AIMS/HYPOTHESIS: Metallothionein 1 (MT1) negatively regulates insulin secretion in pancreatic beta cells, and is upregulated by low non-stimulating glucose levels compared with physiological glucose stimulation in cultured rodent and human islets. Whether this regulation also occurs in vivo and its underlying mechanisms remain unclear. Here we examine whether physiological changes in blood glucose levels in vivo control islet Mt1 gene expression similarly to ex vivo observations, and identify the mechanisms involved.
METHODS: Rats fasted for 1, 2 or 3 days and rats fasted for 2 days and refed

Ischemic stroke (IS) involves persistent oxidative stress, exacerbated inflammation, and neuronal apoptosis. However, the therapeutic efficacy of current neuroprotective agents is severely restricted by the blood-brain barrier, which hinders their effective delivery to ischemic brain tissue. Although xenon (Xe) exhibits well-documented neuroprotective properties, its rapid diffusion and short in vivo retention limit its ability to match the evolving pathology of stroke. Therefore, developing Xe-based delivery systems with enhanced lesion targeting and controllable release is essential to fully

BACKGROUND: Epidemiological studies show an inverse association between cigarette smoking and Parkinson's disease (PD), suggesting a potential protective effect of smoking on PD incidence, despite the well-established and overwhelming harms of smoking to human health. We integrated genomic and proteomic approaches to investigate the causality and molecular basis of this potential relationship.
METHODS: We analyzed summary statistics from genome-wide association studies (GWAS) of smoking initiation (SmkInit), smoking intensity, and PD. Two-sample Mendelian randomization (MR) tested whether gen

UNLABELLED: Non-responsive celiac disease (NRCD) challenges clinicians due to persistent symptoms despite a gluten-free diet (GFD). We present a cross-sectional pilot study including 39 NRCD patients to describe the underlying mechanisms contributing to symptom persistence by integrating different levels of data (fecal shotgun metagenomics, mucosal integrity markers, and metabolomic profiles) and using microbial networks to unravel the community structure of the patient's microbiome. Two distinct clusters of patients were identified based on clinical and demographic variables not influenced by

Alopecia areata (AA) is a chronic autoimmune disorder characterized by non-scarring hair loss, with subtypes ranging from patchy alopecia (AAP) to alopecia totalis and universalis (AT/AU). The aim of this research is to investigate molecular features across AA severity by performing an integrated analysis of scalp transcriptomic datasets (GSE148346, GSE68801, GSE45512, GSE111061) and matched serum proteomic data from GSE148346. Differential expression analysis indicated that, relative to normal scalp, non-lesional AA tissue shows early immune activation-including Type 1 (C-X-C motif chemokine

Phosphatidylinositol phosphates are key phospholipids with a range of regulatory roles, including membrane trafficking and cell polarity. Phosphatidylinositol-4-phosphate [PI(4)P] at the Golgi apparatus is required for the budding-to-filamentous-growth transition in the human-pathogenic fungus Candida albicans; however, the role of plasma membrane PI(4)P is unclear. We have investigated the importance of this phospholipid in C. albicans growth, stress response, and virulence by generating mutant strains with decreased levels of plasma membrane PI(4)P, via deletion of components of the PI-4-kin

Postharvest fruits are highly susceptible to rapid senescence and microbial decay, a process primarily driven by intense respiration and endogenous ethylene signaling. While conventional packaging provides basic physical protection, the shift towards multi-component functional interfaces offers dynamic solutions for physiological regulation. This review provides a critical analysis of advanced functional packaging strategies, focusing on the synergistic integration of natural biopolymers, deep eutectic solvents (DESs), and nanoporous structures. We dissect the intrinsic mechanisms by which the

BACKGROUND: Effective treatment of tuberculosis is often hampered by anti-tuberculosis drug-induced liver injury (ATB-DILI). This study aimed to identify the key targets and mechanisms by which gut microbiota metabolites regulate ATB-DILI.
METHODS: Integrative bioinformatics was used to screen for common targets shared by gut microbiota metabolites and ATB-DILI. The target list was refined through protein-protein interaction network and hub gene analyses. Functional enrichment analysis was performed on the core targets, followed by construction of a microbe-metabolite-target regulatory networ

Feline panleukopenia virus (FPV) causes severe, highly contagious infectious diseases in domestic cats, yet the comprehensive pathogenic mechanisms of circulating field isolates remain incompletely elucidated. Here, two contemporary FPV field strains (F513 and F1105) were isolated and genetically characterized from clinical samples of FPV vaccinated diarrheic cats. In vivo pathogenicity assays demonstrated that both strains induced typical FPV clinical signs, with viral replication predominantly localized to the gastrointestinal tract and shedding primarily occurring via the fecal-oral route.

Alzheimer's disease (AD) is now recognized as a biologically heterogeneous syndrome in which amyloid-β pathology, Tau-related alterations, and neuroinflammation are key interconnected processes. Microglia, astrocytes, oligodendrocytes, complement, inflammasomes, neurovascular cells, and peripheral immunity are implicated as both adaptive and disease-promoting contributors to the pathophysiology of AD. Here, we highlight mechanisms by which innate immunity and peripheral immune cells influence amyloid β-peptide metabolism, Tau pathophysiology, excitatory synaptic function, oxidative stress resp

Hepatocellular carcinoma (HCC) is the most common primary liver cancer and remains a major cause of cancer-related death worldwide. Corosolic acid (CA), an ursane-type pentacyclic triterpenoid with antidiabetic, anti-inflammatory, and antitumour activities, has attracted interest as a potential natural compound for HCC treatment. This review critically evaluates CA as a molecular lead for HCC by integrating its physicochemical characteristics with direct preclinical evidence, cross-model hypotheses, and development constraints. Current studies indicate that CA suppresses HCC cell proliferation

Plasma membrane phospholipid asymmetry is usually described as a homeostatic arrangement that preserves surface identity. In secretory cells, however, this arrangement is briefly and locally relaxed when intracellular calcium rises. Evidence from adrenal chromaffin cells and cerebellar synapses now places this transient remodeling at a specific point in the vesicle cycle: it is largely dispensable for membrane fusion, yet it is required for efficient compensatory endocytosis. In both systems, loss of phospholipid scramblase 1 (PLSCR1) prevents stimulus-dependent phosphatidylserine exposure wit

Synapses serve as the central functional components mediating information transmission, integration, and storage within the central nervous system (CNS). Their functionality depends on the synergistic interplay of the presynaptic membrane, synaptic cleft, and postsynaptic membrane-three structures that collectively sustain neurotransmitter secretion, postsynaptic signaling, and synaptic plasticity. Of note, synaptic impairment represents an early, shared pathological hallmark across aging and age-related neurodegenerative disorders like Alzheimer's disease (AD), Parkinson's disease (PD), Hunti

Nuclear factor κB (NF-κB) is a context-dependent transcriptional network that integrates inflammatory, developmental, metabolic and genotoxic signals. In breast cancer, persistent or stimulus-coupled NF-κB activity can support proliferation, survival, epithelial-mesenchymal plasticity, invasion and therapy resistance, but the magnitude and biological consequences of activation differ across molecular subtypes and cellular compartments. This review critically examines canonical and non-canonical NF-κB signalling, emphasizing IKK/NEMO-dependent IκBα turnover, NIK-IKKα-dependent p100 processing,

Infected burn wounds are characterized by bacterial invasion, oxidative stress, and persistent inflammation, which severely impair tissue regeneration. Herein, we report a crosslinker-free, hydrogen-bonded cationic guar gum hydrogel (CBBM) co-loaded with berberine (BBR) and MnO2-coated black phosphorus nanosheets (BPNS@MnO2) for the microenvironment-adaptive treatment of infected burn wounds. The dynamic guar gum network endowed the hydrogel with injectability, self-healing ability, and conformal adaptability. BPNS@MnO2 exhibited pH-dependent enzyme-like activities, including OXD-like antibact

Recurrent oral ulcer (ROU), a chronic inflammatory disorder of the oral mucosa characterized by recurrent painful ulcerations, significantly compromises patients' dietary intake, speech, and quality of life. While its precise etiology remains elusive, the dynamic interplay between oxidative stress and inflammatory responses has emerged as a pivotal mechanism underlying ROU pathogenesis. Gancao Xiexin Decoction (GCXXD), a traditional Chinese formulation with demonstrated anti-inflammatory and antioxidant properties, has shown clinical efficacy in gastrointestinal disorders such as inflammatory
