Identification of thyroid hormone receptor homologs in the fluke Opisthorchis felineus (Platyhelminthes).

The liver fluke, Opisthorchis felineus of the Opisthorchiidae family, is a well-known causative agent of opisthorchiasis in Russia and Europe. The aim of this work was to identify genes encoding thyroid hormone receptors in O. felineus, and to analyze the expression of possible target genes in response to treatment with exogenous thyroid hormones. We identified two genes encoding thyroid hormone receptors in the O. felineus genome, THRA and THRB. The genes were differentially expressed through the life cycle. The maximal level of mRNA expression of THRA1 and THRB was observed in adult worms. T

SALL4: engine of cell stemness.

The spalt (sal) family is a class of evolutionarily conserved genes originally identified in Drosophila as homeotic genes required for embryonic development. In vertebrates, the expression of sal-like 4 (SALL4) is specifically enriched in both embryonic and adult stem/stem-like cells. SALL4 is a master regulator that contributes to cell stemness in biological development and tumor growth. Thus, Sall4 has emerged as a target for gene therapy. In addition, numerous mutations affecting the Sall4 gene have been discovered and clinically linked to a series of congenital abnormalities, such as Duane

Making high titer retroviral producer cells.

Retroviral vectors are uniquely suitable for high-efficiency gene transfer to a large target cell population and this (among other things) has led to the exploration of previously impractical strategies for somatic gene therapy (1,2). Thus, for example, cultured bone marrow can be used to recolonize mice with hematopoietic progenitors expressing a variety of genes after retroviral gene transfer (3), and efforts to achieve the same with human bone marrow may eventually result in effective therapies for sickle cell anemia, the thalassemias, and a variety of other single-gene disorders. The major

siRNA therapeutics: from hepatic delivery to extrahepatic targeting.

INTRODUCTION: Small interfering RNA (siRNA) has progressed from an experimental gene-silencing tool to an established therapeutic modality through advances in chemical modification and delivery. Lipid nanoparticles (LNP) and N-acetylgalactosamine (GalNAc)-siRNA conjugates have driven this clinical translation, particularly for liver-directed applications. Extending comparable delivery efficiency and selectivity to non-hepatocyte cell populations and extrahepatic tissues remains a major challenge. AREAS COVERED: This review examines approved and late-stage siRNA therapeutics and the delivery p

Calreticulin and myeloproliferative neoplasms: linking biology to targeted therapies.

INTRODUCTION: Calreticulin (CALR) plays important roles in protein homeostasis, immune responses, and cell signaling. Mutations in the CALR gene drive BCR:ABL1-negative classical myeloproliferative neoplasms (MPNs) through constitutive activation of the thrombopoietin receptor and generate a tumor-specific neoepitopes, providing a rationale for selective immunotherapy. AREAS COVERED: This review summarizes the biological functions of wild-type calreticulin and the molecular consequences of mutant calreticulin, including loss of endoplasmic reticulum retention, altered subcellular localization

Targeted pharmacotherapy of NF1-associated plexiform neurofibromas: current evidence and future perspectives.

INTRODUCTION: Neurofibromatosis type 1 (NF1) is a common autosomal dominant tumor-predisposition syndrome caused by loss-of-function NF1 variants. Somatic inactivation of the remaining allele in Schwann cell precursors drives plexiform neurofibromas (PN), benign but often disfiguring tumors that can progress to malignant peripheral nerve sheath tumors. Driven by constitutive RAS-MAPK activation, PN are among the most pharmacologically tractable manifestations of NF1. AREAS COVERED: This review covers the pharmacotherapy of NF1-associated PN: RAS-MAPK activation in Schwann cells; the approved

Hypoxia-Induced MicroRNAs Confer Survival Advantage in Bladder Cancer by Fine-Tuning Oncogenic Pathways.

INTRODUCTION: Bladder cancer (BC) is influenced by hypoxic conditions, which promote tumor progression and resistance to therapy. MicroRNAs (miRNAs) play key roles in regulating gene expression under hypoxia. METHODS: This study utilized an miRNA array to compare expression profiles under normoxic and hypoxic conditions. We analyzed 1,024 miRNAs, focusing on differentially expressed miRNAs. Survival analysis was performed using a cohort of 408 BC patients from TCGA (The Cancer Genome Atlas). Target prediction for hypoxia-inducible factors (HIFs) and cancer-related genes was performed. RESULT

A Rationally Designed AAV9-DM Capsid with Minimal Liver Tropism.

Adeno-associated viral vectors (AAV) are the leading gene therapy in the clinic. AAV9 has been of particular interest due to its wide tropism for multiple tissue types as well as being able to cross the blood-brain barrier and transduce central nervous system tissues. However, effectively and safely targeting extrahepatic tissue following the systemic administration of AAV9 remains a challenge due to high rates of liver transduction and liver toxicity. Thus, a crucial first step in developing a safe AAV9-based vector is to reduce liver targeting. Here we utilized rational design techniques to

Unfolding prion misfolding and the challenge of identifying effective therapeutics: is there hope on the horizon?

INTRODUCTION: Prion diseases are rare, fatal neurodegenerative disorders caused by the conformational conversion of cellular prion protein (PrP) into a pathogenic counterpart called PrPSc, that replicates by templated misfolding. The resulting misfolding cascade leads to synaptic failure and neuronal loss. Despite major advances in understanding prion structure and biology, no therapies exist. The mechanism of prion propagation challenges conventional drug discovery, limiting targets. Small molecules, immunotherapies, and gene silencing, have shown limited clinical success. Increasingly, resea

Engineering precision oncology: Targeting tumors and immune cells with lentiviral vectors.

Lentiviral vectors (LVs) are emerging as versatile tools for the efficient and stable delivery of therapeutic genes. Although VSV-G-pseudotyped LVs remain the standard for ex vivo genetic engineering, their broad tropism and serum sensitivity limit their applicability for in vivo cancer therapy, where precise targeting is essential for efficacy and safety. This review synthesizes two decades of advances in LV pseudotyping, comparing natural viral envelopes, engineered targeting strategies, and multicomponent glycoprotein systems that reshape vector tropism. We discussed donor-derived envelopes

An Integrated Nanozyme-Exosome Platform for Multi-Targeted Therapy of Metabolic-Associated Steatotic Liver Disease.

Metabolic-associated steatotic liver disease (MASLD) is a prevalent chronic liver disorder driven by a complex interplay of lipid accumulation, oxidative stress, and inflammation, for which effective targeted therapies remain limited. To address this multifactorial pathology, we developed an integrated nano-therapeutic platform, termed CMEPA, that unites three complementary components: a copper-based nanozyme with dual superoxide dismutase (SOD)- and catalase (CAT)-like activities, human umbilical cord mesenchymal stem cell-derived exosomes (UC-MSC-Exos) enriched in regulatory microRNAs, and a

Targeting PRKCN, an Essential Driver Orchestrating mTOR-IRF4 Axis Independently of Kinase Activity, in Multiple Myeloma.

Multiple myeloma (MM) remains incurable, necessitating development of novel therapeutic targets. Deregulated PRKCN is implicated in solid tumors, while its role in MM remains elusive. Here, PRKCN is identified as a super-enhancer-driven gene associated with adverse prognosis in MM. PRKCN is transactivated by NF-κB signaling intrinsically existing or exogenously provoked. Constitutive or inducible knockdown of PRKCN significantly impairs cell growth and tumorigenicity, while overcoming drug resistance. PRKCN harnesses IRF4 to exert its effect, and in turn, IRF4 directly induces PRKCN transcript

Development of a DUX4-targeting antibody oligonucleotide conjugate as a therapy for FSHD.

Facioscapulohumeral muscular dystrophy (FSHD) is an autosomal dominant muscular disease in which genetic mutations activate DUX4 expression in skeletal muscle. Currently, there are no approved therapies for FSHD. We developed Delpacibart braxlosiran (del-brax, also known as AOC 1020), an antibody oligonucleotide conjugate (AOC), for the treatment of FSHD that is designed to specifically target and reduce DUX4 mRNA in skeletal muscle. AOC 1020 is composed of DUX4 mRNA-targeting small interfering RNA (siRNA), siDUX4.6, conjugated to a human transferrin receptor 1 (TfR1)-targeting monoclonal anti

Role of Anti-GD2 Targeted PEG-b-PLGA Nanoparticles in the Treatment of MYCN Driven Neuroblastoma.

Neuroblastoma (NB) is an embryonic tumor originating from the neural crest. The most well-defined genetic alteration in NB is the overexpression of the MYCN protein. PI3K/AKT/mTOR and AURORA signaling pathways play a role in MYCN stabilization, and abnormal activation of these pathways has been identified in NB. Nanoparticle (NP) drug delivery systems targeting tumor cells directly assist in the combined delivery of agents and reducing toxicity. In this study, the aim was to develop NPs that reduce the activity of mTOR and AURORA pathways, potentially decreasing MYCN protein enhancement and st

Molecular Mechanisms of Chondrocyte Hypertrophy Mediated by Physical Cues and Therapeutic Strategies in Osteoarthritis.

Osteoarthritis (OA) is a multifactorial degenerative joint disease in which aberrant mechanical cues act in concert with metabolic dysregulation and chronic low-grade inflammation, with chondrocyte hypertrophy representing a key pathological event driving cartilage degeneration. Alterations in extracellular matrix (ECM) properties-including mechanical loading, stiffness and viscoelasticity, topological organization, and surface chemistry-regulate hypertrophic differentiation and matrix degradation in a zone-, stage-, and scale-dependent manner. Microscale measurements often reveal localized st

Inflammatory Bowel Disease: Mechanisms and Therapeutic Advances.

Inflammatory bowel disease (IBD), which includes ulcerative colitis and Crohn's disease, represents a chronic, immune-mediated pathology in which genetic susceptibility, environmental exposures, and microbial perturbations converge to destabilize intestinal homeostasis. Despite a steady global rise in incidence affecting more than 10 million people worldwide, current single-pathway biologic and small-molecule therapies still leave 30-40% of patients without durable remission, with progression to stricturing, fistulizing, or transmural complications. Yet the mechanistic integration of innate an
