Analysis of the tumor reactivity of autologous TILs and allogeneic γδ T cells via tumor organoid-immune cell coculture.

BACKGROUND: The efficacy of conventional αβ T cell-based immunotherapies is often limited by tumor immune evasion. γδ T cells can bypass these limitations through MHC-independent tumor recognition, but their function within the tumor microenvironment (TME) remains poorly characterized due to a lack of relevant preclinical models. This study aims to establish a patient-derived tumor organoid-immune cell coculture system to evaluate γδ T-cell reactivity and cytotoxicity in a human-relevant TME (n = 10 PDTO lines; n = 3 independent experiments). METHODS: We developed an innovative coculture syste

ROSE12, a novel anti-CTLA-4 FcγRs binding-enhanced antibody activated by extracellular adenosine triphosphate, shows tumor-selective regulatory T-cell depletion and antitumor efficacy without systemic immune activation.

BACKGROUND: Intratumoral regulatory T cells (Tregs) are associated with diminished antitumor immunity and poor prognosis in many cancers, with tumor-infiltrating effector Tregs expressing high levels of cytotoxic T-lymphocyte-associated protein 4 (CTLA-4). While Treg depletion is a promising strategy for cancer immunotherapy, systemic Treg depletion may lead to severe autoimmune toxicity. Therefore, to selectively deplete intratumoral Tregs, we used extracellular ATP (exATP), which is highly elevated in solid tumors, as a tumor-selective small molecule. METHODS: We generated ROSE12, a novel a

Prognostic and immunological implications of a mitophagy-associated gene signature in bladder cancer.

BACKGROUND: The pronounced heterogeneity of bladder cancer (BLCA) drives divergent patient outcomes and therapy responses. Mitophagy, a pivotal cellular quality-control and metabolic mechanism, modulates the dynamics of the tumor microenvironment (TME). Its prognostic significance and potential for guiding precision oncology, however, remain incompletely defined. This investigation aimed to evaluate the translational utility of mitophagy-associated signatures for the risk stratification of BLCA, profiling of TME, and prediction of therapeutic response. METHODS: Using The Cancer Genome Atlas (

A myeloid immunosuppressive phenotype defines primary refractoriness to atezolizumab plus bevacizumab in hepatocellular carcinoma.

BACKGROUND & AIMS: Despite improved outcomes with atezolizumab plus bevacizumab (A+B) in hepatocellular carcinoma, primary refractoriness (PRef), characterised by early progression or short-lived disease stabilisation following treatment, remains a significant challenge. METHODS: We analysed 1,296 patients with hepatocellular carcinoma treated with frontline A+B (AB-real) and validated findings in 645 trial participants recruited to IMbrave150 and GO30140. PRef was defined by SITC (Society for the Immunotherapy of Cancer) criteria. We performed a multi-parametric analysis of pre-treatment tum

SPP1+ Macrophages Involved in Juvenile Xanthogranuloma via PPARG Signaling Pathway.

BACKGROUND: Juvenile xanthogranuloma (JXG) is the most common non-Langerhans cell histiocytic disorder in children. However, the pathogenesis remains unclear. METHODS: Lesional and adjacent normal (NOR) skin samples were obtained from patients with histologically confirmed JXG. Each biopsy sample was processed for either bulk or single-cell RNA sequencing (scRNA‑seq). Differentially expressed genes (DEGs) in the bulk transcriptome were identified and subjected to functional enrichment analyses. ScRNA-seq data were analyzed using the Seurat software for cell clustering and annotation. Single-c

Targeting Fibroblast Activation Protein for Precision Medicine.

The advancement of precision medicine depends critically on well-defined molecular targets. Pathologically activated fibroblasts drive disease progression in cancer, fibrosis and chronic inflammation through sustained inflammation, extracellular matrix remodeling and pathological microenvironment formation. Among available markers, fibroblast activation protein (FAP) stands out due to its selective overexpression in diseased tissues versus minimal expression in healthy organs. This unique profile makes FAP a promising target for molecular imaging and targeted therapy, enabling precision theran

Anti-HER2×CCR4 bispecific antibody enhances antitumor immunity in advanced HER2-positive tumors by chemotaxis blockade and depletion of tumor-associated Tregs, without inducing systemic toxicity.

BACKGROUND: CCR4+ regulatory T cells (Tregs), which are widely present in peripheral circulation and tumor microenvironment (TME), promote tumor immune evasion by infiltrating human epidermal growth factor receptor 2-positive (HER2+) tumors in a chemokine-driven manner. However, therapies targeting CCR4 (eg, mogamulizumab) for systemic Treg depletion risk significant toxicity and have far been confined to hematological malignancies. Notably, tumor-chemotactic peripheral Tregs-key precursors to tumor-infiltrating Tregs (tumor-infiltrating lymphocyte (TIL)-Tregs)-remain overlooked. METHODS: We

Follicular lymphoma: The long and winding road leading to your cure?

Follicular lymphoma, the most common indolent lymphoma, though highly responsive to therapy is coupled with multiple relapses for the majority of patients. Advances in biologic understanding of molecular events in lymphoma cells and the tumor microenvironment, along with novel cellular and targeted therapies, suggest this may soon change. Here we first review the development of the molecular concepts and classification of follicular lymphoma, along with therapeutic development of treatments based on chemotherapy plus monoclonal antibodies targeting CD20. We then focus on developments over the

Association of HIV Status With Local Immune Response to Anal Squamous Cell Carcinoma: Implications for Immunotherapy.

IMPORTANCE: The programmed cell death protein 1 (PD-1) and programmed death-ligand 1 (PD-L1) pathway play an important immunosuppressive role in cancer and chronic viral infection, and have been effectively targeted in cancer therapy. Anal squamous cell carcinoma (SCC) is associated with both human papillomavirus and HIV infection. To date, patients with HIV have been excluded from most trials of immune checkpoint blocking agents, such as anti-PD-1 and anti-PD-L1, because it was assumed that their antitumor immunity was compromised compared with immunocompetent patients. OBJECTIVE: To compare

Enhanced anti-tumor efficacy and safety profile of tumor microenvironment-responsive oncolytic adenovirus nanocomplex by systemic administration.

UNLABELLED: Oncolytic adenovirus (Ad) holds great promise as a potential gene therapy for cancer. However, intravenously administered Ad may encounter difficulties due to unfavorable host responses, non-specific interactions, and the heterogeneity of the tumor cell population. As an approach to combine the advantages of oncolytic Ad and synthetic polymers and to address the associated difficulties, Ad was physically complexed with a pH-sensitive block copolymer, methoxy poly(ethylene glycol)-b-poly(l-histidine) (mPEG-b-pHis). Chemical Programming of Intratumoral Boron for Boron Neutron Capture Therapy.

Boron neutron capture therapy (BNCT) is a radiotherapeutic modality in which biological selectivity is dictated not by targeting alone but by the spatial control of nuclear reactions. Following tumor-selective accumulation of 10B, neutron irradiation induces the 10B(n,α)7Li reaction, generating high-linear energy transfer particles with micrometer path lengths that confine cytotoxicity to boron-enriched cells. Despite its intrinsic precision, the clinical translation of BNCT has long been hindered by a fundamental chemical challenge: the inability to achieve sufficient, homogeneous, and durabl

Identification and validation of an intratumor heterogeneity-related prognostic signature in triple-negative breast cancer: a study based on integrative machine learning and single-cell sequencing.

INTRODUCTION: While intratumor heterogeneity (ITH) is recognized as a fundamental driver of treatment resistance and disease progression in triple-negative breast cancer (TNBC), existing prognostic models often fail to quantify ITH as a biomarker robustly. METHODS: This study obtained a training set from the METABRIC, while validation sets were sourced from the GEO database. ITH scores were calculated using the DEPTH2 algorithm, a computational method to infer heterogeneity from bulk transcriptomic data. Patients were stratified into distinct high-ITH and low-ITH groups based on these scores.

In-vitro immune-modulation of triple-negative breast cancer through targeting miR-30a-5p/MALAT1 axis using nano-PDT combinational approach.

BACKGROUND: Triple negative breast cancer (TNBC) is an immunogenic tumor; however, its tumor immune microenvironment (TIME) is densely packed with immune suppressive cytokines and immune checkpoints. The immune-suppressive features of TNBC TIME represent a considerable obstacle to any immunotherapeutic approach. The objective of this study was to develop a multimodal in-vitro strategy to manipulate the TNBC TIME and enhance patients' outcomes by employing carefully tailored hybrid chitosan-lipid Nanoparticles (CLNPs), metformin and chlorin e6 (Ce-6)-mediated PDT, alone or combined. Biomaterial-based 3D human lung models replicate pathological characteristics of early pulmonary fibrosis.

Idiopathic pulmonary fibrosis (IPF) is a progressive and incurable lung disease characterized by tissue scarring that disrupts gas exchange. Epithelial cell dysfunction, fibroblast activation, and excessive extracellular matrix deposition drive this pathology that ultimately leads to respiratory failure. Mechanistic studies have shown that repeated injury to alveolar epithelial cells initiates an aberrant wound-healing response by surrounding fibroblasts through secretion of mediators like transforming growth factor beta (TGF- β), yet the precise biological pathways contributing to disease pro

Schistosoma mansoni infection alters the host pre-vaccination environment resulting in blunted Hepatitis B vaccination immune responses.

UNLABELLED: The impact of endemic infections on protective immunity is critical to inform vaccination strategies. In this study, we assessed the influence of Schistosoma mansoni infection on host responses in a Ugandan fishing cohort given a Hepatitis B (HepB) vaccine. Concentrations of schistosome-specific circulating anodic antigen (CAA) pre-vaccination showed a significant bimodal distribution associated with HepB titers, which were lower in individuals with high CAA. We established that participants with high CAA had significantly lower frequencies of circulating T follicular helper (cTfh)

Natural Killer Cells Promote Fetal Development through the Secretion of Growth-Promoting Factors.

Natural killer (NK) cells are present in large populations at the maternal-fetal interface during early pregnancy. However, the role of NK cells in fetal growth is unclear. Here, we have identified a CD49a+Eomes+ subset of NK cells that secreted growth-promoting factors (GPFs), including pleiotrophin and osteoglycin, in both humans and mice. The crosstalk between HLA-G and ILT2 served as a stimulus for GPF-secreting function of this NK cell subset. Decreases in this GPF-secreting NK cell subset impaired fetal development, resulting in fetal growth restriction. The transcription factor Nfil3, b

Low-dose cisplatin converts the tumor microenvironment into a permissive state for HSVtk-induced antitumor immunity in HPV16-related tonsillar carcinoma.

An adenovirus harboring the HSV thymidine kinase (HSVtk) gene under the regulation of a trans-splicing ribozyme that targets telomerase is cytotoxic to cancer cells because it inhibits DNA replication (Ad5mTR). Furthermore, it induces anti-tumor immunity by activating cytotoxic T cells. Because multiple chemotherapeutic agents also activate cytotoxic T-cell immunity during the direct killing process of tumor cells, we herein explored whether low-dose cisplatin could synergize with cytotoxic Ad5mTR to potentiate its therapeutic effect by boosting anti-tumor immunity in a murine HPV16-associated

[Machine learning-based programmed cell death signature model for precise prediction of prognosis and treatment response in melanoma].

OBJECTIVES: The occurrence, metastasis, and drug resistance of melanoma pose major challenges to patient prognosis, and predictive models capable of accurately forecasting patient outcomes and guiding treatment are still lacking. This study aims to develop predictive models for melanoma prognosis and drug sensitivity based on mechanisms of programmed cell death (PCD). METHODS: Genes related to 19 PCD patterns were collected and integrated from gene set enrichment analysis (GSEA), the Kyoto Encyclopedia of Genes and Genomes (KEGG), relevant reviews, and published studies to establish a compreh
