Role of Hydrazine-Related Chemicals in Cancer and Neurodegenerative Disease.

Hydrazine-related chemicals (HRCs) with carcinogenic and neurotoxic potential are found in certain mushrooms and plants used for food and in products employed in various industries, including aerospace. Their propensity to induce DNA damage (mostly O6-, N7- and 8-oxo-guanine lesions) resulting in multiple downstream effects is linked with both cancer and neurological disease. For cycling cells, unrepaired DNA damage leads to mutation and uncontrolled mitosis. By contrast, postmitotic neurons attempt to re-enter the cell cycle but undergo apoptosis or nonapoptotic cell death. Biomarkers of expo

Abietane-Type Diterpenoids From Salvia kiangsiensis: Isolation, Semisynthesis, and Cytotoxicity of Their Oxime Ester Derivatives.

Systematic phytochemical investigation of whole plant of Salvia kiangsiensis led to the isolation of three undescribed abietane diterpenoids (1-3) and 24 known analogues (4-27), while compounds 2 and 3 belong to the 20-norabietane skeleton. Four derivatives (24a-24d) were also obtained through structural optimization. Their structures were elucidated by a combination of spectroscopic data, chemical methods, and x-ray diffraction. Cytotoxicity screening indicated that 24c, an oxime ester derivative of 24, exerted the most potent activity against the non-small-cell lung cancer (NSCLC) cell line

Lithium-induced kidney injury and fibrosis: A versatile model to explore cellular pathways of injury and repair.

Lithium-induced kidney injury is commonly associated with the development of nephrogenic diabetes insipidus. Longer term lithium exposure is associated with the development of chronic interstitial fibrosis. The mechanisms of lithium-induced kidney injury are multifaceted, affecting many intracellular cell signaling pathways associated with cell cycle regulation, cell proliferation, and subsequent increased extracellular matrix formation and interstitial fibrosis. Amiloride has been demonstrated to have multiple pleiotropic actions, independent of its competitive antagonism of ENaC, in reducing

Advances in understanding the molecular mechanisms of borderline ovarian tumors.

Borderline ovarian tumors (BOTs), which are a special type of epithelial tumor of the ovary, lie between benign and malignant tumors and have low malignant potential. Due to the fact that the early symptoms of these tumors are relatively subtle, they are not easy to diagnose clinically. This study explores advancements in clinical detection methods and provides a comprehensive overview of molecules such as cell migration factors, cell transcription factors, cell damage repair factors, cell cycle regulators, and tumor suppressor genes that are related to the development of BOTs and their relate

Structural optimization elaborates novel potent Akt inhibitors with promising anticancer activity.

Targeting of Akt has been validated as a well rationalized approach to cancer treatment, and represents a promising therapeutic strategy for aggressive hematologic malignancies. We describe herein an exploration of novel Akt inhibitors for cancer therapy through structural optimization of previously described 4-(piperazin-1-yl)-7H-pyrrolo[2,3-d]pyrimidine derivatives. Our studies yielded a novel series of pyrrolopyrimidine based phenylpiperidine carboxamides capable of potent inhibition of Akt1. Notably, 10h exhibited robust antiproliferative effects in both mantle cell lymphoma cell lines and

Molecular Insights into Nutrient-Driven Regulation of Trophic Strategies and Life-Cycle Shifts in Bloom-Forming Mixotrophic Dinoflagellates.

Research into the molecular machinery of eukaryotic unicellular organisms (protists) offers a unique opportunity to unveil the mechanisms of intricate cellular processes and complete life cycles at the level of a single cell operating as a whole organism. Knowledge of protistan molecular biology and life-cycle shifts as adaptations to varying physical-chemical features of the environment helps resolve important issues like evaluation of the balance between external abiotic triggers and internal molecular drivers of cell development. Understanding the molecular mechanisms of these processes is

A Li metal-SiOx hybrid anode enabling synergistic plating/alloying dual-mechanism lithium storage.

Balancing specific energy and cycle durability remains a critical challenge for practical battery systems, as conventional single-mechanism anodes struggle to optimize both simultaneously. Here, we engineer a symbiotic Li-SiOx hybrid anode enabling spatiotemporally coordinated Li metal plating and Li-ion alloying. The preferential lithiated LixSi forms a Li+-conductive network, guiding and confining lithium nucleation beneath the SiOx layer. This precise control over Li plating replenishes active lithium inventory, stabilizing electrochemical reactions and minimizing Li loss. When paired with

Tailoring grain boundary stability of zinc-titanium alloy for long-lasting aqueous zinc batteries.

The detrimental parasitic reactions and uncontrolled deposition behavior derived from inherently unstable interface have largely impeded the practical application of aqueous zinc batteries. So far, tremendous efforts have been devoted to tailoring interfaces, while stabilization of grain boundaries has received less attention. Here, we demonstrate that preferential distribution of intermetallic compounds at grain boundaries via an alloying strategy can substantially suppress intergranular corrosion. In-depth morphology analysis reveals their thermodynamic stability, ensuring sustainable potenc

Subcellular localization of Pseudomonas pyocyanin cytotoxicity in human lung epithelial cells.

The Pseudomonas aeruginosa secretory product pyocyanin damages lung epithelium, likely due to redox cycling of pyocyanin and resultant superoxide and H(2)O(2) generation. Subcellular site(s) of pyocyanin redox cycling and toxicity have not been well studied. Therefore, pyocyanin's effects on subcellular parameters in the A549 human type II alveolar epithelial cell line were examined. Confocal and electron microscopy studies suggested mitochondrial redox cycling of pyocyanin and extracellular H(2)O(2) release, respectively. Pyocyanin decreased mitochondrial and cytoplasmic aconitase activity, A

Secretion of ovarian inhibin and its physiologic roles in the regulation of follicle-stimulating hormone secretion during the estrous cycle of the female guinea pig.

To characterize inhibin secretion during the estrous cycle in guinea pigs, the concentrations of plasma inhibin, estradiol, progesterone, and FSH were determined. A significant positive correlation was observed between inhibin and estradiol throughout the estrous cycle. Plasma inhibin and estradiol started to increase a few days before ovulation (Day 0 = day of estimated ovulation), and decreased after ovulation. These two hormones remained low during the luteal phase. The immunoreactivity of inhibin alpha, betaA, and betaB subunits was colocalized in the granulosa cells of one or two healthy

Impact-Tolerant Batteries Enabled by High-Conductivity Shear Thickening Electrolytes.

The application of shear thickening electrolytes (STEs) in batteries is hindered by the conventional trade-off, in which enhanced impact resistance comes at the cost of reduced ionic conductivity. Here, we overcome this barrier by introducing lithium sulfonate-functionalized mesoporous silica (M-SiO2-SO3Li) as a multifunctional filler. Unlike conventional fillers that impede ion transport, M-SiO2-SO3Li simultaneously induces shear thickening and enhances conductivity. At 15 wt.% loading, the electrolyte achieves a remarkable ionic conductivity of 10.73 mS cm-1, higher than that of filler-free

The use of the same image analysis system to detect genetic damage in human lymphocytes treated with doxorubicin in the Comet and fluoresence in situ hybridisation (FISH) assays.

Two assays, the alkaline single cell gel electrophoresis (Comet) assay and the fluorescence in situ hybridisation (FISH) of a whole chromosome or 'chromosome painting' assay have gained importance in recent years as witnessed by the increasing yield of scientific literature using these techniques. Thus, it would be useful to have one system to measure both endpoints. In the present communication, a cost-effective electronic imaging system developed by Kinetic Imaging Ltd., UK, has been used to measure, after treatment of human lymphocytes with doxorubicin, DNA damage in the Comet assay (using

Integrative Molecular Analysis to Predict Clinical Benefit of Everolimus in Patients With Gastro-Entero-Pancreatic Neuroendocrine Tumors (GEP-NETs).

IntroductionNeuroendocrine tumors (NETs) are rare and heterogeneous. Everolimus, an inhibitor of the mammalian target of rapamycin (mTOR), is widely used in gastrointestinal and pancreatic NETs (GEP-NETs). Owing to the lack of validated biomarkers, exploratory analyses of molecular pathways may help identify subgroups that derive clinical benefit from everolimus.MethodsWe conducted a retrospective observational cohort study of patients with GEP-NETs who received everolimus and underwent tumor next-generation sequencing (NGS). Genomic alterations were categorized into seven predefined signaling

Glycoconjugate strategy for GLUT-driven curcumin delivery and anticancer activity in breast cancer cells.

Selective targeting of cancer cells is a critical strategy in anticancer therapy, and the overexpression of glucose transporters (GLUTs) in malignant cells provides an attractive avenue for achieving this selectivity. In this study, we report the synthesis and biological evaluation of glycoconjugate vanadyl complexes as GLUT-directed anticancer agents against breast cancer. A Schiff base ligand (CG) was synthesized through the condensation of curcumin (C) and glucosamine (G), followed by complexation with vanadyl to form [VO(CG)₂]·5H₂O. The synthesized compounds were comprehensively characteri

Anisakis extracellular vesicles elicit immunomodulatory and potentially tumorigenic outcomes on human intestinal organoids.

BACKGROUND: Anisakis spp. are zoonotic nematodes causing mild to severe acute and chronic gastrointestinal infections. Chronic anisakiasis can lead to erosive mucosal ulcers, granulomas and inflammation, potential tumorigenic triggers. How Anisakis exerts its pathogenic potential through extracellular vesicles (EVs) and whether third-stage infective larvae may favor a tumorigenic microenvironment remain unclear. METHODS: Here, we investigated the parasite's tumorigenic and immunomodulatory capabilities using comparative transcriptomics, qRT-PCR and protein analysis with multiplex ELISA on hum

MCP5, a methyl-accepting chemotaxis protein regulated by both the Hk1-Rrp1 and Rrp2-RpoN-RpoS pathways, is required for the immune evasion of Borrelia burgdorferi.

Borrelia (or Borreliella) burgdorferi, the causative agent of Lyme disease, is a motile and invasive zoonotic pathogen adept at navigating between its arthropod vector and mammalian host. While motility and chemotaxis are well known to be essential for its enzootic cycle, the role of each methyl-accepting chemotaxis proteins (MCPs) in the infectious cycle of B. burgdorferi remains unclear. In this study, we show that mcp5, a gene encoding one of the most abundant MCPs in B. burgdorferi, is differentially expressed in response to environmental signals and at distinct stages of the pathogen's en

Polycomb Group Protein CBX7 Represses Cardiomyocyte Proliferation Through Modulation of the TARDBP/RBM38 Axis.

BACKGROUND: Shortly after birth, cardiomyocytes exit the cell cycle and cease proliferation. At present, the regulatory mechanisms for this loss of proliferative capacity are poorly understood. CBX7 (chromobox 7), a polycomb group (PcG) protein, regulates the cell cycle, but its role in cardiomyocyte proliferation is unknown. METHODS: We profiled CBX7 expression in the mouse hearts through quantitative real-time polymerase chain reaction, Western blotting, and immunohistochemistry. We overexpressed CBX7 in neonatal mouse cardiomyocytes through adenoviral transduction. We knocked down CBX7 by

Chlamydia trachomatis Inhibits Homologous Recombination Repair of DNA Breaks by Interfering with PP2A Signaling.

Cervical and ovarian cancers exhibit characteristic mutational signatures that are reminiscent of mutational processes, including defective homologous recombination (HR) repair. How these mutational processes are initiated during carcinogenesis is largely unclear. Chlamydia trachomatis infections are epidemiologically associated with cervical and ovarian cancers. Previously, we showed that C. trachomatis induces DNA double-strand breaks (DSBs) but suppresses Ataxia-telangiectasia mutated (ATM) activation and cell cycle checkpoints. The mechanisms by which ATM regulation is modulated and its co

Abnormal networks of immune response-related molecules in bone marrow cells from patients with rheumatoid arthritis as revealed by DNA microarray analysis.

INTRODUCTION: Rheumatoid arthritis (RA) is a systemic autoimmune disease characterized by chronic synovitis that progresses to destruction of cartilage and bone. Bone marrow (BM) cells have been shown to contribute to this pathogenesis. In this study, we compared differentially expressed molecules in BM cells from RA and osteoarthritis (OA) patients and analyzed abnormal regulatory networks to identify the role of BM cells in RA. METHODS: Gene expression profiles (GEPs) in BM-derived mononuclear cells from 9 RA and 10 OA patients were obtained by DNA microarray. Up- and down-regulated genes w
