Hydrogen sulfide ameliorates learning memory impairment in APP/PS1 transgenic mice: A novel mechanism mediated by the activation of Nrf2.

Beta-amyloid (Aβ) plaques and oxidative stress are associated with the pathogenesis of Alzheimer's disease (AD). Hydrogen sulfide (H2S) has been recognized as a cytoprotectant, which improves learning memory impairment and exerts antioxidant effects in neurodegenerative disorders, including AD. The experiment was projected to explore the effects of H2S on cognitive deficits, Aβ levels and possible antioxidant mechanisms. Here, APP/PS1 transgenic mice were injected sodium hydrosulfide (NaHS, a H2S donor, 2.8mg/kg) once a day for three months. It was found that APP/PS1 transgenic mice exhibited

Verbal memory declines more rapidly with age in HIV infected versus uninfected adults.

OBJECTIVES: In the current era of effective antiretroviral treatment, the number of older adults living with HIV is rapidly increasing. This study investigated the combined influence of age and HIV infection on longitudinal changes in verbal and visuospatial learning and memory. METHOD: In this longitudinal, case-control design, 54 HIV seropositive and 30 seronegative individuals aged 40-74 years received neurocognitive assessments at baseline visits and again one year later. Assessment included tests of verbal and visuospatial learning and memory. Linear regression was used to predict baseli

Neocortical disruption and behavioral impairments in rats following in utero RNAi of candidate dyslexia risk gene Kiaa0319.

Within the last decade several genes have been identified as candidate risk genes for developmental dyslexia. Recent research using animal models and embryonic RNA interference (RNAi) has shown that a subset of the candidate dyslexia risk genes--DYX1C1, ROBO1, DCDC2, KIAA0319--regulate critical parameters of neocortical development, such as neuronal migration. For example, embryonic disruption of the rodent homolog of DYX1C1 disrupts neuronal migration and produces deficits in rapid auditory processing (RAP) and working memory--phenotypes that have been reported to be associated with developme

Tactile-auditory shape learning engages the lateral occipital complex.

Shape is an object property that inherently exists in vision and touch, and is processed in part by the lateral occipital complex (LOC). Recent studies have shown that shape can be artificially coded by sound using sensory substitution algorithms and learned with behavioral training. This finding offers a unique opportunity to test intermodal generalizability of the LOC beyond the sensory modalities in which shape is naturally perceived. Therefore, we investigated the role of the LOC in processing of shape by examining neural activity associated with learning tactile-shape-coded auditory infor

Amitriptyline rather than lornoxicam ameliorates neuropathic pain-induced deficits in abilities of spatial learning and memory.

BACKGROUND AND OBJECTIVE: Clinical studies have revealed that patients with chronic pain are more likely to have anxiety and depression, which are often associated with cognitive dysfunction. However, whether neuropathic pain can induce cognition dysfunction remains uncertain. Antidepressants and nonsteroidal anti-inflammatory drugs can treat neuropathic pain, but whether they can prevent cognition dysfunction is unknown. The present study was designed to investigate the effects and possible mechanisms of neuropathic pain on learning and memory, and the effects of amitriptyline and lornoxicam

Combined adaptiveness of specific motor cortical ensembles underlies learning.

Learning motor skills entails adaptation of neural computations that can generate or modify associations between sensations and actions. Indeed, humans can use different strategies when adapting to dynamic loads depending on available sensory feedback. Here, we examined how neural activity in motor cortex was modified when monkeys made arm reaches to a visual target and locally adapted to curl force field with or without visual trajectory feedback. We found that firing rates of a large subpopulation of cells were consistently modulated depending on the distance of their preferred direction fro

Reduced water intake: Implications for rodent developmental and reproductive toxicity studies.

In developmental and reproductive toxicity studies, drinking water is a common means of delivering the test agent. Reduced consumption of toxicant-containing water raises questions about indirect effects of reduced maternal fluid consumption resulting from unpalatability, versus direct effects of the test compound. Issues to consider include: objective assessment of dehydration and thirst, the relative contributions of innate and learned behaviors to drinking behavior and flavor preference, and the objective assessment of physiologic stress. Not only do lab animals under ad lib conditions cons

Effects of 5-HT6 receptor antagonism and cholinesterase inhibition in models of cognitive impairment in the rat.

BACKGROUND AND PURPOSE: The beneficial effect of 5-HT6 receptor antagonism in cognition remains controversial. This study has been undertaken to reassess the cognition enhancing properties of acute vs subchronic treatment with the selective 5-HT6 receptor antagonist SB-271046 in unimpaired rats, as well as against scopolamine (cholinergic-) or MK-801 (glutamatergic-mediated) deficits. EXPERIMENTAL APPROACH: The Morris water maze was used, measuring behaviour acquisition and retention, and swim speed. Other behavioural measures included yawning and motor activity. SB-271046 was given acutely b

PTEN-5-HT2C coupling: a new target for treating drug addiction.

It is well known that the ventral tegmental area (VTA) is a brain region in which virtually all abused drugs exert rewarding effects by activating its dopamine neurons. We recently found that the tumour suppressor enzyme phosphatase and tensin homologue deleted on chromosome 10 (PTEN) directly interacts to a region in the third intracellular loop (3L4F) of serotonin 5-HT2C receptors (5-HT2cR) in the rat VTA. PTEN limits agonist-induced 5-HT2cR phosphorylation via its protein phosphatase activity. Systemic or intra-amygdaloid application of the interfering peptide Tat-3L4F is able to disrupt PT

Unilateral lesions of the cholinergic basal forebrain and fornix in one hemisphere and inferior temporal cortex in the opposite hemisphere produce severe learning impairments in rhesus monkeys.

It has been proposed that isolation of the inferior temporal cortex and medial temporal lobe from their cholinergic afferents results in a severe anterograde amnesia. To test this hypothesis directly, seven rhesus monkeys received a unilateral immunotoxic lesion of the cholinergic cells of the basal forebrain with an ipsilesional section of the fornix. In a second surgery, inferior temporal cortex was ablated in the opposite hemisphere. All animals were severely impaired at learning visual scenes and object-reward associations. The impairment in learning scenes was correlated with cholinergic

Excitatory amino acids and memory: evidence from research on Alzheimer's disease and behavioral pharmacology.

The excitatory amino acid transmitter (EAA) system is believed to play a crucial role in a variety of physiological processes related to neuronal plasticity. Substantial neurophysiological evidence suggests that, under normal physiological conditions. EAAs may be involved in mechanisms underlying learning and memory. However, overactivity of this system produces excitotoxic damage to neurons which is believed to be an etiological factor in various neurological conditions, such as epilepsy, and stroke-induced impairments. The fact that EAAs have been implicated in both, normal cognitive functio

On the usage of artificial intelligence for identifying main attributes and predicting neonatal sepsis.

During the neonatal period, newborns are more susceptible to developing conditions and diseases due to their fragility of the transition and adaptation to the extrauterine environment. Neonatal sepsis is one of the leading causes of morbidity and mortality in newborns, particularly among preterm and low birth weight infants, requiring early diagnosis to reduce complications and deaths. In this work, we evaluate the performance of artificial intelligence models in predicting neonatal sepsis and also identify the attributes that most contribute impact on models' learning and their relationship w

Machine learning models using longitudinal laboratory data predict survival in allogeneic stem cell recipients.

Allogeneic hematopoietic stem cell transplantation (HSCT) remains a potentially curative therapy for hematologic malignancies and bone marrow failure syndromes, yet outcomes remain highly heterogeneous and existing prognostic tools rely predominantly on pre-transplant clinical characteristics. We retrospectively analyzed 500 consecutive patients who underwent allogeneic HSCT at a single academic center between 2019 and 2025. Clinical characteristics, transplant variables, comorbidities, medication exposures, and longitudinal laboratory parameters were evaluated for associations with overall su

Identifying effective immune biomarkers in alopecia areata diagnosis based on machine learning methods.

BACKGROUND: Alopecia areata (AA) is a common non-scarring hair loss disorder associated with autoimmune conditions. However, the pathobiology of AA is not well understood, and there is no targeted therapy available for AA.  METHODS: In this study, differential gene expression analysis, immune status assessment, weighted correlation network analysis (WGCNA), and functional enrichment analysis were performed to identify shared genes associated with both immunological response and AA. Machine learning methods were then used to identify three hub genes as potential diagnostic markers for AA. Exter

AI-assisted computational screening and docking simulation prioritize marine natural products for small-molecule PCSK9 inhibition.

SARS-CoV-2 infection has been associated with long-term cardiovascular complications including myocarditis and heart failure, as well as central nervous system sequelae such as cognitive dysfunction and neuropathy. Proprotein convertase subtilisin/Kexin type 9 (PCSK9), a hepatic protease involved in cholesterol regulation, has shown associations with a spectrum of diseases potentially relevant to these Covid-19 complications, such as atherosclerosis. To identify novel human PCSK9 inhibitors, a custom virtual screening pipeline was developed employing (1) a convolutional neural network-based de

Older adults' decision-making following bad advice.

There is minimal research investigating the influence of advice on decision-making in older age. The present study investigated the effect of different types of bad advice, relative to no advice, on young and older adults' decision-making in the Iowa Gambling Task (IGT). Fifty-four older adults and 59 young adults completed the IGT after receiving no advice, or advice to select from disadvantageous deck A (small, high-frequency losses), or disadvantageous deck B (larger, low-frequency losses). Corrugator EMG, memory and fluid intelligence were assessed. Averaged across advice conditions, older

Advanced Sensing System for Sleep Bruxism across Multiple Postures via EMG and Machine Learning.

Diagnosis of bruxism is challenging because not all contractions of the masticatory muscles can be classified as bruxism. Conventional methods for sleep bruxism detection vary in effectiveness. Some provide objective data through EMG, ECG, or EEG; others, such as dental implants, are less accessible for daily practice. These methods have targeted the masseter as the key muscle for bruxism detection. However, it is important to consider that the temporalis muscle is also active during bruxism among masticatory muscles. Moreover, studies have predominantly examined sleep bruxism in the supine po
