The involvement of CYP1A2 and CYP3A4 in the metabolism of clozapine.

AIMS: Clozapine (CLZ), an atypical neuroleptic with a high risk of causing agranulocytosis, is metabolized in the liver to desmethylclozapine (DCLZ) and clozapine N-oxide (CLZ-NO). This study investigated the involvement of different CYP isoforms in the formation of these two metabolites. METHODS: Human liver microsomal incubations, chemical inhibitors, specific antibodies, and different cytochrome P450 expression systems were used. RESULTS: Km and Vmax values determined in human liver microsomes were lower for the demethylation (61 +/- 21 microM, 159 +/- 42 pmol min(-1) mg protein(-1) mean

Small nucleolar RNA host genes in hepatocellular carcinoma: an evidence-weighted and etiology-aware framework for mechanistic and translational interpretation.

Small nucleolar RNA host genes (SNHGs) are a distinctive subgroup of long non-coding RNAs whose loci can generate both host lncRNA transcripts and intronic small nucleolar RNAs. In hepatocellular carcinoma (HCC), dysregulated SNHGs have been linked to tumor growth, epithelial-mesenchymal transition, metastasis, stemness, immune remodeling, extracellular vesicle communication, and therapeutic resistance. However, the clinical meaning of these associations remains uneven because many reported mechanisms are based on limited cell-line experiments, retrospective cohorts, or non-stratified HCC mode

SNX-BAR proteins 5 and 6 are required for NCOA7-AS antiviral activity against influenza A virus.

Interferon-induced antiviral proteins act on the first line of defense against viruses, including influenza A virus (IAV). Among those, the short isoform of nuclear receptor coactivator 7 (NCOA7-AS) has been shown to inhibit IAV entry and more especially the fusion between endosomal and viral membranes. Ectopic expression of NCOA7-AS leads to the increased acidification of the endolysosomal pathway, putatively through NCOA7-AS interaction with the vacuolar ATPase (V-ATPase). However, the precise mechanism of action of NCOA7-AS is not fully understood. Here, we identified the cellular partners

γ-Tocotrienol Sensitises Colorectal Cancer to PD-1 Blockade by Enhancing MHC-I-Associated Tumour Immune Visibility and CD8+ T Cell-Related Antitumour Immunity.

γ-Tocotrienol (γ-T3), a naturally occurring vitamin E isoform from plant-derived sources, has attracted attention as an antitumour agent. However, whether γ-T3 can enhance antitumour immunity and improve immune checkpoint blockade remains unclear. Here, using colorectal cancer (CRC) models, we found that γ-T3 suppressed tumour growth in immunocompetent MC38 and CT26 mouse models, whereas this effect was markedly weakened in immunodeficient hosts, indicating that its in vivo antitumour activity is closely associated with host immunity. Combination treatment with γ-T3 and programmed cell death p

Autoimmune nodopathies: a clinical review.

Autoimmune nodopathies are a subgroup of peripheral neuropathies characterized by autoantibodies targeting nodal and paranodal proteins such as neurofascin-155 (NF155), contactin-1 (CNTN1), and Contactin-associated protein 1 (Caspr1). Unlike chronic inflammatory demyelinating polyradiculoneuropathy (CIDP), these conditions involve direct disruption of the node of Ranvier rather than demyelination, leading to their distinction from CIDP in the 2021 EAN/PNS guidelines. Paranodal autoantibodies are often of the IgG4 subclass, which does not activate complement. Pathogenicity is supported by corre

Targeting splicing-derived neoantigens for precision cancer immunotherapy.

Aberrant pre-mRNA splicing in cancer generates protein sequences that are rare or absent in normal tissues, creating a rich source of tumor-specific neoantigens for immunotherapy. These splicing-derived neoantigens arise through diverse mechanisms, including recurrent somatic mutations in core spliceosome components (SF3B1, SRSF2, U2AF1, and ZRSR2), epigenetic derepression of transposable elements that give rise to chimeric exon-TE junctions, and coordinated dysregulation of splicing regulatory networks in cancers lacking spliceosome coding mutations. These processes produce two major classes

Activation of transforming growth factor beta 1 through integrin alpha V beta 6 and its molecular insights into cancer progression, and future directions.

Cancer metastasis is the last stage of cancer and can often lead to lethality. Cancer cells invade distal organs and establish an entirely new host microenvironment. Among several cancer-influencing mediators, TGF-β has been observed as a potential pro-oncogene to provoke metastasis. Among the 3 isoforms of TGF-β, TGF-β1 is identified as a strong promoter of several cancers, including colorectal, lung, gastric, and prostate cancer. TGF-β1 is always made by cells as an inactive, latent complex, which must be activated to function. A crucial activator of TGF-β is integrin αVβ6, which has a stron

Proteolysis-Based Biomarker Repertoire of the Neurofilament Proteome.

Neurodegeneration presents a significant challenge in ageing populations, often being detected too late for effective intervention. Biomarkers have shown great potential in addressing this issue, with neurofilament (Nf) proteins emerging as validated biomarkers presently transitioning from research to routine laboratory use. Whilst advances in large-scale quantitative analyses have enabled the targeted study of proteolytic Nf fragments in blood, the complete landscape of the Nf proteolytic breakdown remains unknown. This study presents a comprehensive atlas of the human Nf isoform (Z) degradom

Action at a distance: The remarkable coupling of CO2 uptake to electron transfer in specialized cyanobacterial NDH-1 complexes.

Cyanobacteria achieve highly efficient photosynthesis using a CO2-concentrating mechanism relying on specialized Type I (NDH-1) complexes. Among these, NDH-13 and NDH-14 catalyze redox-coupled hydration of CO2 to bicarbonate, supporting carbon fixation in carboxysomes. The mechanism of coupling electron transfer to CO2-hydration by these variant NDH-1 complexes remains unknown. We engineered a Synechococcus PCC7942 strain that expresses exclusively the high flux/low affinity NDH-14 complex, enabling the observation of the coupling of CO2 hydration to cyclic electron flow in isolation from the

CRISPR Disruption of scaRNA1 Reduces Pseudouridylation in Spliceosomal RNA U2 at U89 and Perturbs the Transcriptome in HEK293T Cells.

Small Cajal body-associated RNAs (scaRNAs) are essential for biochemical modification of spliceosomal RNAs and spliceosome function. Changes in scaRNA expression level have been associated with developmental issues, including cancer and congenital heart defects (CHDs), although the mechanism remains unclear. Small Cajal body-associated RNA 1 (scaRNA1) guides pseudouridylation at uridine 89 (Ψ89) of the spliceosomal RNA U2, a highly conserved modification that may be critical for spliceosome function. To investigate the role of scaRNA1 in splicing regulation, CRISPR-Cas9 genome editing was used

Allosteric modulation of NF1 GAP: Differential distributions of catalytically competent populations in loss-of-function and gain-of-function mutants.

Neurofibromin (NF1), a Ras GTPase-activating protein (GAP), catalyzes Ras-mediated GTP hydrolysis and thereby negatively regulates the Ras/MAPK pathway. NF1 mutations can cause neurofibromatosis type 1 manifesting tumors, and neurodevelopmental disorders. Exactly how the missense mutations in the GAP-related domain of NF1 (NF1GRD) allosterically impact NF1 GAP to promote these distinct pathologies is unclear. Especially tantalizing is the question of how same-domain, same-residue NF1GRD variants exhibit distinct clinical phenotypes. Guided by clinical data, we take up this dilemma. We sampled

In vitro and clinical data demonstrate negligible risk of drug-drug interactions with opelconazole, a novel inhaled antifungal agent.

BACKGROUND: Invasive fungal diseases (IFDs) cause high morbidity and mortality among immune-compromised patients. Triazoles, which are recommended to treat IFDs, inhibit and/or are substrates for the phase 1 cytochrome P450 (CYP) pathway that metabolizes many drugs and frequently result in drug-drug interactions (DDIs). Nebulized opelconazole is an investigational inhaled antifungal in development for the treatment of pulmonary aspergillosis. METHODS: In vitro studies first assessed effects of opelconazole on CYP isoforms and human transporter protein interactions. Subsequently, a Phase 1 stu

Exploring the role of glucocorticoid receptors and co-chaperones in Pemphigus foliaceus stratification.

Glucocorticoids (GC) are the main treatment for pemphigus foliaceus (PF). The effects of GC are mediated through glucocorticoid receptors (GR), with GRα and GRβ being the most significant isoforms. Several molecules are involved in mediating the cellular response to GC and can affect the response to treatment. However, the relationship between sensitivity to GC and the expression of GRα, GRβ, FKBP5, FKBP4, HAT1 and HDAC2 in PF disease has not yet been studied. The purpose of this study was to determine the expression of these molecules in patients with different types of response to treatment.

Cholesterol dynamics in rabbit liver: High-fat diet, olive oil, and synergistic dietary effects.

BACKGROUND & AIMS: Lipid metabolism disorders contribute to a range of human diseases, including liver-related pathologies. Rabbits, highly sensitive to dietary cholesterol, provide a model for understanding the development of liver disorders. Sterol regulatory element-binding protein isoform 2 (SREBP2) crucially regulates intracellular cholesterol pathways. Extra-virgin olive oil (EVOO) has shown reducing cholesterol levels and restoring liver parameters affected by HFD. The aim was to investigate the molecular impact of an HFD and supplemented with EVOO on rabbit liver cholesterol metabolism

Aggravated Ulcerative Colitis via circNlgn-Mediated Suppression of Nuclear Actin Polymerization.

Colitis is a chronic bowel disease characterized by damage to the lining of the large intestine, with its precise underlying causes remaining incompletely understood. In this study, we provide evidence that circular RNA circNlgn plays a pivotal role in promoting the development of colitis. Colitis patients produce significant higher levels of circNlgn. Transgenic mice expressing circNlgn exhibit heightened susceptibility to colitis development and progression, primarily attributed to the presence of the protein isoform Nlgn173 encoded by circNlgn. Nlgn173 undergoes translocation into cell nucl

Clusterin expression and distribution in spermatozoa as predictor of male fertility.

Clusterin (CLU), one of the main glycoproteins in mammalian semen and the male reproductive tract, plays a role in spermatogenesis and sperm maturation. Given the poor reliability of classic seminal studies in determining male-fertilizing capacity and the differences in CLU abundance between normal and abnormal spermatozoa, we investigated the potential value of mRNA-CLU levels and protein distribution in spermatozoa as markers of sperm quality and predictors of male fertility. This multicenter study included 90 patients undergoing in vitro fertilization (IVF) treatment with their partners, an

Association of NOX5 Expression with Sperm Activity and Motility in Pathospermic Infertile Men.

BACKGROUND: The newest NOX isoform, NOX5, has been found in mammalian spermatozoa. Many physiological and pathological situations in spermatozoa are mediated by reactive oxygen species (ROS). NOX5 is the main source of ROS in spermatozoa. Our purpose was to investigate the changes in NOX5 expression and the effect of NOX5 expression on sperm motility, chromatin integrity, and oxidative status in oligoasthenozoospermic compared to normozoospermic men. METHODS: Semen samples were collected from 30 normozoospermic (NS) and 30 oligoasthenozoospermic (OAS) men. NOX5 protein expression in sperm sam

Clinical and Molecular Characterization of a Novel Homozygous Frameshift Variant in AEBP1-Related Classical-like Ehlers Danlos Syndrome Type 2 with Comparison to Previously Reported Rare Cases.

Recently, an autosomal recessive subtype of connective tissue disorder within the spectrum of Ehlers-Danlos syndrome (EDS), named classical-like EDS type 2 (clEDS2), was identified. clEDS2 is associated with biallelic variants in the adipocyte enhancer binding protein 1 (AEBP1) gene, specifically, affecting its aortic carboxypeptidase-like protein (ACLP) isoform. We described the 15th patient (13th family) diagnosed with clEDS2. This patient presented with notable similarities in phenotype to the documented cases, along with additional characteristics such as significant prematurity and short
