Mammalian adaptive mutations of the PA protein of highly pathogenic avian H5N1 influenza virus.

UNLABELLED: Highly pathogenic H5N1 influenza A viruses continue to circulate among avian species and cause sporadic cases of human infection. Therefore, the threat of a pandemic persists. However, the human cases of H5N1 infection have been limited mainly to individuals in close contact with infected poultry. These findings suggest that the H5N1 viruses need to acquire adaptive mutations to gain a replicative advantage in mammalian cells to break through the species barrier. Many amino acid mutations of the polymerase complex have been reported to enhance H5N1 virus growth in mammalian cells;

Beyond Antibacterial Activity: Engineering Multifunctional Biomaterials for Multidrug-Resistant Infections.

Antimicrobial resistance is increasingly limiting the effectiveness of conventional antibiotic therapy, particularly in chronic wounds, biofilm-associated infections, and implant-related infections. Antimicrobial biomaterials have emerged as promising alternatives; however, much of the field remains organized around material classes or pathogen killing alone, with comparatively limited consideration of the complex biological environment in which infection develops and persists. This review proposes a function-based framework in which antimicrobial biomaterials are designed as integrated therap

The clinical outcomes and overall survival in patients with mucoepidermoid and adenoid cystic carcinoma of the salivary glands: A comparative analysis of long-term outcomes.

PURPOSE: Mucoepidermoid carcinoma (MEC) and adenoid cystic carcinoma (AdCC) are the two most common salivary gland malignancies, but differ substantially in biological behavior, recurrence patterns and long-term prognosis. Comparative data from patients treated within the same institution using similar management strategies remain limited. This study compared long-term survival, recurrence patterns and prognostic factors between MEC and AdCC to identify histology-specific characteristics that may inform follow-up and treatment strategies. METHODS: We performed a retrospective cohort study of

Impacts and Immune Mechanisms of Pollutant-Mediated Asthma and Airway Inflammation.

Air pollution is a major environmental risk factor contributing to the development, exacerbation, and persistence of asthma. A wide range of airborne pollutants, including particulate matter (PM), ozone (O₃), nitrogen dioxide, sulfur dioxide, diesel exhaust particles (DEPs), volatile organic compounds, and biological contaminants, can interact with the airway epithelium and immune system, leading to dysregulated inflammatory responses. Increasing evidence indicates that pollutant exposure induces epithelial injury, disrupts epithelial barrier integrity, promotes oxidative stress, and activates

Exploratory multi-biofluid proteomic profiling of acute exercise responses in plasma, tear fluid, and nasal secretions.

BACKGROUND: Exercise induces broad physiological adaptations through complex molecular regulation across multiple biological systems. While plasma-based proteomics has substantially advanced our understanding of exercise-associated molecular changes, the proteomic responses of mucosal biofluids, including tear fluid and nasal secretions, remain poorly characterized. A multi-biofluid proteomic approach may provide a more comprehensive characterization of molecular alterations associated with acute exercise. METHODS: Paired samples of plasma, tear fluid, and nasal secretions were collected from

SOMA: a preventive health governance framework for adaptive, consumer-facing digital health guidance.

This paper introduces SOMA, a conceptual preventive health governance framework for adaptive, consumer-facing digital health guidance, designed to support everyday preventive health self-governance. Non-communicable and lifestyle-sensitive chronic diseases account for the majority of global mortality, yet existing consumer-facing digital health tools rarely provide coherent, governed preventive guidance that integrates multimodal personal health data, scientific evidence, user-defined goals, real-world feasibility, relational support, and explicit safety constraints within a unified architectu

GENESIS-SHIELD: an interpretable ensemble for anomaly detection in CRISPR genomic-workflow security.

INTRODUCTION: The rapid advancement of CRISPR-based gene editing has introduced digital-workflow security and integrity challenges: unauthorized modifications, temporal inconsistencies, and duplicated provenance records can compromise the auditability of editing logs. These are distinct from the biological risks of editing itself; our focus is the security of the digital record. Existing single-component detectors capture only one facet of these threats. METHODS: We present GENESIS-SHIELD, a multi-component ensemble whose novelty lies in the integration of established techniques for CRISPR-wo

The relationship between post-occlusive reactive hyperemia and the level of training and lifestyle in young adults.

BACKGROUND: Post-occlusive reactive hyperemia (PORH) is widely used to assess microvascular function. Increasing attention has been directed toward temporal characteristics of the PORH response and the need for individualized arterial occlusion pressure (AOP); however, comparative data across athletes with different training profiles and non-athletic controls remain limited. PURPOSE: To compare PORH-derived microcirculatory parameters among individuals with different training backgrounds using an individualized AOP approach. METHODS: This cross-sectional study included 60 participants (n = 6

Integrated Single-Cell Transcriptomics Identifies γδ T-Cell Heterogeneity and a Candidate HLA-E-NKG2A Regulatory Axis in Pancreatic Ductal Adenocarcinoma.

Background: Pancreatic ductal adenocarcinoma (PDAC) has a profoundly immunosuppressive tumour microenvironment (TME) that limits anti-tumour immunity and contributes to resistance to immunotherapy. Although γδ T-cells can integrate innate and adaptive immune signals, their abundance, transcriptional states and regulatory pathways in PDAC remain incompletely defined. Methods: We performed integrated single-cell RNA sequencing analysis of 39 pancreatic tissue samples, comprising 33 PDAC tumours and 6 adjacent normal tissues. After dataset integration, immune cell annotation, and stringent per-ce

Somatic variant-calling beyond cancer: Repurposing algorithms to map low‑allele‑fraction variants across genomics.

BACKGROUND: Somatic variant callers were originally developed to identify tumour-specific mutations in mixed tumour-normal samples. Increasingly, disciplines such as developmental biology, reproductive medicine, virology and mitochondrial genetics require detection of low-frequency variants from high-depth sequencing. Many laboratories therefore reuse cancer callers without clear guidance on their statistical assumptions or validation in non-cancer contexts. METHODS: We review widely used somatic callers and post-calling classifiers, summarising their underlying models and assumptions. We the

Genomic and phenotypic insights into the novel species Selenomonas lamontii type strain ATCC 33150, currently described as Selenomonas sputigena.

Selenomonas sputigena is an anaerobic, gram-negative bacterium found in the human mouth and upper respiratory tract. This organism is emerging as an important contributor to human health and disease. In the oral cavity, S. sputigena contributes to periodontitis and is associated with early childhood caries. Much of our current understanding of the genus Selenomonas and its relation to human health derives from studies of a single species, S. sputigena, and is further limited to the type strain, ATCC 35185. As S. sputigena is emerging as a significant contributor to human health, we sought to c

VICAST: An Integrated Toolkit for Viral Genome Annotation Curation and Low-Frequency Variant Analysis in Passage Studies.

Cultured virus passage studies are fundamental to understanding viral evolution, attenuation, and host adaptation, yet analyzing genomic changes across passages requires both accurate functional annotation of viral genomes and sensitive detection of low-frequency variants. Existing tools address these needs separately: automated annotation pipelines such as VADR and VIGOR4 perform well for well-characterized virus families but struggle with poorly-annotated or novel genomes, while variant calling pipelines designed for clinical diagnostics focus on consensus sequences rather than the low-frequ

Effect of PEI25k/DOTAP/cholesterol liposomal formulation on the immunogenicity of SARS-CoV-2 spike protein-encoding DNA vaccine.

BACKGROUND: The Coronavirus Disease 2019 (COVID-19) pandemic has highlighted the need to upgrade vaccine strategies. DNA vaccines are advantageous because of their stability and ease of large-scale production; however, they often require adjuvants to boost immune response. Liposomal adjuvants have emerged as effective immune response enhancers, offering efficient delivery and compatibility with biological systems. This study evaluated the immune response induced by a Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) spike protein-encoding DNA vaccine formulated with various liposome

Nutritional status and cancer survival among rural Chinese women: biological mechanisms, health disparities, and translational opportunities.

Cancer remains a major cause of premature mortality among women worldwide, and its burden is particularly pronounced in rural China, where delayed diagnosis, uneven access to oncology services, and nutritional vulnerability may jointly affect survival. This narrative review synthesizes mechanistic, clinical, and population-level evidence on the relationship between nutritional status and cancer survival among rural Chinese women, with a focus on breast, cervical, gastric, and colorectal cancers. It first outlines the epidemiological profile of major female cancers in rural China and summarizes

Restoration interventions enhance plant species diversity and composition in the humid tropical highlands of Ethiopia.

Land degradation in the Ethiopian highlands poses a significant threat to biodiversity and ecosystem services. While large-scale afforestation, reforestation, and natural resource management interventions have been instigated, empirical evidence on their ecological effects remains inadequate. This study aims to examine vegetation diversity and plant community composition patterns associated with land restoration interventions across three agro-ecological zones in the humid tropical highlands of Ethiopia. Vegetation data were systematically collected from 672 plots established across 56 watersh

Masturbation as a sexual and psychological coping strategy in long-distance relationships: a systematic review.

INTRODUCTION: Long-distance relationships (LDRs) reduce opportunities for physical intimacy, often prompting individuals to seek alternative sexual activities such as masturbation. Although common, its influence on sexual satisfaction, sexual health, psychological well-being, and relational stability in LDRs remains understudied. Moreover, cultural and population differences shape diverse meanings and perspectives on masturbation. OBJECTIVES: We employed a systematic review approach to explore the role of masturbation within long-distance relationships. Specifically, this review aims to exami
