Differences in orbitofrontal activation during decision-making between methadone-maintained opiate users, heroin users and healthy volunteers.

OBJECTIVE: Previously, we reported that opiate users enrolled in methadone treatment made 'risky' choices on a decision-making task following a loss of points compared with heroin users and healthy volunteers. One possible explanation for this behaviour is that methadone users were less sensitive to punishment on immediately preceding unsuccessful trials. METHODS: We sought to explore this finding from a neural perspective by performing a post hoc analysis of data from a previous [see text] positron emission tomography study. We restricted the analysis to the opiate groups and controls, asses

Further characterization of oral fentanyl self-administration in male and female Sprague Dawley rats.

Fentanyl is a potent synthetic opioid with high misuse potential and risk for overdose. While primarily associated with the intravenous route of administration, fentanyl is now often found in pills. However, few preclinical studies have assessed oral self-administration of fentanyl. In the present study, different groups of male and female Sprague Dawley rats were tested in a series of paradigms to further characterize oral fentanyl self-administration. Rats were trained to self-administer various doses of fentanyl (2.0-50.0 μg/ml) in the presence or the absence of a drug-paired stimulus and w

Angry and "productive": engagement in gamified precarious work.

We examine the affective engagement of workers in app-mediated food delivery who continue working while explicitly describing the work as harming them financially, emotionally, and bodily, while using the language of betrayed intimacy. Using a mixed-methods design that combines manual coding with eight validated computational instruments and a study-specific linguistic analysis across two corpora-1,385 Glassdoor employer reviews and 1,075 Reddit posts-we identify a sizable minority (n = 338, 34.4% of current-employee reviews) of conflicted but persisting workers. Problematic social media use and cognitive performance in young adults: Associations with selective attention and inhibitory control.

BACKGROUND AND AIMS: Problematic social media use (PSMU) has been associated with emotional and behavioral difficulties, yet its relationship with cognitive performance remains insufficiently understood. This study examined whether PSMU is associated with deficits in selective attention and inhibitory control, and whether sex moderates these associations. METHODS: A total of 943 Spanish young adults (18-35 years) completed the Bergen Social Media Addiction Scale (BSMAS) and two computerized tasks: the Stroop task, assessing selective attention, and the Simon task, assessing inhibitory control

Postoperative factors associated with hazardous alcohol use after metabolic and bariatric surgery.

INTRODUCTION: There is an increased risk of hazardous alcohol use following metabolic and bariatric surgery (MBS). Identifying risk factors of hazardous alcohol use, particularly factors that may differentiate risk among those with regular postoperative alcohol consumption, would inform postoperative monitoring. METHOD: Participants (N = 100) who were up to 5 years post-MBS and were consuming alcohol at least 2-3 times per month completed an assessment as part of a larger study. Measures assessed hazardous alcohol use, drinking motives, psychiatric symptoms, and disordered eating behaviors.

Prevalence and Associated Factors of Codependency Among Wives of Alcohol Users.

BACKGROUND: Codependency refers to an excessive emotional or psychological dependence on a partner, typically one who is struggling with addiction or illness and requires continuous care. Among wives of alcohol users, codependent behaviors are commonly observed and can significantly impact their mental health, social functioning, and overall well-being. Despite the growing recognition of this issue, there is a lack of comprehensive evidence summarizing the prevalence and associated factors of codependency in this population. This scoping review aims to address this gap in the existing literatu

Identity Reconstruction as a Coping Mechanism in Addiction Recovery: A Pilot Stratified Randomized Controlled Trial of Narrative Therapy Group Intervention.

Relapse in substance use disorders remains a persistent clinical challenge. Identity reconstruction, the psychological transition from an internalized "addict" identity to a recovery-oriented self, may be a core coping mechanism for abstinence maintenance. This exploratory pilot stratified randomized controlled trial examined the feasibility and preliminary effects of a Narrative Therapy (NT) group intervention, delivered within an evidence-based practice framework. Forty-five male residents of a closed rehabilitation facility were randomly assigned (n = 15 each) to an 8-week NT intervention,

Neuropeptide S modulation of learning and memory: a systematic review.

BACKGROUND: In the last two decades, Neuropeptide S (NPS) has been identified as a key bioactive peptide in the mammalian brain, influencing fear, anxiety, wakefulness, reward, and learning. While some reviews have addressed its role in reward-seeking and anxiety, few have addressed its particular role in learning and memory. The neuropeptide S receptor 1 is highly expressed in key areas for learning processing, such as the hippocampus, cortex, thalamus, and amygdala. This review aims to examine evidence from human and animal studies that focused on the NPS system's role in modulating learning

Changes in perineuronal net and parvalbumin expression in the orbitofrontal cortex of male Wistar rats following repeated fentanyl administration.

The misuse of opioid medications is a significant health issue in the United States. Very few studies have investigated the effect of opioids on perineuronal nets (PNNs), scaffold-like structures that surround neurons and are involved in the regulation of plasticity-dependent mechanisms such as development, learning and memory, and acquisition of addiction-like phenotypes. Regulation of PNNs in the orbitofrontal cortex (OFC) during periods of drug intoxication or withdrawal is widely unknown. In this study, male Wistar rats were injected with fentanyl (0.125 mg/kg, s.c.) or 0.9% saline twice d

Specific genes of the dopaminergic (dop-3) and serotonergic (tph-1) pathways contribute to the effects of ethanol consumption in Caenorhabditis elegans.

BACKGROUND: Excessive alcohol consumption is a global health issue and a leading cause of disease, disability, and mortality. This study aimed to determine the effects of a 24-hour ethanol exposure, post-exposure withdrawal (cessation of alcohol intake), and post-exposure withdrawal relief on the sensorimotor performance of the nematode Caenorhabditis elegans. METHODS: A modified kinetic chemotaxis assay (commonly referred as "diacetyl race") was conducted with worm populations subjected to three different doses of ethanol pre-exposure to assess the impact of ethanol on locomotion. Alcohol-associated liver disease in Asian Americans: the case for disaggregated analysis.

Alcohol-associated liver disease (ALD) is an increasingly important contributor to liver-related morbidity and mortality in the United States, yet its burden among Asian Americans remains poorly defined. Most national datasets aggregate Asian Americans into a single racial category, obscuring important differences across East Asian, Southeast Asian, and South Asian populations. This approach reinforces assumptions that Asian Americans uniformly have low-risk alcohol use and low rates of alcohol-related disease, potentially concealing vulnerable subgroups. Emerging evidence suggests considerabl

Increased Risk of Alcohol Use Disorder Following Bariatric Surgery.

BACKGROUND: Obesity affects over 40% of adults in the U.S., and bariatric surgery, particularly Roux-en-Y gastric bypass (RYGB), is among the most effective treatments for sustained weight loss and comorbidity reduction. However, emerging evidence has linked bariatric surgery to an increased risk of alcohol use disorder (AUD), particularly in the years following surgery. OBJECTIVE: To review the evidence surrounding the incidence, timing, mechanisms, and risk factors associated with AUD following bariatric surgery, with an emphasis on RYGB. METHODS: A comprehensive review of the literature e

The Increased Activity of CaV3.2 Channels Contributes to Nociceptors' Hyperexcitability and Chronic Neuropathic Pain Following Spinal Cord Injury in Mice.

Following spinal cord injury (SCI), up to 60-70% of patients develop chronic neuropathic pain. SCI-induced neuropathic pain (SCI-NP) is often lifelong and therapeutically intractable, leading to a severe decline in quality of life and increased risk for depression, anxiety, and addiction. Studies in preclinical rat models of SCI support the contribution of nociceptors' hyperexcitability and their spontaneous activity (SA) in driving the development/maintenance of SCI-NP and suggest that reducing nociceptors' hyperexcitability could provide a therapeutic strategy to treat SCI-NP. Previous data

Impact of environmental enrichment on heroin-induced neuroadaptations in the insula, nucleus accumbens and ventral tegmental area.

The ventral tegmental area (VTA), nucleus accumbens (NAc) and insular cortex (IC) are brain regions implicated in addiction. However, the involvement of heroin-induced neuroadaptations in these regions is not fully uncovered. Here, we used male and female Long Evans rats to investigate the causal roles of two parallel pathways: VTA→IC and VTA→NAc in opioid-driven behaviors, related neuroadaptations and neural mechanisms by which environmental enrichment (EE) attenuates drug taking and seeking. Our findings are: 1) confirmation that the VTA→IC and VTA→NAc pathways consist of dopamine (DA) and n

Sinomenine alleviates chronic inflammatory pain through DNA methyltransferase 1-mediated DNA methylation of the P2X2 purinergic receptor: an epigenetic mechanism of analgesia.

BACKGROUND: Chronic inflammatory pain lacks effective and non-addictive therapeutic options. Sinomenine (SIN), a non-opioid alkaloid derived from the traditional Chinese medicine Sinomenium acutum, has demonstrated analgesic potential in clinical practice, yet its precise mechanism remains incompletely understood. PURPOSE: This study aimed to investigate the analgesic mechanism of SIN and to identify a novel epigenetic pathway involved. METHODS: The effects of SIN were evaluated in complete Freund's adjuvant-induced chronic inflammatory pain models in rats and lipopolysaccharide stimulated B

The gut-liver-brain axis in alcohol use disorder: The emerging role of bile acid dysregulation.

Alcohol use disorder (AUD) is a chronic, relapsing neuropsychiatric disorder with limited treatment options. The gut-liver-brain axis has emerged as an important contributor to alcohol addiction. Within this axis, bile acids, important signaling molecules that connect the gut, liver, and central nervous system (CNS), have received increasing attention, as their metabolic dysregulation is closely associated with AUD. This review systematically elucidates the physiological metabolic processes of bile acids and the mechanisms by which chronic alcohol consumption disrupts their homeostasis. We pro

The Concept of Recovery in Compulsive Sexual Behavior Disorder - A Scoping Review.

Recovery is a central concept in the fields of mental health and addiction, yet its definition remains a subject of debate. Compulsive Sexual Behavior Disorder (CSBD), classified in the ICD-11 as an impulse control disorder, is a relatively prevalent mental health concern. This scoping review mapped and summarized the state of art of the concept of recovery in CSBD, including study characteristics, terminology and measures used, main recovery approach (deficit-based versus strengths-based, or mixed), and the key aims of these studies. Hormetic pathways in environmental enrichment in animal models and humans.

Environmental Enrichment (EE), which provides enhanced sensory, cognitive, motor, and social stimulation, has emerged as a powerful paradigm for investigating neuroplasticity and stress resilience. This chapter explores how EE functions through hormetic mechanisms-where multiple mild stimuli trigger adaptive responses that promote beneficial outcomes. Since Hebb's pioneering work on neuronal ensembles, research has demonstrated that EE enhances neurogenesis, synaptic plasticity, and neurotrophic factor expression (BDNF and NGF) while modulating inflammatory processes, epigenetic pathways, and
