An optimized geo-hierarchical ensemble model to forecast hospitalizations from respiratory viruses in the United States.

Accurate forecasting of infectious diseases is crucial for timely public health response. Ensemble frameworks have shown promising outcomes in short-term forecasting of COVID-19, among other respiratory viruses, however, there is a need to further improve these frameworks. Here, we propose the generalized Optimized Geo-Hierarchical Ensemble Model (OGEM), a novel forecasting machine learning framework to forecast state-level hospitalizations of influenza, COVID-19, and RSV in the U.S. independently. This framework is multi-resolution: it integrates state, regionally-trained, and nationally-trai

Recent Advances in Paediatric Vesicoureteral Reflux: Risk Assessment and Management Strategies.

Recent advances in paediatric vesicoureteral reflux (VUR) have focused on risk-stratification models to better identify patients at risk for complications such as renal scarring, persistent VUR, or breakthrough urinary tract infections (UTIs). By addressing critical clinical questions-such as predicting VUR presence, assessing the likelihood of spontaneous resolution, and determining the risk of breakthrough UTIs despite antibiotic prophylaxis-these tools enhance decision-making. Predictive models like the VURx score and ureteral diameter ratio integrate clinical, demographic, and imaging data

In-silico study of approved drugs as potential inhibitors against 3CLpro and other viral proteins of CoVID-19.

The global pandemic, due to the emergence of COVID-19, has created a public health crisis. It has a huge morbidity rate that was never comprehended in the recent decades. Despite numerous efforts, potent antiviral drugs are lacking. Repurposing of drugs presents a low-cost and rapid solution for finding new drugs by exploiting known drugs. In this study, we employed an integrated In-Silico approach using molecular docking and machine learning regression models to explore the potential inhibitors against key proteins of SARS-CoV-2. A library of 5903 drugs from the ZINC database was retrieved an

Artificial Intelligence Approach for Severe Dengue Early Warning System.

Dengue fever is a viral infectious disease transmitted through mosquito bites, and has symptoms ranging from mild flu-like symptoms to deadly complications. Dengue fever is one of the global burden diseases which annually have 50-100 million cases with 500,000 cases of severe dengue fever, of which 22,000 deaths occur mostly in children. Despite the discovery of vaccines, vector control is still the main approach for prevention efforts. Early detection and accessibility to medical care can reduce severe Dengue mortality rate from 50% to 2%. In the previous study, both statistical and machine l

AI-driven model for early failure prediction after HIFU integrating immediate post-ablation ultrasound and clinical data.

PURPOSE: High-Intensity Focused Ultrasound (HIFU) is an emerging focal therapy for localized prostate cancer, offering an alternative to radical prostatectomy and radiotherapy while balancing oncological control and functional preservation. However, early treatment failure remains a critical challenge, and tools to support early risk stratification are lacking. We developed a two-stage model integrating immediate post-ablation ultrasound with routinely available clinical variables to predict early recurrence (≤ 12 months) after HIFU. METHODS: Seventy-two patients treated with HIFU were analyz

A hybrid physics-informed neural and explainable AI approach for scalable and interpretable AQI predictions.

Air Pollution is a critical environmental issue affecting public health, climate, and ecosystems. However, accurately predicting and classifying Air Quality Index (AQI) levels across different regions remains a challenging task due to the complex nature of air pollution patterns. Conventional and ensemble ML and DL models often fail to capture the physical laws goverming the air pollution, which leads to inaccurate predictions. This study addresses these issues by introducing an approach that employs Physics-Informed Neural Networks (PINN) with Explainable AI (XAI) techniques for AQI classific

Predicting humoral responses to primary and booster SARS-CoV-2 mRNA vaccination in people living with HIV: a machine learning approach.

BACKGROUND: SARS-CoV-2 mRNA vaccines are highly immunogenic in people living with HIV (PLWH) on effective antiretroviral therapy (ART). However, whether viro-immunologic parameters or other factors affect immune responses to vaccination is debated. This study aimed to develop a machine learning-based model able to predict the humoral response to mRNA vaccines in PLWH and to assess the impact of demographic and clinical variables on antibody production over time. METHODS: Different machine learning algorithms have been compared in the setting of a longitudinal observational study involving 497

Machine learning approaches for prediction of early death among lung cancer patients with bone metastases using routine clinical characteristics: An analysis of 19,887 patients.

PURPOSE: Bone is one of the most common sites for the spread of malignant tumors. Patients with bone metastases whose prognosis was shorter than 3 months (early death) were considered as surgical contraindications. However, the information currently available in the literature limits our capacity to assess the risk likelihood of 3 month mortality. As a result, the study's objective is to create an accurate prediction model utilizing machine-learning techniques to predict 3 month mortality specifically among lung cancer patients with bone metastases according to easily available clinical data.

Prospective contingency explains behavior and dopamine signals during associative learning.

Associative learning depends on contingency, the degree to which a stimulus predicts an outcome. Despite its importance, the neural mechanisms linking contingency to behavior remain elusive. In the present study, we examined the dopamine activity in the ventral striatum-a signal implicated in associative learning-in a Pavlovian contingency degradation task in mice. We show that both anticipatory licking and dopamine responses to a conditioned stimulus decreased when additional rewards were delivered uncued, but remained unchanged if additional rewards were cued. These results conflict with con

Innate biosignature of treatment failure in human cutaneous leishmaniasis.

The quality and magnitude of the immune and inflammatory responses determine the clinical outcome of Leishmania infection, and contribute to the efficacy of antileishmanial treatments. However, the precise immune mechanisms involved in healing or in the chronic immunopathology of human cutaneous leishmaniasis (CL) are not well understood. Through sequential transcriptomic profiling of blood monocytes, neutrophils, and eosinophils over the course of systemic treatment with meglumine antimoniate, we revealed that a heightened and sustained Type-I interferon response signature is a hallmark of tr

Prediction of bloodstream infection using machine learning based primarily on biochemical data.

Early diagnosis of bloodstream infection (BSI) is crucial for informed antibiotic use. This study developed a machine learning approach for early BSI detection using a comprehensive dataset from Rigshospitalet, Denmark (2010-2020). The dataset included 144,398 samples from adult patients, containing blood culture results, demographics, and up to 36 biochemical variables. Positive blood culture was observed in 6.4% of samples, mostly caused by Staphylococcus aureus, Escherichia coli, and Enterococcus faecium. 80% of the samples (N = 43,351 patients) were used for ML model development and five-f

Predicting non-chemotherapy drug-induced agranulocytosis toxicity through ensemble machine learning approaches.

Agranulocytosis, induced by non-chemotherapy drugs, is a serious medical condition that presents a formidable challenge in predictive toxicology due to its idiosyncratic nature and complex mechanisms. In this study, we assembled a dataset of 759 compounds and applied a rigorous feature selection process prior to employing ensemble machine learning classifiers to forecast non-chemotherapy drug-induced agranulocytosis (NCDIA) toxicity. The balanced bagging classifier combined with a gradient boosting decision tree (BBC + GBDT), utilizing the combined descriptor set of DS and RDKit comprising 237
