Unpacking the daily dynamics of parenting strain: A 15-day diary study of caregiving role overload, depressive symptoms, and parental burnout among parents of autistic children.

OBJECTIVES: To explore the day-to-day dynamics between daily caregiving role overload and daily depressive symptoms among parents of autistic children, and to examine gender differences in these variables' average levels and day-to-day dynamics, as well as whether such differences help explain parental burnout. METHODS: We recruited parents of autistic children from autism intervention centers using convenience sampling (N = 210; 81.9% mothers) in mainland China. Parents completed a brief online diary survey each evening across 15 weekdays over three consecutive weeks to assess daily depressi

Gut microbiome alterations are sex-dependently associated with brain abnormalities in a mouse model of Neurofibromatosis type I.

Neurofibromatosis type 1 (NF1) is a genetic condition presenting with variable symptomatology, however most individuals will demonstrate cognitive and behavioural difficulties, including autism. Using a heterozygous germline knockout mouse model of NF1 (Nf1 +/-), we performed in-depth behavioural evaluations encompassing learning and memory, stereotypy, social interaction, anxiety- and depression-like behaviour. Anatomical and functional studies of the brain and gastrointestinal tract were followed by the first investigation of gut microbiota composition (via full-length 16S rRNA sequencing) i

Sex differences in ferroptosis-related vulnerability to autism-like deficits in the adolescent medial prefrontal cortex following embryonic valproic acid exposure.

BACKGROUND: Prenatal valproic acid (VPA) exposure is an important environmental risk factor for autism spectrum disorder (ASD), but the mechanisms underlying VPA-induced ASD-like phenotypes remain unclear. Emerging evidence implicates neuronal ferroptosis and neurodevelopmental disturbances, yet whether these alterations vary across developmental stages and between sexes is unknown. METHODS: Adolescent male and female C57BL/6 mice exposed to VPA at embryonic day 12.5 (E12.5) or postnatal day 14 (PND14) were used to examine behavioral and neurobiological effects of VPA exposure across developm

Competing programs shape cortical sensorimotor-association axis development.

The cerebral cortex is organized along a dominant sensorimotor-to-association (S-A) axis, anchored by modality-specific primary sensorimotor areas at one end and transmodal association areas forming distributed networks that support abstract cognition at the other1-11. The developmental mechanisms shaping this axis remain unclear9-24. Here we present converging multispecies evidence supporting the multinodal induction-exclusion in network development (MIND) model, in which S-A patterning is governed by competing processes of induction and exclusion driven by two opposing transcriptomically def

The impact of maternal vitamin D deficiency during pregnancy and lactation on autism-like behavior in offspring.

Autism spectrum disorder (ASD) is a neurodevelopmental disorder characterized by social impairments and stereotyped behaviors. Many epidemiological studies have found a potential relationship between vitamin D deficiency (VDD) and ASD. However, the mechanism remains unclear. In this study, a VDD model was established by feeding a vitamin D-depleted diet to 5-week-old female mice, from their adulthood through pregnancy to end of lactation. Social deficits, repetitive stereotyped behaviors and anxiety-like behaviors were evaluated in the offspring. The results showed the number of buried marbles

Psychosocial factors associated with the trajectories of interparental conflict for Australian fathers of autistic children: A longitudinal study across 10 years of child development.

Limited research exists on fathers' experiences of interparental conflict (IPC) in families with autistic children. We aimed to identify: (1) the extent to which these fathers report IPC across 10 years of child development (4-14 years) and how this compares to fathers of non-autistic children; (2) distinct trajectories of IPC for fathers of autistic children and (3) factors associated with trajectories of IPC among fathers of autistic children. This is a retrospective study using a national dataset. Participants were 281 fathers of autistic children and 7046 fathers of non-autistic children.

"There is No Help:" Caregiver Perspectives on Service Needs for Adolescents and Adults with Profound Autism.

The underrepresentation of individuals with profound autism (who require 24/7 access to care) in autism research has resulted in limited knowledge about their service needs and a lack of evidence-based practices tailored to those needs. This study explored caregiver perspectives on service needs, barriers to accessing care, and treatment priorities to guide treatment development and improvement of service delivery. A sequential mixed-methods design integrated quantitative survey data (n = 423; Mage = 18.89 years; 26.7% female) with qualitative interviews (n = 20) with caregivers of adolescents

GABA and Glx predict EEG responses of visual sensitivity in autism.

The mechanisms underlying atypical sensory processing in autism remain to be elucidated, but research points toward a role of the glutamatergic/GABAergic balance. To investigate the potential relationships between visual sensitivity and its molecular correlates in autism, we combined data from electroencephalography (EEG) and magnetic resonance spectroscopy (MRS) studies. Twenty autistic adults and sixteen neurotypical adults (NT) participated in both an EEG study assessing visual sensitivity (Sapey-Triomphe et al., Autism Research, 2023) and in an MRS study measuring Glx and GABA+ concentrati

TAU ablation in excitatory neurons and postnatal TAU knockdown reduce epilepsy, SUDEP, and autism behaviors in a Dravet syndrome model.

Intracellular accumulation of TAU aggregates is a hallmark of several neurodegenerative diseases. However, global genetic reduction of TAU is beneficial also in models of other brain disorders that lack such TAU pathology, suggesting a pathogenic role of nonaggregated TAU. Here, conditional ablation of TAU in excitatory, but not inhibitory, neurons reduced epilepsy, sudden unexpected death in epilepsy, overactivation of the phosphoinositide 3-kinase-AKT-mammalian target of rapamycin pathway, brain overgrowth (megalencephaly), and autism-like behaviors in a mouse model of Dravet syndrome, a sev

Environmental Enrichment Rescues Social Behavioral Deficits and Synaptic Abnormalities in Pten Haploinsufficient Mice.

Pten germline haploinsufficient (Pten+/-) mice, which model macrocephaly/autism syndrome, show social and repetitive behavior deficits, early brain overgrowth, and cortical-subcortical hyperconnectivity. Previous work indicated that altered neuronal connectivity may be a substrate for behavioral deficits. We hypothesized that exposing Pten+/- mice to environmental enrichment after brain overgrowth has occurred may facilitate adaptation to abnormal "hard-wired" connectivity through enhancing synaptic plasticity. Thus, we reared Pten+/- mice and their wild-type littermates from weaning under eit

Transcutaneous Electrical Acupoint Stimulation in Early Life Changes Synaptic Plasticity and Improves Symptoms in a Valproic Acid-Induced Rat Model of Autism.

Autism spectrum disorder (ASD) is a developmental disorder characterized by social behavior deficit in childhood without satisfactory medical intervention. Transcutaneous electrical acupoint stimulation (TEAS) is a noninvasive technique derived from acupuncture and has been shown to have similar therapeutic effects in many diseases. Valproic acid- (VPA-) induced ASD is a known model of ASD in rats. The therapeutic efficacy of TEAS was evaluated in the VPA model of ASD in the present study. The offspring of a VPA-treated rat received TEAS in the early life stage followed by a series of examinat

"In Their Own Time": Parents Gently Push Their Autistic Youth Towards Independent Community Mobility and Participation.

Autistic adults have decreased independence in community mobility and driving, which is associated with decreased participation in work, education and community participation. This is the first exploration of the development of community mobility, driving and participation skills over adolescence and emerging adulthood. Interviews with 15 mothers of autistic youth, capable of independence, were qualitatively analysed using grounded theory. Four major themes emerged: mothers gently pushing, teaching, letting go and working towards hopes and dreams. These results suggest earlier intervention acr

Restricted, Repetitive, and Stereotypical Patterns of Behavior in Autism-an fMRI Perspective.

The main objective of this paper is to determine whether resting-state fMRI can identify functional connectivity differences between individuals with autism who experience severe issues with restricted, repetitive, and stereotypical behaviors, those who experience only mild issues, and controls. We use resting-state fMRI data from the ABIDE-I preprocessed repository, with participants grouped according to their ADI-R Restricted, Repetitive, and Stereotyped Patterns of Behavior Subscore. Three processing methods are used for analysis. A time-correlation approach establishes a basic baseline, an

Default mode-visual network hypoconnectivity in an autism subtype with pronounced social visual engagement difficulties.

Social visual engagement difficulties are hallmark early signs of autism (ASD) and are easily quantified using eye tracking methods. However, it is unclear how these difficulties are linked to atypical early functional brain organization in ASD. With resting state fMRI data in a large sample of ASD toddlers and other non-ASD comparison groups, we find ASD-related functional hypoconnnectivity between 'social brain' circuitry such as the default mode network (DMN) and visual and attention networks. An eye tracking-identified ASD subtype with pronounced early social visual engagement difficulties

Integration of therapies in autistic children; a survey based in Karachi, Pakistan.

BACKGROUND: Concomitant spinal canal stenosis and degenerative spondylolisthesis have a wide variation in surgical choices. There is consensus regarding performing fusion for unstable degenerative spondylolisthesis; however, for stable degenerative spondylolisthesis, consensus does not exist. This study aims to compare laminectomy only using spinous process splitting approach and laminectomy and transforaminal lumbar interbody fusion (TLIF) in the treatment of lower lumbar canal stenosis with low-grade stable degenerative spondylolisthesis. METHODS: Forty-two patients with single-level lumbar
