Intestinal mucus trapping in the rapid expulsion of Trichinella spiralis by rats: induction and expression analyzed by quantitative worm recovery.

Rats were immunized with a Trichinella spiralis infection restricted by chemotherapy to the intestine (the T/M regime) or with a complete infection that resulted in the deposition of muscle larvae. After an oral challenge infection, rapid expulsion could be demonstrated in both groups within 20 min and with 100% recovery of the infectious dose from the stomachs and intestines of infected animals. Immune and nonimmune groups were distinguished by the large numbers of worms in the intestinal lumens of immune rats and large numbers of worms in the intestinal walls in nonimmune rats. Infectious la

Listeriosis in beige mice and their heterozygous littermates.

The ability to resist the facultative intracellular bacterium Listeria monocytogenes was not impaired in the beige mutants of C57BL/6J mice which are known to be deficient in a number of immune functions. The intravenous LD50 of Listeria in beige (bg/bg) mice and their normal heterozygous (bg +) littermates was approximately 5 X 10(5). Growth of Listeria in the spleen and liver during primary and secondary infections was similar in the two groups of mice, and each was able to act efficiently in adoptive transfer of immunity. Histological examination showed a normal accumulation of polymorphonu

The fetal exposome: cellular and epigenetic programming of neonatal immune function.

Fetal life represents a critical period in the development of the neonatal immune system. During this period of high epigenomic plasticity, environmental exposures, maternal health conditions and perinatal factors can have a lifelong impact on gene expression and function, influencing neonatal immune function and contributing to the later development of immune-mediated disease. We conducted a narrative review of human and translational evidence linking antenatal exposures with cellular and epigenetic regulation of neonatal immune function. Evidence was synthesised across maternal health, envir

Quantitative characterization of immune cells by measuring cellular signal transduction pathway activity.

For many diseases, including cancer, infections, and auto-immune diseases, the immune response is a major determinant of disease progression, response to therapy, and clinical outcome. Innate and adaptive immune responses are controlled by coordinated activity of different immune cell types. The functional activity state of immune cells is determined by Signal Transduction Pathways (STPs). A recently developed technology (Simultaneous Transcriptome-based Activity Profiling of Signal Transduction Pathways, STAP-STP) enables simultaneous and quantitative activity measurement of relevant STPs in

Efferent ductule epithelium orchestrates reproductive immune homeostasis under inflammatory stress.

Immune dysregulation is increasingly recognized as a key contributor to male infertility, yet the role of the efferent ductules in reproductive immune homeostasis remains unexplored. Located between the immune-privileged testis and the epididymis, efferent ductules are essential for male fertility by reabsorbing over 90% of testicular fluid and maintaining sperm in suspension through the coordinated activity of reabsorptive primary ciliated cells (RPCCs) and multiciliated cells (MCCs). Using complementary in vivo and in vitro approaches, we investigated the contribution of efferent ductule epi

Immune Dysregulation in Sepsis. A Narrative Review for the Clinicians.

Immune dysregulation presents a significant clinical challenge due to its rapid progression and complex interplay between hyperinflammatory and immunosuppressive responses. Different responses from the innate and adaptive immune systems can result in diseases such as immunoparalysis, cytokine storms, and secondary infections. Current diagnostic methods remain non-specific and time-consuming, delaying targeted interventions. A compartmentalized approach to immune monitoring, distinguishing innate and acquired immune response functional differentiation, is essential for distinguishing between hy

Understanding COVID-19 in children: immune determinants and post-infection conditions.

Coronavirus disease 2019 in children presents with milder clinical manifestations than in adults. On the other hand, the presence of a wide range of inflammatory manifestations, including multisystem inflammatory syndrome in children (MIS-C), in the period after infection suggests a particular susceptibility of some children toward severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). Both protective factors that prevent evolution to severe forms and risk factors for post-infectious conditions are likely to be found in age-related differences in the immune system. The prompt innate res

Immune Functions of Astrocytes in Viral Neuroinfections.

Neuroinfections of the central nervous system (CNS) can be triggered by various pathogens. Viruses are the most widespread and have the potential to induce long-term neurologic symptoms with potentially lethal outcomes. In addition to directly affecting their host cells and inducing immediate changes in a plethora of cellular processes, viral infections of the CNS also trigger an intense immune response. Regulation of the innate immune response in the CNS depends not only on microglia, which are fundamental immune cells of the CNS, but also on astrocytes. These cells align blood vessels and ve

Activating the Antitumor Immune Response in Non-Hodgkin Lymphoma Using Immune Checkpoint Inhibitors.

Non-Hodgkin lymphomas comprise a heterogenous group of disorders which differ in biology. Although response rates are high in some groups, relapsed disease can be difficult to treat, and newer approaches are needed for this patient population. It is increasingly apparent that the immune system plays a significant role in the propagation and survival of malignant cells. Immune checkpoint blocking agents augment cytotoxic activity of the adaptive and innate immune systems and enhance tumor cell killing. Anti-PD-1 and anti-CTLA-4 antibodies have been tested as both single agents and combination t

Influence of host genetic variation on rubella-specific T cell cytokine responses following rubella vaccination.

The variability of immune response modulated by immune response gene polymorphisms is a significant factor in the protective effect of vaccines. We studied the association between cellular (cytokine) immunity and HLA genes among 738 schoolchildren (396 males and 342 females) between the ages of 11 and 19 years, who received two doses of rubella vaccine (Merck). Cytokine secretion levels in response to rubella virus stimulation were determined in PBMC cultures by ELISA. Cell supernatants were assayed for Th1 (IFN-gamma, IL-2, and IL-12p40), Th2 (IL-4, IL-5, and IL-10), and innate/proinflammator

Fusarium pseudograminearum Effector FpSP5 Is Recognized as a PAMP and Also Contributes to Virulence during Infection.

Fusarium pseudograminearum, the dominant causal agent of Fusarium crown rot (FCR), secretes numerous effectors to modulate host immunity during infection, but the functions of most remain unknown. In this study, we identified FpSP5, an effector from F. pseudograminearum, as a pathogen-associated molecular pattern (PAMP) that triggers plant immunity. FpSP5 induces cell death, oxidative burst, and PTI in Nicotiana benthamiana, conferring resistance against multiple pathogens. Furthermore, its perception specifically depends on the coreceptor BAK1 in N. benthamiana. Interestingly, FpSP5 is not on

Anti-inflammatory mechanisms of breast milk exosomes in neonatal immune development.

Breast milk exosomes (ΒΜΕs) are extracellular vesicles of 30-150 nm that have currently attracted research attention due to their protective role in neonatal immune development. Their bioactive cargos, including miRNAs, proteins and lipids, have been shown to facilitate cellular communication and regulate various physiological processes. In the present article, we explore known molecular mechanisms through which ΒΜΕs affect innate and adaptive immunity as well as the integrity of the intestinal epithelial barrier. BMEs can reduce symptoms of inflammatory intestinal diseases, such as necrotizin

Immune cells in liver transplantation: Dual roles, mechanistic insights, and targeted therapeutic strategies.

Liver transplantation is the most effective treatment for end-stage liver disease and acute liver failure; however, organ shortage and transplant rejection remain key challenges limiting its development. In recent years, the unique role of immune cells in liver transplantation has provided new insights into the mechanisms of immune rejection and tolerance. The liver's unique immune microenvironment, dominated by innate immune cells, maintains immune tolerance through complex cell-cell interactions and cytokine networks. This article systematically reviews the dynamic roles of macrophages, neut

The insect effector CSPs activate an atypical plant TNL immune receptor.

CSP-mediated activation of RCSP involves noncanonical structural and catalytic mechanisms-including BB-loop deletion, α5α6-loop insertion, and atypical catalytic residues within the RCSP-TIR domain. Chemosensory proteins (CSPs), a conserved effector family in phloem-feeding insects such as aphids and the brown planthopper (Nilaparvata lugens), induce dwarfism in Nicotiana benthamiana through recognition by the Toll-interleukin-1 receptor (TIR) domain-containing nucleotide-binding leucine-rich repeat receptor (TNL) RCSP. However, CSP-mediated RCSP activation and enzymatic mechanism remain unkno

Role of E5 from HPV16 in the Evasion of the Immune Response.

Human papillomavirus type 16 (HPV16) persistence relies on early viral mechanisms that synchronize oncogenic signaling with immune evasion, with the E5 oncoprotein serving as a central regulator of the viral cycle and the initiation of cell transformation. This review integrates current evidence on how E5 reconfigures host cell dynamics: first, by hijacking signaling pathways such as EGFR, MAPK/ERK, and PI3K/AKT to drive keratinocyte proliferation and survival; and second, by organizing a multi-step immune evasion strategy. We detail how E5 suppresses innate antiviral responses, specifically b

Extracellular vesicles in malaria pathogenesis and Therapy: Emerging insights and future perspectives.

Malaria remains a major global health challenge, responsible for millions of cases and substantial mortality each year. The emergence of drug-resistant strains and the lack of an effective vaccine underscore the urgent need for novel therapeutic strategies. Recent research has highlighted the pivotal role of extracellular vesicles (EVs), particularly small EVs derived via the endosomal pathway-commonly known as exosomes-in the progression of malaria and modulation of immune responses. These exosomes carry bioactive molecules that influence both innate and adaptive immunity. The biogenesis of e

Segmented regression modeling of a phytogenic blend on growth and immune function in broilers: a dose-response study.

This study investigated the dose-dependent effects of a phytogenic feed additive, on growth performance, immune response, hematology, and organ development in broiler chickens. A total of 300-day-old broiler male chicks (Ross 308) with the average weight of 41.2 ± 2.32 g were randomly allocated into 4 treatment groups (0.0, 0.25, 0.50, and 1.00 mL/L phytogenic blend in drinking water) with 5 replications per groups (15 birds per pen) in a completely randomized design. Growth performance showed a clear bell-shaped response, with the 0.50 mL/L dose optimizing feed intake, body weight gain, and f
