Maternal obesity induces activator protein 1-mediated inflammatory response to impair embryonic neurogenesis.

Maternal obesity (MO) is a growing global problem, which poses significant risks to fetal neurodevelopment and long-term neurological functions of offspring, but the underlying molecular mechanisms remain to be established. To address this female mice were fed either a control diet or a high-fat diet (HFD) for 2 months to induce obesity, and the same dietary treatments were maintained during pregnancy. Embryos were sampled at E11.5 and E13.5. Single-cell RNA sequencing revealed reduced proportions of neurons and neural progenitors in embryos from obese mothers. The downregulation of neurogenes

Systematic discovery of retina-enriched Rik genes identifies 1190005I06Rik as a novel modulator of visual signalling.

BACKGROUND: High‑throughput transcriptome projects have revealed thousands of mammalian genes with little or no functional annotation. Among these are hundreds of loci assigned provisional “Rik” identifiers following discovery in the RIKEN cDNA annotation effort. Although often dismissed as genomic dark matter, such genes may encode tissue‑restricted proteins that modulate physiologic functions and influence disease. The retina is a highly specialised neural tissue and a common site of inherited disorders; understanding its molecular repertoire could illuminate novel therapeutic avenues. METHO

RAGE/NF-κB signaling mediates paraquat-induced alveolar-capillary barrier injury in a human alveolar microphysiological system.

Paraquat (PQ), a highly toxic herbicide that causes fatal pulmonary injury, has no specific clinical antidotes, mainly due to incomplete understanding of its pathogenic mechanisms. Inhalational exposure to PQ is the primary route of toxicity among agricultural workers. However, conventional experimental models fail to accurately replicate the physiological characteristics of the alveolar-capillary barrier and the air-liquid interface (ALI). In this study, we developed an Alveolar Microphysiological System (AMS) that co-cultures alveolar epithelial cells (EPCs) and pulmonary microvascular endot

METTL16 S-glutathionylation-triggered RNA m6A modification of IGF2BP3 inhibits CFTR expression and promotes chemoresistance in colorectal cancer.

5-Fluorouracil (5-FU) resistance in colorectal cancer (CRC) involves oxidative stress mechanisms, but the role of epitranscriptomic regulation remains unclear. This study investigates how oxidative post-translational modifications of the N6-methyladenosine (m6A) methyltransferase METTL16 contribute to 5-FU resistance. Parental (HCT8, HCT15) and 5-FU-resistant CRC cells were compared using redox proteomics, m6A-seq, RNA-seq, and functional assays. METTL16 S-glutathionylation was assessed via streptavidin pulldown and mass spectrometry. IGF2BP3's role was validated through knockdown/overexpressi

Aberrant splicing of MBD1 reshapes the epigenome to drive convergent myeloerythroid defects in MDS.

Myelodysplastic neoplasms (MDS) feature hematopoietic deficits driven, in part, by transcript splicing abnormalities. To date, such disease-driving transcripts have been identified in association with specific splicing factor mutations. However, conserved aberrant splicing-derived transcripts that drive MDS independently of mutational status remain poorly studied, despite representing global therapeutic targets. In this study, we characterize an MDS-associated isoform of methyl-binding-domain 1 (MBD1; MBD1-L) as a novel member of this class of transcripts. Rather than originating from a mutant

Lipotoxicity Amplifies IL-6 Trans-Signaling in Sinusoidal Endothelial Cells to Promote Endotheliopathy and Liver Injury.

BACKGROUND AND AIMS: Metabolic dysfunction-associated steatotic liver disease (MASLD) affects approximately 25% of the global population and is a major health concern. Liver sinusoidal endothelial cells (LSECs) undergo morphological and functional alterations during MASLD progression, resulting in endotheliopathy characterized by pro-inflammatory phenotypes. We hypothesize that lipotoxicity synergistically exacerbates LSEC dysfunction in MASLD via IL-6 trans-signaling. METHODS: Human adipocytes were treated with palmitic acid (PA). Human LSECs were treated with PA and the IL-6/soluble IL-6 re

IL-23-engineered oncolytic vaccinia virus enhances pancreatic cancer control through inflammasome-driven Th17-skewed CD4+ T-cell responses.

BACKGROUND: Oncolytic viruses represent a promising immunotherapeutic strategy for converting pancreatic and other immunologically cold tumors. While their ability to stimulate innate immune signaling and promote antitumor immunity is established, mechanisms enabling durable adaptive responses remain poorly defined. METHODS: We used CRISPR-Cas9-based homologous recombination to generate a thymidine kinase (TK) and B2R double-deleted backbone virus, VV∆TK∆B2R (VV-DD). On this backbone, we engineered VV∆TKmp40∆B2Rmp19 (VV-DDIL23), which expresses the p40 and p19 subunits of interleukin-23 (IL-2

Multi-omics integration identifies VIM and LGALS3 as apoptosis-associated genes in astrocytes after traumatic brain injury.

Apoptosis contributes substantially to neurological deficits after traumatic brain injury (TBI), yet the molecular drivers and cellular regulation of astrocyte-associated apoptosis remain poorly defined. We integrated bulk RNA-seq, single-cell RNA-seq, and spatial transcriptomics to identify apoptosis-related hub genes in TBI. Differentially expressed genes were intersected with apoptosis-related genes and ranked by protein-protein interaction network topology. Astrocyte heterogeneity was examined through sub-clustering, pseudotime, regulon, and in silico transcription-factor perturbation anal

Kidney Transcriptome Sequencing Improves Molecular Diagnosis and Reveals Splicing Complexity Across the Alport Spectrum.

INTRODUCTION: Hereditary glomerular basement membrane (GBM) disorders caused by pathogenic variants in COL4A 3/A4/A5 are increasingly recognized as Alport spectrum. Disease severity varies widely and is influenced by variant type, allelic dosage, sex, and genetic modifiers. DNA-based testing incompletely captures this heterogeneity, particularly splice-altering and regulatory variants, limiting precise molecular diagnosis and prognostic stratification. METHODS: We performed whole-transcriptome sequencing on 93 kidney biopsies from 93 patients (90 families) with ultrastructural GBM abnormaliti

Cistanche tubulosa phenylethanol glycoside liposome ameliorates oxaliplatin-induced peripheral neuropathy by promoting RNA N6-methyladenosine modification.

BACKGROUND: Oxaliplatin-induced peripheral neuropathy (OIPN) is a frequent dose-limiting toxicity in cancer treatment, with no effective FDA-approved therapy available. Dysregulated RNA N6-methyladenosine (m6A) modification and impaired PI3K/AKT signaling have been implicated in neuronal injury and regeneration. Phenylethanol glycosides (CPhGs), the main bioactive compounds of Cistanche tubulosa, possess neuroprotective and anti-inflammatory properties, but their role in OIPN remains unclear. The current research aims to investigate the potential therapeutic effect in OIPN. METHODS: Mice were

Single cell analysis of muscle contracture in cerebral palsy reveals profibrotic and antimyogenic stem cell populations with altered cell-cell interactions.

Development of muscle contractures is common in cerebral palsy (CP) and is characterized by high muscle stiffness that limits function and mobility. However, the state of stem cells within contracture, particularly muscle stem cells and fibroadipogenic progenitors, is largely unknown. This study leverages single cell RNA sequencing technology to determine how specific cell types are altered in the contracture environment. Skeletal muscle biopsies were collected from children with CP or typically developing (TD) children undergoing surgery. The 10X Genomics platform was used on tissue from n =
