Imidacloprid exposure is associated with brown adipose tissue impairment in high-fat diet-fed C57BL/6 mice.

Imidacloprid, a broad-spectrum neonicotinoid insecticide widely used in agriculture, may impair mitochondria-rich brown adipose tissue (BAT), increase the risk of obesity, and alter systemic metabolic homeostasis. This study investigated the obesogenic effects of imidacloprid in a mouse model. The results revealed that imidacloprid intervention resulted in increased body weight, impaired glucose tolerance, altered lipid profiles, and a reduced mitochondrial membrane potential in brown adipocytes, particularly upon high-fat diet. Additionally, analysis using the Seahorse XF24 Analyzer also indi

Dermal exposure represents a major pathway of chlorpyrifos uptake in insectivorous bats: Insights from a toxicokinetic model.

Bats help to control populations of agricultural pests by eating large amounts of insects; however, foraging in croplands can present a threat to bats through exposure to pesticides via contaminated prey or through their skin and lungs. Although exposure to pesticides is known to occur in bats, it is unclear which routes predominate, and how toxic compounds are distributed among different tissues after being taken up from the environment. A non-invasive approach to evaluating the risk of exposure is the use of toxicokinetic modeling. Here, we developed a physiologically-based toxicokinetic mod

Transforming agricultural waste into intelligent biopesticide delivery: soil-disintegrable tea saponin microcapsules for sustainable crop protection.

BACKGROUND: In order to achieve intelligent and sustainable delivery of tea tree oil (TTO) for tea pest control, we developed a soil-disintegrable, pH-responsive microcapsule system (TMS) with an average particle size of 2.05 μm using 15.6% tea saponin as the shell precursor via interfacial polymerization. RESULTS: Optimized TMS (1-2 μm) achieved 97.4% encapsulation efficiency. The system exhibited accelerated TTO release under acidic conditions (78.8% at pH 5.0), enabling targeted delivery to fungal infection sites. It demonstrated superior foliar adhesion and rainfastness (84.5% retention)

Biosynthesis and heterologous production of the α-agarofuran scaffold of Celangulin V from Celastrus angulatus.

Celangulin V is a widely used biopesticide derived from Celastrus angulatus, and features antifeedant and insecticidal properties as a dihydro-β-agarofuran (DHβAF) sesquiterpenoid. Its biosynthesis remains largely unexplored. Here, we assemble a chromosome-level and haplotype-resolved reference genome of C. angulatus, with each haplotype assembled into 23 pseudochromosomes and achieving scaffold N50 of 14.31 and 14.01 Mb, respectively. This high-quality genome reveals that a recent β whole-genome triplication (β-WGT) event occurred ~34.3 million years ago, and that the expansion of sesquiterpe

Effect of the neonicotinoid acetamiprid on molecular events during in vitro boar spermatozoa capacitation.

Acetamiprid (Ace) is a widely used neonicotinoid insecticide whose intensive application in agriculture has raised concerns about its potential effects on the reproductive health of animals and humans. Successful fertilization depends on the integrity of the plasma membrane (PM) of the spermatozoa and the proper progression of the series of events involved in capacitation. The aim of this study was to evaluate the effects of Ace on the capacitation of boar spermatozoa and several associated functional parameters: membrane carbohydrate redistribution, membrane microdomains (MDM), cholesterol ef

Entomopathogenic Fungi as Orchestrators of Plant Defense, Growth, and Stress Tolerance-A Comprehensive Review.

Entomopathogenic fungi (EPF), including Beauveria spp., Metarhizium spp., Lecanicillium spp., and Isaria spp., help manage pest populations through direct infestation and insecticidal toxin production. Beyond pest control, they enhance plant growth and overall health, which makes them essential components of sustainable agriculture. EPF orchestrates systemic acquired resistance (SAR) and induced systemic resistance (ISR) via salicylic acid (SA) and jasmonic acid (JA) signalling pathways, respectively. Through these mechanisms, EPF promotes plant resilience against both biotic (pathogens and pe

Receptor-mediated membrane insertion of Cry1Ac triggers cell lysis and oligomeric state remodeling.

The precise insecticidal mechanism of Cry toxins remains elusive, and the structural architecture of their transmembrane pores has yet to be elucidated. In this study, we constructed a C-terminally truncated Cry1Ac mutant, termed Cry1AcNC, which retains the N-terminal domain and toxin core. Using negative-stain electron microscopy, we visualized a tetrameric pore-ring structure formed by Cry1AcNC. To examine the functional role of helix flexibility in pore formation, we introduced disulfide bonds between the α-1 and α-2 helices to restrict their mobility. Cytotoxicity assays using Sf9 cells he

Toxicological effects of a difenoconazole fungicide on a non-target butterfly.

Agricultural intensification, driven by increasing food demand, is one of the main drivers of global biodiversity loss. Pesticides, including fungicides, may increase crop yield but pose substantial threats to the environment. However, our current knowledge on the effects of fungicides on biodiversity suffers from less research interest compared to other pesticides and a neglect of insect herbivores. We here investigate the effects of a commercially available, ready-to-use difenoconazole fungicide on a non-target insect, the butterfly Pieris napi, under controlled laboratory conditions. When b

Effects of quercetin in male rats' testis after consumption of permethrin: a transmission electron microscopy (TEM) study.

Permethrin, a widely used pyrethroid insecticide, impairs male reproductive function through oxidative stress and endocrine disruption. This study evaluated the protective effects of quercetin against permethrin-induced testicular toxicity in adult male Wistar rats. Thirty rats were divided into five groups (n = 6): control, DMSO vehicle, permethrin (35 mg/kg/day orally), permethrin + quercetin (15 mg/kg/day orally), and quercetin alone, for 60 days. Permethrin exposure induced severe ultrastructural damage including mitochondrial swelling, nuclear chromatin condensation, and Sertoli cell deta

Alpha- and beta-pinene alter the morphology of oocytes in Rhipicephalus microplus ticks (Acari: Ixodidae).

Pinenes are bicyclic monoterpenes that contain two constitutional isomers, alpha- and beta-pinene. Both exhibit a wide range of biological activities, including fungicidal, insecticidal, antimicrobial and antibacterial effects, and are found in the essential oil of numerous plant species with acaricidal properties. Thus, the objective of this research was to evaluate the effect of sublethal concentrations of alpha- and beta-pinene, diluted in 2 % Tween 80, on the morphology of the ovary of semi-engorged Rhipicephalus microplus female ticks. For this, the ticks were immersed in different concen

Mitochondria-targeted therapy with metformin and MitoQ reduces oxidative stress, improves mitochondrial function, and restores metabolic homeostasis in a murine model of Gulf War Illness.

Gulf War Illness (GWI) is a cluster of medically unexplained chronic symptoms, including neurological and gastrointestinal impairments, and muscle fatigue, suffered by veterans of the Persian Gulf War. A GWI model in C57BL/6 mice exposed to the nerve gas prophylactic pyridostigmine-bromide (PB) and the insecticide permethrin (PER) was used to test the effect of mitochondria-potentiating agents, metformin and MitoQ on chronic fatigue, observed in GWI. The exposure of mice to PB/PER resulted in enhanced oxidative stress, impaired mitochondrial function, and reduced autophagy. Treatment with the

High throughput screening and metabolic engineering of Saccharopolyspora spinosa for spinosad production.

The macrolide antibiotics spinosad, synthesized by Saccharopolyspora spinosa, is a highly effective, environmentally-friendly insecticide. However, due to poor fermentation performance and difficulties in engineering S. spinosa strains, the production cost of spinosad is still high, which restricts its industrial application. The industrial strains used for antibiotic production primarily originate from mutagenic screening, whereas traditional screening methods are time-consuming and laborious. Here, an in vitro detection method for spinosad was established to accelerate the breeding of mutate

Semi-field studies on biochemical markers of honey bee workers (Apis mellifera) after exposure to pesticides and their mixtures.

Due to the fact that many different pesticides are used in crop production and their residues can accumulate in the environment, bees are in contact with various pesticides at the same time. Most studies on their influence on honey bees focus on single substances in concentrations higher than those found in the environment. Our study assessed the chronic effects of commonly used pesticides and their mixtures on selected biochemical markers in worker bee hemolymph. Workers developed in the hive and were provisioned with to pesticides in concentrations corresponding to residues detected in polle

A comparative genotoxicity study of agrochemicals: nuclear abnormalities, comet assay, and gene expression alterations.

Agrochemicals (AGs) are known for their ability to have a negative impact on the health of non-target species, despite the fact that they are meant to protect agricultural plants from harmful pests. Catla catla (Hamilton, 1822) gill cells (ICG) were exposed to four AGs: insecticide (Imidacloprid (IMI)), fungicide (Curzate (CZ)), herbicide (pyrazosulfuron ethyl (PE)), and fertilizer micronutrients (MN) with sublethal concentrations 1/20th, 1/10th, and 1/5th of IC50, described here as low dose (LD), medium dose (MD), and high dose (HD), respectively. A rapid and simple micro-assay to assess catalase activity in individual mosquito tissues.

Oxidative stress generated as a normal byproduct of aerobic metabolism is minimized by the enzyme catalase (CAT; EC 1.11.1.6), which reduces hydrogen peroxide to molecular oxygen and water. In various mosquitoes, hydrogen peroxide and/or CAT activity have been implicated in oxidative responses to viral and protozoal pathogens as well as in ovarian maturation and insecticide resistance. We combined features of various CAT assays to develop a simple micro-assay that enables comparison of enzyme activities in individual mosquito tissues on a microscope slide. Activity recovered in the supernatant
