Killing of Cryptococcus neoformans by human peripheral blood mononuclear cells stimulated in culture.

Since cell-mediated immunity (CMI) is critical for host defenses against the encapsulated fungus Cryptococcus neoformans, the production of human antifungal effector cells as a consequence of the CMI response was investigated. Peripheral blood mononuclear cells (PBMC) were stimulated in culture with killed C. neoformans. Stimulated (but not unstimulated) PBMC killed a subsequent inoculum of live encapsulated organisms, with maximal killing seen after 3-7 days. Killing required the presence of both adherent and nonadherent stimulated PBMC and was enhanced by anticapsular antibody. In contrast,

Factors responsible for preterm delivery of the immature newborn infant (less than or equal to 1000 gm).

Although there are excellent studies examining factors responsible for preterm delivery in general, there is a scarcity of data describing factors specifically associated with delivery of the immature newborn infant. Our purpose is to characterize these factors in order to determine what may be done to limit preterm delivery rate of extremely low birth weight infants. Obstetric variables responsible for the birth of 338 live in-born immature neonates in a large single perinatal center were determined. We were unable to affect 65% of these pregnancies because there was no chance for interventio

Selectivity in the responses of hamsters to conspecific vocalizations.

Lordosis responses by estrous hamsters were triggered by brief manual stimulation. Lordosis durations then were timed, as manual stimulation was discontinued, and subjects were exposed to tape-recordings of conspecific vocalizations reproduced at intensities of 0-60 dB SPL. Relative to the durations observed in the presence of ambient noise alone, recordings of stress-elicited screams failed to facilitate lordosis regardless of stimulus intensity. In contrast, ultrasounds by male or female hamsters did prolong lordosis, and to extents that were directly related to intensity but unrelated to th

Functional and metabolic consequences of vitamin B-6 deficiency in the rat heart.

The cardiac functional and metabolic consequences of pyridoxine deficiency were studied in rats maintained on a pyridoxine-deficient diet for 10 weeks. Because food intake was diminished in the pyridoxine-deficient rats, a second group of animals was fed a diet restricted to the intake of the pyridoxine-deficient animals. The inotropic response (developed pressure) to an isoproterenol or Ca2+ concentration response curve was measured simultaneously with high energy phosphate levels using a modified Langendorf apparatus and 31P nuclear magnetic resonance spectroscopy. The inotropic response to

Ophthalmic manifestations of infantile phytanic acid storage disease.

Two patients had infantile phytanic acid storage disease. Patient 1 had nystagmus from early infancy, epicanthal folds, esotropia, and a pigmentary retinopathy. The second case had similar manifestations; however, no nystagmus was present. Both patients were hypotonic as infants, had a severe hearing impairment, and were moderately severely developmentally delayed. Serum phytanic acid levels in both cases were clearly elevated. The fundus and fluorescein angiogram showed macular and diffuse retinal pigment epithelial defects, vascular attenuation, and pigmentary dispersion. The electroretinogr

Transient hypogammaglobulinemia, elevated immunoglobulin E levels, and food allergy.

Four infants presented with the combination of food allergy, transient hypogammaglobulinemia (THI), and elevated serum IgE levels. Food allergy was documented by history, positive skin tests for immediate hypersensitivity, radioallergosorbent test, histamine release studies, and lymphocyte transformation in response to food allergens. THI was probably secondary to decreased production since there was no evidence of protein loss from the gastrointestinal tract. Immunologic studies revealed normal B cell number and function in vitro. T cell number and proliferative response to mitogens and antig

Interlocking schedules: the relationship between response and time requirements.

Rats were exposed to an interlocking fixed-ratio 150 fixed-interval 5-minute schedule of food reinforcement and then to yoked variable-ratio schedules in which individual ratios corresponded exactly to the ratios of responses to reinforcement obtained on the interlocking schedule. After additional training with the interlocking schedule, the rats were exposed to yoked variable-interval schedules in which intervals corresponded to the intervals between successive reinforcements obtained on the second interlocking schedule. Response rates were highest in the yoked VR condition and lowest in the

Detection and localization of occult lesions with brainstem auditory responses.

Seven vertex-positive potentials--the brainstem auditory response--can be recorded from the human scalp within 10 milliseconds of an appropriate acoustic stimulus. The first of these potentials is generated in the acoustic nerve, the third in the pons, and the fifth in the midbrain. Measurement of the relative latencies and amplitudes of these potentials allowed detection of subclinical lesions in 37 (53 percent) of 70 patients with suspected multiple sclerosis who had no signs or symptoms of brainstem involvement by the disease. Abnormalities in the brainstem auditory response provided the fi

Humoral and cellular immunogenicity of COVID-19 vaccine boosters in participants with advanced HIV disease.

BACKGROUND: People living with HIV (PLWH) who experience advanced immunosuppression are susceptible to severe COVID-19 and demonstrate compromised vaccine responses due to low CD4 counts and uncontrolled HIV viral load. Although vaccine boosters enhance immunity in the general population, their immunogenicity in individuals with advanced HIV remains inadequately characterised. METHODS: This study evaluated the humoral and cellular immunogenicity of COVID-19 vaccine boosters in 41 individuals with advanced HIV at baseline and 4 weeks post-vaccination. Binding antibodies, neutralising antibodie

Investigation of Emotion Regulation Skills in Migraine Patients Using Electroencephalography (EEG) Method.

Emotion regulation is essential for maintaining daily functioning. Previous studies indicate that individuals with migraine have difficulty identifying emotions and exhibit higher alexithymia scores. The P300 and N400 components, associated with attention and semantic processes, provide insights into neural changes during emotion regulation. The aim of this study is to investigate the relationship between migraine and emotion regulation by evaluating the P300 and N400 responses obtained during emotion regulation tasks. Participants included individuals with and without a migraine diagnosis. Th

Responses of neurons in macaque V4 to object and texture images.

Humans and monkeys can rapidly recognize objects in everyday scenes. While it is known that this ability relies on neural computations in the ventral stream of visual cortex, it is not well understood where this computation first arises. Previous work suggests selectivity for object shape first emerges in area V4. To explore the mechanisms of this selectivity, we generated a continuum of images between "scrambled" textures and photographic images of both natural and man-made environments, using techniques that preserve the local statistics of the original image while discarding information abo

AI-assisted decision-making in mild traumatic brain injury.

OBJECTIVE: This study evaluates the potential use of ChatGPT in aiding clinical decision-making for patients with mild traumatic brain injury (TBI) by assessing the quality of responses it generates for clinical care. METHODS: Seventeen mild TBI case scenarios were selected from PubMed Central, and each case was analyzed by GPT-4 (March 21, 2024, version) between April 11 and April 20, 2024. Responses were evaluated by four emergency medicine specialists, who rated the ease of understanding, scientific adequacy, and satisfaction with each response using a 7-point Likert scale. Evaluators were

Polyelectrolyte Multilayer Coatings As a Tunable Tool to Control Inflammatory Response to Biomaterials in Vitro.

The immune response to implanted medical devices can impact their performance and longevity in the body. Instead of designing implants to produce the desired immune responses, this might more easily be achieved with the use of surface modification technology, which can alter the biological profile of the implant without compromising function or performance. In the present study, we generated two polyelectrolyte (PE) multilayer (PEM) coatings from polystyrenesulfonate (PSS) and poly(allylamine hydrochloride) (PAH). These two coatings had identical chemical compositions, but the wettability and

mTOR inhibition impacts the flagellin-augmented inflammatory and antimicrobial response of human airway epithelial cells to Pseudomonas aeruginosa.

OBJECTIVE: The airway epithelium provides a first line of defense against pathogens by release of antimicrobial factors and neutrophil-attracting chemokines. Pseudomonas (P.) aeruginosa, a Gram-negative bacterium that expresses flagellin as an important virulence factor, is a common cause of injurious airway inflammation. The aim of our study was to determine the contribution of flagellin to the inflammatory, antimicrobial, and metabolic responses of the airway epithelium to P. aeruginosa. Furthermore, as we previously showed that targeting mTOR limited the glycolytic and inflammatory response

A nursing-led sepsis response team guiding resuscitation with point-of-care ultrasound: A review and model for improving bundle compliance while individualizing sepsis care.

A dysregulated host response to infection resulting in life-threatening organ dysfunction defines the onset of sepsis. Unfortunately, sepsis is common, costly, and deadly. The Surviving Sepsis Campaign publishes regularly updated, evidence-informed, detection, and treatment guidelines culminating in time-sensitive care "bundles." The goal of these bundles is to expedite sepsis recognition because it is widely held that early treatment is life-saving. Hospitals are mandated to publicly report their bundle compliance, and this will soon be tied to hospital reimbursement. For these reasons, hospi

Distinct response properties between the FFA to faces and the PPA to houses.

INTRODUCTION: The object recognition system involves both selectivity to specific object category and invariance to changes in low-level visual features. Mounting neuroimaging evidence supports that brain areas in the ventral temporal cortex, such as the FFA and PPA, respond preferentially to faces and houses, respectively. However, how regions in human ventral temporal cortex partitioned and functionally organized to selectively and invariantly respond to different object categories remains unclear. What are the changes of response properties at the intersection of adjacent but distinctively-

Label-free multiphoton imaging to assess neoadjuvant therapy responses in breast carcinoma.

Neoadjuvant chemotherapy has been used increasingly in patients with early-stage or locally advanced breast carcinoma, and has been recommended as a general approach in locally advanced-stage diseases. Assessing therapy response could offer prognostic information to help determine subsequent nursing plan; particularly it is essential to identify responders and non-responders for the sake of helping develop follow-up treatment strategies. However, at present, diagnostic accuracy of preoperative clinical examination are still not satisfactory. Here we presented an alternate approach to monitor t

Reward improves response inhibition by enhancing attentional capture.

Reward plays a crucial role in enhancing response inhibition. While it is generally assumed that the process of response inhibition involves attentional capture and the stopping of action, it is unclear whether this reflects a direct impact of reward on response inhibition or rather an indirect mediation via attentional capture. Here, we employed a revised stop-signal task (SST) that separated these two cognitive elements, by including a continue signal that required the same motor response as in go trials, but also attention to a cue, as in stop trials. We first confirmed the engagement of th

Brain responses and approach bias to social alcohol cues and their association with drinking in a social setting in young adult males.

Alcohol is mainly consumed in social settings, in which people often adapt their drinking behaviour to that of others, also called imitation of drinking. Yet, it remains unclear what drives this drinking in a social setting. In this study, we expected to see stronger brain and behavioural responses to social compared to non-social alcohol cues, and these responses to be associated with drinking in a social setting. The sample consisted of 153 beer-drinking males, aged 18-25 years. Brain responses to social alcohol cues were measured during an alcohol cue-exposure task performed in an fMRI scan

CD83 Modulates B Cell Activation and Germinal Center Responses.

CD83 is a maturation marker for dendritic cells. In the B cell lineage, CD83 is expressed especially on activated B cells and on light zone B cells during the germinal center (GC) reaction. The function of CD83 during GC responses is unclear. CD83(-/-) mice have a strong reduction of CD4(+) T cells, which makes it difficult to analyze a functional role of CD83 on B cells during GC responses. Therefore, in the present study we generated a B cell-specific CD83 conditional knockout (CD83 B-cKO) model. CD83 B-cKO B cells show defective upregulation of MHC class II and CD86 expression and impaired
