Selective enhancement of the interneuron network and gamma-band power via GluN2C/GluN2D NMDA receptor potentiation.

N-Methyl-d-aspartate receptors (NMDARs) are a family of ligand-gated ionotropic glutamate receptors that mediate a slow, calcium-permeable component to excitatory neurotransmission. The GluN2D subunit is enriched in GABAergic inhibitory interneurons in cortical tissue. Diminished levels of GABAergic inhibition contribute to multiple neuropsychiatric conditions, suggesting that enhancing inhibition might have therapeutic utility, thus making GluN2D modulation an attractive drug target. Here, we describe the actions of a GluN2C/GluN2D-selective positive allosteric modulator, (+)-EU1180-453, whic

Lesion-specific circular RNA and mRNA signatures in secondary progressive multiple sclerosis.

Circular RNAs (circRNAs) are single-stranded RNAs with a covalently closed loop structure which have been increasingly recognised as stable and functionally relevant regulators of gene expression via sponging miRNAs. Recently, studies have revealed their roles in human diseases, subsequently emerging as potential biomarkers. Multiple sclerosis (MS) is an inflammatory, neurodegenerative, and immune-related disease of the central nervous system characterised by distinct lesion types that reflect different stages of brain tissue injury and repair. Our study conducted a comprehensive transcriptomi

The Potential Role of SGLT2 Inhibitors in the Treatment of Depression: Mechanisms and Clinical Prospects.

Major depressive disorder (MDD) is a high-burden psychiatric illness characterized by persistent low mood, diminished interest, and cognitive impairment. Its global prevalence continues to rise, accompanied by a significant risk of suicide. Traditional first-line treatments, such as selective serotonin reuptake inhibitors (SSRIs), are limited by delayed onset, limited response rates, and adverse effects, which has spurred the exploration of novel therapeutic strategies. A growing body of research indicates that the pathogenesis of MDD extends beyond the monoamine hypothesis, also involving neu

Astrocyte-related proteins mediate the association of YWHAG with Alzheimer's pathology and enhance its diagnostic value.

Recent advances have identified YWHAG as a promising synaptic biomarker, with evidence showing that the YWHAG:NPTX2 ratio strongly predicts cognitive decline and Alzheimer's disease (AD) progression independent of amyloid and tau pathology. However, the links between YWHAG and astrocytic processes-key regulators of amyloid clearance, tau phosphorylation, and neuroinflammation-remain poorly understood. A total of 530 participants were included. Levels of YWHAG and a panel of biologically relevant astrocyte-derived proteins were measured using a proximity extension assay and validated immunoassa

Effects of Maternal Tetramethyl Bisphenol F Exposure on Neurodevelopment and Behavior in Mouse Offspring.

Bisphenol A (BPA) has long been used in plastics, resins, and food packaging materials; however, extensive research has demonstrated its reproductive, developmental, and endocrine-disrupting effects. Consequently, BPA has been increasingly restricted and replaced with structural analogues. Among these, tetramethyl bisphenol F (TMBPF) has emerged as one of the most widely used substitutes, particularly in epoxy resins and food-can coatings. Although initially regarded as a safer alternative, accumulating evidence suggests that TMBPF may exert multiple toxicological effects, raising concerns abo

Hippocampal network activity changes during early epileptogenesis predict subsequent epilepsy.

OBJECTIVE: Despite decades of research, the circuit mechanisms that underlie focal epileptogenesis remain incompletely understood. In this study, we aimed to characterize the changes in hippocampal network activity induced by an epileptogenic insult. METHODS: In rats, prolonged electrical perforant pathway stimulation leads in a high percentage of animals to the development of temporal lobe epilepsy. However, a subset of animals remained apparently non-epileptic for unknown reasons. We monitored alterations of neuronal activity by chronically recording the local field potential (LFP) from the

Distinct excitability of thalamocortical neurons correlates with the presence of cerebellar afferents.

UNLABELLED: Thalamocortical (TC) neurons within the ventrolateral thalamus (VL) receive projections from the cerebellum and the basal ganglia (BG) to facilitate motor and non-motor functions. Tonic and rebound firing patterns in response to excitatory cerebellar and inhibitory BG inputs, respectively, are a canonical feature of TC neurons and plays a key role in signal processing. The intrinsic excitability of TC neurons has a strong influence on how they respond to synaptic inputs, however, it is unknown whether their afferents influence their firing properties. Understanding the input-specif

Chronic pregabalin treatment protects against spreading depolarization and alters hippocampal synaptic characteristics in a model of familial hemiplegic migraine-type 1.

Familial hemiplegic migraine type-1 (FHM-1) is a form of migraine with aura caused by mutations in the P/Q-type (Cav2.1) voltage-gated calcium channel. Pregabalin, used clinically in the treatment of chronic pain and epilepsy, inhibits P/Q-type calcium channel activity and recent studies suggest that it may have potential for the treatment of migraine. Spreading Depolarization (SD) is a neurophysiological phenomenon that can occur during migraine with aura by propagating a wave of silenced neuronal function through cortex and sometimes subcortical brain structures. Here, utilizing an optogenet

Typhaneoside Suppresses Glutamate Release Through Inhibition of Voltage-Dependent Calcium Entry in Rat Cerebrocortical Nerve Terminals.

Glutamate is the major excitatory neurotransmitter in the brain and is involved in many brain functions. In this study, we investigated whether typhaneoside, a flavonoid from Typhae angustifolia pollen, affects endogenous glutamate release from rat cortical synaptosomes. Using a one-line enzyme-coupled fluorometric assay, glutamate release stimulated by the K+ channel blocker 4-aminopyridine was monitored to explore the possible underlying mechanisms. The vesicular transporter inhibitor bafilomycin A1 and chelation of extracellular Ca2+ ions with EGTA suppressed the effect of typhaneoside on t

Melittin, the Major Pain-Producing Substance of Bee Venom.

Melittin is a basic 26-amino-acid polypeptide that constitutes 40-60% of dry honeybee (Apis mellifera) venom. Although much is known about its strong surface activity on lipid membranes, less is known about its pain-producing effects in the nervous system. In this review, we provide lines of accumulating evidence to support the hypothesis that melittin is the major pain-producing substance of bee venom. At the psychophysical and behavioral levels, subcutaneous injection of melittin causes tonic pain sensation and pain-related behaviors in both humans and animals. At the cellular level, melitti

Drug Refraining and Seeking Potentiate Synapses on Distinct Populations of Accumbens Medium Spiny Neurons.

Cocaine-associated cues and contexts can precipitate drug seeking in humans and in experimental animals. Glutamatergic synapses in the core subcompartment of the nucleus accumbens (NAcore) undergo transient potentiation in response to presenting drug-associated cues. The NAcore contains two populations of medium spiny neurons (MSNs) that differentially express D1 or D2 dopamine receptors. By recording the ratio of AMPA and NMDA glutamate receptor currents (AMPA/NMDA ratio) from MSNs in NAcore tissue slices, we endeavored to understand which subpopulation of MSNs was undergoing transient potent

Neural network-enhanced investigation of ferroptosis and druggability in early-onset alzheimer's disease.

Alzheimer's disease (AD) is a complex neurodegenerative disorder which is multifactorial in nature. Some of its characteristics are slow cognitive decline, memory problems and behavioral changes. AD patient brains show a progressive synaptic toxicity, autophagy, neuroinflammation, excess generation of reactive oxygen species (ROS), neuronal death and oxidative stress, which occurs due to disrupted metal homeostasis along with tau and amyloid-β protein deposition. Notably, lipid peroxidation, iron buildup and elevated oxidative stress in AD brains suggest a possible molecular connection between

HAT regimen attenuates NF-κb-driven megakaryocyte apoptosis and neuronal cell death in sepsis: convergent mechanisms protecting thrombocytopenia and cognitive function.

BACKGROUND: Sepsis-induced NF-κB hyperactivation drives two devastating cell death cascades: megakaryocyte apoptosis causing thrombocytopenia (incidence 35%-59%), and neuronal apoptosis with microglial-mediated neuroinflammation causing cognitive dysfunction in up to 70% of survivors. Mechanistically, NF-κB-driven upregulation of pro-apoptotic mediators (cleaved caspase-3, cytochrome c release) impairs megakaryopoiesis while simultaneously inducing hippocampal neuronal death and synaptic loss. The HAT regimen (hydrocortisone, ascorbic acid, and thiamine) modulates complementary nodes of this N

Cognitive Impairment in Asthma: Emerging Evidence, Clinical Consequences, and Future Directions.

Asthma is increasingly recognised as a systemic inflammatory disease with effects extending beyond the respiratory system. Emerging evidence suggests that cognitive impairment may represent an under-recognised extrapulmonary manifestation with important implications for disease management and long-term neurological health. This narrative review critically synthesises current evidence regarding the epidemiology, clinical consequences, and potential mechanisms linking asthma with cognitive dysfunction. Observational and longitudinal studies indicate that asthma is associated with an increased ri

Electromagnetic field induced activation of amyloid-β degrading enzyme, neprilysin, for accelerated Alzheimer's disease therapy.

BackgroundAmyloid-β accumulation and misfolding are key features of Alzheimer's disease (AD), contributing to synaptic dysfunction, neuroinflammation, and neuronal loss. Normally, amyloid-β is cleared by chaperone-assisted folding, enzymes such as neprilysin and insulin-degrading enzyme, and autophagy pathways. In AD, these mechanisms become impaired, leading to toxic aggregation. Enhancing the activity of endogenous degrading enzymes offers a promising therapeutic approach, with electromagnetic (EM) fields emerging as a potential noninvasive method to promote amyloid-β clearance.ObjectiveThis

Identification of Functional lncRNAs in Alzheimer Disease by Integrative Analysis of lncRNA-mRNA Network Based on Competing Endogenous RNA Mechanism.

BACKGROUND: Emerging evidence highlights the critical involvement of noncoding RNAs (ncRNAs) in the pathologic process of Alzheimer disease (AD). However, the precise functional contributions and regulatory landscapes of long noncoding RNAs (lncRNAs) in AD remain unclear. OBJECTIVE: This study aims to delineate the regulatory roles of functional lncRNAs in AD by systematically analyzing lncRNA-mRNA interactions within an AD-associated network, leveraging the competing endogenous RNA (ceRNA) framework. METHODS: We constructed an AD-specific lncRNA-mRNA network by integrating a probe reannotat
