whether sevoflurane impairs oligodendrocyte myelination by promoting microglial phagocytosis of oligodendrocyte precursor cells (OPCs). Mice received either a single 2-h exposure to 3.3% sevoflurane on postnatal day 2 (P2) or repeated 2-h exposures

Towards a Closed-Loop Bioengineering Framework for Immersive VR-Based Telerehabilitation Integrating Wearable Biosensing and Adaptive Feedback.

Telerehabilitation-the remote delivery of rehabilitation services-is undergoing a paradigm shift with the convergence of immersive virtual reality (VR) and wearable biosensor technologies. This perspective article outlines a vision for home-based motor and cognitive rehabilitation that is engaging, personalized, and data-driven. We describe how immersive VR environments (for example, simulations of home settings or supermarkets) coupled with wearable sensors can address current challenges in rehabilitation by increasing patient motivation, enabling real-time biofeedback, and supporting remote

Effects of plyometric training on physical performance and mental well-being in young soccer players.

Plyometric training (PT) improves performance in young soccer players, but its psychological effects remain underexplored. This is especially relevant for young elite players facing early selection pressures. The aim was to evaluate the effects of unilateral and bilateral PT on performance and mental well-being in elite young soccer players according to their maturation level. Sixty-seven tier-3 male athletes (12.03±2.34 years; pre- and post-peak heigh velocity groups) were randomised into unilateral PT (n= 23), bilateral PT (n= 23), or active control (n= 21), with all groups maintaining stand

Traumatic Brain Injury and Chronic Traumatic Encephalopathy: Not Only Trigger for Neurodegeneration but Also for Cerebral Amyloid Angiopathy?

Traumatic brain injury (TBI) has been linked to the development of neurodegenerative diseases, particularly Alzheimer's disease (AD) and chronic traumatic encephalopathy (CTE). This review critically assesses the relationship between TBI and cerebral amyloid angiopathy (CAA), highlighting the complexities of diagnosing CAA in the context of prior head trauma. While TBI has been shown to facilitate the accumulation of amyloid plaques and tau pathology, the interplay between neurodegenerative processes and vascular contributions remains underexplored. Epidemiological studies indicate that TBI in

The induction of a specific mental set for problem solving is accompanied by increased neural representational similarities.

A mental set is a cognitive bias that results from repeatedly applying a stereotypical problem-solving strategy, which can hinder new and creative ideas. However, the neural mechanisms underlying mental set induction remain unclear. We hypothesized that repeated use of a strategy increases neural representation similarity, thus producing a mental set. To test this, we conducted representational similarity analysis across continuously presented mindset-induction trials using the same loose chunk decomposition problem-solving strategy. This induction was expected to impair the solving of subsequ
