MicroRNA-mediated responses to magnesium-induced stress coordinate target genes involved in porphyrin and biosynthesis of secondary metabolites metabolisms in mulberry (Morus alba L.).

BACKGROUND: MicroRNAs (miRNAs) play essential regulatory roles in magnesium (Mg) imbalance by targeting key defense genes for cleavage or translational repression. miRNA targets in response to Mg deficiency and toxicity remain uncharacterized in mulberry (Morus alba) in Mg-induced stress. We, for the first time, investigated Mg-stress-responsive miRNAs and their targets in mulberry leaves using transcriptome-wide sequencing to identify miRNAs putatively involved in Mg-stress tolerance. In this study, Mg (MgSO4) treatment applied in six concentration ranging from 0 mM/L (T0) as Mg deficiency, 1

microRNA miR-31-5p confers protection to melanin-deficient vitiligo keratinocytes against UV-B induced DNA damage by activating autophagy machinery.

Despite lacking melanin, vitiligo skin paradoxically exhibits a lower incidence of non-melanoma skin cancers, indicating alternative epidermal defense mechanisms. We investigated the protective role of microRNA-31-5p (miR-31-5p), which is elevated in vitiligo lesions, against UV-B-induced DNA damage in keratinocytes. Primary human keratinocytes exposed to UV-B were treated with miR-31-5p mimic or antagomiR. miR-31-5p mimic significantly reduced DNA damage and increased the expression of nucleotide excision repair genes (DDB1, ERCC2, XPC), while alleviating replication stress. Longitudinal and Stability-Aware Analysis Reveals Treatment-Specific MicroRNA Response Signatures Following Immune-Reconstitution and B-Cell-Targeted Therapies in Multiple Sclerosis.

Disease-modifying therapies (DMT)s) for relapsing-remitting multiple sclerosis (RRMS) act through distinct immunological mechanisms, yet the within-patient molecular response programs associated with these therapies remain incompletely defined. Here, we reanalyzed publicly available peripheral blood mononuclear cell (PBMC) miRNA microarray data (GSE230064) using a longitudinal, robustness-focused framework to compare therapy-associated miRNA response patterns following cladribine versus ocrelizumab treatment. Baseline (t0) and 6-month post-treatment (t1) samples were paired within individuals

Alterations of Inflammatory Mediators (TNF-α and HMGB1) and Circulating microRNA-22 Expression in Pre- and Post-Menopausal Women with Early-Stage Epithelial Ovarian Cancer.

BACKGROUND: Ovarian cancer is an important cause of cancer-related mortality among women, disease severity increases due to hormonal changes, which modify the ovarian environment and may contribute to the development of ovarian cancer (OC). Inflammatory mediators play a central role in the enhancement of OC progression. Several microRNA play central role in OC; microRNA-22 (miR-22) is a biomarker associated with hormonal status regarding menopause and the development of ovarian cancer. Evaluating these biomarkers and assess the interaction among them, may contribute to a deeper understanding o

MicroRNA Expression Profiles Before and After Neoadjuvant Chemotherapy in Breast Cancer: Correlations with Molecular Subtypes, Pathological Response, and Clinical Timing-A Pilot Translational Study.

Neoadjuvant chemotherapy (NAC) is the standard of care for locally advanced breast cancer, yet the molecular predictors of pathological response remain incompletely defined, particularly regarding microRNA (miRNA) dynamics. We investigated paired pre- and post-NAC miRNA expression profiles in relation to molecular subtype, residual cancer burden (RCB), and clinical timing parameters. Seven patients with invasive breast cancer (Luminal A n = 3, Luminal B n = 1, TNBC n = 2, HER2+ n = 1) who received NAC (AC-T or TCHP) were included in this pilot study. Extracellular Vesicle-Associated MicroRNA-25-3p Derived from Pancreatic Cancer Cells Promotes Hepatic Stellate Cell Activation and Enhances Cancer Progression.

Pancreatic cancer is a highly aggressive malignancy with a poor prognosis, mainly due to late diagnosis and early metastatic spread. The underlying mechanisms of pancreatic cancer metastasis, particularly the role of hepatic stellate cell (HSC) activation, are not fully understood. This study tried to find potential biomarkers for pancreatic cancer progression and prognosis by analyzing extracellular vesicle (EV)-associated miRNA profiles in plasma from pancreatic cancer patients and non-cancer controls. Functional assays, including transfection, western blotting, immunofluorescence, and enzym

The microRNA Expression in Crypt-Top and Crypt-Bottom Colonic Epithelial Cell Populations Demonstrates Cell-Type Specificity and Correlates with Endoscopic Activity in Ulcerative Colitis.

BACKGROUND AND AIMS: Colonic epithelial barrier dysfunction is one of the early events in ulcerative colitis [UC], and microRNAs [miRNAs] participate in its regulation. However, the cell type-specific miRNome during UC remains unknown. Thus, we aimed to explore miRNA expression patterns in colon tissue and epithelial cells during active and quiescent UC. METHODS: Small RNA-sequencing in colon tissue, crypt-bottom [CD44+], and crypt-top [CD66a+] colonic epithelial cells from two cohorts of UC patients [n = 74] and healthy individuals [n = 50] was performed. Data analysis encompassed differenti

Regulatory axis of circular RNA DTNB, microRNA-485-5p, and myeloid cell leukemia 1 attenuates inflammation and apoptosis in caerulein-treated AR42J cells.

Acute pancreatitis (AP) is an inflammatory disease of the pancreas and the main cause of hospital admissions for gastrointestinal diseases. Here, the work studied the circular RNA DTNB/microRNA-485-5p/MCL1 axis in AP and hoped to unravel the related mechanism. Caerulein exposure replicated an AP model in AR42J cells, and caerulein-mediated expression of circDTNB, miR-485-5p, and MCL1 was recorded. After exposure, cells were intervened with transfection plasmids and tested for LDH release, apoptosis, and inflammation. To determine the interwork of circDTNB, miR-485-5p, and MCL1, prediction resu

Targeting microRNA-190a halts the persistent myofibroblast activation and oxidative stress accumulation through upregulation of Krüppel-like factor 15 in oral submucous fibrosis.

BACKGROUND/PURPOSE: Oral submucous fibrosis (OSF) is a condition characterized by inflammation and excessive collagen deposition, which has been identified as a potentially malignant disorder. Recently, several microRNAs (miRNAs) have been shown to be implicated in various disorders associated with fibrosis. However, how these miRNAs modulate OSF development is poorly understood. Therefore, the study aimed to identify the specific miRNAs that contribute to the progression of OSF and to investigate their molecular mechanisms in promoting fibrosis. MATERIALS AND METHODS: The expression and clin

MicroRNA-194-5p/Heparin-binding EGF-like growth factor signaling mediates dexamethasone-induced activation of pseudorabies virus in rat pheochromocytoma cells.

Pseudorabies virus (PRV) is a neurotropic virus, which infects a wide range of mammals. The activity of PRV is gradually suppressed in hosts that have tolerated the primary infection. Increased glucocorticoid levels resulting from stressful stimuli overcome repression of PRV activity. However, the host cell mechanism involved in the activation processes under stressful conditions remains unclear. In this study, infection of rat PC-12 pheochromocytoma cells with neuronal properties using PRV at a multiplicity of infection (MOI) = 1 for 24 h made the activity of PRV be the relatively repressed s

Stratification of acute myocardial and endothelial cell injury, salvage index and final infarct size by systematic microRNA profiling in acute ST-elevation myocardial infarction.

AIM: Acute injury and subsequent remodelling responses to ST-segment elevation myocardial infarction (STEMI) are major determinants of clinical outcome. Current imaging and plasma biomarkers provide delayed readouts of myocardial injury and recovery. Here, we sought to systematically characterize all microRNAs (miRs) released during the acute phase of STEMI and relate miR release to magnetic resonance imaging (MRI) findings to predict acute and late responses to STEMI, from a single early blood sample. METHODS AND RESULTS: miRs were quantified in blood samples obtained from patients after pri

Eupatilin ameliorates postmenopausal osteoporosis via elevating microRNA-211-5p and repressing Janus kinase 2/Signal transducer and activator of transcription 3 pathway.

This study explored the effect of Eupatilin on postmenopausal osteoporosis and explored the mechanisms associated with miR-211-5p. First, the rats were given intragastric administration of Eupatilin every day and subcutaneously injected once a week with oligonucleotides or plasmids that interfered with the expression of miR-211-5p or Janus kinase 2 (JAK2). After 4 weeks, a rat model of osteoporosis was established. Then, serum alkaline phosphatase, calcium and phosphorus levels were detected, as well as femur bone mineral density and biomechanical parameters. HE staining and Masson staining we

MicroRNA-122-5p Inhibition Improves Inflammation and Oxidative Stress Damage in Dietary-Induced Non-alcoholic Fatty Liver Disease Through Targeting FOXO3.

Misregulated microRNA network has been emerging as the main regulator in non-alcoholic fatty liver disease (NAFLD). The deregulation of miR-122-5p is associated with the liver disease. However, the specific role and molecular mechanism of miR-122-5p in NAFLD remain unclear. In this study, we have reported that the high-fat diet (HFD) or palmitic acid (PA) significantly upregulated the hepatic miR-122-5p expression in vivo and in vitro. Inhibition of miR-122-5p suppressed accumulation-induced inflammation of lipids and oxidative stress damage in PA-treated L02 cells and HFD-induced fatty liver.

MicroRNA Profiling Shows a Time-Dependent Regulation within the First 2 Months Post-Birth and after Mild Neonatal Hypoxia in the Hippocampus from Mice.

Brain development occurs until adulthood, with time-sensitive processes happening during embryo development, childhood, and puberty. During early life and childhood, dynamic changes in the brain are critical for physiological brain maturation, and these changes are tightly regulated by the expression of specific regulatory genetic elements. Early life insults, such as hypoxia, can alter the course of brain maturation, resulting in lifelong neurodevelopmental conditions. MicroRNAs are small non-coding RNAs, which regulate and coordinate gene expression. It is estimated that one single microRNA

Extracellular vesicles derived from cancer-associated fibroblasts carry tumor-promotive microRNA-1228-3p to enhance the resistance of hepatocellular carcinoma cells to sorafenib.

Cancer-associated fibroblasts (CAFs)-derived extracellular vesicles (EVs) can promote tumor progression by delivering microRNA (miRNA). Whether EVs could transfer miR-1228-3p into hepatocellular carcinoma (HCC) cells to affect chemoresistance was discussed in this study. Normal fibroblasts (NFs) and CAFs were isolated from tissue samples of HCC patients. We assessed the functions of HCC cells after co-culturing with NFs and CAFs. miR-1228-3p gain-of-function experiments were conducted. Next, functional assays were carried out to determine the binding of miR-1228-3p to placenta associated 8 (PL

Inhibition of microRNA-448 suppresses CD4+ T cell inflammatory activation via up-regulating suppressor of cytokine signaling 5 in systemic lupus erythematosus.

Systemic lupus erythematosus (SLE) is a chronic autoimmune disease. MicroRNA-448 (miR-448) has a pro-inflammatory effect in various inflammation-related diseases and is up-regulated in serum of patients with SLE. However, the role of miR-448 in SLE development remains elusive. In our study, we found high expression of miR-448 in peripheral blood mononuclear cells (PBMCs) of SLE patients, and miR-448 level was positively associated with disease severity. Besides, miR-448 level was up-regulated during the growth of MRL/lpr mice. To investigate the function of miR-448 in SLE, we subjected 8-week
