The Proteus syndrome is a rare congenital hamartomatous condition that is characterized by a wide range of malformations, sometimes involving the face. Common manifestations include partial gigantism, congenital lipomas, and plantar hyperplasia. In this report we describe the craniofacial clinicopathological development in a girl with Proteus syndrome from age 6 to 20 years. The patient had pronounced hemifacial hypertrophy, exostoses in the left parietal region, and enlargement of the inferior alveolar nerve and mandibular canal in the affected region. The dental development of the affected l

Case reports of mandibular molars with iatrogenic perforations are presented. Such perforations have been treated with internal amalgam repair or by surgical intervention. However, the potential exists for tissue migration and pocket formation. The use of guided tissue regeneration as a barrier for apical migration is beneficial in surgical treatment of perforations. Follow-up examination of perforated teeth treated with resorbable barriers showed successful repair. Results obtained with this technique offer the possibility of a successful repair for an otherwise hopeless complication of endod

Myelinating cells wrap axons with multi-layered myelin sheaths for rapid impulse propagation. Dysfunctions of oligodendrocytes or Schwann cells are often associated with neuroinflammation, as observed in animal models of leukodystrophies and peripheral neuropathies, respectively. The neuroinflammatory response modulates the pathological changes, including demyelination and axonal injury, but also remyelination and repair. Here we discuss different immune mechanisms as well as factors released or exposed by myelinating glia in disease conditions. The spectrum of inflammatory mediators varies wi

BACKGROUND: The clinical spectrum of autosomal recessive spastic ataxia of Charlevoix–Saguenay (ARSACS) in Asian populations remains incompletely defined. We aimed to characterize the clinical, radiological, and genetic features of Japanese patients with ARSACS and to expand the mutational and phenotypic spectrum of this disorder. METHODS: We conducted a retrospective case series of patients diagnosed with ARSACS in our department between January 2016 and December 2023. Five patients from four families with biallelic SACS variants were identified. RESULTS: Genetic analysis revealed seven patho

Periodontal disease is a chronic inflammatory disorder that gradually destroys tooth-supporting tissues including the alveolar bone, periodontal ligament, and cementum. In recent years, interest has grown in repurposing metformin, a well-known oral antidiabetic drug, as an adjunct to periodontal therapy. Experimental studies have indicated that metformin can aid periodontal healing by promoting stem cell-driven regeneration, enhancing osteoblast activity, and reducing osteoclast-mediated resorption. Clinical trials have further reported that both systemic and local applications of metformin, w

OBJECTIVES: External cervical resorption (ECR) is a progressive condition that may compromise both periodontal and pulpal tissues. This report describes the outcome of a single-session combined endodontic-periodontal approach for the management of ECR in a previously treated maxillary central incisor.
CASE PRESENTATION: A 41-year-old male presented with ECR affecting the maxillary left central incisor, associated with previous trauma, a sinus tract, and defective root canal treatment. Cone-beam computed tomography (CBCT) was used for diagnosis, classification, and treatment planning. A single

Background/Objectives: Although gingivitis is the most common oral disease in children, periodontitis accompanied by alveolar bone resorption may develop in patients with severe congenital neutropenia. However, no reports to date have focused on changes in the alveolar bone of these patients during long-term follow-up. Case Summary: A girl aged 8 years and 5 months who developed leukemia due to severe neutropenia was admitted to the hospital and referred to the pediatric dentistry department for oral care. Panoramic radiographs at the first visit revealed significant alveolar bone resorption a

Periodontitis constitutes the primary cause of tooth loss among adults in China. The disease is characterized by the high morbidity, which significantly impairs both oral and systemic health. As the key insights of initial periodontal therapy, subgingival scaling and root planing (SRP) have been considered as simple, effective, and cost-efficient treatment approaches for managing periodontal inflammation. Substantial published evidences indicated that the majority of patients diagnosed with periodontitis could achieve lifelong retention of their natural teeth through standardized initial perio

Alveolar ridge resorption following tooth extraction or periodontal disease often necessitates bone augmentation to restore sufficient volume for stable and esthetic implant placement. However, conventional guided bone regeneration using collagen membranes demonstrates limited space maintenance and graft stability, particularly in complex defects. Additionally, harvesting autogenous bone requires a secondary surgical site, increasing morbidity and prolonging recovery. To address these limitations, we present the modified bone ring tenting technique, which utilizes autogenous bone blocks to ach

This report evaluated the impact of using multiple individualized endodontic access guides in the management of obliterated root canals in maxillary molars. A 70-year-old female patient presented with pain in the upper left first molar. Intraoral examination revealed an extensive resin composite restoration, with no signs of tooth mobility or marginal periodontal disease. The clinical diagnosis was symptomatic apical periodontitis. Cone beam computed tomography (CBCT) revealed a hypodense area in the mesiobuccal and palatal (P) roots, associated with signs of apical periodontitis and obliterat

Over the past few decades, the scientific community has been highly concerned about the obesity epidemic. Artificial sweeteners are compounds that mimic the sweet taste of sugar but have no calories or carbohydrates; hence, they are very popular among patients suffering from diabetes or obesity, aiming to achieve glycemic and/or weight control. There are four different types of sweeteners: artificial, natural, rare sugars, and polyols. Artificial and natural sweeteners are characterized as non-nutritional sweeteners (NNSs) since they do not contain calories. The extended use of sweeteners has

Periodontitis is one of the most prevalent inflammatory disease worldwide, which affects 11% of the global population and is a major cause of tooth loss. Recently, oxidative stress (OS) has been found to be the pivital pathophysiological mechanism of periodontitis, and overactivated OS will lead to inflammation, apoptosis, pyroptosis and alveolar bone resorption. Interestingly, heme oxygenase-1 (HO-1), a rate-limiting enzyme in heme degradation, can exert antioxidant activites through its products-carbon monoxide (CO), Fe2+, biliverdin and bilirubin in the inflammatory microenvironment, thus e

Periodontitis is a chronic inflammatory disease caused by the local microbiome and the host immune response, resulting in periodontal structure damage and even tooth loss. Scaling and root planning combined with antibiotics are the conventional means of nonsurgical treatment of periodontitis, but they are insufficient to fully heal periodontitis due to intractable bacterial attachment and drug resistance. Novel and effective therapeutic options in clinical drug therapy remain scarce. Nanotherapeutics achieve stable cell targeting, oral retention and smart release by great flexibility in changi

UNLABELLED: We report a case of severe central sleep apnea incidentally diagnosed during polysomnography for suspected obstructive sleep apnea. Characteristic clinical features included episodic hyperventilation followed by apnea from hypocapnia, which did not follow a Cheyne-Stokes pattern. Combined with the identification of cerebellar and brainstem malformations known as the "molar tooth sign" on a brain magnetic resonance imaging, developmental delay, and motor coordination problems, Joubert syndrome (a congenital disease) was first diagnosed at the age of 50 years. Central apneas were als

Microrchidia CW-type zinc finger protein 2 (MORC2) is an ATPase-containing nuclear protein which regulates transcription through chromatin remodelling and epigenetic silencing. MORC2 may have a role in the development of neurones, and dominant variants in this gene have recently been linked with disorders including Charcot-Marie-Tooth type 2Z disease, spinal muscular atrophy and, more recently, a neurodevelopmental syndrome consisting of developmental delay, impaired growth, dysmorphic facies, and axonal neuropathy (DIGFAN), presenting with hypotonia, microcephaly, brain atrophy, intellectual

Microrchidia CW-type zinc finger 2 (MORC2) gene encodes a protein expressed in all tissues and enriched in the brain. MORC2 protein is composed of a catalytic ATPase domain, three coil-coiled domains allowing dimerization or protein complex interaction, a zinc-finger CW domain allowing DNA interaction, and a CHROMO-like (CHRromatin Organization Modifier) domain. Recently, de novo or dominantly inherited heterozygous mutations have been associated with a spectrum of disorders affecting the peripheral nervous system such as the Charcot-Marie-Tooth disease, spinal muscular atrophy-like phenotype

Approximately 1 in 36 children are diagnosed with autism spectrum disorder (ASD). The disorder is four times more common in males than in females. Zinc deficiency and mutations in SHANK2 and SHANK3 (members of a family of excitatory postsynaptic scaffolding proteins) are all risk factors that may contribute to the pathophysiology of the disease. The presence of shankopathies (loss of one copy of the SHANK3 gene) can lead to the development of Phelan-McDermid syndrome (PMDS)-a rare genetic disorder characterized by developmental delay, intellectual disability, poor motor tone, and ASD-like symp

The pediatric Charcot-Marie-Tooth (CMT) specific quality of life (QOL) outcome measure (pCMT-QOL) is a recently developed and validated patient-reported measure of health QOL for children with CMT. The aim of this study was to provide and validate an Italian version of the pCMT-QOL. The original English version was translated and adapted into Italian using standard procedures. pCMT-QOL was administered to patients genetically diagnosed with CMT, aged 8 to 18 years. A retest was given 2 weeks later to assess reliability in all patients. A total of 22 patients (median age 14 years, DS 2.5; M:F 1
