Cow's milk protein enteropathy is a symptom complex that composed of severe diarrhoea and malnutrition. This disorder is caused by non-immunoglobulin E-mediated food allergy. Its clinical features and natural course have been explained in many reports, of different types of cow's milk and soy reactions. In the present article, we describe a newborn patient who presented with chronic diarrhoea and failure to thrive diagnosed as cow's milk protein enteropathy. The duodenal biopsy revealed granulomatous duodenitis which has not been described before. Her clinical and pathological findings respond

The small G protein Rheb (Ras homologue enriched in brain) is known to promote mammalian target of rapamycin (mTOR) signaling. In this study, we show that Rheb like-1 protein (RhebL1) rescues mTOR signaling during nutrient withdrawal and that tuberous sclerosis complex-1 (TSC) and TSC2 impairs RhebL1-mediated signaling through mTOR. We identify critical residues within the switch I region (N41) and 'constitutive' effector (Ec) region (Y/F54 and L56) of Rheb and RhebL1, which are required for their efficient activation of mTOR signaling. Mutation of Rheb and RhebL1 at N41 impaired their interac

Psoriasis vulgaris is one of the most prevalent T cell-mediated inflammatory diseases in humans. It is multifactorial in origin and shows polygenic inheritance. Systemic immunosuppressive therapies play an important role in management of severe disease cases but are associated with variable response and toxicity. Within the last decade there have been significant advances in our understanding of the genetic basis of this complex disease and polymorphic candidate genes have been proposed. Susceptibility gene characterization together with an explosion in knowledge of its primary immune basis wi

Polyclonal hypergammaglobulinemia is a characteristic of chronic inflammatory conditions, including persisting viral infections and autoimmune diseases. Here we have studied hypergammaglobulinemia in mice infected with lymphocytic choriomeningitis virus (LCMV), which induces nonspecific immunoglobulins as a result of switching natural IgM specificities to IgG. The process is dependent on help from CD4+ T cells that specifically recognize LCMV peptides presented by B cells on major histocompatibility complex class II molecules. Thus, hypergammaglobulinemia may arise when specific helper T cells

Reactive angioendotheliomatosis (RA) is a rare self-limited skin condition characterized histopathologically by a proliferation of endothelial cells within vascular lumina, usually as a result of different stimuli such as systemic infections, cryoproteinemias, monoclonal gammopathies, allergic conditions, severe peripheral vascular atherosclerotic disease, and iatrogenic arteriovenous fistulas. We report on a 67-year-old woman with a 20-year history of seropositive rheumatoid arthritis who presented with violaceous swelling of her left forearm. A skin biopsy revealed the histopathologic findin

Primary biliary cirrhosis (PBC) has been classified as an autoimmune disease. It is characterized by a multitude of immune-mediated symptoms and phenomena. Humoral autoimmunity directed against mitochondrial auto-antigens or structures cross-reactive with these proteins represents the basis of one of the most specific tests in any autoimmune disease: the detection of auto-antibodies against pyruvate dehydrogenase. The molecular cloning and expression of antigens of the ketoacid dehydrogenase complex has led to the establishment of highly specific and reliable testing systems for anti-mitochond

The human deafness dystonia syndrome results from the mutation of a protein (DDP) of unknown function. We show now that DDP is a mitochondrial protein and similar to five small proteins (Tim8p, Tim9p, Tim10p, Tim12p, and Tim13p) of the yeast mitochondrial intermembrane space. Tim9p, Tim10p, and Tim12p mediate the import of metabolite transporters from the cytoplasm into the mitochondrial inner membrane and interact structurally and functionally with Tim8p and Tim13p. DDP is most similar to Tim8p. Tim8p exists as a soluble 70-kDa complex with Tim13p and Tim9p, and deletion of Tim8p is synthetic

Proteolytic activation of receptor-bound protective antigen (PA) at the cell surface removes PA20, allowing PA63 to oligomerize and form a ring-shaped heptameric prepore. The prepore binds edema factor (EF) and lethal factor (LF) and, after endocytosis and trafficking of the complex to an acidic, vesicular compartment, it undergoes membrane insertion and mediates translocation of EF/LF to the cytosol. Data from membrane conductance experiments support a model of membrane insertion in which the 2beta2-2beta3 loop of PA, which is disordered in native PA and the prepore, forms a 14-stranded trans

The classification of immune-mediated demyelinating polyneuropathies has become more complex in recent years. Initial definitions of chronic inflammatory demyelinating polyneuropathy (CIDP) were often broad enough to include almost any acquired polyneuropathy with demyelinating features. However, subdivision of acquired demyelinating polyneuropathies into different categories seems justified because 1) several distinct clinical syndromes have been identified, 2) characteristic and distinctive patterns of serum antibody binding are associated with each specific clinical syndrome and, 3) the syn

Therapeutic effectiveness of ursodeoxycholic acid (UDCA) for primary biliary cirrhosis strongly indicates that UDCA possesses immunomodulatory activities. In order to further investigate mechanical background of such UDCA action, we first asked whether UDCA modulates glucocorticoid-mediated signal transduction. Using electrophoretic mobility-shift assay, we demonstrated that treatment with UDCA promoted the specific complex formation between the cytosol protein and the glucocorticoid-response element DNA in a dose-dependent fashion in vitro, and also nuclear translocation of the glucocorticoid

Activation of transducin-GDP by NaF is mainly mediated by aluminofluorde or beryllofluoride complexes acting as GTP gamma-phosphate analogs. In millimolar magnesium, NaF at concentrations above 3 mM is active even in the absence of aluminium or beryllium. This activation has a Hill coefficient of 3 with respect to F-, and its rate is linear with respect to Mg2+ concentrations above 2 mM. Upon fluoride dilution, inactivation rate is hundreds of times faster than for aluminofluoride-activated T alpha GDP. We propose that at high NaF concentrations, 3 hydrogen-bonded fluorides in the gamma-phosph

Murine A strain (KkDdLd) sarcoma I (SaI) tumor cells have been transfected with a cloned H-2Kb gene. The resulting clones (SKB clones) stably express high levels of a molecule that is serologically and biochemically indistinguishable from the H-2Kb antigen. SKB clones are not susceptible to cytotoxic T lymphocyte-mediated lysis by H-2Kb-specific bulk, cloned, or H-2Kb-restricted lymphocytic choriomeningitis virus-specific effectors. Survival times of A/J and B10.A mice challenged i.p. with the H-2Kb-expressing transfectants and the parental SaI cells are similar, suggesting that the presence o

In addition to an earlier review [Z. ges. inn. Med. 35 (1980) 58] newer concepts in the pathogenesis of autoimmune diseases are presented. Acantholysis in pemphigus is caused by proteases released from cells of stratified squamous epithelium stimulated by antibodies to cell membrane antigens. The dermoepidermal cleavage in pemphigoid is presumably due to proteases from granulocytes accumulating at the dermoepidermal junction by antibody-mediated complement activation. Receptor autoantibodies can directly stimulate or block receptor function, increase cell membrane receptor degradation, or acti

Cutaneous vasculitis may present as an urticaria-like eruption in association with viral illness, systemic lupus erythematosus, Sjögren's syndrome, or serum sickness. Often, however, urticarial vasculitis presents as an immune complex-mediated disease without other associated connective-tissue disease or associated infection. Many forms of therapy have been tried, including antihistamines, prednisone, and immunosuppressive drugs without success in every patient. Because colchicine had been effective in some patients with non-urticarial cutaneous vasculitis, we decided to use it in two patients

Using antagonists with selectivity for the delta (ICI 154,129) and mu (beta-funaltrexamine) binding sites, evidence was obtained to indicate that delta receptors within the brain mediate the endogenous opioid component of endotoxic hypotension. The therapeutic actions of intravenous ICI 154,129 were dose related, with effective doses between 15-60 mg/kg. Evidence for a functional interaction between mu and delta binding sites was obtained: prior occupancy of the mu binding site by beta-funaltrexamine prevented the usual therapeutic response to the delta antagonist ICI 154,129 in endotoxemic ra

Clinical fever seems clearly associated with those diseases in which inflammation, in one form or other, plays a dominant role. The actual contribution of the various listed factors to the febrile response associated with any specific disease may, of course, be complex. For instance in a gram-positive infection, inflammatory cells may be activated to produce endogenous pyrogen at the site of bacterial multiplication by any or all of the following mechanisms, most of which have been demonstrated experimentally: phagocytosis of bacteria, chemotactic factors produced by bacteria or released from

Rabbits infected with Trypanosoma congolense were tested for immediate and delayed-type hypersensitivity responses to parasite antigens. Positive skin-test reactions were exclusively of the Arthus type, as shown by sequential histological analyses which revealed that more than 90% of the infiltrating cells at 24 hr postinjection were polymorphonuclear neutrophils. Skin reactions could be elicited in normal rabbits by intradermal injection of trypanosome antigen-antibody complexes. The absence of cell-mediated hypersensitivity to trypanosome antigens in infected rabbits was demonstrated by skin

UNLABELLED: The global spread of multidrug-resistant tuberculosis (MDR-TB) remains a major challenge to tuberculosis control programs, particularly in resource-limited settings where access to rapid molecular drug-susceptibility testing is limited. Resistance to isoniazid and rifampicin is most commonly associated with mutations in the katG and rpoB genes, respectively, making these loci valuable molecular markers for early MDR-TB screening. In this study, we developed a dual loop-mediated isothermal amplification assay coupled with lateral flow detection for the simultaneous identification of

BACKGROUND: Despite advances in small-molecule inhibitors (SMIs), the clinical outcomes for glioblastoma (GBM) remain bleak. Recently, polymerase I and transcript release factor (PTRF/Cavin1) has emerged as a promising therapeutic target, with its inhibitor EPIC-1042 demonstrating preclinical antitumor activity. However, the therapeutic limitations of SMIs necessitate alternative strategies to achieve enduring target suppression.
METHODS: EPIC-0726, a proteolysis-targeting chimera (PROTAC) degrader of PTRF, was developed through computer-aided drug design (CADD). Target engagement and degrada
