Cartilage oligomeric matrix protein (COMP) is a member of the thrombospondin family of extracellular matrix glycoproteins. All members of the family contain a highly conserved region of thrombospondin type 3 sequence repeats that bind calcium. A mutation in COMP previously identified in a patient with pseudoachondroplasia resulted in abnormal sequestration of COMP in distinctive rER vesicles. The mutation, Asp-446 --> Asn, is located in the type 3 repeats of the molecule. This region was expressed in a mammalian culture with and without the mutation to study the structural or functional proper

Neovascularization frequently accompanies chronic immune responses characterized by T cell infiltration and activation. Angiogenesis requires endothelial cells (ECs) to penetrate extracellular matrix, a process that involves matrix metalloproteinases (MMPs). We report here that activated human T cells mediate contact-dependent expression of MMPs in ECs through CD40/CD40 ligand signaling. Ligation of CD40 on ECs induced de novo expression of gelatinase B (MMP-9), increased interstitial collagenase (MMP-1) and stromelysin (MMP-3), and activated gelatinase A (MMP-2). Recombinant human CD40L induc

Most epithelial cells secrete a glycoprotein-rich apical extracellular matrix that can have diverse but still poorly understood roles in development and physiology. Zona Pellucida (ZP) domain glycoproteins are common constituents of these matrices, and their loss in humans is associated with a number of diseases. Understanding of the functions, organization and regulation of apical matrices has been hampered by difficulties in imaging them both in vivo and ex vivo. We identified the PAN-Apple, mucin and ZP domain glycoprotein LET-653 as an early and transient apical matrix component that shape

Intervertebral disc degeneration (IVDD) is a leading cause of chronic disabling musculoskeletal disorders, with its incidence rising annually among the aging global population. This degenerative process is strongly linked to persistent functional impairment and reduced quality of life in patients, while placing a growing socioeconomic burden on global healthcare systems. Previous studies have demonstrated that kaempferol exerts potent anti-inflammatory effects by modulating pro-inflammatory cytokine expression and attenuating extracellular matrix degradation in degenerated intervertebral discs

Injury to articular cartilage remains a major clinical challenge owing to its limited self-healing capacity. Current cell-based therapies are limited by issues such as dedifferentiation and regulatory hurdles, whereas existing scaffold systems often lack the essential biochemical signals necessary for effective tissue regeneration. Consequently, biomimetic acellular strategies capable of recruiting endogenous stem cells while establishing a conducive biochemical microenvironment to direct their chondrogenic differentiation are needed. In this study, we developed a bioactive, hierarchical porou

INTRODUCTION: Wooden breast disease is a myopathy of the skeletal muscle in chickens of commercial breeding. Although the underlying pathophysiology remains unknown, we and others have previously shown that affected broilers display varying degrees of fibrosis, extracellular matrix (ECM) remodeling, inflammation, and alterations in various molecular signaling pathways. Other myopathy conditions, such as Duchenne muscular dystrophy, also affect the cardiac muscle and are associated with fibrosis and reduced cardiac function. To determine potential cardiac implications of wooden breast disease a

Chondroitin sulfate (CS) is a symptomatic slow acting drug for osteoarthritis (OA) widely used for the treatment of this highly prevalent disease, characterized by articular cartilage degradation. However, little is known about its mechanism of action, and recent large scale clinical trials have reported variable results on OA symptoms. Herein, we aimed to study the modulations in the intracellular proteome and the secretome of human articular cartilage cells (chondrocytes) treated with three different CS compounds, with different origin or purity, by two complementary proteomic approaches. BACKGROUND: Gastric cancer (GC) is traditionally classified into intestinal (IGC), diffuse (DGC), and mixed (MGC) types based on pathological features, with each subtype exhibiting distinct clinical outcomes. Among these, DGC is associated with poor prognosis, characterized by low cell adhesion and a high stromal component. Recent proteomic studies have revealed significant differences in extracellular matrix (ECM) composition between DGC and IGC, highlighting the critical role of ECM in tumor biology. MGC, which combines both intestinal and diffuse characteristics, presents substantial hetero

BACKGROUND: Biotechnological advances in tissue engineering increasingly exploit naturally derived scaffolds as bioactive delivery systems. Acellular fish skin (AFS) provides a collagen-rich extracellular matrix favorable for tissue regeneration but lacks intrinsic antimicrobial activity. Propolis, a polyphenol-rich natural resin, exhibits antimicrobial, antioxidant, and immunomodulatory properties, yet its direct application is limited by burst release and local cytotoxicity. In this study, we aimed to develop and characterize a propolis-functionalized AFS scaffold as a multifunctional biomat

Doxycycline (DOX) is a second-generation tetracycline antibiotic extensively used in clinical practice, not only for its antimicrobial activity but also for its emerging therapeutic potential in modulating extracellular matrix (ECM) in remodeling processes. Increasing evidence supports its role in attenuating pathological conditions associated with ECM degradation by targeting matrix metalloproteinases type 2 (MMP-2), including cancer progression and abdominal aortic aneurysm (AAA). However, DOX exhibits limited stability in aqueous and semi-physiological environments, undergoing hydrolysis an

Uterine fibroids or leiomyomas are the most common benign neoplasms in women and a major source of morbidity, often impairing quality of life and overall well-being. Current management options-ranging from medical therapy to surgery-may be limited by adverse effects, incomplete symptom control, recurrence, or loss of uterine integrity. Given their dense, collagen-rich extracellular matrix, uterine fibroids may be amenable to enzymatic degradation using Clostridium histolyticum collagenase (CCH), a minimally invasive, non-hormonal therapeutic candidate. This scoping review maps the current lite

INTRODUCTION: Colorectal cancer (CRC) remains one of the most common solid malignancies in the Western population. Despite advances in screening, diagnostics, and treatment, there are still no highly specific tests for early diagnosis or biomarkers that can reliably identify high-risk patients or monitor disease progression more effectively than currently established tumor markers. The pathogenesis of CRC is closely associated with chronic inflammation, oxidative stress, and remodeling of the tumor microenvironment. The present study focuses on several biomarkers selected based on available li

Alendronate (ALN) is widely employed against skeletal disorders, but its low oral bioavailability and prolonged skeletal retention necessitate high systemic doses that carry significant risk. The research team previously developed a novel bioceramic drug-delivery system (DDS) that sequentially releases gentamicin during the first two weeks post-implantation (with reported therapeutic concentrations released daily) and ALN thereafter to be used as a bone substitute. The main objective of this study was to validate the DDS's osteogenic capacity ex vivo, once gentamicin had been depleted and only

Osteogenesis imperfecta (OI) is a family of rare heritable skeletal disorders associated with dominant mutations in the collagen type I encoding genes and recessive defects in proteins involved in collagen type I synthesis and processing and in osteoblast differentiation and activity. Historically, it was believed that the OI bone phenotype was only caused by abnormal collagen type I fibrils in the extracellular matrix, but more recently it became clear that the altered bone cell homeostasis, due to mutant collagen retention, plays a relevant role in modulating disease severity in most of the

BACKGROUND: Tenocytes derived from tendons have been reported to be effective in the treatment of rotator cuff tears through the expression of extracellular matrix proteins. Human dermal fibroblasts, known to express collagen types I and III as tenocytes do, may likely be substitutes for tenocytes to enhance healing rotator cuff tears.
PURPOSE: To demonstrate the capability of human dermal fibroblasts to enhance healing of rotator cuff tears.
STUDY DESIGN: Controlled laboratory study.
METHODS: The cellular properties and expression profiles of growth factors were compared between human derm

Autoimmune-mediated dry eye disease is a pathological feature of multiple disorders including Sjögren's syndrome, lupus and rheumatoid arthritis that has a life-long, detrimental impact on vision and overall quality of life. Although late stage disease outcomes such as epithelial barrier dysfunction, reduced corneal innervation and chronic inflammation have been well characterized in both human patients and mouse models, there is little to no understanding of early pathological processes. Moreover, the mechanisms underlying the loss of cornea homeostasis and disease progression are unknown. He

Mechanical stress damage and insufficient self-repair can contribute to osteoarthritis (OA) in the affected joint. As the effects of stress on chondrocyte metabolism can regulate cartilage homeostasis, the specific stress-response condition is therefore a key to the generation of an OA disease model. We aimed to produce a specific stress- and cell-based OA model after evaluating the metabolic responses of chondrocytes in response to a series of static and cyclic compression stressors. A static load exceeding 40 psi initiated extracellular matrix (ECM) degradation through a decrease in the sulp

INTRODUCTION: Xenogeneic extracellular matrix (ECM) hydrogels have shown promise in remediating cardiac ischemia damage in animal models, yet analogous human ECM hydrogels have not been well development. An original human placenta-derived hydrogel (hpECM) preparation was thus generated for assessment in cardiomyocyte cell culture and therapeutic cardiac injection applications.
METHODS AND RESULTS: Hybrid orbitrap-quadrupole mass spectrometry and ELISAs showed hpECM to be rich in collagens, basement membrane proteins, and regenerative growth factors (e.g. VEGF-B, HGF). Human induced pluripoten

Cartilage is an important tissue in skeletogenesis, in the growth of long bones, and as a flexible component of the mature skeleton. The extracellular matrix proteins type II collagen and aggrecan comprise 90% of the matrix and are characteristic of cartilage. Type II collagen provides structural integrity to the tissue, while aggrecan confers resiliency. The quantity of type II procollagen is controlled at the level of transcription of mRNA from the COL2A1 gene. In addition, type II procollagen can be expressed in two isoforms by differential splicing of the primary gene transcript, a post-tr

BACKGROUND CONTEXT: The nonunion rate after posterolateral spinal fusion can be as high as 35%. This has stimulated interest in the development of techniques for enhancing new bone formation, including the addition of bioactive peptides or the use of cell-based therapies, including genetically modified cells. In previous studies we have demonstrated that exposing autologous, marrow-derived osteoprogenitor cells to a recombinant human bone morphogenetic protein-6 (rhBMP-6) containing extracellular matrix induces osteoblastic differentiation, and that these cells are capable of increasing new bo
