[Effect of acupuncture intervention on neurological function, cerebral microglia activation and secondary nerve damage in traumatic brain injury rats].

OBJECTIVE: To observe the effect of acupuncture on activities of microglia in traumatic brain injury (TBI) rats. METHODS: Fifty-four male SD rats were randomly and equally divided into normal control, model and acupuncture groups according to the random number table (n=18 rats in each group). The TBI model was established by using a free fall brain injury striking device after exposing the local cranial bone (to induce the left parietal cerebral contusion). Acupoints "Baihui" (GV20), "Shuigou" (GV26), "Fengfu" (GV16), "Yamen" (GV15) and bilateral "Hegu" (LII4) were stimulated intensively by t

Small-molecule hypoxia therapy in mitochondrial disease.

In this issue of Cell, Blume et al. provide compelling rationale for pursuing pharmacologic optimization of a small-molecule "HypoxyStat," which left-shifts the oxyhemoglobin dissociation curve in red blood cells in an attempt to induce an effective and sustained reduction of chronic tissue hyperoxia in primary mitochondrial disease (PMD) and was well-tolerated and effective for both pre-symptomatic and advanced disease treatment to extend survival and improve neurologic outcomes in a mouse model of Leigh syndrome spectrum.

Varicella zoster virus transcription in latently-infected human ganglia.

Varicella zoster virus (VZV) causes childhood chickenpox, becomes latent in cranial, dorsal root and autonomic ganglia, and can reactivate decades later to cause shingles and other serious neurological complications. Herein, we summarize investigations conducted over the past decade that have identified virus genes expressed in latently-infected human ganglia. A model of VZV gene regulation during latent infection was tested and future directions in the study of VZV latency are discussed.

Altered circadian rhythmicity is an early sign of murine dietary thiamine deficiency.

To determine if circadian clock function is affected by thiamine deficiency, the rhythm of locomotor (wheel-running) activity was measured in a murine model of dietary thiamine deficiency. About 1 month before the expected onset of overt neurological illness, locomotor rhythmicity in deficient animals exhibited a shortened free-running period without a change in amplitude. This effect was fully reversible by thiamine administration. Disordered circadian timekeeping may contribute to altered physiological responses in Wernicke's encephalopathy.

Multidisciplinary management of dystonia misdiagnosed as hysteria.

Dystonia in the pediatric age group can be confused with hysteria, particularly when it occurs in an emotionally disturbed child with a negative family history of dystonia. A 20-year-old girl with a 12 year history of DMD is described. From age 12 to 17 she was housed in a mental institution after a misdiagnosis of hysteria was made. The progressive nature of DMD and the important emotional components are stressed. The multidisciplinary management model is discussed as a valuable method in the treatment of this chronic neurological disorder.

Machine learning approaches for risk prediction in aortic dissection: a systematic review and meta-analysis.

BACKGROUND: Aortic dissection (AD) is a life-threatening cardiovascular emergency with high morbidity and mortality. Accurate risk prediction is essential for timely intervention, yet traditional statistical models often fail to capture the complex, nonlinear interactions inherent in AD pathophysiology. In recent years, machine learning (ML) has emerged as a promising approach to improve prognostic accuracy. However, the overall performance, methodological quality, and clinical applicability of ML-based prediction models for AD have not been comprehensively evaluated. OBJECTIVE: This systemat

Improving outcomes in patients with bipolar depression: a comprehensive review.

Only 3 medications are currently approved in the US for acute bipolar depression: 2 atypical antipsychotics and a combination atypical antipsychotic-selective serotonin reuptake inhibitor. Metabolic, neurologic, and hormonal adverse events are associated with all of the atypical antipsychotics approved for this indication. However, these agents differ in their propensity to cause weight gain or other side effects that significantly impact a patient's physical health and ability to function, and the selection of medication-which may also include a mood stabilizer-as well as other forms of treat

Iron supplementation aggravates periventricular cystic white matter lesions in newborn mice.

Periventricular leukomalacia (PVL) is the main cause of neurological impairment in premature newborns. The pathogenesis of PVL remains unclear but may involve glutamate excitotoxicity and free radical production. Oxygen and iron, which are widely used in premature newborns, are oxidizing agents with a potential for promoting free radical production. We previously described a mouse model of excitotoxic neonatal white matter lesions mimicking several aspects of human PVL. In the present study, we used this mouse model to investigate whether iron pretreatment or 100% oxygen exposure worsened exci

A mouse model reproducing the pathophysiology of neonatal group B streptococcal infection.

Group B streptococcal (GBS) meningitis remains a devastating disease. The absence of an animal model reproducing the natural infectious process has limited our understanding of the disease and, consequently, delayed the development of effective treatments. We describe here a mouse model in which bacteria are transmitted to the offspring from vaginally colonised pregnant females, the natural route of infection. We show that GBS strain BM110, belonging to the CC17 clonal complex, is more virulent in this vertical transmission model than the isogenic mutant BM110∆cylE, which is deprived of hemoly

Correlation of reaching and grasping kinematics and clinical measures of upper extremity function in persons with stroke related hemiplegia.

Timed measures of standardized functional tasks are commonly used to measure treatment effects in persons with upper extremity (UE) paresis due to stroke. The effectiveness of their ability to measure motor recovery has come into question because of their inability to distinguish between motor recovery and compensations. This paper presents three linear regression models generated from twelve kinematic measures collected during the performance of a two phase reach/grasp and transport /release activity as performed by 21 persons with upper extremity hemiparesis due to chronic stroke. One of the

Characterizing brain network alterations in cervical spondylotic myelopathy using static and dynamic functional network connectivity and machine learning.

BACKGROUND: Cervical spondylotic myelopathy (CSM) is a debilitating condition that affects the cervical spine, leading to neurological impairments. While the neural mechanisms underlying CSM remain poorly understood, changes in brain network connectivity, particularly within the context of static and dynamic functional network connectivity (sFNC and dFNC), may provide valuable insights into disease pathophysiology. This study investigates brain-wide connectivity alterations in CSM patients using both sFNC and dFNC, combined with machine learning approaches, to explore their potential as biomar

Comparative assessment of Ca2+ oscillations in 2- and 3-dimensional hiPSC derived and isolated cortical neuronal networks.

Human induced Pluripotent Stem Cell (hiPSC) derived neural cells offer great potential for modelling neurological diseases and toxicities and have found application in drug discovery and toxicology. As part of the European Innovative Medicines Initiative (IMI2) NeuroDeRisk (Neurotoxicity De-Risking in Preclinical Drug Discovery), we here explore the Ca2+ oscillation responses of 2D and 3D hiPSC derived neuronal networks of mixed Glutamatergic/GABAergic activity with a compound set encompassing both clinically as well as experimentally determined seizurogenic compounds. Both types of networks a

Rapid Dynamic Naturalistic Monitoring of Bradykinesia in Parkinson's Disease Using a Wrist-Worn Accelerometer.

Motor fluctuations in Parkinson's disease are characterized by unpredictability in the timing and duration of dopaminergic therapeutic benefits on symptoms, including bradykinesia and rigidity. These fluctuations significantly impair the quality of life of many Parkinson's patients. However, current clinical evaluation tools are not designed for the continuous, naturalistic (real-world) symptom monitoring needed to optimize clinical therapy to treat fluctuations. Although commercially available wearable motor monitoring, used over multiple days, can augment neurological decision making, the fe

A Population Model of Deep Brain Stimulation in Movement Disorders From Circuits to Cells.

For more than 30 years, deep brain stimulation (DBS) has been used to target the symptoms of a number of neurological disorders and in particular movement disorders such as Parkinson's disease (PD) and essential tremor (ET). It is known that the loss of dopaminergic neurons in the substantia nigra leads to PD, while the exact impact of this on the brain dynamics is not fully understood, the presence of beta-band oscillatory activity is thought to be pathological. The cause of ET, however, remains uncertain, however pathological oscillations in the thalamocortical-cerebellar network have been l

Sex differences in the associations between lifestyle, intestinal permeability and brain health in middle-aged adults.

BACKGROUND: Evidence suggests that brain health is related to intestinal health. Increased intestinal permeability (IP) biomarkers as risk factors for reduced brain health, considering sex differences in the gut-brain axis, in middle-aged adults has not been studied. AIM: To evaluate the associations between IP biomarkers and brain health in a middle-aged cohort of healthy women and men. METHODS: 827 participants from the Barcelona Brain Health Initiative (BBHI) study with a complete neuropsychological evaluation, neurofilament light-chain (NfL), and brain-MRI data were studied (median age:

IFITM3 knockout alleviates neuronal parthanatos by restoring astrocytic glutamate uptake in intracerebral hemorrhage mice.

INTRODUCTION: Intracerebral hemorrhage (ICH) causes severe neurological deficits mainly attributable to secondary brain injury. Parthanatos is a subtype of regulated cell death triggered by glutamate excitotoxicity. Interferon-induced transmembrane protein 3 (IFITM3) is an immune regulatory molecule involved in neuronal death in various neurodegenerative disorders. However, it remains unclear whether and how IFITM3 and parthanatos participate in secondary brain injury after ICH. OBJECTIVES: This study aims to investigate whether IFITM3 aggravates neuronal parthanatos by impairing astrocytic g
