Anterograde and retrograde degenerative reactions in caudate nucleus and putamen after experimental lesion of the pallidum in squirrel monkey (Saimiri sciureus).

After stereotactic lesions in the pallidum in 4 squirrel monkeys, electron microscopic material from the striatum was examined for anterograde and retrograde degenerative changes. In the experiment with pallidum internum lesion, only degenerated striatal fibers were observed, more than likely thalamostriatal fibers that pass through the site of the lesion. The three experiments with pallidum externum lesion revealed that the two types of striatal aspiny neurons react with a penumbral degeneration to interruption of their axons. Also, the axospinous type IV striatal synapses, which originate in

Harvest and Application of Bone Marrow Aspirate Concentrate to Address Acetabular Chondral Damage During Hip Arthroscopy.

BACKGROUND: During hip arthroscopy, managing concomitant cartilage damage and chondrolabral junction breakdown remains an ongoing challenge for orthopaedic surgeons, as previous studies have associated such lesions with inferior postoperative outcomes1-7. Although higher-level studies are needed to fully elucidate the benefits, recent literature has provided supporting preliminary evidence for the utilization of bone marrow aspirate concentrate (BMAC) in patients with moderate cartilage damage and full-thickness chondral flaps undergoing acetabular labral repair7,8. Thus, as the incorporation

Stromal cell-derived factor-1 regulates expression of vascular endothelial growth factor and osteopontin after tooth extraction.

BACKGROUND: The present study aimed to investigate the expression of stromal cell-derived factor-1 (SDF-1) after tooth extraction in rats and its regulatory effect on the expression of osteopontin (OPN) and vascular endothelial growth factor (VEGF). METHODS: The first mandibular molar of the rats on the left was extracted, the histopathological changes of the extraction sockets were observed, and the SDF-1 protein and mRNA expression were measured at 0, 1, 4, 7, 10, and 14 days. Furthermore, we used AMD3100 to block SDF-1/CXC chemokine receptor 4 (CXCR4) signaling, and the expression changes

Short-Chain Fatty Acids: Bridging Gut Microbiota and Systemic Aging-Mechanisms, Interventions, and Current Challenges.

Aging is a systemic degenerative process that can lead to functional decline in multiple organs, such as skeletal muscles and the heart, and accelerates the overall aging process through organ-to-organ interactions mediated by metabolites such as short-chain fatty acids (SCFAs). SCFAs serve as a crucial link connecting intestinal health and anti-aging, and their levels and functions undergo significant changes with aging. However, current research lacks understanding of the downstream molecular mechanisms of SCFAs, and intervention methods are mostly limited to simple regulation. This article

Electron microscopy reveals water networks inside hydrated cellulose fibers.

Cellulose is a highly abundant linear glucose polymer that exists in amorphous and fibrillar forms 1-3 . Primarily produced by vascular plants but also microbes and tunicates, cellulose predominantly performs architectural functions by stabilizing cell walls, tissues, and multicellular communities 4-9 . Owing to its association with other cell surface materials, physiological higher-order structures of cellulose have been difficult to obtain. Here, we describe a never-dried tunicate cellulose fibril structure by cryogenic electron microscopy, which resolves a fiber of more than 360 cellulose s

Water-Driven Oxygen and Nitric Oxide Release from Porphyrin- and Mn-Based Metal-Organic Framework Enables Accelerated Acute Wound Healing.

Medical gases, particularly oxygen and nitric oxide (NO), have attracted significant interest in clinical applications, notably wound healing, due to their role in enhancing cell proliferation, angiogenesis, and collagen deposition. However, the use of these gases has been limited by challenges such as inefficient gas delivery and potential toxicity to normal tissues. In this study, we elucidate a feasible approach using a porphyrin-based metal-organic framework (MOF), a unique material that shows immense potential for dual-gas-assisted wound healing. The MOF nanorods, meticulously designed, c

Macro- and Microanatomy of the Sympathetic Innervation of the Spleen in Rodents.

In recent years, several studies have demonstrated the crucial role played by the sympathetic spleen innervation in regulating immune cell function involving fighting pathogens or tissue injury. These findings have sparked interest across different research fields with a common goal of understanding and manipulating splenic sympathetic activity to modulate immune function and inflammation. However, the anatomical identification of spleen-projecting neurons in rodents presents a considerable challenge, given the multi-compartmentalized location of their cellular components. This article address

Corneal injury is associated with stromal and vascular alterations within cranial dura mater.

The cornea and cranial dura mater share sensory innervation. This link raises the possibility that pathological impulses mediated by corneal injury may be transmitted to the cranial dura, trigger dural perivascular/connective tissue nociceptor responses, and induce vascular and stromal alterations affecting dura mater blood and lymphatic vessel functionality. In this study, using a mouse model, we demonstrate for the first time that two weeks after the initial insult, alkaline injury to the cornea leads to remote pathological changes within the coronal suture area of the dura mater. Specifical

Vascular Cast to Program Antistenotic Hemodynamics and Remodeling of Vein Graft.

The structural stability of medical devices is established by managing stress distribution in response to organ movement. Veins abruptly dilate upon arterial grafting due to the mismatched tissue property, resulting in flow disturbances and consequently stenosis. Vascular cast is designed to wrap the vein-artery grafts, thereby adjusting the diameter and property mismatches by relying on the elastic fixity. Here, a small bridge connection in the cast structure serves as an essential element to prevent stress concentrations due to the improved elastic fixity. Consequently, the vein dilation is

Dysregulation of Histone Deacetylases Inhibits Trophoblast Growth during Early Placental Development Partially through TFEB-Dependent Autophagy-Lysosomal Pathway.

Dysregulated biological behaviors of trophoblast cells can result in recurrent spontaneous abortion (RSA)-whose underlying etiology still remains insufficient. Autophagy, a conserved intracellular physiological process, is precisely monitored throughout whole pregnancy. Although the exact mechanism or role remains elusive, epigenetic modification has emerged as an important process. Herein, we found that a proportion of RSA patients exhibited higher levels of autophagy in villus tissues compared to controls, accompanied with impaired histone deacetylase (HDAC) expression. The purpose of this s

Quaternary structure of patient-homogenate amplified α-synuclein fibrils modulates seeding of endogenous α-synuclein.

Parkinson's disease (PD) and Multiple System Atrophy (MSA) are progressive and unremitting neurological diseases that are neuropathologically characterized by α-synuclein inclusions. Increasing evidence supports the aggregation of α-synuclein in specific brain areas early in the disease course, followed by the spreading of α-synuclein pathology to multiple brain regions. However, little is known about how the structure of α-synuclein fibrils influence its ability to seed endogenous α-synuclein in recipient cells. Here, we aggregated α-synuclein by seeding with homogenates of PD- and MSA-confir

Is essential tremor a degenerative disorder or an electric disorder? Degenerative disorder.

Essential tremor (ET) is a highly prevalent neurologic disease and is the most common of the many tremor disorders. ET is a progressive condition with marked clinical heterogeneity, associated with a spectrum of both motor and non-motor features. However, its disease mechanisms remain poorly understood. Much debate has centered on whether ET should be considered a degenerative disorder, with underlying pathological changes in brain causing progressive disease manifestations, or an electric disorder, with overactivity of intrinsically oscillatory motor networks that occur without underlying str

Expression and potential role of CXCL5 in the pathogenesis of intrauterine adhesions.

OBJECTIVE: C-X-C motif chemokine ligand 5 (CXCL5), a member of the chemokine family, is associated with remodeling of connective tissues. However, its role in formation of intrauterine adhesions (IUA) remains unclear. We aimed to investigate the expression and mechanism underlying the role of CXCL5 in IUA. METHODS: Expression of CXCL5 in IUA was detected by immunohistochemistry in a rat model of IUA and by real-time PCR and western blotting in patients with IUA. The protein levels of matrix metalloproteinase 9 (MMP9) and transcription factor p65 in human endometrial cells were assessed by wes

Mutations with pathogenic potential in proteins located in or at the composite junctions of the intercalated disk connecting mammalian cardiomyocytes: a reference thesaurus for arrhythmogenic cardiomyopathies and for Naxos and Carvajal diseases.

In the past decade, an avalanche of findings and reports has correlated arrhythmogenic ventricular cardiomyopathies (ARVC) and Naxos and Carvajal diseases with certain mutations in protein constituents of the special junctions connecting the polar regions (intercalated disks) of mature mammalian cardiomyocytes. These molecules, apparently together with some specific cytoskeletal proteins, are components of (or interact with) composite junctions. Composite junctions contain the amalgamated fusion products of the molecules that, in other cell types and tissues, occur in distinct separate junctio

Non-destructive mechanical characterisation of UVA/riboflavin crosslinked collagen hydrogels.

AIMS: To establish a non-destructive method of characterising the mechanical properties of collagen hydrogels to model corneal tissue and to examine the effect of photochemical crosslinking on their mechanical properties. METHODS: Collagen hydrogels were manufactured, submerged in 0.1% riboflavin solution and crosslinked using two UVA tube bulbs with an intensity of between 2.8 and 3.2 mW/cm(2). The hydrogels were clamped around their outer edge and deformed using a sphere. The deformation was measured in situ using a long-working-distance microscope connected to a CCD camera, and the deforma

Intercellular septate-like junction of neoplastic cells in myxopapillary ependymoma of the filum terminale.

Septate junction, a common intercellular feature of invertebrate epithelium, is absent in most vertebrate tissues. In an ultrastructural study of three cases of myxopapillary ependymoma of the filum terminale, structures similar to septate junction were observed in two cases. They were circumferential bands around the finger-like processes of neoplastic cells in which slightly widened 30-40-nm intercellular spaces (as compared to 15-20 nm in non-junctional region) were transversed at regular intervals by parallel septa resulting in characteristic ladder-like appearance. The electron-dense sept

Anterior proliferative vitreoretinopathy. Clinicopathologic, light microscopic, and ultrastructural findings.

Proliferative vitreoretinopathy (PVR) involving the posterior and equatorial retina is an established clinicopathologic entity. Clinically, a similar process, anterior PVR (APVR), results in anterior dragging of the peripheral retina by membranes which connect to the ciliary body or iris and cause circumferentially and radially fixed retinal folds. The pathology of APVR, however, has not been reported. The authors describe pathologic findings in 28 cases of APVR and ultrastructural pathologic findings in 6 surgical APVR specimens. Anterior PVR was frequently associated with retinal detachment

[Clinical aspects and differential diagnosis of occipital bone processes].

Tumours and tumour-like processes of the cranial bones do not occur very frequently; 2.5 per cent of all bone tumours are found in the calotte. On the other hand, the pre-operative and operative error possibilities regarding these processes are considerable, leaving multiple bone metastases out of consideration. The situation is complicated by many processes in the vicinity of the bone which affect or alterate the bone secondarily. On the basis of two better known tumours, the giant cell tumour and the aneurysmatic bone cyst, the diagnostic possibilities and difficulties are discussed in great
