Fruit color, a primary quality affecting consumer and breeder preferences, depends on a coordinated regulation of numerous genes. Nuclear factor Y (NFY) is an evolutionarily conserved heterotrimeric transcription factor complex that plays crucial roles in diverse developmental processes in plants. However, little attention has been given to members of the large NFY gene family in the pepper (Capsicum annuum) genome. In this study, 43 CaNFY genes (15 CaNFYAs, 16 CaNFYBs, and 12 CaNFYCs) were identified. Their gene structure, genomic mapping, motif composition, phylogenetic relationships, and ge

This study explored the effects of dihydromyricetin (DHM) on lipopolysaccharide (LPS)-induced intestinal injury in weaned piglets and also investigated its possible molecular mechanism. The results showed that dietary supplementation of DHM could improve the jejunum morphological structure of piglets induced by LPS, reduce jejunum mucosa inflammation and endoplasmic reticulum stress, increase jejunum mucosa antioxidant capacity and the expression of nuclear factor erythroid 2-related factor 2 (Nrf2) and improve jejunum mucosa permeability. In addition, DHM downregulated the expression of toll-

The first project of the Action Network on Measuring Population and Organizational Health Literacy was the Health Literacy Population Survey 2019-2021 (HLS19). Based on the HLS19's experience, improvements were needed to the communicative health literacy questionnaire before it could be used in the second wave of the HLS survey. Our aim was to test the comprehensibility of the Hungarian version of the revised questionnaire with cognitive interviews. Two questions caused comprehension problems: (1) discussing health information with your doctor from other sources such as internet, family and fr

The 'Viroporin' family comprises a number of mostly small-sized, integral membrane proteins encoded by animal and plant viruses. Despite their sequence and structural diversity, viroporins share a common functional trend: their capacity to assemble transmembrane channels during the replication cycle of the virus. Their selectivity spectrum ranges from low-pH-activated, unidirectional proton transporters, to size-limited permeating pores allowing passive diffusion of metabolites. Through mechanisms not fully understood, expression of viroporins facilitates virion assembly/release from infected

As an important branch of the covalent organic frameworks (COFs) family, one-dimensional COFs (1D COFs), which are formed by the ordered arrangement of confined covalent bonds in one dimension and non-covalent interactions (van der Waals force, π-π interactions, and hydrogen bonds) in the vertical two and three dimensions has aroused much attention. Compared with two-dimensional (2D)/three-dimensional (3D) COFs, 1D COFs behaved more easily dispersing and had more opportunities for active sites exposure due to their weaker interchain/interlayer interaction, modified nonlinear edge, and pore str

G protein-coupled receptors (GPCRs) are the largest and most pharmacologically important family of membrane proteins, responsible for a wide range of physiological responses through the transmission of extracellular signals. Their vital role in human health makes them key targets, with many approved drugs acting by modulating GPCRs. Natural products, known for their complex structures and biological activity, remain a major source of GPCR-targeting agents. Recent advances in computer-aided drug design (CADD) have improved the discovery of GPCR ligands, including natural compounds, through both

The histone deacetylases (HDACs) family plays a critical role in tumorigenesis and has been identified as having a significant impact on tumor immunity. Herein, we employed a fragment-centric structure-based design platform, leading to the discovery of SDFZ-8 as a highly potent HDAC1 inhibitor (IC50 = 0.4 nM). SDFZ-8 exhibits strong antiproliferative effects by enabling histone acetylation and inducing cell apoptosis. Crucially, SDFZ-8 treatment led to a significant enhancement of antitumor immunity, as evidenced by increased activation of T cells, enhanced polarization of M1-type macrophages,

Synthetic clay analogues (SCAs) of a new organosilicate layered material family, in contrast to common clays, are produced via an in situ room-temperature sol-gel route, providing the possibility for the design and synthesis of diverse, tailor-made functional groups on the surface and interior of the synthetic clay sheets. In this work, we introduce organophyllosilicates bearing different functional end groups, which are synthesized by a magnesium metal salt precursor and organosilanes such as (3-aminopropyl)-triethoxysilane (APTEOS), N-[3-(trimethoxysilyl)-propyl]-ethylenediamine (EDAPTEOS),

Azetidine, a four-membered aza-cycle, is a crucial structure in many bioactive compounds and drugs. However, their biosynthesis is frequently enigmatic. Here we report the mechanism of azetidine amino acid (polyoximic acid) biosynthesis in the polyoxin antifungal pathway. Genetic, enzymological and structural experiments revealed that PolF is a member of haem-oxygenase-like dimetal oxidase and/or oxygenase (HDO) superfamily, and this enzyme alone is sufficient for the transformation of L-isoleucine (L-Ile) and L-valine to their azetidine derivatives via a 3,4-desaturated intermediate. Mechanis

Human wingless-type MMTV integration site family member 10A (WNT10A) is a secreted glycoprotein that is involved in signaling pathways essential to ectodermal organogenesis and tissue regeneration. WNT10A was first linked to human disorders in 2006, demonstrating a WNT10a variant to be associated with cleft lip with/without cleft palate. Numerous publications have since then identified the importance of WNT10A in the development of ectodermal appendages and beyond. In this review, we provide information on the structure of the WNT10A gene and protein, summarize its expression patterns in diffe

Nephronophthisis (NPHP) is a recessive tubulointerstitial nephropathy and a leading genetic cause of kidney failure in children and young adults. The most common genetic cause is a homozygous deletion of NPHP1, which encodes nephrocystin-1, a protein essential for primary cilium structure and cell junctions. Using personalized medicine and deep phenotyping, we investigated a family with three siblings carrying a homozygous NPHP1 deletion. We compared kidney biopsy tissue and human urine-derived renal epithelial cells (hURECs) from these individuals. Bulk RNA-seq on patient hURECs revealed alte

Quinones, a ubiquitous class of organic compounds produced in all kingdoms of life, are recognized for their diverse biological functions. In plants, quinones are implicated in allelopathy, where they act as allelochemicals influencing the growth and development of neighboring plants, of which one well-documented role of quinone is in the induction of haustoria of parasitic plant roots. In this study, through an immune marker gene-based chemical biology screening, we identified Lapachol, a naphthoquinone originally isolated from species of the Bignoniaceae family. Lapachol suppresses elicitor-

Spider venom has been shown to possess a variety of pharmacological activities with promising applications in biomedical and agricultural sciences. However, the molecular structure and functions of these toxins have only been revealed for a few species. In this study, we unveiled the venom composition of Macrothele washanensis venom for the first time using transcriptomics, proteomics, and bioinformatic analyses. A total of 147,492,716 Illumina valid reads were obtained from the transcriptome of the venom glands, and 3,010,024 unigenes were assembled to predict 69 putative toxin sequences, whi

Peripheral T-cell lymphoma (PTCL) is a heterogeneous group of mature T-cell malignancies with poor prognosis. Therefore, improved therapies are urgently required to improve patient outcomes. In this study, metabolic inhibitor drug screening reveals that quinacrine elicits excellent antitumor activity both in vitro and in vivo by downregulating intracellular arginine levels in PTCL. Single-cell transcriptomic analyses reveal aberrant arginine metabolism in patients with PTCL, characterized by excessive solute carrier family 3 member 2 (SLC3A2) mediated arginine uptake preferentially in tumor ce

BACKGROUND: Marfan syndrome (MFS) is a genetic disorder affecting connective tissue, with variable incidence rates. A significant portion of cases stems from novel genetic variants, while others inherit it from affected parents.
OBJECTIVE: This study focuses on identifying the genetic cause of MFS in a specific family, using whole-exome sequencing (WES).
METHODS: A 15-year-old male with confirmed MFS was examined, showing symptoms of palpitations and severe mitral valve regurgitation. WES was performed, followed by confirmation with Sanger sequencing. Variants were assessed for pathogenicity

Crumbs homolog 1 (CRB1) is one of the key genes linked to retinitis pigmentosa and Leber congenital amaurosis, which are characterized by a high clinical heterogeneity. The Crumbs family member CRB2 has a similar protein structure to CRB1, and in zebrafish, Crb2 has been shown to interact through the extracellular domain. Here, we show that CRB1 and CRB2 co-localize in the human retina and human iPSC-derived retinal organoids. In retina-specific pull-downs, CRB1 was enriched in CRB2 samples, supporting a CRB1-CRB2 interaction. Furthermore, novel interactors of the crumbs complex were identifie

Despite their lack of a defined 3D structure, intrinsically disordered regions (IDRs) of proteins play important biological roles. Many IDRs contain short linear motifs (SLiMs) that mediate protein-protein interactions (PPIs), which can be regulated by post-translational modifications like phosphorylation. 20% of pathogenic missense mutations are found in IDRs, and understanding how such mutations affect PPIs is essential for unraveling disease mechanisms. Here, we employ peptide-based interaction proteomics to investigate 36 disease-associated mutations affecting phosphorylation sites. Our re

The photoreceptor cilium is vital for maintaining the structure and function of the retina. However, the molecular mechanisms underlying the photoreceptor cilium integrity and retinal homeostasis are largely unknown. Herein, it is shown that kinesin family member 11 (KIF11) localizes at the transition zone (connecting cilium) of the photoreceptor and plays a crucial role in orchestrating the cilium integrity. KIF11 depletion causes malformations of both the photoreceptor ciliary axoneme and membranous discs, resulting in photoreceptor degeneration and the accumulation of drusen-like deposits t
