Pemphigus is a group of autoimmune bullous diseases characterized by a loss of cell adhesion and blister formation within the epidermis in the skin and/or the mucosal surfaces. Pemphigus develops due to the interaction between predisposing genetic factors and environmental factors. Among the latter, infectious agents such as viruses and bacteria were reported as preceding or exacerbating pemphigus disease. In this article we review the literature concerning the relationship between pemphigus, viral and bacterial pathogens.

Epidermolysis bullosa (EB) pruriginosa is a distinctive clinical subtype of dystrophic EB. We report a patient with dominant dystrophic EB pruriginosa, who had an excellent response to systemic thalidomide treatment. The mechanism of action of thalidomide in the management of pruriginous disorders is not yet completely understood. Most recent studies point towards an immunomodulatory action of thalidomide that may suppress excessive production of tumour necrosis factor-alpha and may downregulate certain cell surface adhesion molecules involved in leucocyte migration.

Immunity to genital tract infection with Chlamydia trachomatis is mediated by type 1 CD4+ T lymphocytes. To define the signals that govern lymphocyte trafficking to the genital mucosa, integrins expressed by infiltrating T cells and endothelial addressins displayed on local vasculature were characterized during the course of infection. All T cells expressed the alphaLbeta2 heterodimer that binds vascular ICAM-1, and most displayed enhanced levels of the alpha4beta1 integrin that interacts with VCAM-1. AlphaE and beta7(low) integrin chains were detected on approximately 15 and 30% of infiltrati

The relationship between acute endothelial dysfunction and the extravasation of leukocytes was studied in vivo with intravital microscopy of the rat mesenteric microvasculature. Acute endothelial dysfunction of the rat mesenteric microvasculature was induced in vivo by superfusing the mesentery for 90 min with one of three different stimulating agents: NG-nitro-L-arginine methyl ester (L-NAME, 50 microM), thrombin (0.5 U/mL), or hydrogen peroxide (H2O2, 50 microM). All three agents induced a similar increase in leukocyte rolling and adherence, which was significantly greater than that observed

We have demonstrated that human brain capillary endothelial (HBCE) cells, unlike umbilical or aortic endothelial cells are permissively infected by HIV. HIV infection of HBCE cells is noncytolytic and is mediated by a CD4- and GalCer-independent mechanism, implying that HBCE cell tropic strains utilize a unique receptor. The V3 loop of gp120 appears to be important in this reaction. T-cell tropic but not brain-derived macrophage tropic HIV strains selectively infect brain endothelium suggesting that T-cell tropism is important for HIV entry through the blood-brain barrier (BBB). The ability of

This in vitro study investigated the adhesion of human human monocytes / macrophages (MΦ) to poly(styrene-block-isobutylene-block-styrene) (SIBS)-based PreserFlo® MicroShunts, as macrophage interactions can significantly influence microimplant success. For this, purified human MΦ were cultured for 35 days on SIBS-based PreserFlo® MicroShunts and compared against control intraocular lens (IOL) materials (PMMA and hydrophilic acrylate). We found significantly higher MΦ adhesion on the PreserFlo® MicroShunts than on PMMA or hydrophilic acrylic IOLs (p < 0.05). These results indicate that the surf

The mesenchymal subtype of the WHO grade IV Glioblastoma (GBM) is the most common and lethal form of adult brain cancer, characterised by its rapid growth, invasiveness and colonisation. Current standard of care includes surgical resection, chemotherapy and radiotherapy. Despite this, mortality remains dismal with median patient survival of less than 15 months after treatment. For these reasons, the World Health Organisation (WHO) classifies GBM as an incurable disease. To address this challenge, the GBM scientific community are investigating new approaches to gain insights into GBM function a

Cardiovascular disease (CVD) remains one of the leading causes of death worldwide. Aberrant platelet function mediates fibrin(ogen)-rich thrombi that lead to occlusive thrombi associated with mortality. The receptor, TREM-like transcript-1 (TLT-1), stored in the platelet α-granules and released upon platelet activation, binds fibrinogen and von Willebrand factor. Once it is released from platelets, TLT-1 is a potential therapeutic target to prevent the thrombosis associated with CVD. Here we designed an assay to screen a compound library of small molecules inhibitors. Human embryonic kidney (H

Cell directed therapy is an evolving therapeutic approach to treat organ dysfunction arising from hyperinflammation and cytokine storm by processing immune cells in an extracorporeal circuit. To investigate the mechanism of action of the Selective Cytopheretic Device (SCD), in vitro blood circuits were utilized to interrogate several aspects of the immunomodulatory therapy. SCD immunomodulatory activity is due to its effects on circulating neutrophils and monocytes in a low ionized calcium (iCa, Ca2+) blood circuit. Activated neutrophils adhere to the SCD fibers and degranulate with release of

Cellular senescence is a hallmark of advanced age and a major instigator of numerous inflammatory pathologies. While endothelial cell (EC) senescence is aligned with defective vascular functionality, its impact on fundamental inflammatory responses in vivo at single-cell level remain unclear. To directly investigate the role of EC senescence on dynamics of neutrophil-venular wall interactions, we applied high resolution confocal intravital microscopy to inflamed tissues of an EC-specific progeroid mouse model, characterized by profound indicators of EC senescence. Progerin-expressing ECs suppo

CONTEXT: Pterygium, meaty eyes, is a disease that produces a triangular, conjunctival-epithelial, neovascularized overgrowth covering the cornea, which can cause vision loss. Histological characterization of Pterygium reveals the presence of proliferating fibroblasts (FBs) that remodel the extracellular matrix, with infiltration of immune cells, causing chronic inflammation. The fresh juice of Echeveria pallida E. Walther (Crassulaceae), mechanically extracted from the leaves, can be used to lubricate the eyes and remove Pterygium, even in advanced, degenerative ocular disease.
OBJECTIVE: Thi

PURPOSE OF REVIEW: This study aims to gather the current state of the literature about anti-Nectin-4 innovative associations in solid tumors and to investigate underlying resistance mechanisms.
RECENT FINDINGS: Antibody-drug conjugate (ADC) targeting Nectin-4 efficacy gained attention and offers a promising association with other antineoplastic drugs especially in urothelial carcinoma. The heterogeneity of Nectin-4 expression across the molecular subtypes was highlighted especially in urothelial cancers. A unique study using preclinical models demonstrated an upregulation of P-gp expression,

Psoriasis is a common chronic inflammatory skin disease. The diversity and heterogeneity of immune cells in human skin have been studied in recent years, but the spatial distribution of immune cells at the single-cell level in the human psoriatic epidermis and dermis remains unclear. In this study, we mapped psoriatic skin immune cells from paired lesional, perilesional, and nonlesional skin samples using mass cytometry. Phenotypic dendritic cells (DCs) were found in the psoriatic epidermis and dermis. Psoriatic dermal CD1c+CD11b+ cDC2s migrated to the epidermis in the perilesional skin during

INTRODUCTION: Anti-IgLON5 disease is a rare autoimmune disease of the central nervous system. Different from the previous autoimmune encephalitis, the disease is a chronic progressive disease characterized by abnormal sleep, sleep apnea and motor disorders, which is prone to misdiagnosis and missed diagnosis.
METHODS: We report a unique case of anti-IgLON5 disease in a pediatric patient with Langerhans cell histiocytosis (LCH). He gradually developed increased muscle tone and nystagmus during chemotherapy and showed signs of meningeal enhancement on cranial imaging. Due to insufficient eviden

The advent of new technologies in recent years has revolutionized the methods by which pathogens are studied and at the same time it has provided new tools to design vaccines against infections for which vaccine development has so far been unsuccessful. The availability of genomic data provides the basis for the reverse vaccinology approach, a biotechnological strategy that uses bioinformatics analysis of microbial genome data for the in silico selection of potential vaccine candidates for the development of protein-based vaccines. The antigens selected by reverse vaccinology can be produced a

This Viewpoint gives an overview of the inherited disorders that are characterized by defects of leukocyte trafficking. Three paradigmatic diseases have been selected: warts, hypogammaglobulinemia, infections, and myelokathexis; Wiskott-Aldrich; and leukocyte adhesion deficiency syndromes. These conditions encompass defects in the steps that are required for leukocyte motility: from the response to chemokines, which is altered in warts, hypogammaglobulinemia, infections, and myelokathexis syndrome, to the impairment of leukocyte adhesion and migration found in leukocyte adhesion deficiency syn

The immune system maintains the homeostasis of the biliary epithelium, where bile and cholangiocytes play an important role in modulating immune responses. Furthermore, cholangiocytes represent the target of the immune system in several hepatobiliary diseases (e.g., primary biliary cirrhosis). This article is organized into two parts. The first section reviews the role of cholangiocytes in the immune defense and the mechanisms by which cholangiocytes regulate immune responses. The expression of adhesion molecules on the cholangiocyte surface allows the interaction with immune cells, thus local

OBJECTIVE: To investigate the effects of steady shear flow on the deformation properties of an adherent leukocyte and its nucleus.
METHOD: A compound drop model was developed to simulate the leukocyte adhered to the inner surface of a blood vessel, and a two dimensional computational fluid dynamics (CFD) was conducted to solve the model equations.
RESULT: The results show: 1) The Reynolds number of the external shear flow plays a crucial role in leukocyte deformation. Leukocyte deformation increases with Reynolds number; 2) The nucleus deforms together with the leukocyte, and the deformation

Prostate cancer metastasis to the bone occurs at high frequency in patients with advanced disease, causing significant morbidity and mortality. Over a century ago, the "seed and soil" theory was proposed to explain organ-specific patterns of metastases. Today, this theory continues to be relevant as we continue to discover factors involved in the attraction and subsequent growth of prostate cancer cells to the bone. These include the accumulation of genetic changes within cancer cells, the preferential binding of cancer cells to bone marrow endothelial cells, and the release of cancer cell che
