Impairment of Hypoxia-Induced CA IX by Beta-Blocker Propranolol-Impact on Progression and Metastatic Potential of Colorectal Cancer Cells.

The coexistence of cancer and other concomitant diseases is very frequent and has substantial implications for treatment decisions and outcomes. Beta-blockers, agents that block the beta-adrenergic receptors, have been related also to cancers. In the model of multicellular spheroids formed by colorectal cancer cells we described a crosstalk between beta-blockade by propranolol and tumour microenvironment. Non-selective beta-blocker propranolol decreased ability of tumour cells to adapt to hypoxia by reducing levels of HIF1α and carbonic anhydrase IX in 3D spheroids. We indicated a double actio

Osteocyte Perilacunar/canalicular Remodeling (PLR) Drives Spatially Heterogeneous Lacunar Remodeling During and After Lactation.

BACKGROUND: Pregnancy and lactation impose substantial demands on maternal calcium homeostasis, leading to pronounced skeletal remodeling during lactation followed by recovery after weaning. Although bone mass is largely returned at the tissue level after weaning, it remains unclear whether osteocyte-level remodeling exhibits a similarly reversible pattern. Osteocytes regulate mineral mobilization through perilacunar/canalicular remodeling (PLR), which is elevated during lactation. However, its spatial and temporal regulation in response to reproduction remains poorly defined. OBJECTIVE/HYPOT

Novel Pt@PCN-Cu-induced cuproptosis amplifies αPD-L1 immunotherapy in pancreatic ductal adenocarcinoma through mitochondrial HK2-mediated PD-L1 upregulation.

BACKGROUND: Copper accumulation triggers mitochondrial-driven cell death, known as cuproptosis, offering a promising mechanism for targeted cancer therapy. Recent studies have highlighted the critical role of intratumoral copper levels in regulating the expression of programmed cell death ligand-1 (PD-L1), suggesting that copper-induced cuproptosis not only enhances cancer cell death but may also amplify the effects of anti-PD-L1 antibodies (αPD-L1). However, in tumors where monotherapy with αPD-L1 shows limited efficacy, particularly in pancreatic ductal adenocarcinoma (PDAC), the role of cop

Current Anti-Myeloma Chimeric Antigen Receptor-T Cells: Novel Targets and Methods.

Multiple myeloma (MM) treatment becomes a major challenge once triple-class or penta-refractoriness develops. Emerging immunotherapies, including bispecific antibodies or chimeric antigen receptor (CAR)-T cell therapy, are promising options for such patients. However, the requirement for specialized expertise and staff under stringent manufacturing conditions results in high costs and restricted production. This article explores the manufacturing and clinical application of CAR T-cells in MM, highlighting their potential, limitations, and strategies to enhance efficacy. CAR-T can be manufactur

Zinc peroxide-coated gold nanorods as the therapeutic adjuvants for photothermal enhanced chemodynamic therapy and metalloimmunotherapy against tumor.

INTRODUCTION: Traditional tumor therapies have limited efficacy, leading to a high mortality rate. Although immunotherapy holds considerable therapeutic promise, clinical immunotherapy for solid tumors only elicits durable responses in a minority of patients. This limitation is largely attributed to the highly immunosuppressive nature of the tumor microenvironment (TME). Notably, conventional chemodynamic therapy (CDT) is hampered by endogenous hydrogen peroxide (H2O2) shortage in tumor cells, and AZP compensates this defect via in-situ H2O2 self-supplementation to amplify hydroxyl radical gen

Loading tea polyphenols enhances the repair of human umbilical cord mesenchymal stem cell sheet after spinal cord injury.

BACKGROUND: Spinal cord injury (SCI) is a devastating central nervous system disorder that remains a global health challenge. SCI-induced oxidative stress in the postinjury microenvironment limits tissue repair by provoking the excessive production of reactive oxygen species (ROS). Tea polyphenols (TP), as a natural plant polyphenol, could effectively reduce ROS. In recent years, stem cell-based therapy combined with cell sheet technology has been widely used in the treatment of SCI. Therefore, we constructed human umbilical cord mesenchymal stem cell sheet loaded with TP (CS-TP) and evaluated

Remodeling lesions locate at sites of strong extravillous trophoblast invasion and are associated with neutrophil presence in the human first-trimester decidua.

STUDY QUESTION: Does the degree of extravillous trophoblast (EVT) invasion influence decidual tissue architecture and immune cell distribution in first-trimester decidua? SUMMARY ANSWER: Areas of pronounced morphological changes have been identified at sites of strong EVT invasion in the decidua basalis-defined as 'remodeling lesions'-and are associated with a substantially reshaped immune cell landscape. WHAT IS KNOWN ALREADY: During early human placental development, EVTs invade the decidua to facilitate placental attachment and nutrient supply to the fetus. EVT-driven decidual tissue rest

An Optimized Conversion of Spermatogonial Stem Cells Into Spinal Cord Neurons Enhances Functional Recovery in Rats After Spinal Cord Injury.

BACKGROUND: The inflammatory response following spinal cord injury (SCI) is a critical factor contributing to neural dysfunction. This response exacerbates neural damage in the injured area by facilitating the infiltration of inflammatory cells and the release of cytokines, thereby remarkably impeding neural repair and functional recovery. Spermatogonial stem cells (SSCs) have the potential for spinal cord nerve regeneration. Concurrently, olfactory ensheathing cells (OECs), a unique cell type with neurorepair potential, can effectively enhance nerve regeneration through the secretion of vario

Piezo1 mediates hypoxia-induced endometriosis fibrosis via the mtDNA -dependent cGAS-STING pathway.

BACKGROUND: Endometriosis (EMs) is a prevalent gynecological disorder characterized by the ectopic growth of functional endometrial tissue, and its fibrotic pathology represents a primary contributor to chronic pain and infertility in affected patients. Hypoxia, a hallmark of the endometriotic microenvironment, is a well-established driver of fibrogenesis. However, the precise molecular mechanisms translating hypoxic signaling into pro-fibrotic responses remain elusive. The mechanosensitive ion channel Piezo1 is recognized as a contributor to fibrosis in various diseases; whereas its specific

Zhuyu pill mitigates NAFLD through activation of IL22/JAK1/STAT3 signaling pathway.

This study aims to investigate the lipid-lowering, anti-inflammatory, and antioxidant effects of Zhuyu Pill in non-alcoholic fatty liver disease (NAFLD), and to determine whether its therapeutic action is mediated through modulation of the IL-22/JAK1/STAT3 signaling pathway. In animal experiments, C57BL/6 mice were fed a high-fat diet for 17 weeks to induce a NAFLD model. They were randomly divided into a control group, a Zhuyu Pill treatment group, and a positive control group, and the intervention lasted for 6 weeks. Mouse liver tissue and serum were collected for biochemical analysis of blo

Systematic review of gallbladder disorders: Molecular pathogenesis, biomarkers, diagnostic approaches, and emerging therapeutic strategies.

BACKGROUND: Gallbladder cancer (GBC) is the most aggressive and lethal malignancy of the biliary tract, characterized by rapid disease progression, early invasion, and poor survival rates. The unfavorable prognosis is primarily attributed to delayed diagnosis, nonspecific clinical manifestations, and limited effective therapeutic options. Although GBC is relatively uncommon worldwide, its incidence demonstrates considerable geographic variation, with particularly high prevalence observed in South Asian countries, especially northern India, Chile, and certain regions of East Asia. Chronic galls

Tau Oligomers Induce Brain Endothelial Cell Hyperpermeability and Increase NLRP3 Inflammasome Signaling and MMP-9 Activity.

OBJECTIVE: Microvascular hyperpermeability and blood-brain barrier (BBB) dysfunction is a key consideration in neurological disorders, particularly tauopathies, a group of neurodegenerative disorders driven by misfolded and aggregated tau protein. Tau pathology has been shown to activate microglial NLRP3 inflammasome, an innate immune system sensor that responds to changes in the microenvironment, including cellular stress. Increases in reactive oxygen species (ROS), for example, activate NLRP3 inflammasome signaling, which provides a platform for the maturation of caspase-1 enzyme. Mature cas

Cucurbitacin I induces immunogenic cell death and synergistically potentiates cisplatin efficacy in epithelial ovarian cancer.

BACKGROUND: Epithelial ovarian cancer (EOC) is characterized by a suppressive tumor immune microenvironment (TIME) that undermines therapeutic efficacy. Immunogenic cell death (ICD) is a promising immunotherapy strategy attributed to ameliorating the suppressive TIME. Cisplatin (CDDP), the first-line chemotherapy for EOC, is hindered by cumulative toxicity. Cucurbitacin I (CuI), a bioactive plant-derived compound, exhibits remarkable antitumor activity in preclinical models. However, its ICD-inducing capability and cisplatin-sensitizing potential in EOC remain uninvestigated. PURPOSE: This st

The Role of Iron-Overloaded Macrophages in Mesenchymal Stem Cell Senescence and Anemia in Myelodysplastic Syndromes: Protocol for an In Vitro Study.

BACKGROUND: Myelodysplastic syndromes (MDSs) are a group of highly heterogeneous myeloid clonal diseases. Anemia is the most common clinical symptom, yet its pathogenesis remains incompletely understood. Preliminary evidence suggests an increase in macrophage infiltration and iron load in the bone marrow of patients with an MDS, alongside elevated interleukin-6 (IL-6) expression in bone marrow mesenchymal stem cells (BMSCs). The Kelch-like ECH-associated protein 1 (Keap1)-nuclear factor erythroid 2-related factor 2 (Nrf2)-antioxidant response element (ARE) pathway is a critical regulator of ce

Can treatment with human mesenchymal stem cells rescue the degenerative disc phenotype? An in vitro pilot study of induced cytokine expression.

BACKGROUND CONTEXT: Given the relatively low cell density in degenerative discs, strategies intended to bolster disc cellularity through stem cell injections have come into clinical use. Stem cell therapy is meant to provide a source of viable disc cells that can promote a healthy disc phenotype. Nevertheless, there is a limited understanding of the mechanisms through which stem cell therapy impacts degeneration. PURPOSE: The objectives of this pilot study were: 1) to evaluate gene expression changes associated with an in vitro induced degenerative phenotype in human nucleus pulposus (NP) cel

Actin-binding protein drebrin E is involved in junction dynamics during spermatogenesis.

The actin-based cytoskeleton plays a critical role in the seminiferous epithelium during spermatogenesis by conferring cell shape, adhesion, structural support and cell polarity to both Sertoli and developing germ cells, which are essential for spermatogonial stem cell renewal, maintenance of the stem cell niche, cell cycle progression, mitosis, meiosis, spermiogenesis and spermiation. However, few functional studies are found in the literature, which explore the functional significance of actin dynamics in these events. This by and large is due to a lack of information on the proteins that re
