Synergistic protein-reinforced DNA hydrogels with tunable biomechanics for mechanoresponsive drug release.

DNA hydrogels are widely explored in biomedical research for their programmability and soft tissue-mimicking mechanics. However, their application in load-bearing implants is restricted by insufficient mechanical robustness, especially for mimicking the biomechanics of the nucleus pulposus, which is a naturally occurring functional tissue essential for spinal flexibility and shock absorption in the intervertebral disk. Here, we report a novel single-pot, two-step fabrication strategy in which long DNA strands produced by rolling circle amplification form an initial viscoelastic network that is

Supramolecular DNA/amino acid-based oxidase-mimetic nanocatalysts exhibiting drug degradation capability.

Developing efficient and environmentally benign approaches for the remediation of antibiotic pollutants has become a paramount research imperative, since the extensive use of antibiotics has raised serious concerns due to their potential to induce antibiotic resistance and disrupt the ecological balance. In this work, we report the self-assembly of fluorenylmethyloxycarbonyl-lysine (Fmoc-K) aggregates with natural calf thymus DNA (CT-DNA) and Cu2+ to construct a catalyst that possesses copper-dependent active sites, mirroring the catalytic function of laccase, an oxidase known for its ability

DNA methylation profiles aid to identify putative genome activation histories along lymphomagenesis.

DNA methylation (DNAm) is widely used to leverage biological information in the context of cancer. Active regulatory elements marked by increased chromatin accessibility and specific transcription factor (TF) binding, such as enhancers, show tumor-specific DNAm patterns reflecting the biological forces shaping tumorigenesis. However, DNAm changes also occur at regions that are inactive at histone-mark level in fully developed tumors. Beyond hypermethylation of promoters, these changes are often overlooked, leaving their functional relevance poorly understood. By analyzing DNAm and chromatin st

Blood DNA Methylation Predicts Long-Term Risk of Dementia in Prospective Cohorts.

Blood-based DNA methylation may help identify biological changes related to dementia risk before clinical symptoms appear. We conducted an epigenome-wide association study of incident all-cause dementia in 5,999 cognitively healthy women from the Women's Health Initiative Memory Study, 777 of whom developed dementia over up to 25 years of follow-up. Baseline blood DNA methylation was tested for association with time to dementia. One CpG site, cg05917797, was significantly associated with dementia risk, with higher methylation linked to lower risk. This association was only minimally changed af

Predominant Merkel Cell Polyomavirus DNA Detection in Essential Thrombocythemia within Myeloproliferative Neoplasms.

UNLABELLED: Acute thrombocythemic myeloproliferative disease in mice has been reported upon introduction of middle T gene expression of mouse polyomavirus. Merkel cell polyomavirus (MCPyV) is an oncogenic human polyomavirus that accounts for approximately 80% of all Merkel cell carcinomas. In this study, we assessed the presence of MCPyV DNA in fresh bone marrow (BM) aspirates from patients with myeloproliferative neoplasms (MPN) using MCPyV-specific DNA polymerase chain reaction. MCPyV DNA prevalence was significantly higher in 78 BM samples from patients with MPNs (17.9%, 14/78) than in 66 B

Mendelian randomization reveals DNA methylation-related pyroptosis genes associated with psoriasis risk.

BACKGROUND: Research has indicated a connection between pyroptosis and psoriasis, yet the specific genes involved remain largely unidentified. This study employed Mendelian randomization (MR) to evaluate the potential causal impact of both pyroptosis-related genes and DNA methylation signatures on psoriasis risk. METHODS: Pyroptosis-related genes were sourced from the GeneCards database. We integrated quantitative trait locus (QTL) data, including expression (eQTLs), DNA methylation (mQTLs), and protein expression (pQTLs). The GCST90014456 database provided genome-wide association study (GWAS

Rapid Discovery of CD38 Inhibitor via DNA-Encoded Natural Product Library Screening.

CD38 is a multifunctional enzyme that plays a pivotal role in NAD+ metabolism and calcium signaling, and its abnormal activity is closely associated with multiple myeloma, age-related metabolic decline, neurodegenerative diseases, and other disorders. Although monoclonal antibodies such as daratumumab have been approved for clinical application, their inherent limitations necessitate the development of novel small-molecule CD38 inhibitors. In this study, we employed DNA-encoded library (DEL) technology for the high-throughput screening of CD38 inhibitors, using a DEL library containing more th

Electroacupuncture confers neuroprotection against ischemic stroke via the DNA methylation-RhoGDIα axis.

Electroacupuncture (EA) has emerged as a promising alternative therapy for ischemic brain injury, yet its underlying molecular mechanisms remain incompletely understood. This study investigated the neuroprotective effects and epigenetic mechanisms of EA in a mouse model of ischemic stroke. Cerebral ischemia was induced by middle cerebral artery occlusion (MCAO) in mice. EA was applied at Baihui (GV20) and left Zusanli (ST36) acupoints following surgery. Neurological deficits were assessed, infarct volume was measured by TTC staining, and neuronal damage was evaluated by Nissl staining. RhoGDIα

CYLD-mediated DNA damage coordinates pathological cardiac hypertrophy via RIPK1-dependent signaling.

Cardiac remodeling under pressure overload involves maladaptive changes that lead to heart failure. This study investigated the role of the deubiquitinating enzyme CYLD in this process. We found that CYLD expression was significantly upregulated in hypertrophic mouse myocardium and cardiomyocytes. Silencing CYLD markedly attenuated pathological remodeling, improved cardiac function, and reduced DNA damage. Mechanistically, CYLD drives apoptosis by deubiquitinating RIPK1, a key signaling kinase. Inhibition of RIPK1 synergized with CYLD silencing to alleviate hypertrophy-associated DNA damage. O

Biomarking MELAS with neurofilament light chain and circulating cell free mitochondrial DNA.

Mitochondrial diseases are genetic disorders caused either by nuclear or mitochondrial DNA (mtDNA) alterations and characterized by high genetic and phenotypic variability. The common mtDNA m.3243 A > G variant in the MT-TL1 gene leads to clinical manifestations ranging from the classical MELAS (myopathy, encephalopathy, lactic acidosis and stroke-like episodes) syndrome to milder phenotypes such as MIDD (maternally inherited diabetes and deafness) or a spectrum of clinical features of intermediate severity defined as MELAS-Spectrum. The heterogeneous disease course makes the identification of

DNA barcoding and mitochondrial genetic diversity of mosquito vectors from central India.

BACKGROUND OBJECTIVES: Accurate discrimination of mosquito vectors is essential for entomological surveillance in India, where multiple Anopheles, Aedes and Culex species transmit malaria, lymphatic filariasis and arboviral diseases. However, routine species identifications are challenged by cryptic and morphologically similar taxa. Furthermore, population genetic information of major mosquito vectors remains elusive. METHODS: A total of 265 adult mosquitoes representing 21 species from four mosquito genera and one non-mosquito dipteran genus ( Chironomus ) used as an outgroup, were collected

Mitophagy mitigates mitochondrial DNA-induced activation of cGAS-STING in autoimmune thyroiditis.

Autoimmune thyroiditis arises from disrupted homeostasis of thyroid follicular epithelial cells and coordinated immune cell activation within the microenvironment. However, its pathogenesis is not fully understood. Here, we identify a mitochondrial (mt) DNA-cGAS-STING inflammatory axis as a driver of autoimmune thyroiditis in mice. By contrast, ubiquitin-dependent mitophagy mediated by PINK1 and Parkin was found to protect mice from disease. Mechanistically, mitochondrial dysfunction elevates mitochondrial reactive oxygen species levels, activating the ATM-CHK2 DNA damage response pathway, whi

Viral manipulation of DNA damage response signaling: Molecular mechanisms and therapeutic implications.

The DNA damage response (DDR) network maintains genomic integrity, functions as a signaling hub that viruses exploit to drive pathogenesis, and constitutes a central interface between viral infection and host cell fate. This review discusses the molecular mechanisms by which viruses subvert host DDR pathways, with an emphasis on viral replication, latency, immune evasion, and host genomic instability. Representative examples include PARP1-dependent ccDNA stabilization by hepatitis B virus (HBV), MRN complex sequestration by herpes simplex virus type 1 (HSV-1) for immune evasion, ATM signaling

APOBEC3B regulates HPV replication by inducing R-loop formation and DNA damage.

APOBECs are cytidine deaminases whose levels are increased in cells with high-risk HPV genomes and are responsible for most mutations in HPV associated cancers. APOBEC3B is a nuclear member of this family and is shown to be a positive regulator of HPV replication as well as expression. The proviral effects of A3B found in HPV positive cells contrast with its role as a restriction factor for many other viruses. Studies demonstrated that A3B can bind and regulate the formation of R-loops, which are trimeric nucleic acid structures consisting of an RNA paired with its complementary DNA strand, di

Evaluation of Artificial Intelligence-Generated DNA Aptamers Against Treponema pallidum Surface Proteins.

DNA aptamer sequences were selected in silico with assistance from artificial intelligence (AI) using the new Xelari.com platform against three known immunogenic surface proteins of Treponema pallidum designated Tpp17, Tpp47 and Tp0751. The in silico aptamers were synthesized with 5' Alexa Fluor 647 labels and shown to bind live Treponema pallidum by fluorescence microscopy and spectrofluorometry. While AI-predicted Kd values differed somewhat from measured Kd values, it is clear that the aptamers bound the T. pallidum surface which is consistent with the molecular docking models for each apta

DNA-Targeted Sonodynamic Activation Enhances Antibacterial Efficacy in Deep and Hypoxic Infections.

Deep-seated bacterial infections remain difficult to eradicate because both antibiotic delivery and photodynamic therapy are intrinsically constrained by tissue depth, biofilm protection, and hypoxic microenvironments. Sonodynamic therapy (SDT) offers noninvasive activation with clinically relevant penetration depth, but its antibacterial application has been limited by the scarcity of sonosensitizers that combine high activity, target specificity, and clinically relevant molecular scaffolds. Here, we establish TLD1433, a clinically advanced Ru(II) photosensitizer, as a DNA-targeting sonosensi

Circulating Tumor DNA in Breast Cancer: Diagnostic Insights From a Case Series.

Circulating tumor DNA (ctDNA), the tumor-derived fraction of cell-free DNA, has emerged as a valuable biomarker for monitoring treatment response, detecting minimal residual disease (MRD), and identifying early cancer recurrence. While histologic tissue diagnosis remains the gold standard for confirming malignancy, guidelines from organizations such as the National Comprehensive Cancer Network (NCCN) and the European Society for Medical Oncology (ESMO) acknowledge that ctDNA may serve as a supplemental tool in rare instances where tissue is unobtainable. In such cases, results should be interp

DNA Vaccination Against Macrophage Migration Inhibitory Factor Protecting Mice From Experimental Pancreatitis.

Objective Macrophage migration inhibitory factor (MIF) has a pivotal role in the development of gastroenterological diseases, including pancreatitis. In this study, we aimed to explore the effect of deoxyribonucleic acid (DNA) vaccination producing an auto-MIF antibody on experimental pancreatitis and to provide additional evidence that MIF affects the development of pancreatitis. Methods Mice were treated with an MIF-DNA vaccine by introducing oligonucleotides encoding a helper T epitope into the cDNA sequence of murine MIF by in vivo electroporation. Thereafter, experimental pancreatitis was

Using cell-free DNA tumor methylation score to detect and monitor cholangiocarcinoma.

BACKGROUND: Cholangiocarcinoma (CCA) is characterized by late or unclear diagnosis, high recurrence rates, and limited options for noninvasive disease monitoring. The widely used serum biomarker CA 19-9 has suboptimal sensitivity and specificity, underscoring the need for more accurate and reliable quantitative measures of tumor burden. Cell-free DNA (cfDNA) methylation represents a promising approach for longitudinal disease monitoring. METHODS: In this single-center cohort study, we evaluated a cfDNA methylation-based Tumor Methylation Score (TMS) for detecting and monitoring CCA. Twenty-fi
