Microfluidic ocular formulation of voriconazole designed for hospital production.

Fungal keratitis is a serious corneal infection with a limited therapeutic panel. Voriconazole, an azole antifungal, has been shown to be effective against many of the fungal strains involved in this type of infection. The absence of an ocular dedicated formulation on the market has led to the use of the sterile commercial cyclodextrin based formulation as a hospital preparation with some drawbacks. The aim of this study was to design a dedicated formulation for ocular delivery using oil-in-water nanoemulsion. A 0.4% voriconazole nanoemulsion was formulated with an oil and two surfactants, com

Rutin-luteolin flavonoid-encapsulated chitosan nanoparticles: a novel strategy to combat multidrug-resistant Pseudomonas aeruginosa.

Pseudomonas aeruginosa (P. aeruginosa) is a major opportunistic pathogen with strong biofilm-forming ability and high resistance to antibiotics. Natural flavonoids such as rutin and luteolin possess antimicrobial potential, but their poor solubility and bioavailability limit therapeutic applications. This study evaluated the antibacterial and anti-biofilm efficacy of rutin–luteolin encapsulated chitosan nanoparticles (RUT-LUT-CS) against clinical and reference strains of P. aeruginosa. RUT-LUT-CS nanoparticles were spherical with an average size of 285 nm (FESEM), hydrodynamic diameter of 470

Antimicrobial strategies of nanoparticles and chelating agents for mitigating Staphylococcus spp. contamination on slaughterhouse surfaces.

The rise of multidrug-resistant (MDR) staphylococci in the food industry, particularly in slaughterhouses, necessitates the development of innovative disinfection strategies. To address the challenge of antimicrobial resistance, this study investigated formulations based on the combination of nanoparticles (NPs) with chelating agents as a robust alternative to conventional disinfectants. The synergistic antimicrobial potential of NPs combined individually with chelating agents, specifically EDTA or a disinfection formulation (HLE, based on hydrogen peroxide, lactic acid and EDTA), against plan

Discovery, evolution, and environmental co-selection of novel msrA-harboring plasmids in multidrug-resistant Staphylococcus from retail foods.

The escalating prevalence of antimicrobial resistance in food-related pathogens constitutes a critical challenge to global public health and food safety. To elucidate the mechanisms underlying the high erythromycin resistance rate observed in our previous surveillance of Staphylococcus in retail foods, we conducted a comprehensive genomic investigation of msrA-positive Staphylococcus from a nationwide retail food collection in China. We identified 102 msrA-positive isolates, all of which exhibited a multidrug-resistant (MDR) phenotype, with a significantly higher prevalence in aquatic products

Co-occurrence of biofilm formation, acid tolerance, and antibiotic resistance in environmental Escherichia coli associated with lettuce.

BACKGROUND: Environmental niches represent important reservoirs of Escherichia coli with stress-adaptation traits that support persistence outside the host. Contaminated irrigation water and soils can facilitate transfer to fresh produce, where bacterial survival may reduce the effectiveness of downstream control measures. This study investigated the co-occurrence of biofilm formation, acid tolerance, and antibiotic resistance (AR) in environmental E. coli and their contribution to persistence along the farm-to-produce continuum. RESULTS: Eighteen E. coli isolates recovered from irrigation wa

Candida (Candidozyma) auris strains exposed to azole fungicides become less susceptible to manogepix.

Candida auris is a multidrug-resistant fungal pathogen responsible for invasive nosocomial infections with high mortality rates. Recent detection of C. auris in natural environments suggests the existence of an environmental reservoir, prompting investigation into the potential role of azole fungicides used for plant protection in the development of resistance to antifungals used in human medicine. Here, we assessed the impact of azole fungicides on in vitro resistance development in two C. auris strains (B11220 and B11221) through sequential exposure to epoxiconazole, propiconazole, or tebuco

In vitro antibiofilm and intracellular activity of delafloxacin against Staphylococcus aureus and Staphylococcus epidermidis in bone and joint infections.

Staphylococcal bone and joint infections (BJIs) represent significant challenges for treatment. Fluoroquinolone-based combination therapies, especially involving levofloxacin, are pivotal antimicrobials in their management. However, resistance to levofloxacin is prevalent in methicillin-resistant strains. Delafloxacin, a novel broad-spectrum fluoroquinolone, may offer a therapeutic alternative. This study aimed to investigate the in vitro antibiofilm and intracellular activities of delafloxacin in a bone context. The antibiofilm activity of delafloxacin was assessed using two pairs of levoflox

A light-activated nickel(II)-antibiotic conjugate for synergistic antibacterial therapy.

Herein, we report a molecular conjugate, Ni-1-Cip, by linking the antibiotic ciprofloxacin to a Ni(II) scaffold for treatment of multidrug-resistant (MDR) bacterial infections using photothermal therapy (PTT). The localized hyperthermia generated by the Ni(II) core potentiates the antibacterial activity of the ciprofloxacin, leading to eradication of methicillin-resistant S. aureus (MRSA) as demonstrated in a murine model.

Antimicrobial Stewardship in Veterinary Ophthalmology: Making Antimicrobial Resistance Visible.

Antimicrobial resistance is increasingly recognized in veterinary ophthalmology, yet its development remains largely unnoticed in daily clinical practice. Although resistant ocular pathogens are reported worldwide, antimicrobial prescribing decisions are frequently made without feedback on local resistance patterns. This Viewpoint discusses aggregated antibiograms as a practical tool to improve situational awareness and support antimicrobial stewardship in veterinary ophthalmology. While antibiograms do not quantify reductions in antimicrobial resistance, their systematic use can provide conte

Multidrug-Resistant Escherichia coli in the Swine Production Chain: Genomic Evidence of Abattoir-Associated Dissemination.

Antimicrobial resistance (AMR) is a rising public health issue that compromises human, animal, and environmental health. This study assessed AMR in the swine production chain in Costa Rica using commensal E. coli as an indicator microorganism. Sampling of feces, carcass, and lymphoid tissue at exporting swine abattoirs yielded 269 presumed E. coli isolates identified by lactose-positive growth on MacConkey agar and indole production. A subset of isolates (n = 40) was confirmed as E. coli by VITEK® 2 GN testing. Antimicrobial susceptibility testing against 18 antibiotics was performed using the

Biosynthesis mechanisms, antimicrobial effects, and characterization of silver nanoparticles produced by bacterial and actinomycete isolates derived from a gold mine.

BACKGROUND: Silver nanoparticles (AgNPs) are widely studied for their antimicrobial properties, and microbial synthesis offers an eco-friendly and scalable alternative to conventional methods. While Streptomyces spp. are well-known producers, little is known about AgNP biosynthesis by microorganisms adapted to metal-rich environments such as gold mines, which may represent a largely unexplored source of microorganisms with potential for AgNP biosynthesis. This study investigated the biosynthetic potential of bacterial and actinomycete isolates recovered from the Sukari Gold Mine, Egypt, with a

Differential membrane lipid disruption by lipopeptide antibiotics, colistin and turnercyclamycins.

Lipopeptide natural products are essential agents against multidrug-resistant bacteria, but their clinical utility is often constrained by toxicity and resistance. Here, we compare the mechanisms of action of two superficially similar lipopeptide antibiotics: colistin, a last-line treatment for Gram-negative infections, and turnercyclamycins, a new class active against certain colistin-resistant strains. Both antibiotics require lipopolysaccharide (LPS) biosynthesis, even when LPS transport to the outer membrane (OM) is impaired. Colistin rapidly disrupts both the OM and the cytoplasmic membra

Broad spectrum antimicrobial nanoparticles with low toxicity to prevent biofilm formation on urologic devices.

Antimicrobial coatings for medical implants are critical in preventing device failures and infections. Antibiotics are often used as prophylactic or coatings but fail to prevent biofilm formation and drive antibiotic resistance. Herein, the antibacterial and antibiofilm activities of different polyhydroxy fullerene-based metal nanoparticle coatings on polyurethane discs were quantified after exposure to Escherichia coli. Gold-silver nanoparticles (GSNP) exhibited superior antibacterial activity compared to other silver-containing nanoparticles. GSNPs were evaluated against Escherichia coli, En
