Competitive binding and characteristic properties of pea 7S protein with three phenolic compounds: Implications for digestion and intestinal transport.

The delivery of phenols by proteins as carriers is a current research hotspot, but single phenol can no longer sufficient to meet people's health needs. In this study, a protein-phenol triligand complex of curcumin, resveratrol, caffeic acid and pea 7S protein was constructed. Fluorescence spectroscopy confirmed that quenching constants (Ksv) of resveratrol, curcumin and caffeic acid binding to protein were 2.73 × 104 M-1, 3.35 × 104 M-1and 3.38 × 104 M-1, respectively, indicating that caffeic acid has the strongest binding affinity. Discovery of Novel Cyclic Peptides as SMAD2-SMAD4 Interaction Inhibitors for the Treatment of Hepatic Fibrosis.

Hepatic fibrosis, characterized by the excessive deposition of the extracellular matrix, represents a common consequence of various chronic liver disorders. However, no specific drugs are available for antifibrotic therapy to date. SMAD2 is phosphorylated by transforming growth factor-β and subsequently binds to SMAD4 to generate a heteromeric complex, which then translocates into the nucleus and aggravates liver fibrosis. Herein, based on molecular docking simulation and structure-activity relationship study, we report the discovery of a novel cyclic peptide CMF9 that targets SMAD2 and potent

Humanization and characterization of a murine monoclonal neutralizing antibody against human adenovirus 7.

Human adenovirus type 7 (HAdV7) is commonly associated with febrile acute respiratory disease (ARD) outbreaks. We have reported that 10G12, a mouse monoclonal antibody (mAb) specifically recognizing and neutralizing HAdV7, is a promising candidate for humanization. In this study, we engineered the six variants of 10G12 with increased degree of humanization and investigated their biological activity. The humanized monoclonal antibody (mAb) 10G12-M2 was shown to retain the parental antibody's high binding affinity, specificity and potent efficacy of viral suppression. The mAb 10G12-M2 recognized

Structure-activity relationship study of hydroxyethylamine isostere and P1' site structure of peptide mimetic BACE1 inhibitors.

An aromatic substituent has been introduced into a known hydroxyethylamine (HEA)-type BACE1 inhibitor containing the superior substrate sequence to enhance inhibitory activity. The HEA-type isosteres bearing different hydroxyl group and methyl group configurations were prepared through a branched synthesis approach using intra- and inter-molecular epoxide opening reactions. The effect of their configuration was evaluated, showing that an R-configuration improved the inhibitory activity, while introduction of a methyl group on the isostere decreased the activity. Based on the non-substituted is

A synthetic diterpene analogue inhibits mycobacterial persistence and biofilm formation by targeting (p)ppGpp synthetases.

Bacterial persistence coupled with biofilm formation is directly associated with failure of antibiotic treatment of tuberculosis. We have now identified 4-(4,7-DiMethyl-1,2,3,4-tetrahydroNaphthalene-1-yl)Pentanoic acid (DMNP), a synthetic diterpene analogue, as a lead compound that was capable of suppressing persistence and eradicating biofilms in Mycobacterium smegmatis. By using two reciprocal experimental approaches - ΔrelMsm and ΔrelZ gene knockout mutations versus relMsm and relZ overexpression technique - we showed that both RelMsm and RelZ (p)ppGpp synthetases are plausible candidates f

Identification and evaluation of bioactive natural products as potential inhibitors of human microtubule affinity-regulating kinase 4 (MARK4).

Microtubule affinity-regulating kinase 4 (MARK4) has recently been identified as a potential drug target for several complex diseases including cancer, diabetes and neurodegenerative disorders. Inhibition of MARK4 activity is an appealing therapeutic option to treat such diseases. Here, we have performed structure-based virtual high-throughput screening of 100,000 naturally occurring compounds from ZINC database against MARK4 to find its potential inhibitors. The resulted hits were selected, based on the binding affinities, docking scores and selectivity. Further, binding energy calculation, L

In silico discrimination of nsSNPs in hTERT gene by means of local DNA sequence context and regularity.

Understanding and predicting the significance of novel genetic variants revealed by DNA sequencing is a major challenge to integrate and interpret in medical genetics with medical practice. Recent studies have afforded significant advances in characterization and predicting the association of single nucleotide polymorphisms in human TERT with various disorders, but the results remain inconclusive. In this context, a comparative study between disease causing and novel mutations in hTERT gene was performed computationally. Out of 59 missense mutations, five variants were predicted to be less sta

Discovery of Novel N-Phenylpyrazole Derivatives as Protoporphyrinogen IX Oxidase Inhibitors.

Protoporphyrinogen oxidase (PPO) is one of the major targets for herbicide discovery. N-Phenylpyrazole derivatives bearing ester, oxime, and isoxazoline moieties were synthesized via isomerization, derivatization and cyclization. Bioassays showed C34 potently inhibited Nicotiana tabacum PPO (NtPPO) (Ki = 15.6 nM), with superior activity to fluazolate (Ki = 18.3 nM). Compound C34 showed excellent postemergence herbicidal efficacy to fluazolate at 37.5 g a.i./ha and broader-spectrum herbicidal activity than fluazolate at 75-150 g a.i./ha. Meanwhile, C34 produced only 10-20% injury in peanut and

Discovery of Pyrimidinedione Derivatives Containing Oxime Ester/Carbamate Fragments as Potent Protoporphyrinogen IX Oxidase Inhibitors.

To continue our research on developing PPO inhibitors, we replaced the triazinone core of compounds B, G, and H from our previous work with pyrimidinediones, employing a scaffold-hopping strategy to design and synthesize compounds I, J, and K. Compound I18 exhibited potent NtPPO inhibition (Ki = 10.9 nM) and broad-spectrum herbicidal activity comparable to saflufenacil, providing 100% control of 29 weeds at 75 g a.i./ha. It also showed improved crop safety, causing only 30% peanut injury and thus being safer than saflufenacil (80% injury), while exhibiting low toxicity to zebrafish and earthwo
