Structure-based modeling of critical micelle concentration (CMC) of anionic surfactants in brine using intelligent methods.

Critical micelle concentration (CMC) is one of the main physico-chemical properties of surface-active agents, also known as surfactants, with diverse theoretical and industrial applications. It is influenced by basic parameters such as temperature, pH, salinity, and the chemical structure of surfactants. Most studies have only estimated CMC at fixed conditions based on the surfactant's chemical parameters. In the present study, we aimed to develop a set of novel and applicable models for estimating CMC of well-known anionic surfactants by considering both the molecular properties of surfactant

Focus on the road to modelling cardiomyopathy in muscular dystrophy.

Alterations in the DMD gene, which codes for the protein dystrophin, cause forms of dystrophinopathies such as Duchenne muscular dystrophy, an X-linked disease. Cardiomyopathy linked to DMD mutations is becoming the leading cause of death in patients with dystrophinopathy. Since phenotypic pathophysiological mechanisms are not fully understood, the improvement and development of new disease models, considering their relative advantages and disadvantages, is essential. The application of genetic engineering approaches on induced pluripotent stem cells, such as gene-editing technology, enables t

Cardiometabolic Syndrome: An Update on Available Mouse Models.

Cardiometabolic syndrome (CMS), a disease entity characterized by abdominal obesity, insulin resistance (IR), hypertension, and hyperlipidemia, is a global epidemic with approximately 25% prevalence in adults globally. CMS is associated with increased risk for cardiovascular disease (CVD) and development of diabetes. Due to its multifactorial etiology, the development of several animal models to simulate CMS has contributed significantly to the elucidation of the disease pathophysiology and the design of therapies. In this review we aimed to present the most common mouse models used in the res

Microtubule-associated proteins (MAPs) in microtubule cytoskeletal dynamics and spermatogenesis.

The microtubule (MT) cytoskeleton in Sertoli cells, a crucial cellular structure in the seminiferous epithelium of adult mammalian testes that supports spermatogenesis, was studied morphologically decades ago. However, its biology, in particular the involving regulatory biomolecules and the underlying mechanism(s) in modulating MT dynamics, are only beginning to be revealed in recent years. This lack of studies in delineating the biology of MT cytoskeletal dynamics undermines other studies in the field, in particular the plausible therapeutic treatment and management of male infertility and fe

The role of oligodendrocyte gap junctions in neuroinflammation.

Gap junctions (GJs) provide channels for direct cell-to-cell connectivity serving the homeostasis in several organs of vertebrates including the central (CNS) and peripheral (PNS) nervous systems. GJs are composed of connexins (Cx), which show a highly distinct cellular and subcellular expression pattern. Oligodendrocytes, the myelinating cells of the CNS, are characterized by extensive GJ connectivity with each other as well as with astrocytes. The main oligodendrocyte connexins forming these GJ channels are Cx47 and Cx32. The importance of these channels has been highlighted by the discovery

Irritability in Huntington's Disease: Factor Analysis of Snaith's Irritability Scale.

BACKGROUND: Elevated levels of irritability are reported to occur in a number of neurological conditions, including Huntington's disease (HD), a genetic neurodegenerative disorder. Snaith's Irritability Scale (SIS) is used within HD research, but no psychometric evaluation of this instrument has previously been undertaken. Therefore, the current study aimed to analyze the factor structure of this scale among an HD population. METHODS: Exploratory and confirmatory factor analysis were used to examine the structural properties of SIS using responses from 1,264 HD gene expansion carriers, across

Transgenic animal models of neurodegenerative diseases and their application to treatment development.

Neurodegenerative disorders of the aging population affect over 5 million people in the US and Europe alone. The common feature is the progressive accumulation of misfolded proteins with the formation of toxic oligomers. Previous studies show that while in Alzheimer's disease (AD) misfolded amyloid-beta protein accumulates both in the intracellular and extracellular space, in Lewy body disease (LBD), Parkinson's disease (PD), Multiple System Atrophy (MSA), Fronto-Temporal dementia (FTD), prion diseases, amyotrophic lateral sclerosis (ALS) and trinucleotide repeat disorders (TNRD), the aggregat

Mathematical prediction in infection.

It is now increasingly common for infectious disease epidemics to be analysed with mathematical models. Modelling is possible because epidemics involve relatively simple processes occurring within large populations of individuals. Modelling aims to explain and predict trends in disease incidence, prevalence, morbidity or mortality. Models give important insight into the development of epidemics. Following disease establishment, epidemic growth is approximately exponential. The rate of growth in this phase is primarily determined by the basic reproduction number (R0), the number of secondary ca

Alzheimer's disease polygenic risk, APOE ε4 dose, and accelerometer-derived circadian rest-activity rhythms in relation to incident dementia and Alzheimer's disease in UK Biobank: a prospective cohort study.

BACKGROUND: Circadian rest-activity rhythm disruption and sleep-wake fragmentation are common in Alzheimer's disease (AD) and may precede clinical diagnosis. Wrist-worn accelerometry enables objective, scalable circadian phenotyping in population cohorts. We examined whether AD polygenic risk and APOE ε4 dose are reflected in accelerometry-derived circadian phenotypes and whether these phenotypes predict subsequent incident dementia/AD. METHODS: We analyzed 94,241 UK Biobank participants with 7-day wrist accelerometry and genetic data. Circadian phenotypes included relative amplitude (RA), L5

Genetics of murine type 2 diabetes and comorbidities.

Type 2 diabetes (T2D) is a polygenic and multifactorial complex disease, defined as chronic metabolic disorder. It's a major global health concern with an estimated 463 million adults aged 20-79 years with diabetes and projected to increase up to 700 million by 2045. T2D was reported to be one of the four leading causes of non-communicable disease (NCD) deaths in 2012. Environmental factors play a part in the development of polygenic forms of diabetes. Polygenic forms of diabetes often run-in families. Fortunately, T2D, which accounts for 90-95% of the entire four types of diabetes including,

Genetic exploration of the relationship between liability to psychiatric disorders and acne vulgaris.

Observational epidemiology suggests a link between the dermatological disorder acne vulgaris and several psychiatric disorders. However, the biological mechanisms that underlie the relationship between acne and mental health are poorly characterised. Here, we employed a genetic approach using large-scale genome-wide association studies of acne and ten psychiatric disorders to both estimate causal effects and uncover potential shared genetic risk factors. Multiple psychiatric disorders displayed evidence of small-to-moderate genetic correlations with acne. However, only genetic liability to sch

Genetic and environmental covariation between autistic traits and behavioral problems.

Our objective was to examine the overlap between autistic traits and other behavioral problems in a general population sample, and explore the extent to which this overlap is due to genetic or environmental factors. Youth Self Report (YSR) data were collected in a general population sample of 424 twin pairs at 18 years of age, and their nontwin siblings. In 197 of these twin families, self-report ratings on the Autism-spectrum Quotient (AQ) were collected. Stepwise backward regression analyses revealed that of all 8 YSR syndrome scales, the Withdrawn Behavior (WB) and Social Problems (SOC) sca

HMC3 revealed: how much do these "Microglia" really tell us?

Microglia are a key driver of neurodegenerative disease, orchestrating inflammatory signaling, metabolic stress responses, synaptic remodeling, and neuronal fate within the central nervous system (CNS). Among experimental models, the human microglial cell line, HMC3, is one of the most widely used models for mechanistic investigation and pharmacological screening of microglial dysfunction, particularly in neurodegenerative contexts. Nevertheless, a key question remains: how faithfully does HMC3 reflect human microglial biology? This review integrates current evidence on HMC3 cells, including t

Genetic barcoding uncovers the clonal makeup of solid and liquid biopsies and their ability to capture intra-tumoral heterogeneity.

Intratumoral heterogeneity (ITH) is fueling tumor progression in breast cancer, as specific clones present within a tumor may have a selective advantage to colonize distant organs and escape therapy. Accurate sampling of ITH is therefore a pressing challenge in clinical oncology to adequately predict recurrence and inform rational and personalized therapies. Here, we used genetic barcoding to track the spatiotemporal composition of human breast cancer clones in six preclinical models-across two cell lines and four patient-derived xenografts (PDXs). This allowed a direct side-by-side quantitati

Concerted Action of Targeted Nucleic Acid Therapeutics as Flexible, Precision and Personalized Cancer Treatment.

Nucleic acid therapeutics offer unique opportunities to develop personalized precision therapies by responding specifically and effectively at the gene level to evolving pathogens. However, many nucleic acid drugs employ a single-target strategy, which is effective in monogenic disorders, yet can be ineffective in treating diseases characterized by complex and interacting molecular events. Nucleic acid combinations capable of simultaneously modulating multiple pathways and harnessing concerted effects constitute a promising approach to combatting complex, dynamically evolving and heterogeneous

Inhibiting 15-PGDH restores redox homeostasis and confers neuroprotection in Parkinson's disease.

The prostaglandin- and autocoid-degrading enzyme 15-hydroxyprostaglandin dehydrogenase (15-PGDH) is shown here to be pathologically elevated in Parkinson's disease (PD) patients and mouse models of PD in the substantia nigra, the region of the brain where dopaminergic neurons are lost in PD. Inhibiting 15-PGDH by pharmacologic blockade or partial genetic reduction restores redox homeostasis and mitigates microglial and astrocyte activation, dopaminergic neuron loss, and motor impairment across three mouse models of PD. These models included systemic 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine

Patient-Derived Organoids in Gastrointestinal Disease: Current Applications, Limitations, and Future Perspectives.

The development of patient-derived organoids (PDOs) has substantially advanced the study of gastrointestinal diseases by providing three-dimensional human models that faithfully recapitulate the structural, molecular, and functional characteristics of native tissues. Unlike conventional two-dimensional cultures and animal models, intestinal organoids preserve epithelial architecture, cellular heterogeneity, and patient-specific genetic features, enabling more physiologically relevant investigations of gastrointestinal physiology and disease. Recent technological advances, including co-culture

Generation of C9orf72 repeat knock-in iPSC lines for modelling ALS and FTD.

Induced pluripotent stem cell (iPSC) models are powerful tools for neurodegenerative disease modelling, as they allow mechanistic studies in a human genetic environment and they can be differentiated into a range of neuronal and non-neuronal cells. However, these models come with inherent challenges due to line-to-line and clonal variability. To combat this issue, the iPSC Neurodegenerative Disease Initiative (iNDI) has generated an iPSC repository using a single clonal reference line, KOLF2.1J, into which disease-causing mutations and revertants are introduced via gene editing. Here we descri
