Fetal cell/tissue therapy in adult disease: a new horizon in regenerative medicine.

Fetal tissue is the richest source of primordial stem cells and has several properties that make it particularly useful for transplantation. It is superior to adult (mature) tissue in certain respects. First, fetal cells are capable of proliferating faster and more often than mature, fully differentiated cells. This means that these donor cells are able to quickly reverse the lost function of the host. In addition, these fetal cells can often differentiate in response to the environmental cues around them. This is because of their location--they can grow, elongate, migrate, and establish funct

Degradation and repair of articular cartilage.

Approximately 95,000 total knee replacements and 41,000 other surgical procedures to repair cartilaginous defects of the knee are performed annually in the United States (1). The response of normal articular cartilage to injury or arthritic degeneration is often a sub-optimal repair; the biochemical and mechanical properties of the new tissue differ from the native cartilage, resulting in inadequate or altered function. It is believed that the chondrocytes from the surrounding areas, although perhaps capable of some limited migration at the damaged site, are not able to proliferate and produce

Bio-inspired synthesis of a hierarchical self-assembled zinc phosphate nanostructure in the presence of cowpea mosaic virus: in vitro cell cycle, proliferation and prospects for tissue regeneration.

Self-assembly is an important auto-organization process used in designing structural biomaterials which have the potential capability to heal tissues after traumatic injury. Although various materials having the ability to heal after injury are available, there is still a substantial need to develop new improved materials. To address this issue, we have developed hierarchical three-dimensional (3D) self-assembled zinc phosphate (Zn3(PO4)2) in the presence of cowpea mosaic virus (CPMV). Zn3(PO4)2 nanoparticles are self-assembled into nanosheets with a high degree of isotropy and then self-organ

Osteogenic differentiation of human bone marrow-derived mesenchymal cells cultured on alumina ceramics.

Gout is the most common autoinflammatory arthritis characterized by elevated serum urate and recurrent attacks of intra-articular crystal deposition of monosodium urate (MSU). Although the pathogenesis of gout is still unclear, accumulated studies indicate that genetic factors trigger gout development, including some susceptibility genes that control the production and clearance of urate and lead to hyperuricemia. However, the epidemiological evidence suggests that only less than 10% of hyperuricemia patients develop gout, indicating that other genes unrelated to the urate metabolism may also

Chimeric Antigen Receptor T-cell (CAR-T) Therapy in Autoimmune Diseases: A Narrative Review.

Autoimmune diseases are chronic immune-mediated disorders that, in their systemic forms, such as systemic lupus erythematosus (SLE), systemic sclerosis (SSc), idiopathic inflammatory myopathies, and antineutrophil cytoplasmic antibody (ANCA)-associated vasculitis (AAV), remain associated with progressive organ damage, impaired quality of life, and excess mortality despite conventional immunosuppression. A substantial proportion of patients experience refractory disease, recurrent flares, and glucocorticoid dependence, underscoring the need for therapies that address the underlying disease proc

Rotating strain field: Development of a novel mechanical stimulation device and orthogonal alignment of MC3T3-E1 cells in response to anisotropic stimulation.

Cyclic deformation of cell culture substrates induces various cellular responses, including alignment and cytoskeletal remodeling. However, most studies have focused on deformation in a fixed direction, which does not adequately replicate the complexity of in vivo mechanical environments. In this study, we propose a rotating strain field (RSF), in which the direction of extension continuously changes over time. We developed a device and culture chamber capable of applying this stimulus to cells and evaluated its performance. Deformation analysis showed that the RSF generated on the chamber bot

From Retina to Brain: Viral Tools for Targeting Retinal Cells and Mapping Visual Circuits.

The retina serves as a uniquely accessible entry point to the central nervous system, enabling both cell type-specific manipulation and circuit-level interrogation of visual pathways. Viral vectors have emerged as indispensable tools in this context, facilitating precise gene delivery, functional perturbation, and connectivity mapping across retinal and brain circuits. Among retinal neurons, retinal ganglion cells (RGCs) represent a major focus due to their role as the sole output neurons conveying visual information to central targets. Accordingly, recent advances in viral engineering includi

Breaking the Surface: Buoyant Metal-Polymer Open-Cell Hybrid Lattice Metamaterials.

Metallic lattice metamaterials have demonstrated transformative potential across biomedical, aerospace, defense, and thermal engineering. Yet their application in buoyant marine structures has remained elusive, as their open-cell architectures inherently prevent flotation, even at densities below that of water. Here, we present the first buoyant metal-polymer open-cell hybrid lattice metamaterial. The design is guided by a novel skeletal density framework, built around an environment-independent buoyancy parameter that excludes externally accessible porosity. The resulting hybrid architecture

From Degeneration to Regeneration: The Evolving Landscape of Cell-Based Tendon Repair.

Tendinopathies represent a major clinical challenge. Vasculature, neuromuscular junctions, low cellularity, and slow extracellular matrix (ECM) turnover restrict endogenous repair and predispose injured tendons to fibrosis, mechanical weakness, and reinjury. Current therapeutic strategies including rehabilitation protocols, anti-inflammatory medications, platelet-rich plasma (PRP) injections, and surgical repair primarily address symptoms or structural deficits without correcting the underlying biological limitations of tendon healing. Cell-based therapies have emerged as a promising regenerat

Biological Functions and Applications of Exosomes from Periodontal Ligament Stem Cells.

Periodontal regeneration requires the reconstruction of root cementum, periodontal ligament, and alveolar bone, which are frequently disrupted by periodontitis, trauma, and other inflammatory conditions. Although periodontal ligament stem cells (PDLSCs) have been considered promising for periodontal tissue engineering because of their multipotency, self-renewal capacity, and immunomodulatory activity, direct cell-based therapy still faces various limitations, including donor variability, culture-related changes, delivery difficulties, and safety concerns. Drug and stem cells delivery to salivary glands - a concise review.

INTRODUCTION: Systemic equilibrium, oral health, and digestion depend on the health of the salivary glands (SGs). SG disorders, such as Sjögren's syndrome, oral and maxillofacial cancer, and radiation-induced damage, are usually associated with xerostomia, which severely impacts the patient's quality of life. Current therapeutic modalities primarily provide symptomatic therapy without addressing the basic underlying tissue damage or promoting regeneration. Emerging pharmacological and stem cell treatments may restore SG function, but tailored delivery, effectiveness, and safety issues restrict

Melt electrowriting of hydrophilic/hydrophobic multiblock copolymers for bone tissue regeneration.

Bone-healing complications can occur due to large bone defects or an insufficient bone regeneration capacity. Melt electrowriting (MEW) is a potential candidate for manufacturing synthetic scaffolds that may resolve bone-healing complications. MEW can exploit various biocompatible polymers with a wide range of tissue engineering applications. Poly (ethylene oxide terephthalate)-poly(butylene terephthalate) (PEOT/PBT), a multiblock copolymer family, has emerged as a promising biomaterial to guide cell behavior, particularly in promoting bone differentiation. The polymer is known for its tunabil

Living and Injectable Porous Hydrogel Microspheres Promoting Inflammation Modulation and Extracellular Matrix Remodeling for Intervertebral Disc Regeneration.

Intervertebral disc degeneration (IDD) remains a major cause of chronic low back pain, with limited options for effective disease modification. The harsh microenvironment of the degenerated disc, characterized by excessive oxidative stress and inflammation, severely impairs the survival and reparative potential of transplanted stem cells. Here, we present an injectable, porous and multifunctional hydrogel microsphere platform (GMPB) designed to remodel the local niche and enhance stem cell-based disc regeneration. Microspheres were fabricated from a composite matrix of gelatin methacryloyl (Ge

Yes-associated protein-mediated melatonin regulates the function of periodontal ligament stem cells under oxidative stress conditions.

BACKGROUND: Human periodontal ligament stem cells (PDLSCs) regenerate oral tissue. In vitro expansion causes replicative senescence in stem cells. This causes intracellular reactive oxygen species (ROS) accumulation, which can impair stem cell function. Tissue engineering efficiency is reduced by exogenous ROS stimulation, which causes premature senescence under oxidative stress. Melatonin (MT), a powerful free radical scavenger, can delay PDLSCs senescence but may not maintain stemness under oxidative stress. This experiment examined the effects of hydrogen peroxide-induced oxidative stress o

N-acetylglucosamine sensor, Ngs1 contributes to Beauveria bassiana vegetative growth, oxidative phosphorylation, fungal development, and cell wall integrity during entomopathogen-insect interaction.

N-acetylglucosamine (GlcNAc), a key component of fungal cell walls and insect cuticles, is an important signal to activate fungal response during entomopathogen-insect interactions. Research on Ngs1, the only identified GlcNAc sensor and transducer, has been primarily restricted to Candida species. Although our previous work identified an Ngs1 homology in Beauveria bassiana, its physiological functions in entomopathogenic fungi remain largely unexplored. In this study, we unveiled the sub-localization of Ngs1 in the nucleolus. Further transcriptomic analysis revealed that Ngs1 plays a crucial

The effect of epigenetic reprogramming using MI192 HDAC inhibitor on enhancing the osteogenesis of human adipose-derived stem cells in vitro.

The ability to control stem cell function is the key to stem cell-based therapy and living tissue regeneration. In natural conditions, histone deacetylases (HDAC) are regarded as the important defining epigenetic reprogramming for stem cell differentiation. To date, human adipose-derived stem cells (hADSCs) have been widely utilised for bone tissue engineering applications. The present study aimed to examine the effect of a novel HDAC2&3-selective inhibitor, MI192, on hADSCs epigenetic reprogramming for regulating its osteogenic potential in vitro. The results confirmed that MI192 treatment re

Comprehensive Transcriptome Analysis of mRNA Expression Patterns Associated With Enhanced Biological Functions in Periodontal Ligament Stem Cells Subjected to Short-Term Hypoxia Pretreatment.

Short-term hypoxia pretreatment significantly enhances periodontal ligament stem cell (PDLSC)-based periodontal tissue regeneration by improving various cellular biological functions, but the underlying mechanisms remain unclear. In this study, based on RNA sequencing (RNA-seq), we comprehensively analyzed the possible regulatory mechanisms of the short-term hypoxic effects on the biological functions of healthy and inflammatory PDLSCs. A total of 134 and 164 differentially expressed genes (DEGs) were identified under healthy and inflammatory conditions, respectively. Functional enrichment ana
