Dual action of the cyclodiene insecticide dieldrin on the gamma-aminobutyric acid receptor-chloride channel complex of rat dorsal root ganglion neurons.

The gamma-aminobutyric acidA (GABAA) receptor-chloride channel complex is known to be the target site of dieldrin, a cyclodiene insecticide, and lindane. In order to elucidate the mechanisms of dieldrin interaction with the GABA system, whole-cell patch clamp experiments were performed with rat dorsal root ganglion neurons in primary culture. When co-applied with GABA, dieldrin exerted a dual effect on the GABA-induced chloride current. The chloride current induced by 10 microM GABA was greatly enhanced by the first 20-sec co-application with 1 microM dieldrin, but the enhancement subsided dur

Prunus domestica L. Fruit Extract Mitigates the Depletion of Rat Hippocampal Neurogenesis and Memory Deficits Observed in a D-Galactose-Induced Aging Rat Model.

BACKGROUND/OBJECTIVES: Accumulation of oxidative damage and brain inflammation plays a role in aging-associated memory impairment. D-galactose (D-gal) is a monosaccharide used to induce aging in rat models by promoting oxidative damage and neuroinflammation. Prunus domestica L. (PD) contains high levels of flavonoid compounds, which possess antioxidant, anti-inflammatory, neuroprotective and anti-aging properties. This study investigated the effects of PD fruit extract on memory, vascular-associated cell proliferation, immature neuronal cells, and survival of neurons in the dentate gyrus (DG)

Adhesion molecule with Ig-like domain 1 regulates stability of carotid plaque via TGFβ/Smad signaling pathway by interaction with TGFRII.

Carotid atherosclerosis is a significant risk factor for cardiovascular and cerebrovascular diseases. Maintaining plaque stability can prevent plaque rupture and thrombus formation, slow disease progression, and is critically important for preventing cerebrovascular events (such as stroke, transient ischemic attack (TIA), and similar events). Mechanisms influencing plaque stability are still unclear. In this study, stable plaques (n = 5) and unstable plaques (n = 5) were collected from patients and analyzed using RNA-sequencing. 594 differently expressed genes were found by RNA-seq. Pathways e

Immunomodulatory roles of the HLA family and B2M in atherosclerosis: Diagnostic biomarkers and thrombosis risk.

Atherosclerosis (AS), a chronic inflammatory disease driven by lipid accumulation and immune dysregulation, predisposes to acute cardiovascular events via plaque rupture and thrombosis. The molecular mechanisms linking immune microenvironment imbalance to plaque instability remain incompletely defined. Utilizing the GSE100927 dataset (69 AS patients vs 35 controls), we identified differentially expressed genes (DEGs) and constructed co-expression networks via weighted gene co-expression network analysis. Hub genes were intersected with DEGs to determine core genes. Immune cell infiltration was

Differential expression of gene co-expression networks related to the mTOR signaling pathway in bipolar disorder.

Bipolar disorder (BPD) is a severe mental illness characterized by episodes of depression and mania. To investigate the molecular mechanisms underlying the pathophysiology of bipolar disorder, we performed transcriptome studies using RNA-seq data from the prefrontal cortex (PFC) of individuals with BPD and matched controls, as well as data from cell culture and animal model studies. We found 879 differentially expressed genes that were also replicated in an independent cohort of post-mortem samples. Genes involving the mechanistic target of rapamycine (mTOR) pathway were down-regulated, while

Adoptive transfer of Pfkfb3-disrupted hematopoietic cells to wild-type mice exacerbates diet-induced hepatic steatosis and inflammation.

BACKGROUND AND OBJECTIVES: Hepatic steatosis and inflammation are key characteristics of non-alcoholic fatty liver disease (NAFLD). However, whether and how hepatic steatosis and liver inflammation are differentially regulated remains to be elucidated. Considering that disruption of 6-phosphofructo-2-kinase/fructose-2,6-biphosphatase 3 (Pfkfb3/iPfk2) dissociates fat deposition and inflammation, the present study examined a role for Pfkfb3/iPfk2 in hematopoietic cells in regulating hepatic steatosis and inflammation in mice. METHODS: Pfkfb3-disrupted (Pfkfb3 +/-) mice and wild-type (WT) litter

Disruption of FOXP3-EZH2 Interaction Represents a Pathobiological Mechanism in Intestinal Inflammation.

BACKGROUND & AIMS: Forkhead box protein 3 (FOXP3)+ regulatory T cell (Treg) dysfunction is associated with autoimmune diseases; however, the mechanisms responsible for inflammatory bowel disease pathophysiology are poorly understood. Here, we tested the hypothesis that a physical interaction between transcription factor FOXP3 and the epigenetic enzyme enhancer of zeste homolog 2 (EZH2) is essential for gene co-repressive function. METHODS: Human FOXP3 mutations clinically relevant to intestinal inflammation were generated by site-directed mutagenesis. T lymphocytes were isolated from mice, hu

Functional activity of monocytes and macrophages in HTLV-1 infected subjects.

The Human T lymphotropic virus type-1 (HTLV-1) infects predominantly T cells, inducing proliferation and lymphocyte activation. Additionally, HTLV-1 infected subjects are more susceptible to other infections caused by other intracellular agents. Monocytes/macrophages are important cells in the defense against intracellular pathogens. Our aims were to determine the frequency of monocytes subsets, expression of co-stimulatory molecules in these cells and to evaluate microbicidal ability and cytokine and chemokine production by macrophages from HTLV-1 infected subjects. Participants were 23 HTLV-

Curcumin delivery system based on biodegradable polyhydroxybuterate Chitosan copolymer and Cobalt oxide nanoparticles against colorectal cancer.

In this study, a biodegradable nanocomposite composed of polyhydroxybutyrate-co-chitosan and cobalt oxide (PHB-co-CS/Co3O4) was developed for targeted and sustained in-vitro release of curcumin (CUR). The nanocomposite was prepared by varying the concentrations of Co3O4 nanoparticles (NPs) and using chitosan with different molecular weights (LCS, HCS) to optimize the release profile. The PHB-co-CS copolymer was synthesized through a coupling reaction between PHB-diol and the terminal isocyanate groups of chitosan, using stannous octanoate as a catalyst. PHB-diol was obtained via transesterific

Co-exposure to arsenic and nickel induces oxidative stress and mineral imbalance, impairing male reproductive parameters in Wistar rats.

Simultaneous exposure to multiple toxic metals is a common scenario in environmental contamination. Among these metals, arsenic and nickel are widely distributed pollutants with well-established toxic effects. However, their combined impact on male reproductive physiology and micromineral homeostasis remains poorly understood. This study evaluated the effects of subchronic co-exposure to arsenic (1 mg L-1) and nickel (7 mg L-1) in drinking water for 70 days on the hypothalamus, testis, and epididymis of adult Wistar rats. Exposed rats showed arsenic and nickel retention in all analyzed tissues

Molecular co-alteration patterns of RICTOR-mutant metastatic lung adenocarcinomas: a single-center cohort study.

RICTOR, a core component of the mTORC2 complex, regulates AKT signaling and has been implicated in tumor biology. While RICTOR amplification and overexpression have been reported, the mutational landscape of RICTOR and its molecular context in non-small cell lung cancer (NSCLC) remain insufficiently characterized. This study aimed to describe the prevalence and co-alteration patterns of RICTOR mutations in metastatic lung adenocarcinoma. We retrospectively analyzed 137 patients diagnosed with metastatic lung adenocarcinoma between 2018 and 2024. Genomic profiling was performed using next-gener

Carbon dioxide gas-induced pneumothorax versus one-lung ventilation in minimally invasive esophagectomy: a multicenter propensity score matching cohort study.

BACKGROUND: Carbon dioxide gas-induced pneumoperitoneum might be the reason for the shorter postoperative survival of patients with malignant tumors. Whether CO 2 gas-induced pneumothorax has unfavorable impacts on the surgical and oncological outcomes of minimally invasive esophagectomy remains unclear. METHODS: Between 2010 and 2016, a total of 998 patients with squamous cell carcinoma of the esophagus who received video-assisted surgery were registered from three large-volume medical centers. The overall survival (OS) and disease-free survival (DFS) were compared after using propensity sco

Enhanced pathogenicity and synergistic effects of co-infection with bovine viral diarrhea virus 1 and HoBi-like virus in cattle and guinea pigs.

INTRODUCTION: The Bovine Viral Diarrhea Virus 1 (BVDV1) and HoBi-like virus (BVDV3), both within the same genus, share genomic homology and exhibit low antigenic cross-reactivity despite presenting similar clinical manifestations. In 2021, a bovine respiratory disease complex (BRDC) outbreak on two cattle farms in the Inner Mongolia Autonomous Region of China resulted in ten fatalities. METHODS: Metagenomic and metatranscriptomic analyses were used to identify viral agents, including a co-infection case. A genetic evolution analysis assessed the relationships with related strains. Coronavirus M Protein Trafficking in Epithelial Cells Utilizes a Myosin Vb Splice Variant and Rab10.

The membrane (M) glycoprotein of coronaviruses (CoVs) serves as the nidus for virion assembly. Using a yeast two-hybrid screen, we identified the interaction of the cytosolic tail of Murine Hepatitis Virus (MHV-CoV) M protein with Myosin Vb (MYO5B), specifically with the alternative splice variant of cellular MYO5B including exon D (MYO5B+D), which mediates interaction with Rab10. When co-expressed in human lung epithelial A549 and canine kidney epithelial MDCK cells, MYO5B+D co-localized with the MHV-CoV M protein, as well as with the M proteins from Porcine Epidemic Diarrhea Virus (PEDV-CoV)

Co-interventions with Clostridium butyricum and soluble dietary fiber targeting the gut microbiota improve MAFLD via the Acly/Nrf2/NF-κB signaling pathway.

Purpose: The pathogenesis of metabolic associated fatty liver disease (MAFLD) is complex. Lipid metabolic disorder, chronic inflammation, and oxidative stress are the core events for MAFLD. Dietary intervention is an important treatment strategy for preventing the onset and progression of MAFLD. Clostridium butyricum (CB) and soluble dietary fiber (SDF) are often considered beneficial for health. We explored how two microbiota-targeted interventions (SDF and CB) influence the hepatic immune system, oxidative stress, and lipid metabolism in MAFLD mice. Methods: To explore the role of SDF and CB

A stimuli-responsive drug release nanoplatform for kidney-specific anti-fibrosis treatment.

The renoprotective effects of hypoxia inducible-factor (HIF) activators have been demonstrated by improving renal hypoxia in chronic kidney disease. Cobalt chloride is one of the most widely used HIF activators in biomedicine; however, poor kidney targeting and undesirable side effects greatly limit its clinical applications. Here, we report a novel stimuli-responsive drug release nanoplatform in which glutathione (GSH)-modified Au nanoparticles (GLAuNPs) and Co2+ self-assemble into nanoassemblies (GLAuNPs-Co) through coordination interactions between empty orbitals of Co2+ and lone pairs of G

Murine cytomegalovirus strains co-replicate at multiple tissue sites and establish co-persistence in salivary glands in the absence of Ly49H-mediated competition.

Infection with multiple genetically distinct strains of pathogen is common and can lead to positive (complementation) or negative (competitive) within-host interactions. These interactions can alter aspects of the disease process and help shape pathogen evolution. Infection of the host with multiple strains of cytomegalovirus (CMV) occurs frequently in humans and mice. Profound, NK-cell-mediated (apparent) competition has been identified in C57BL/6 mice, and prevented the replication and shedding of certain co-infecting CMV strains. However, the frequency of such strong competition has not bee

Suppressive effect of CORM-2 on LPS-induced platelet activation by glycoprotein mediated HS1 phosphorylation interference.

In recent years, it has been discovered that septic patients display coagulation abnormalities. Platelets play a major role in the coagulation system. Studies have confirmed that carbon monoxide (CO) has important cytoprotective and anti-inflammatory function. However, whether CO could alter abnormal activation of platelets and coagulation and thereby reduce the incidence of mortality during sepsis has not been defined. In this report, we have used CO-releasing molecules (CORM-2) to determine whether CO inhibits LPS-induced abnormal activation of platelets and have explored the potential mecha
