Cortical Correlates of Balance During Single and Dual Tasks in Huntington's Disease: A Preliminary fNIRS Study.

ObjectiveExamine cortical activation patterns in Huntington's disease (HD) under single-task (ST) and dual-task (DT) balance conditions compared to controls using portable functional near-infrared spectroscopy (fNIRS).BackgroundIndividuals with HD have difficulty multitasking while performing balance tasks, so previously automatic tasks may require more attentional resources to maintain stability and prevent falls. Our understanding of the neural mechanisms underlying the relationship between impaired cognition and balance in HD is minimal. fNIRS provides a noninvasive means to functionally im

Claustrum and dorsal endopiriform cortex complex cell-identity is determined by Nurr1 and regulates hallucinogenic-like states in mice.

The Claustrum/dorsal endopiriform cortex complex (CLA) is an enigmatic brain region with extensive glutamatergic projections to multiple cortical areas. The transcription factor Nurr1 is highly expressed in the CLA, but its role in this region is not understood. By using conditional gene-targeted mice, we show that Nurr1 is a crucial regulator of CLA neuron identity. Although CLA neurons remain intact in the absence of Nurr1, the distinctive gene expression pattern in the CLA is abolished. CLA has been hypothesized to control hallucinations, but little is known of how the CLA responds to hallu

TREK-1 inhibition promotes synaptic plasticity in the prelimbic cortex.

Synaptic plasticity is one of the putative mechanisms involved in the maturation of the prefrontal cortex (PFC) during postnatal development. Early life stress (ELS) affects the shaping of cortical circuitries through impairment of synaptic plasticity supporting the onset of mood disorders. Growing evidence suggests that dysfunctional postnatal maturation of the prelimbic division (PL) of the PFC might be related to the emergence of depression. The potassium channel TREK-1 has attracted particular interest among many factors that modulate plasticity, concerning synaptic modifications that coul

Body mass variability is represented by distinct functional connectivity patterns.

Understanding weight-related differences in functional connectivity provides key insight into neurocognitive factors implicated in obesity. Here, we sampled three groups from human connectome project data: 1) 47 pairs of BMI-discordant twins (n = 94; average BMI-discordancy 6.7 ± 3.1 kg/m2), 2) 47 pairs of gender and BMI matched BMI-discordant, unrelated individuals, and 3) 47 pairs of BMI-similar twins, to test for body mass dependent differences in between network functional connectivity. Across BMI discordant samples, three networks appeared to be highly sensitive to weight status; specific

Resting-state functional connectivity between right anterior insula and right orbital frontal cortex correlate with insight level in obsessive-compulsive disorder.

Few studies have explored the neurobiological basis of insight level in obsessive-compulsive disorder (OCD), though the salience network (SN) has been implicated in insight deficits in schizophrenia. This study was then designed to investigate whether resting-state (rs) functional connectivity (FC) of SN was associated with insight level in OCD patients. We analyzed rs-functional magnetic resonance imaging (fMRI) data from 21 OCD patients with good insight (OCD-GI), 19 OCD patients with poor insight (OCD-PI), and 24 healthy controls (HCs). Seed-based whole-brain FC and ROI (region of interest)

Anatomy and white matter connections of the orbitofrontal gyrus.

OBJECTIVE The orbitofrontal cortex (OFC) is understood to have a role in outcome evaluation and risk assessment and is commonly involved with infiltrative tumors. A detailed understanding of the exact location and nature of associated white matter tracts could significantly improve postoperative morbidity related to declining capacity. Through diffusion tensor imaging-based fiber tracking validated by gross anatomical dissection as ground truth, the authors have characterized these connections based on relationships to other well-known structures. METHODS Diffusion imaging from the Human Conne

Increased binding of 5-HT1A receptors in a dissociative amnesic patient after the recovery process.

Dissociative amnesia is characterized by an inability to retrieve information already saved in memories. 5-HT has some role in neural regulatory control and may be related to the recovery from dissociative amnesia. To examine the role of 5-HT1A receptors in the recovery from dissociative amnesia, we performed two positron emission tomography (PET) scans on a 30-year-old patient of dissociative amnesia using [(11)C]WAY-100635, the first at amnesic state, and the second at the time he had recovered. Exploratory voxel-based analysis (VBA) was performed using SPM software. 5-HT1A BPND images were

Activation of the prefrontal, occipital and parietal cortices during go/no-go discrimination tasks in the monkey as revealed by positron emission tomography.

The regions of the monkey brain involved in cognito-behavioral tasks were mapped using the H2(15)O positron emission tomography (PET) activation technique. Monkeys performed hand movements in an asymmetrically rewarded go/no-go reaction-time task with discrimination between green (go) and red (no-go) light signals. Regional cerebral blood flow (rCBF) was measured during the go/no-go task and a control task in which only go signals were presented. In the go/no-go task, when compared with the control, a significant increase in rCBF was noted in the following regions: (1) the principal sulci; (2)

Interactive representations of power and status signals in learning social hierarchies.

Social hierarchies, which organize our social relationships, have heterogeneous effects on choice behavior. Two central dimensions of social hierarchies, status, which is earned through recognition of competence, and power, which is the capacity to control the outcomes of others, may contribute to different behavioral profiles. Yet, the neurocomputational processes by which status- and power-related signals are learned based on social feedback, and influence subsequent decision-making, remain unclear. Here, we combine functional neuroimaging with computational modeling to examine how individua

From peripheral neurotoxicity to central dysfunction: linking neuropathic pain and cognition in chemotherapy-induced peripheral neuropathy.

Chemotherapy-induced peripheral neurotoxicity (CIPN) represents a significant clinical burden, affecting 70-80 % of patients during treatment and persisting chronically in 20-30 % of survivors. While peripheral nerve injury is the primary pathological hallmark, emerging evidence demonstrates that central nervous system (CNS) dysregulation plays a crucial role in pain chronification and associated cognitive impairment. This review synthesizes recent findings on cortical and subcortical alterations that drive neuropathic pain processing in CIPN, examining dysregulated glutamatergic and GABAergic

Neural correlates linked to developmental and affective disorder-related self-referential processing: an ALE meta-analysis.

Recent models of self-development highlight different rates of development in brain networks underlying emotion and cognition, which may contribute to increased vulnerability to depression and social anxiety disorder during adolescence. Functional magnetic resonance imaging (fMRI) studies suggest age- and affective disorder-related differences in neural activation during self-referential processing (SRP)-a cognitive process central to typical and atypical development. However, findings are mixed, with high methodological variability. We conducted the first activation likelihood estimation meta

Alterations in self-related brain networks in autism spectrum disorder: a systematic review of functional and structural magnetic resonance imaging studies.

Autism spectrum disorder (ASD) involves alterations in social communication and restricted, repetitive behaviors. Emerging evidence highlights atypical self-awareness as a key factor in ASD-related social impairments. However, the neural mechanisms underlying differences in self-processing remain fragmented. This systematic review synthesizes findings from 49 functional magnetic resonance imaging (fMRI) and structural magnetic resonance imaging (sMRI) studies (2000-2025) to explore self-related brain networks in ASD, based on the hierarchical self-model comprising interoceptive, exteroceptive,

Gamma-band transcranial alternating current stimulation restores motor function in a Parkinson's disease mouse model.

Parkinson's disease (PD) is associated with abnormal neural oscillations within the cortico-striatal-thalamo-cortical (CSTC) circuit, and current therapeutic options remain limited in their ability to restore physiological network dynamics. Transcranial alternating current stimulation (tACS) has recently emerged as a promising non-invasive approach. However, its optimal frequency and underlying mechanisms in PD remain incompletely understood. Here, we compared theta (6 Hz), alpha (10 Hz), beta (20 Hz), and gamma (40 Hz) tACS in 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP)-treated mice a

Modeling single nucleus microglia across species identifies immune pathways and therapeutic candidates in Alzheimer's disease.

Alzheimer's disease (AD) is a progressive neurodegenerative disease characterized by memory loss and behavioral changes. A pivotal influence on AD pathology is the dysregulation of microglia in the brain. Despite promising findings in mouse models, there are limitations to the translatable biological information across species due to differences in the physiology, timeline of disease, and human heterogeneity. To address these interspecies discrepancies, we developed a novel implementation of the Translatable Components Regression (TransComp-R) framework, which integrated microglial single-nucl

Paving the way for precision treatment of psychiatric symptoms with functional connectivity neurofeedback.

Treatment for Major Depressive Disorder (MDD) remains relatively homogeneous, despite patients having heterogeneous subsets of symptoms with differing underlying neural aberrations. Demonstrating potential for a more individualised treatment approach, we recently showed that normalisation of a neural network and a corresponding reduction in related symptoms can be achieved using real-time fMRI functional connectivity neurofeedback (FCNef). Specifically, we showed that brooding rumination but not anxiety symptoms decreased as functional connectivity between the dorsolateral prefrontal cortex (D

Transcranial direct current stimulation selectively improves temporal working memory.

Temporal working memory (WM)-the ability to temporarily store and manipulate temporal information-is constrained by a storage capacity threshold of approximately 2-3 s. Transcranial direct current stimulation (tDCS), an emerging non-invasive intervention, holds potential for enhancing temporal WM. We investigated whether anodal tDCS over key WM regions (right prefrontal cortex, PFC; posterior parietal cortex, PPC) could improve temporal WM capacity specifically for durations exceeding the critical threshold. We used a matching-to-sample paradigm with sample durations encompassing the threshold

Distinct neuronal vulnerability and metabolic dysfunctions are characteristic features of fast-progressing Alzheimer's patients with Lewy bodies.

Tau protein accumulation is linked to dementia progression in Alzheimer's disease (AD), with potential co-pathologies contributing to it. The progression of dementia in patients with AD varies between individuals, and the association between co-pathology and heterogeneity in dementia progression rate remains unclear. We used longitudinal cohort data, postmortem brain tissues, and biochemical methods such as immunoassays and proteomic profiling to investigate the molecular components associated with progression rate. We report that AD with comorbidities, such as dementia with Lewy bodies (DLB)

Abnormal phase-amplitude coupling in patients with first-episode schizophrenia during the auditory steady-state response and resting states.

The auditory steady-state response (ASSR) is a robust index for schizophrenia. Abnormal phase-amplitude coupling (PAC) in schizophrenia might be influenced by the confounding factors of illness stages and antipsychotic treatments. To exclude the effects of confounding factors, we examined abnormal PAC in antipsychotic-naïve patients with first-episode schizophrenia (FES) during the ASSR. 67 FES and 84 healthy controls (CON) were recruited and their EEG data were collected during the 20 Hz ASSR, 30 Hz ASSR, 40 Hz ASSR, and resting states. All the possible PAC patterns at Fz were compared betwee
