Neurofibromatosis type 1-associated unusual pleomorphic astrocytoma displaying continual malignant progression.

Patients with neurofibromatosis type 1 (NF1) often have gliomas as a complication, most of which are benign pilocytic astrocytomas which have arisen in optic pathways. In the present case, a 17-year-old girl (at death) with stigmata of NF1, initially had a bulky tumor mass in the left thalamus, developing into the lateral ventricle, at 13 years of age. Partially resected tissue samples showed pleomorphic astrocytoma with abundant xanthoma cells and degenerative structures such as Rosenthal fibers (RF) and eosinophilic granular bodies. Fine eosinophilic granules identical to RF, both immunophen

Spatiotemporal metabolic networks in a preclinical model of pancreatic cancer and associated cachexia.

Cachexia or body wasting is a significant contributing factor to the morbidity and mortality of pancreatic cancer. Despite an unmet need, its metabolic underpinnings remain poorly understood. Here, we characterize a mouse model that recaptures the longitudinal progression of human cachexia during pancreatic cancer development. We profile the metabolomes of pre-, early- and late-stage cancer-associated cachexia in male and female pancreas, its interstitial fluid (IF), plasma, liver, adipose tissue and skeletal muscle. We find that each tissue has a unique metabolome and trajectory across stages

ANKRD49 promotes immune evasion in lung adenocarcinoma by activating the MEF2A/PD‑L1 axis.

Non‑small cell lung cancer (NSCLC), particularly lung adenocarcinoma (LUAD), exhibits a high rate of primary resistance to immune checkpoint inhibitors, underscoring the need to decipher the molecular mechanisms of immune evasion. Here, we identify ANKRD49 as a critical regulator of PD‑L1 expression and immunosuppression in LUAD. Immunohistochemical analysis of clinical LUAD specimens revealed that high ANKRD49 expression correlates with reduced CD4+ and CD8+ T cell infiltration, increased FOXP3+ regulatory T cells, and elevated PD‑L1 levels. Functional experiments demonstrated that ANKRD49 ov

Transgelin Expression in Activated Cancer-Associated Fibroblasts Regulates Stromal Contractility and Promotes Colon Cancer Progression.

Transgelin is an actin-binding protein that promotes cancer progression via activation of cancer-associated fibroblasts and has been identified as a prognostic marker. However, its distribution and functional role in colon cancer remain unclear. In this study, we aimed to elucidate the mechanistic role of transgelin in colon cancer progression by focusing on its functional impact in cancer-associated fibroblasts. Tissue microarrays from 359 human colon cancer tissues were investigated to elucidate the clinical importance of transgelin expression in cancer stroma. We focused on transgelin in fi

Impact of CT-Based Body Composition Analysis on Postoperative Survival in Patients Undergoing Colorectal Cancer Surgery.

BackgroundColorectal cancer (CRC) remains a leading cause of cancer-related mortality worldwide, underscoring the need for reliable prognostic biomarkers. Emerging evidence suggests that body composition parameters may influence cancer outcomes. This study aimed to evaluate whether low skeletal muscle mass, high visceral fat, and their coexistence predict long-term outcomes in patients with CRC.MethodsThis retrospective study included 103 patients with pathological stage III CRC who underwent curative resection. Skeletal mass index (SMI) and visceral fat index (VFI) were calculated from preope

Potential biomarkers and preventative interventions for muscle wasting in adolescent and young adults with cancer: A translational study.

Adolescent and young adults (AYAs) with sarcoma and our mouse model were used to evaluate early and late effects of doxorubicin (Dox) on skeletal muscle and the impact of physical activity on preventing Dox-induced damage. Skeletal muscle changes in 46 AYAs were quantified using computed tomography scans pre- and post- high-dose Dox treatment. Physical activity was monitored via wearable device. Patient blood samples were analyzed for myo-miRNA changes. In mice, we analyzed acute and late effects of Dox vs. Dox + exercise on skeletal muscle histology, Z-disc morphology, collagen deposition, ch

Cancer IDO1-Mediated Tryptophan-Kynurenine Metabolic Reprogramming to Drive Skeletal Muscle Atrophy and Cachexia Acceleration.

BACKGROUND: Cancer cachexia is a debilitating syndrome characterized by severe skeletal muscle wasting, which significantly impairs patient quality of life and survival. Indoleamine 2,3-dioxygenase 1 (IDO1), a key enzyme in tryptophan (Trp) metabolism, is often upregulated in cancers, but its specific role in driving lung cancer-associated cachexia remains inadequately defined. This study investigated the mechanistic role of Ido1 in cancer cachexia and evaluated the therapeutic potential of its inhibition. METHODS: We established Lewis lung carcinoma (LLC) models in C57BL/6 mice using wild-ty

Cachexia-induced alterations of miR-27a-3p drive cell-type specific effects in FAPs and tumor cells that coincide with muscle wasting.

The contribution of muscle-resident stromal cells to cancer-associated muscle wasting is poorly understood. We characterized the role of fibroadipogenic progenitors (FAPs) in pancreatic cancer-associated cachexia and investigated how dysregulated microRNAs in the tumor and FAPs drive muscle wasting. In cancer-bearing mice, FAPs engage in a chronic pro-inflammatory and pro-adipogenic program coinciding with muscle wasting. Additionally, in vitro and in vivo findings suggest that FAPs from cachectic mice cause muscle wasting, indicating that they may release pro-atrophic factors. Cancer-induced

Therapeutic Gfral silencing via antisense oligonucleotides ameliorates cancer-associated cachexia and extends survival in tumor-bearing mice.

Cancer cachexia is a life-threatening wasting syndrome that can be driven by tumor-derived GDF15 signaling through the brainstem-restricted receptor GFRAL. Despite its clinical importance, central genetic interventions targeting this axis remain limited. Here, we develop a potent antisense oligonucleotide (Gfral-ASO, A427) that specifically silences Gfral expression in the central nervous system. Using orthotopic mouse models of severe colon (MC38) and pancreatic (KPC and MPC1) cancers, we show that a single intracerebroventricular administration of Gfral ASO markedly blunts GFRAL expression a

The association between body composition and overall survival in patients with advanced non-small cell lung cancer.

Nutritional status is associated with prognosis in a variety of cancers. Studies analyzing the association between the measurements of skeletal muscle and adipose tissue obtained from Computerized Tomography (CT) images at the time of diagnosis of advanced non-small cell lung cancer (NSCLC) and overall survival (OS) are relatively few. Data from 425 patients diagnosed with advanced NSCLC between January 2016 and December 2017 were retrospectively analyzed, with an average follow-up of 15.3 months. To outline the patient's chest CT plain image at the time of diagnosis, skeletal muscle and subcut

Nutritional status in retroperitoneal sarcoma: Implication of prognostic Nutritional Index (PNI) and skeletal muscle index (SMI) on postoperative and oncological outcomes.

BACKGROUND AND AIMS: Nutritional status is a critical yet underexplored factor in retroperitoneal sarcoma (RPS) care. Traditional anthropometric indicators such as BMI are unreliable in this setting due to tumor-related weight masking. This study aimed to assess the prognostic impact of preoperative nutritional status in RPS patients using the Prognostic Nutritional Index (PNI) and Skeletal Muscle Index (SMI). METHODS: A retrospective cohort study was conducted on 250 RPS patients treated at a high-volume sarcoma centre between 2010 and 2023. Nutritional status was assessed preoperatively via
